A formulated red ginseng extract inhibits autophagic flux and sensitizes to doxorubicin-induced cell death

  • Background: Ginseng is believed to have antitumor activity. Autophagy is largely a prosurvival cellular process that is activated in response to cellular stressors, including cytotoxic chemotherapy; therefore, agents that inhibit autophagy can be used as chemosensitizers in cancer treatment. We examined the ability of Korean Red Ginseng extract (RGE) to prevent autophagic flux and to make hepatocellular carcinoma (HCC) cells become more sensitive to doxorubicin. Methods: The cytotoxic effects of total RGE or its saponin fraction (RGS) on HCC cells were examined by the lactate dehydrogenase assay in a dose- or time-dependent manner. The effect of RGE or RGS on autophagy was measured by analyzing microtubule-associated protein 1A/1B-light chain (LC)3-II expression and LC3 puncta formation in HCC cells. Late-stage autophagy suppression was tested using tandem-labeled green fluorescent protein (GFP)-monomeric red fluorescent protein (mRFP)-LC3. Results: RGE markedly increased the amount of LC3-II, but green and red puncta in tandem-labeled GFP-mRFP-LC3 remained colocalized over time, indicating that RGE inhibited autophagy at a late stage. Suppression of autophagy through knockdown of key ATG genes increased doxorubicin-induced cell death, suggesting that autophagy induced by doxorubicin has a protective function in HCC. Finally, RGE and RGS markedly sensitized HCC cells, (but not normal liver cells), to doxorubicin-induced cell death. Conclusion: Our data suggest that inhibition of late-stage autophagic flux by RGE is important for its potentiation of doxorubicin-induced cancer cell death. Therapy combining RGE with doxorubicin could serve as an effective strategy in the treatment of HCC.

  • 부패된 홍미삼엑기스로부터 분리동정한 내삼투압성 효모인 C. parapsilosis가 홍미삼엑기스의 성분에 미치는 영향을 알아보기 위하여 C. parapsilosis를 배양하면서 pH의 변화, ginsenoside의 함량, 지방산, phenol 화합물 등의 변화를 조사하였다. 엑기스를 소량 첨가하였을 때는 배지의 pH에 다소 영향을 주었으나 엑기스를 다량(10~20%) 첨가하였을 때는 pH에 큰 영향을 미치지 못하였다. C. parapsilosis는 홍삼엑기스의 사포닌의 함량에 영향을 주지 않았고 사포닌을 탄소원으로 이용하지 못하였으며 사포닌 중량 첨가에 따른 생육 촉진경향 혹은 저해현상도 없었다. C. parapsilosis의 홍미삼엑기스의 유리당 이용 패턴은 glucose, fructose 등 단당류를 먼저 흡수 이용하고 다음에 sucrose, maltose 등을 2차적으로 이용하였다. 홍미삼엑기스의 대부분의 지방산은 C. parapsilosis 생육에 의하여 현저히 감소하였으며 이중결합이 없는 stearic acid, palmitic acid는 변화가 거의 없었다. 또한 페놀화합물 중 maltol은 현저히 감소하였고 그 외에 vanillic acid, m-coumaic acid의 함량은 거의 변화가 없었다. 비휘발성 유기산의 함량은 거의 변화가 없었으며 아미노산은 배양 전에 비하여 현저히 감소하였고 특히 arginine은 완전 소진되었다.

진공력적방식(Vacuum Impulse Stem)을 이용한 홍삼의 추출 방법 (Extraction and Concentration Method of Red Ginseng by Vacuum Impulse System)

  • 추출 시간을 5시간으로 단축하고, 추출 온도를 $33\~50^{\circ]C$로 설정한 진공력적방식 조건과 $80^{\circ]C$의 고온의 다중단 추출방식 간의 조사포닌 함량비교 결과, 고온의 다중단 추출방법으로 제조한 엑스의 조사포닌 함량이 $8\~9\%$인데 비하여 저온 및 단시간 추출인 진공력적 추출방법은 조사포닌 함량이 $11.5\%$$41.9\%$ 높았다. 또한 진공력적 추출방법이 엑스 수득율비교에서도 $13.6\%$ 증가하였다. 향미는 종합적으로 진공력적엑스가 강하였으며 이를 선호하는 경향이었다. 진공력적추출 엑스의 색상은 기존 제품 엑스와 유사하였고 유동성이 좋았다. 진공력적 추출법은 짧은 추출 시간과 공정 자동화로 인건비 감소를 기대할 수 있으며, HPLC 패턴으로 사포닌 분해가 거의 없음을 확인 할 수 있었다.

Immunostimulatory Effect of Fermented Red Ginseng in the Mouse Model

  • In this study, Woongjin fermented red ginseng extract (WFRG) was evaluated for its potential ability to act as an adjuvant for the immune response of mice. For the in vitro study, macrophages were treated with serial concentrations ($1{\mu}g/mL$, $10{\mu}g/mL$, and $100{\mu}g/mL$) of WFRG. For in vivo studies, mice were administered different concentrations (10 mg/kg/day, 100 mg/kg/day, and 200 mg/kg/day) of WFRG orally for 21 days. In vitro, the production of nitric oxide and TNF-${\alpha}$ by RAW 264.7 cells increased in a dose-dependent manner. In vivo, WFRG enhanced the proliferation of splenocytes induced by two mitogens (i.e., concanavalin A and lipopolysaccharide [LPS]) and increased LPS-induced production of TNF-${\alpha}$ and IL-6, but not IL-$1{\beta}$. In conclusion, WFRG has the potential to modulate immune function and should be further investigated as an immunostimulatory agent.

인삼이 토끼 적혈구막의 $Na^{+}-K^{+}-ATPase$의 활성도에 미치는 영향 (Effect of Ginseng on Sodium-Potassium activated ATPase in Rabbit Red Cell Membrane)

  • The effect of ginseng on the ATPase activity of rabbit ref cell membrane has been investigated. The experiments were also designed to determine whether the components of ginseng could be attributed to the effect on ATPase activity which dependent upon sodium plus potassium and is sensitive to ouabain. The following results were observed. 1. The activity of the $Na^{+}-K^{+}-ATPase$ from red cell membrane is stimulated by ginseng, and the concentration of ginseng for half-maximal activity is about 15 mg%. The pH optimum for the ginseng sensitive component is 7.6. 2. The portion of the enzyme activity stimulated by ginseng is completely abolished by ouabain. 3. The activating effect of ginseng on the ATPase, with a given concentration of sodium in the medium, is increased by raising the potassium concentration but activity ratio is decreased. 4. The activating effect of ginseng on the ATPase, with a given concentration of potassium in the medium, is increased by raising the sodium concentration but the activity ratio is decreased. 5. The ATPase activity is increased by small amounts of calcium but inhibited by larger amounts and the rate of activity by ginseng is constant. 6. The action of ginseng on the ATPase activity was not related to the sulfhydryl group of cysteine, the amino group of lysine, the imidazole group of histidine, the quanidinium group of arginine, the carboxyl group of aspartic acid, or the hydroxyl group of threonine. 7. The activating effect of ginseng on the ATPase activity may be not due to a saponin which is contained in ginseng.

홍삼박 효소분해물을 이용하여 제조한 홍삼주의 발효특성 변화 (Changes in Fermentative Characteristics of Red Ginseng Wine using Enzymatically Hydrolyzed Red Ginseng Marc)

  • 복합 탄수화물 분해효소와 단백질 분해효소로 처리하여 제조한 홍삼박 효소분해물에 효모를 발효시킨 홍삼주의 발효특성을 조사하였다. 초기 가용성 고형분 함량을 $20^{\circ}Bx$$30^{\circ}Bx$로 조정한 후 효모를 접종하고 $25^{\circ}C$에서 발효시험을 실시한 결과, 발효 9일 후 가용성 고형분 함량은 $7.5^{\circ}Bx$$13.1^{\circ}Bx$로, 환원당 함량은 18 g/L와 24 g/L까지 감소하였다. 에탄올 함량은 당 소비량과 비례하여 최종 에탄올 함량은 각각 65 g/L와 106 g/L까지 증가하였다. pH는 모든 실험구에서 발효 2일째까지 급격하게 감소하여 pH 4.73에서 pH 3.99로 낮아졌으며, 그 이후 9일째까지 pH의 변화는 미미하였다. 적정산도는 pH 변화와 비례하여 증가하였다. 7일간 발효시킨 후 홍삼주의 조사포닌 함량은 효소 처리하지 않은 대조구에 비하여 100~200% 향상되었다. 그러므로 홍삼박 효소분해물은 홍삼 발효주의 원료로 이용 가능함을 확인하였다.

Saponin을 함유한 생약추출물의 거담작용에 관한 연구 (Study on the Expectorant Activity of Ethanol Extracts from Some Crude Drugs Containing Saponins)

  • The effect of ethanol extracts of Gilkyung(Platicodi Radix, Platicodongrandiflorum A, DC), Onji(Polygalae Radix, Polygala tennuifolia Willdenow) and Deoduk(Codonopsis lanceolate Radix, Codonopsis lanceolata) on expectorant activity of rat trachea was investigated. Following treatment of 50% ethanol extract of these medicinal plants (25 mg/rat), the content of neurotransmitters such as acetylcholine and histamine in tissue was significantly increased. The secretions of acid glycoproteins and the artificially injected phenol red were also increased. However, there was no significant difference except Onji From the histological study through periodic acid Schiff and alcian blue stain, the thickness of inner membrane of acinar glands and the stained glycoproteins on surface of epithelium and on the glands were observed in all the rats trachea treated with extract of medicinal plants. In vitro, the viscosity of mucin solution was slightly decreased with an addition of the extracts. Onji showed the most effective expectorant activity among them at the identical conditions. The mechanism of expectorant activity of these medicinal plants seems to be due to stimulation of secretion and changes of rheological properties of mucus.

서양삼 추출물의 초단파 및 식초 처리에 의한 인삼 사포닌 성분 변화 (The Change of Ginsenoside Composition in American Ginseng (Panax quinquefolium) Extract by the Microwave and Vinegar Process)

  • The purpose of this study is to develop a new preparation process of American ginseng (Panax quinquefolium) extract featuring high concentration of ginsenoside $Rg_3$, $Rg_5$, and $Rk_1$, Red ginseng special components. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of American ginseng were processed under several treatment conditions of microwave and vinegar (about 14% acidity). The results showed that the quantity of ginsenoside $Rg_3$ increased by over 0.9% at the 20 minutes of the pH 2~4 vinegar and microwave American ginseng ethanol extract compared with other process times. The result of MAG-20 indicates that the American ginseng microwave and vinegar-processed American ginseng extracts (about 14% acidity) treated for 20 minutes produced the highest amount of ginsenoside $Rg_3$ (0.969%), $Rg_5$ (1.071%), and $Rk_1$ (0.247%). Besides, MAG-15 indicates that the microwave - and vinegar-processed American ginseng extracts (about 14% acidity) treated for 15 minutes produced the highest amount of ginsenoside $Rg_3$ (0.772%), $Rg_5$ (1.330%), and $Rk_1$ (0.386%). This indicates that American ginseng treated with microwave and vinegar had the quantity of the ginsenoside $Rg_3$ over 32 times the amount of the ginsenoside $Rg_3$ (which was not found in raw and American ginsengs) in the average commercial Red ginseng.

Molecular mechanism of protopanaxadiol saponin fraction-mediated anti-inflammatory actions

  • Background: Korean Red Ginseng (KRG) is a representative traditional herbal medicine with many different pharmacological properties including anticancer, anti-atherosclerosis, anti-diabetes, and anti-inflammatory activities. Only a few studies have explored the molecular mechanism of KRG-mediated anti-inflammatory activity. Methods: We investigated the anti-inflammatory mechanisms of the protopanaxadiol saponin fraction (PPD-SF) of KRG using in vitro and in vivo inflammatory models. Results: PPD-SF dose-dependently diminished the release of inflammatory mediators [nitric oxide (NO), tumor necrosis factor-${\alpha}$, and prostaglandin $E_2$], and downregulated the mRNA expression of their corresponding genes (inducible NO synthase, tumor necrosis factor-${\alpha}$, and cyclooxygenase-2), without altering cell viability. The PPD-SF-mediated suppression of these events appeared to be regulated by a blockade of p38, c-Jun N-terminal kinase (JNK), and TANK (TRAF family member-associated NF-kappa-B activator)-binding kinase 1 (TBK1), which are linked to the activation of activating transcription factor 2 (ATF2) and interferon regulatory transcription factor 3 (IRF3). Moreover, this fraction also ameliorated HCl/ethanol/-induced gastritis via suppression of phospho-JNK2 levels. Conclusion: These results strongly suggest that the anti-inflammatory action of PPD-SF could be mediated by a reduction in the activation of p38-, JNK2-, and TANK-binding-kinase-1-linked pathways and their corresponding transcription factors (ATF2 and IRF3).

Anxiolytic-like Effects of Panax ginseng on the Elevated Plus-maze Model in Mice

  • This study was performed to investigate the anxiolytic-like effects Panax ginseng in mice using the elevated plus-maze model. Furthermore, the anxiolytic-like effects of Panax ginseng were compared to a known active anxiolytic drug, diazepam. Ginseng total saponin (GTS, 100 mg/kg) from red ginseng (RG), sun ginseng (SG) total extract (50 mg/kg), butanol fraction of SG(25 and 50 mg/kg) and ginsenosides ($Rb_1,\;Rg_1,\;and\;Rg_5$ and Rk mixture) significantly increased the number of open arm entries and the time spent on the open arm, compared with that of control. However, Red ginseng (RG) total extract (l00 mg/kg), GTS (25, 50 mg/kg), SG total extract (25 mg/kg) and ginsenosides ($Rg_{3}-R\;and\;Rg_{3}-S$) did not increase the number of open arm entries and the time spent on the open arm. On the other hand, butanol fraction of RG (l00 mg/kg), total extract of SG (50 mg/kg), butanol fraction of SG (50 mg/kg), ginsenosides ($Rb_{1},\;and\;Rg_{5}$ and Rk mixture) decreased the locomotor activity, in a similar fashion to diazepam. These data support that ginseng has the anxiolytic-like effects and the anxiolytic potential of SG was stronger than that of RG. Ginsenosides $Rb_{1},\;Rg_{1},\;and\;Rg_{5}$ and Rk mixture play important role on the anxiolytic-like effects of Panax ginseng. We provide evidence that ginseng and some ginsenosides may be useful for the treatment of anxiety.
