음나무 배발생 캘러스의 증식 및 체세포배 발달을 위한 액체 현탁 배양조건 확립 (Establishment of suspension culture condition for embryogenic callus proliferation and somatic embryo development of Kalopanax septemlobus)

  • 본 실험은 음나무 배발생 세포의 증식 및 체세포배 발달을 위한 액체 현탁 배양조건의 확립을 위해 수행되었다. 배발생 세포의 생장율은 접종 밀도가 증가할수록 감소하였다. 배발생 세포의 증식에 가장 효과적인 접종 밀도는 0.1 g/100 ml 로서 이 농도에서 세포의 생장율이 가장 높았다. 배양기간에 따른 배발생 세포의 생장 패턴 및 세포 주기 (G1, S, G2/M) 분석 결과, 세포의 생장은 배양 5일 후부터 증가가 시작되어 15일 까지 급격히 생장하였으며 그 이후에는 점차 감소하였다. 배양 기간 별 세포 주기 (cell cycle)의 변화가 명확하게 관찰되어 배양 5일째 5기는 초기의 5.5% 에서 11.7%로 두 배정도 증가하였으며, 배양 15일 이후부터는 다시 초기의 세포 주기로 되돌아가면서 안정화되는 것으로 나타났다. 따라서 음나무 배발생 세포의 현탁배양은 15일의 주기로 배양하는 것이 증식에 가장 효과적인 것으로 생각되었다. 한편 배발생 세포에서 체세포배의 유도는 배양초기의 접종 밀도가 중요한 것으로 나타났다. 0.5 g/L 의 낮은 밀도로 접종 시에는 65% 이상의 어뢰형 배가 유도된 반면 접종 밀도가 높아질수록 어뢰형으로의 배발달은 급격히 감소하였다. 초기의 접종 밀도가 증가할수록 특히 어뢰형 배의 발달은 지연되었으나 구형 및 심장형 배의 유도는 초기 접종밀도에 영향을 받지 않았다. 이상의 실험결과로 음나무 액체 배양 시 초기 접종 밀도를 조절함으로써 배발생 캘러스의 증식 및 체세포배를 효과적으로 유도할 수 있었으며 이는 체세포배 생산을 위한 배양 기간의 단축이 가능함을 보여주는 결과이다.

Substrate Selection for Larval Settlement and Spat Growth in the Purple Clam, Saxidomus purpuratus (Sowerby) in Laboratory Culture

  • The purpose of this study is to determine the appropriate substrate for larval settlement and spat growth in the purple clam, Saxidomus purpuratus in laboratory culture. Larvae were reared with 3 different types of sediments (mud, sand, and mixed) for 46 days in settlement experiment, and settled spats were further grown in 3 types of sediments for 36 weeks in growth experiment. The density of settled spats in muddy sediments was more than 2 times higher than those in mixed or sandy sediments. But, the average size of settled spats in muddy sediments was smaller than those in mixed or sandy sediments. After 36 weeks of growth period, growth rate decreased as shell length increased. When shell length was less than 2 mm, growth rate in mixed sediments was significantly higher than that in sandy sediments. When shell length was more than 2 mm, there was no significant difference in growth rate among different substrates. Sediment type affected growth rate only when the spats were relatively small (less than 2 mm). Muddy sediments seems better for larval settlement, while mixed sediments is best for spat growth. We suggest the laboratory procedure for enhancing seedling production of S. purpuratus.

조선전기 고문헌에 수록된 식품저장법에 대한 문헌고찰 (Review of Literature on Food Preservation of the Early Joseon Dynasty)

  • This study aims to investigate the food preservation methods adopted by the Joseon Dynasty, which existed before the 17th century. A total of 232 food preservation methods were discovered in 25 books, and could be classified by their targeted food types: vegetables (84), sea foods (60), meats (41), fruits (37), and others (10). Depending on the preservation method applied, they are classified as food drying, soaking, mud cellar preservation, and other preservation. Food drying is further classified into 8 sub-types: drying, sun-drying, shadow-drying, wind-drying, dry heat, combined drying, smoking, and others. Soaking could be sub-divided into using salt, ash, dry sand, bran, fermented paste, wet distillers grains, oil, and others. Mud cellar preservation is sub-classified into installing shelf inside the mud cellar, making the mud cellar for food preservation, and making hole or underground tunnel for food preservation. Other food preservation methods include minimizing moisture loss by applying beeswax on a section of the vegetable stem, and cutting the vegetables or fruits with their branches and leaves for food preservation.

Direct Somatic Embryogenesis of Curculigo orchioides Gaertn., an Endangered Medicinal Herb

  • In vitro multiplication of Curculigo orchioides was achieved by direct somatic embryogenesis in young leaf segments. Immature leaf segments of about 0.5 cm in length were cultured on MS medium supplemented with different concentrations of BAP (2-10 $\mu{M}$) or Kin (2-10 $\mu{M}$). Optimum response in terms of per cent cultures responding (89%) and the number of embryos per explant (16) were observed on MS medium supplemented with 8 $\mu$M BAP. The emergence of several somatic embryos on the adaxial side of the leaf segments was observed one month after the culture. Germinated somatic embryos were grown up to about 1.5 cm length before transferring to maturation medium. For maturation, the individual embryos were isolated and transferred to MS medium supplemented with BAP (5 $\mu{M}$) and NAA (0.5 $\mu{M}$). The plantlets emerged from the embryos were transferred to soil containing 1 peat: 1 sand with 90% success. The embryos were formed directly on the leaf segments without any callus phase. Direct regeneration of somatic embryos is important for the conservation of this endangered species, as rare somaclonal variants are likely to arise than from indirect regeneration.

Factors affecting the urease activity of native ureolytic bacteria isolated from coastal areas

  • Coastal erosion is becoming a significant problem in Greece, Bangladesh, and globally. For the prevention and minimization of damage from coastal erosion, combinations of various structures have been used conventionally. However, most of these methods are expensive. Therefore, creating artificial beachrock using local ureolytic bacteria and the MICP (Microbially Induced Carbonate Precipitation) method can be an alternative for coastal erosion protection, as it is a sustainable and eco-friendly biological ground improvement technique. Most research on MICP has been confined to land ureolytic bacteria and limited attention has been paid to coastal ureolytic bacteria for the measurement of urease activity. Subsequently, their various environmental effects have not been investigated. Therefore, for the successful application of MICP to coastal erosion protection, the type of bacteria, bacterial cell concentration, reaction temperature, cell culture duration, carbonate precipitation trend, pH of the media that controls the activity of the urease enzyme, etc., are evaluated. In this study, the effects of temperature, pH, and culture duration, as well as the trend in carbonate precipitation of coastal ureolytic bacteria isolated from two coastal regions in Greece and Bangladesh, were evaluated. The results showed that urease activity of coastal ureolytic bacteria species relies on some environmental parameters that are very important for successful sand solidification. In future, we aim to apply these findings towards the creation of artificial beachrock in combination with a geotextile tube for coastal erosion protection in Mediterranean countries, Bangladesh, and globally, for bio-mediated soil improvement.

유색칼라 기내배양묘의 순화재배에 관한 연구 (A Study on acclimatization culture of the tissue cultured seedings in Zantedschia spp.)

  • 유색칼라 조직배양묘의 순화재배 용토선발실험에서 초기생육 출엽율은 큰 차이가 없었으나 생육 6개월 후의 괴경비대는 모래(50%)+Peatmoss(50%) 처리에서 가장 좋았다. 순화재배 시기별 순화율은 8월 20일 정식에서 64%~70.4%로 가장 저조하였고, 3월 20과 4월 20일은 90%이상의 순화율을 나타냈다. 괴경비대은 3월 20일 정식에서 구중이 가장 무거웠다. 순화 재배시 배양묘의 재식거리 10×10cm에서 괴경중이 가장 양호했다.

  • Collection, evaluation and preservation of tropical forage grasses, napier grass breeding, tissue culture of pangola grass and alfalfa selection were conducted in Taiwan. The results showed that some species such as pangola grass (Digitaria decumbens) with good performance and wide adaptability were selected and released. Forage yield and quality of napier grass have been improved and have good results in animal performance. Embryogenic callus cultures from young inflorescences and stem segments of pangola grass could provide an alternative method for rapid propagation and improvement. Three better varieties of alfalfa were selected. They maintain satisfactory stands for two or three years on well-drained sand loam and loam soil, and used only as annual crop in flat area and acid soil. However, more studies in forage breeding for acid soil arc required to maximise the forage quality and animal production. Thus, animal fed with forages of high quality and a few grains supplement will be the future achivement in livestock industry.

  • The author introduces the preventive and therapeutic methods of diseases in Japanese flounder which have been conducted by the Fish Disease Laboratory, the Mikamewan Fishery Cooperative Union, Ehime Prefecture, since 1982. Prevention 1. Addition of a sand substrate in the culture pond was effective to prevent ulcer disease. 2. Bathing in 0.5ppm of copper ion was effective to prevent some parasites. 3. Low stocking density of the fish reduced an incidence of edwardsiellosis or gliding bacterial disease. 4. Removal of the diseased fish prevented thd spread of lyphoeystis. 5. So-called $\pi$-water was effective to prevent the fry from some diseases. 6. Immersion of the juvenile in a sodium nifrusylate solution during transportation was effective to prevent gliding bacterial disease. Therapy 1. Sodium nifrustylate or oxytetracycline was effective to cure gliding bacterial disease. 2. Bathing in formalin(150ppm) or freshwater was effective to cure scuticociliatidosis. 3. Erythromycin was effective to cure $\beta$-hemolytic Streptococcus sp. infection. 4. Bathing in a hydrogen peroxide solution(1.5%) was effective to cure white spot disease.

Characterization of Two Urease-Producing and Calcifying Bacillus spp. Isolated from Cement

  • Two bacterial strains designated as CT2 and CT5 were isolated from highly alkaline cement samples using the enrichment culture technique. On the basis of various physiological tests and 16S rRNA sequence analysis, the bacteria were identified as Bacillus species. The urease production was 575.87 U/ml and 670.71 U/ml for CT2 and CT5, respectively. Calcite constituted 27.6% and 31% of the total weight of sand samples plugged by CT2 and CT5, respectively. Scanning electron micrography analysis revealed the direct involvement of these isolates in calcite precipitation. This is the first report of the isolation and identification of Bacillus species from cement. Based on the ability of these bacteria to tolerate the extreme environment of cement, they have potential to be used in remediating the cracks and fissures in various building or concrete structures.

In Vitro Production of Indian Citrs Ringspot Virus-Free Plants of Kinnow Mandarin (Citrus nobilis Lour X C. deliciosa Tenora) by Ovule Culture

  • Indian citrus ringspot virus (ICRSV)-free plants of Kinnow mandarin (Citrus nobilis Lour x C. deliciosa Tenora) were raised from virus-infected plants using unfertilised ovules as explants. Plants were tested by indirect ELISA and RT-PCR before using their explant. An amplified product of 539 bp was obtained by RT- PCR in ICRSV infected plants. Unfertilized ovules were excised from unopened flower buds of plants tested postive for virus and were cultured on Murashige and Skoog's (MS) basal medium supplemented with various concentrations of kinetin (KN) or malt extract (ME). Maximum induction (31.94%) of embryogenic callus was observed on MS medium supplemented with KN ($9.29\;{\mu}M$). Transfer of embryogenic calli to similar media composition resulted in somatic embryogenesis in all cultures, with an average number of 60.36 globular, 17.39 heart and 7.71 cotyledonary-shaped somatic embryos per culture. All cotyledonary shaped embryos developed into complete plantlets within 60 days on transfer to similar medium. Embryogenic callus induction, somatic embryo formation, maturation, germination and plantlet formation were achieved on MS medium supplemented with KN ($9.29\;{\mu}M$) alone. The plantlets derived from somatic embryos were transferred to sterilized soil, sand and vermiculite (3:1:1) mixture. After acclimatization, the plantlets were transferred to screen house and were indexed for ICRSV employing indirect ELISA and RT-PCR and found free of virus. A distinct feature of this study is the induction of somatic embryogenesis from unfertilised ovules to produce virus-free plants.
