Salvianolic acid B가 고강도 운동부하에 의한 흰쥐 골격근과 뇌조직의 Oxidative Stress에 미치는 영향 (Effects of Salvianolic Acid B Against Oxidative Stress in Skeletal Muscle and Brain Tissue following Exhaustive Exercise in Rats)

  • Objectives : Salvianolic acid B (SAB) is an active ingredient in Salvia miltiorrhiza frequently used for cardiovascular and cerebrovascular diseases. The present study investigated the antioxidant effects of SAB on the skeletal muscle and the brain tissue of rats following exhaustive exercise.Methods : The rats were treated with oral administration of SAB (30 mg/kg) daily for 5 days prior to the exhaustive exercise. The exhaustive exercise was performed as swimming for 150 min with 5% body weight attached to the tail on the 5th day. The antioxidant effects of SAB was evaluated by measuring the superoxide generation in the gastrocnemius and the 4-HNE expression in the hippocampal tissue. In addition, c-Fos-expressing cells in the brain tissue was observed using immunohistochemistry.Results : Histological features and muscle fiber type composition were not different between the SAB group and the exhaustive exercise group. SAB significantly reduced the upregulation of superoxide generation in the muscle tissue. SAB significantly reduced the increase of c-Fos-expressing cells in the cerebral cortex, paraventricular thalamic nucleus, dorsomedial hypothalamic nucleus, the CA1, CA3, and DG regions of hippocampus. SAB significantly reduced the upregulation of 4-HNE expression in the CA1 and DG regions of hippocampus caused by the exhaustive exercise.Conclusions : The results suggest that SAB exerts antioxidative effect against oxidative stress in the skeletal muscle and the brain tissue following exhaustive exercise, while SAB may has an anti-stress effect on stress responses in the brain.

랫드 두개골 결손부에서 단삼 수용성 추출물의 골형성 효과 (Effect of water extract of Danshen on bone regeneration of rat calvarial defect model)

  • The purpose of this study was to evaluate the osteogenic capacity of water extract of danshen (Salvia miltiorrhiza Bunge). We have established in rat critical-sized calvarial defect model using the combination with collagen scaffold and danshen hydrophilic extract. All rats were extinguished at 8 weeks after bone graft surgery, and the bone regeneration ability of bone grafting sides was evaluated by plain radiography and micro-CT. These results revealed water extract of danshen had the potential to promote osteogenesis especially continuous oral administration with local treatment compared to one-shot local treatment. This compound may provide a new alternative agent for growth factors to promote bone healing and bone regeneration. In conclusion, these results suggest that danshen hydrophilic extract have the potential to promote osteogenesis in bone defects. Further studies about fusion technology with salvianolic acid B, peptides, growth factors, and scaffolds using of the combination of tissue engineering, cell engineering and mechanical engineering are needed.

중국 재배지역별 단삼의 내부형태 및 이화학적 패턴 연구 (A Study on Internal Morphology and Physicochemical Patterns of Salvia miltiorrhiza Bunge Root According to Cultivation Regions in China)

  • Objectives : This study aimed to be used as basic data for obtaining excellent quality of Salvia miltiorrhiza (SM) root. Methods : SM cultivated in Anhui Bozou, Henan, Shandong, Henan Wenxian, Zhejiang, Hebei, Liaoning, Shanxi, Shaanxi Shangluo, Sichuan Zhongjiang, Sichuan Minyang and Jiangsu in China were gathered and its internal morphology and physicochemical patterns were analyzed. Results : Cross-section of sang plants according to cultivated regions in China showed that cork layers consisted of cells with four to six layers, cortex width was wide, and phloem was semilunar-shaped. Although there are forms layers, yet boundary was vague. It was observed that eight to ten vascular bundles were arranged in wooden parts in all directions and as a single line of vascular was distributed nearby form layers as it nears to the center. Tanshinon IIA identified a spot in the same location with standard product near by Rf 0.9 in the test identifying TLC of SM according to cultivated areas in China. The largest amount was 0.535% identified in Shandong and the lowest amount of 0.05% in Hebei. Salvianolic acid B identified a spot in the same location with standard product near by Rf 0.7 in the test identifying TLC of SM according to cultivated areas in China. The largest amount was 2.72% identified in Liaoning and the lowest amount of 0.825% in Jiangsu. Conclusions : The results are assumed to be used as basic data for obtaining excellent quality of SM oriental medical hers and distinguishing authentic or fake herbs.

천연방부제 개발을 위한 자원식물을 활용한 발효 전·후 물질 변화와 항균활성 비교 (Comparison of Substance Change and Antibacterial Activity Before and After Fermentation Using Resource Plants for The Development of Natural Preservatives)

  • 화학방부제는 항균활성은 좋지만 많은 부작용이 보고되고 있어 최근에는 인체에 해가 없으며 방부 기능과 자체 효능이 있는 천연방부제의 개발이 활발하다. 또한 천연물의 흡수율을 높이기 위한 방법으로 분자량이 커서 인체에 잘 흡수되지 않아 저분자화를 시키기 위한 방법으로 균주를 이용하여 발효하는 것도 늘어나고 있는 추세이다. 이에 천연방부제가 방부 효과는 지속되어야 하고 첨가했을 때 품질에 손상을 주지 않아야 하며, 향, 색과 성분에 영향을 끼치지 않아야 한다는 요건들을 충족시켜야 한다. 따라서 본 연구는 인체에 해가 없으며 항균활성이 좋은 황금, 단삼, 관중을 선정하였다. 식물은 모두 에탄올로 추출하였고, 발효균주로는 B. subtilis, L. casei, S. cerevisiae와 누룩에서 분리한 Asp. oryzae로 액체와 고체발효를 하여 발효 전과 발효 후의 유효성분의 함량을 비교하였다. 항균활성을 알아보기 위해 C. albicans, S. aureus, P. aeruginosa 세균 3종과 진균은 E. coli로 억제력을 탐색하였다. 천연방부제 개발로 사용하기 위해서는 항균활성이 있고 이러한 실험을 수행하여 다음과 같은 결과를 얻었다. 1. 원산지, 굵기와 용매에 따른 수율은 황금은 중국산이며 굵기는 2,800㎛로 50% ethyl alcohol 추출물에서 21.88%, 단삼은 국내산으로 굵기는 2,800㎛이며 50% ethyl alcohol 추출물에서 25.62%, 관중은 중국산으로 굵기는 2,800㎛, 70% ethyl alcohol 추출물에서 6.50%의 수율을 얻었다. 2. L. casei로 1일 액체 발효했을 때 수율은 황금 66.8%, 단삼 84.8%, 관중 73.0%였다. B. subtilis로 고체발효한 황금과 단삼은 24.40%, 39.30%, 관중은 L. casei로 발효 했을 때 11.10%의 수율을 얻었다. 3. 액체발효 후 황금에서 S. aureus 균주에 대하여 9mm 정도의 clear zone이 확인되었고, 단삼과 관중 발효에서는 항균력이 확인되지 않았다. 4. 황금을 L. casei로 고체발효 했을 때 C. albicans와 S. aureus에서 높은 항균활성을 보였다. 단삼은 S. cerevisiae로 고체발효 했을 때 S. aureus에서 항균활성을 보였다. 관중은 L. casei, S. cerevisiae로 고체발효 했을 때 E. coli와 S. aureus에서 항균활성이 높게 나타났다. 발효하지 않았을 때보다 발효 후의 항균활성이 매우 높았다. 5. 유효성분의 변화는 고체발효하지 않은 황금에서는 baicalin 101.57, baicalein 28.26, wogonin 5.33mg/g이었고, S. cerevisiae로 고체발효 했을 때는 baicalin 94.31, baicalein 30.41, wogonin 3.57mg/g로 baicalein의 함량은 증가된 것으로 확인되었다. 발효하지 않은 단삼에서는 salvianolic acid A가 1.82mg/g 이었으며, S. cerevisiae로 고체발효했을 때는 5.70mg/g으로 증가하였다. 관중의 유효성분은 flavaspidic acid AP, flavaspidic acid PB, flavaspidic acid AB, flavaspidic acid BB로 나타났다.

Could Natural Products Confer Inhibition of SARS-CoV-2 Main Protease? In-silico Drug Discovery

  • In December 2019, the COVID-19 epidemic was discovered in Wuhan, China, and since has disseminated around the world impacting human health for millions. Herein, in-silico drug discovery approaches were utilized to identify potential candidates as Severe Acute Respiratory Syndrome coronavirus 2 (SARS-CoV-2) main protease (Mpro) inhibitors. We investigated several databases including natural and natural-like products (>100,000 molecules), DrugBank database (10,036 drugs), major metabolites isolated from daily used spices (32 molecules), and current clinical drug candidates for the treatment of COVID-19 (18 drugs). All tested compounds were prepared and screened using molecular docking techniques. Based on the calculated docking scores, the top ones from each project under investigation were selected and subjected to molecular dynamics (MD) simulations followed by molecular mechanics-generalized Born surface area (MM-GBSA) binding energy calculations. Combined long MD simulations and MM-GBSA calculations revealed the potent compounds with prospective binding affinities against Mpro. Structural and energetic analyses over the simulated time demonstrated the high stabilities of the selected compounds. Our results showed that 4-bis([1,3]dioxolo)pyran-5-carboxamide derivatives (natural and natural-like products database), DB02388 and Cobicistat (DB09065) (DrugBank database), salvianolic acid A (spices secondary metabolites) and TMC-310911 (clinical-trial drugs database) exhibited high binding affinities with SARS-CoV-2 Mpro. In conclusion, these compounds are up-and-coming anti-COVID-19 drug candidates that warrant further detailed in vitro and in vivo experimental estimations.

효소 처리에 따른 단삼 추출물의 이화학적 특성 (Physicochemical properties of Salvia miltiorrhiza Bunge following treatment with enzymes)

  • 본 연구는 국내산 단삼을 활용하여 amylase계, cellulase계, pectinase계 및 protease계 효소제를 이용한 저온 추출법에 따른 단삼 추출물의 이화학적 품질 변화를 조사하였다. 단삼 추출물의 수율, pH, 당도 및 색도 변화는 amylase계 효소를 이용하여 $60^{\circ}C$, 4시간 처리구에서 58.3%, pH 6.04, $5.97^{\circ}Brix$와 65.06(L) 및 35.13(b)로 가장 높게 나타났으며, a값은 protease계 효소를 이용하여 $60^{\circ}C$에서 처리구에서 14.88로 붉은색을 나타내었다. 효소제를 이용한 단삼 추출물의 항산화능은 추출농도 55 ppm에서 DPPH 및 ABTs 소거능 84.25 및 74.11%로 amylase계 효소를 이용하여 $60^{\circ}C$, 4시간 처리구에서 가장 높은 소거능을 나타내었다. 총 페놀성 화합물 함량은 $60^{\circ}C$ 효소 처리구간들에서 비교적 높은 함량을 나타내었다. 단삼 추출물의 salvianolic acid B 함량은 $60^{\circ}C$에서 4시간, amylase계 효소구에서 3,002 mg%로 가장 높은 함량을 나타내었으며, cryptotanshinone 함량은 $60^{\circ}C$ 4시간 amylase 및 protease계 효소 처리구에서 3.8 mg%으로 가장 높게 나타났으며, tanshinon I함량은 $60^{\circ}C$ 4시간 protease계 효소구에서 14.2 mg%으로 가장 높게 나타났다. 이상의 결과로 amylase계 효소를 이용하여 단삼의 저온 효소 추출시 단삼 추출물의 지표성분의 안정적 추출이 가능한 바, 기능성 소재로서 대량생산을 통한 산업적 이용이 가능할 것으로 판단되며, 효소제를 이용한 저온 효소추출법은 천연물의 다양한 원료에 접목시킬 수 있을 것으로 생각된다.

Effects of compound traditional Astragalus and Salvia Miltiorrhiza extract on acute and chronic hepatic injury

  • Previous reports showed that Compound Astragalus and Salvia miltiorrhiza extract (CASE), which was mainly composed of astragalosides, astragalus polysaccharide and salvianolic acids, inhibited hepatic fibrosis by mediating transforming growth factor-${\beta}$ (TGF-${\beta}$)/Smad signaling. Our aim was to examine the effects of CASE on D-galactosamine (D-GalN) treated liver injury in mice and carbon tetrachloride ($CCl_4$)-induced liver fibrosis in rats. CASE was administered to mice with D-GalN-induced liver injury and to rats with $CCl_4$-induced liver fibrosis, respectively. Liver injury was routinely evaluated by relative liver weight, serum levels of ALT, AST, hyaluronic acid (HA), hepatic malondialdehyde (MDA) content, superoxide dismutase (SOD) activity, hydroxyproline (HYP) and histopathologic changes. Treatment of mice with CASE (60, 120, and 240 mg/kg, ig) significantly lowered ALT, relative liver weight, and MDA levels when compared with D-GalN treated mice. CASE (120, 240 mg/kg) significantly lowered ALT, AST, HA, HYP, and MDA levels against $CCl_4$ treated rats. Decreased SOD level was reversed with CASE treatment. Upon histopathological examination, CASE treatment had significantly inhibitory effect on the progression of hepatic fibrosis in rats. These results indicate that CASE might be effective in treatment and prevention of acute and chronic hepatic injury due to its antioxidant activity.

Quality Evaluation of Modified Bo-Yang-Hwan-O-Tang by Capillary Electrophoresis and High-performance Liquid Chromatography

  • High-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) were used to identify five active components in the modified herbal decoction Bo-Yang-Hwan-O-Tang (mBHT), i.e., amygdalin, decursin, paeoniflorin, salvianolic acid B, and calycosin-7-O-${\beta}$-D-glycoside. These components were identified by comparing their retention times and mass spectra with those of reference compounds. The conditions of both analytical methods were optimized and validated. Sufficient separation of target analytes was achieved using a buffer consisting of 40 mM sodium borate and 60 mM sodium dodecylsulfate (SDS) containing 10% methanol (pH 9.5) at 250 nm for CE analysis and gradient elution with a water-methanol mobile phase and ultraviolet (UV) photodiode array detector (DAD) at 250 nm for HPLC analysis. The mBHT components were determined within 65 min by HPLC and 16 min by CE. All calibration curves showed high linearity (R > 0.999) within the ranges tested. Intra-day and inter-day precision were less than 1.6% and 1.8% for HPLC and 2.5% and 4.8% for CE, respectively. The accuracy of the methods ranged from 98.8% to 102.3% for HPLC and from 95.9% to 108.2% for CE.

단삼의 수확시기에 따른 생육 특성 및 주요 성분 변이 (Changes of Major Components and Growth Characteristics According to Harvesting Times of Salvia miltiorrhiza Bunge)

  • Background : This study examined the effect of harvesting time on the growth, yield characteristics, and major beneficial components in Salvia miltiorrhiza. Methods and Results : Although plant height, stem diameter and branch length were not affected by harvesting time, the number of stems was highest when harvested in mid October. There were no differences in root length and thickness, however, the rhizome was thicker when it was harvested at the end of October or early November than when it was harvested in early and mid October. The dried root weight also showed a similar pattern. However, there was a statistically significant increase to 408 kg (16%) in the rhizome weight when in late October and a rise to 455 kg (29%) when harvested in early November. Harvest time had little effect on the content of the major component of S. miltiorrhiza. For example, salvianolic acid content rose from 9.42 to 9.64% with later harvest times, and tanshinone ${\prod}A$ content was tended to be slightly more increased in mid October which S. miltiorrhiza has 0.22% tanshinon ${\prod}A$ than in early October. Conclusions : According to these results, the optimum harvest time for S. miltiorrhiza is early November when plant or major component yields are hightest. There were no significant harvest time effects on the major beneficial components.
