구리이온(II)이 존재할 때 Salvianolic acid B에 의한 DNA 절단 (DNA Breakage by Salvianolic acid B in the Presence of Cu (II))

  • 단삼의 성분인 salvianolic acid B는 다양한 생리활성이 알려져 있다. 특히 항산화 효과는 간세포, 신경세포를 포함한 다양한 세포유형에서 보호효과가 있다고 보고되었다. 하지만 ferulic acid와 같이 항산화제로 여겨지는 몇몇 페놀성 물질은 특정 전이 금속이 있으면 산화작용을 하며 이것이 항암 효능을 설명하기도 한다. 본 실험에서 salvianolic acid B가 $Cu^{2+}$ 환경에서 산화작용을 하는지 알아보았다. salvianolic acid B와 $Cu^{2+}$를 동시 처리하면 supercoilded 형태의 DNA가 open circular 혹은 linear 형태로 바뀌었으나 salvianolic acid B 혹은 $Cu^{2+}$를 단독 처리 했을 때는 그렇지 않았다. $Cu^+$에만 특정적인 킬레이터 neocuproine을 이용하여 salvianolic acid B가 $Cu^{2+}$$Cu^+$로 환원시킴을 알았으며 $H_2O_2$를 물과 산소로 분해하는 catalase를 처리하면 DNA 분해가 일어나지 않았다. 활성산소종 중 하나인 $H_2O_2$는 생체분자 특히 DNA를 공격하여 정상기능을 수행하지 못하게 한다. 정리하면 salvianolic acid B에 의한 $Cu^{2+}$의 환원은 $H_2O_2$를 생성하며 $H_2O_2$는 DNA 분해를 일으킨다. 이런 결과는 salvianolic aicd B의 항암효과가 salvianolic acid의 $H_2O_2$ 생성 때문일 수 있다는 작은 단서를 줄 수 있으며 이는 좀 더 실험이 이뤄져야 한다.

Marker compound contents and antioxidant capacities of the taproot and lateral root of Danshen (Salvia miltiorrhiza Radix)

  • In this study, the marker compound contents of both the taproot and lateral root of Danshen (Salvia miltiorrhiza Radix), which is cultivated in Korea, were investigated. The salvianolic acid B content was the highest in the taproot (5.17-6.75%) and lateral root (3.99-5.69%). The cryptotanshinone, tanshinone I, and tanshinone IIA contents were higher in the lateral root than in the taproot of Danshen (p <0.05). Principal component analysis results revealed that the taproot was correlated to the salvianic acid A, rosmarinic acid, salvianolic acid B, and salvianolic acid A contents, whereas the lateral root was correlated to the cryptotanshinone, tanshinone I, and tanshinone IIA contents. The total phenolic content and total flavonoid content of the taproot were higher than those of the lateral root (p <0.05); however, the antioxidant activities of the taproot and lateral root of Danshen were similar. The salvianolic acid B content was correlated to the TPC of the taproot (r=0.748) and the 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity of the lateral root (r=0.847). This study could provide useful information for the classification of Danshen as a herbal medicinal product.

Highly-sensitive Detection of Salvianolic Acid B using Alumina Microfibers-modified Electrode

  • Alumina microfibers with porous structures were prepared through hydrothermal reaction, and then used to modify the surface of carbon paste electrode (CPE). After modification with alumina microfibers, the electrochemical activity of CPE was found to be greatly improved. On the surface of alumina microfibers-modified CPE, the oxidation peak current of salvianolic acid B, a main bioactive compound in Danshen with anti-oxidative and anti-inflammatory effects, was remarkably increased compared with that on the bare CPE surface. The influences of pH value, amount of alumina microfibers and accumulation time were studied. Based on the strong signal amplification effects of alumina microfibers, a novel electrochemical method was developed for the detection of salvianolic acid B. The linear range was from 5 ${\mu}gL^{-1}$ to 0.3 mg $L^{-1}$, and the detection limit was 2 ${\mu}gL^{-1}$ (2.78 nM) after 1-min accumulation. The new method was successfully used to detect salvianolic acid B in ShuangDan oral liquid samples, and the recovery was over the range from 97.4% to 102.9%.

고강도 운동부하에 의한 흰쥐의 심장근육 손상에 대한 Salvianolic Acid B의 효능에 대한 연구 (Effect of Salvianolic Acid B on Cardiac Muscle Damage Following Exhaustive Exercise in Rats)

  • Objectives: This study examined the effects of salvianolic acid B (SAB) on exhaustive exercise-induced cardiac muscle damage to rats. Methods: The study was carried out with 12-week-old young adult male Sprague-Dawley rats. Thirty-six rats were divided into 3 groups; normal (n=12), exhaustive exercise group (ExS, n=12) and exhaustive exercise with SAB (ExS+SAB, n=12). Five days before exhaustive exercise, SAB were medicated for 5 days in ExS+SAB group. Rats in ExS and ExS+SAB group were forced to swim for 150 minutes and then they were sacrificed, while rats in normal group were sacrificed at rest. After that, blood was collected and cardiac muscle tissue damage indices were analyzed. Results: Serum aspartate transaminase activity and lactate dehydrogenase activity were significantly lower in ExS+SAB group than in ExS group. Serum creatine phosphokinase activity of ExS+SAB was significantly lower than ExS group. However, the content of serum creatinine had no difference between ExS and ExS+SAB group. In the H&E stained left ventricle myocardium, ExS group showed signs of myocardial damage such as sporadic fragmentation of myocardial fibers, interstitial edema, cytoplasmic eosinophilia and neutrophils infiltration. However, ExS+SAB group alleviated the severity of the signs of myocardial damage. In the myocardial dihydroethidium staining, optical density was remarkably decreased in ExS+SAB group compared to ExS group. Furthermore, the up-regulation of Bax/Bcl-2 ratio was observed in ExS+SAB group compared with ExS group. Conclusions: The above results suggest that SAB may protect cardiac muscle damage via antioxidant activity and prevention of apoptosis.

온도에 따른 단삼의 광합성 특성 및 수확시기가 품질에 미치는 영향 (Effect of temperature on photosynthetic capacity and influence of harvesting time on quality of Salvia miltiorrhiza Bunge)

  • 본 연구에서는 고품질 단삼을 생산하기 위한 재배지대 구분과 수확시기에 따른 단삼의 생산성 및 성분함량에 대한 기초자료를 확보하기 위하여 온도조건별 생리반응과 수확시기별 품질을 조사하였다. 동화산물의 축적과 수분이용 효율에 영향을 미치는 광합성율, 증산율 및 엽록소 형광반응을 조사한 결과 $20^{\circ}C$에서 광합성율, 기공전도도, 수분이용 효율이 가장 좋았고 광보상점이 가장 낮아 약광조건에서도 광합성 효율이 높았으며, 최대광합성속도도와 순양자수율도 가장 높았다. 수확시기에 따른 단삼뿌리의 생육 및 수량은 큰 차이가 없었으나 salvianolic acid B, cryptotanshinone, tanshinone I, tanshinone IIA의 함량은 수확시기가 늦을수록 유의하게 증가하였으며 salvianolic acid B 함량이 가장 많았으며 수확시기별로는 11월 상순에 비해 이듬해 3월 중순 이후에 수확시료에서 약25.5배 증가하였다. Cryprotanshinone, tanshinone I, tanshinone IIA 함량 또한 2015년 3월 20일 수확한 시료에서 가장 높은 함량을 나타내었다. 따라서 단삼재배에 적합한 온도는 $20^{\circ}C$전후가 적절한 것으로 사료되며 상대적으로 고온조건인 $30^{\circ}C$에서 광합성율, 증산율, 엽록소 형광반응이 크게 저하되므로 서늘한 기후조건인 준고랭지 이상의 지역에서 단삼을 재배하는 것이 유리할 것으로 판단된다. 또한 고품질 단삼 유통을 위한 적정 수확시기는 단삼을 파종한 다음 해 3월 중순 이후에 수확을 하는 것이 합리적인 것으로 사료된다.

달임 조건에 따른 Salvianolic Acid B의 구조변환 차이 (Chemical Conversion Pattern of Salvianolic Acid B in Aqueous Solution under Different Decoction Conditions)

  • 식품 및 한약재는 수용액 중에서 가열 및 달임을 행한 후에 섭취하는 경우가 매우 많다. 그러나 가열 및 달임에 의한 소재 중에 함유된 성분변화 연구는 매우 미흡한 실정이다. 이에 본 연구에서는 순환기계질환 치료를 위해 많이 이용되고 있는 단삼(Salvia miltiorrhiza Bunge)이라는 한약재에 다량 함유된 salvianolic acid B(Sal B)를 대상으로 가열조건에 따른 성분변화 양상을 HPLC 분석을 통해 비교하였다. 그 결과 $100^{\circ}C$의 동일 온도에서는 상압에서보다 가압조건 하에서 Sal B 변환 생성물들의 함량이 더 높아짐을 알 수 있었다. 그리고 가압조건 하에서 온도를 $100^{\circ}C$로 동일하게 설정하고 달임시간에 따른 변화를 비교한 결과, 매우 한정적인 성분들을 제외하고, 대부분의 성분들이 가열시간의 경과와 함께 그 생성물들의 함량이 증가하는 경향을 보였다. 한편 가열 온도와 압력 조건을 달리하여 각각 4시간동안 가열을 행한 결과, 가열온도와 압력이 높을수록 생성률이 특징적으로 높아지는 생성물들이 있는 반면, 오히려 그 반대의 경우도 관찰되었다. 이 결과들로부터 과도한 가열 온도와 지나치게 높은 가압 조건에서는 상압조건과는 다른 가열생성물들의 조성이 형성됨을 알 수 있었다. 이에 따라 가열 방법 및 가열 조건에 따라 생리활성의 발현 양상에도 영향이 초래될 가능성이 시사되었다. 본 연구결과는 한의학뿐만이 아니라 조리학적 측면에 있어서도 매우 중요한 성과라 판단되는 바 관련 연구의 기초자료로 활용되길 기대한다.

Marker compounds contents of Salvia miltiorrhiza Radix depending on the cultivation regions

  • Salvia miltiorrhiza Radix is cultivated in Korea and China and is traditionally used to treat cardiovascular diseases. In this study, we developed and validated a quantitative analysis method for S. miltiorrhiza Radix using high-performance liquid chromatography (HPLC). Identification was performed using ultra performance liquid chromatography-tandem mass spectrometry. For quantitative analysis, we used seven marker compounds. Separation conditions for HPLC were optimized using an ODS column with gradient conditions of 1% formic acid in distilled water and 1% formic acid in acetonitrile, with a flow rate of 0.8 mL/min and a detection wavelength of 280 nm. This method showed good linearity ($R^2=0.9998$), precision (relative standard deviation ${\leq}3.3%$), accuracy (recovery of 94.16-102.89%), limit of detection ($7.53{\mu}g/mL$), and limit of quantification ($23.71{\mu}g/mL$). This approach successfully quantified marker compounds in S. miltiorrhiza Radix. The individual marker compounds were identified by comparing the molecular masses and retention times with does standard compounds. Marker compound contents of S. miltiorrhiza Radix were investigated with different cultivation regions. Seven marker compounds were detected and quantified in all samples. Among them, salvianolic acid B showed the highest contents and it ranged from 4.13 to 7.15%. The salvianolic acid B content (7.15%) of marker compound was the highest in Bonghwa, and the tanshinone IIA content (1.90%) was the highest in Pohang. The results of marker compounds and developed method were intended to provide a favorable reference for the study of S. miltiorrhiza Radix from different regions of Korea.

Salvianolic acid B ameliorates psoriatic changes in imiquimod-induced psoriasis on BALB/c mice by inhibiting inflammatory and keratin markers via altering phosphatidylinositol-3-kinase/protein kinase B signaling pathway

  • Salvianolic acid B (SAB) is an active phytocomponent of a popular Chinese herb called Radix Salvia militiorrhiza with numerous biological properties. The anti-psoriasis activity of SAB was examined by evaluating various psoriasis inflammatory and keratin markers against imiquimod (IMQ)-induced psoriasis on BALB/c mice. Totally 50 healthy BALB/c mice were evenly divided into 5 groups including control, drug control (SAB; 40 mg/kg), IMQ-induced psoriasis (5%), IMQ exposure and treated with SAB (40 mg/kg), or standard methotrexate (MTX; 1 mg/kg). Mice supplemented with either SAB or MTX significantly lowered the values of psoriasis area severity index (PASI), erythema, scaling, skin thickness, inflammatory markers (interleukin [IL]-22/23/17A/1β/6) and lipid peroxidation product (malondialdehyde). Also, IMQ exposed BALB/c mice treated with SAB or MTX display lesser histopathological changes with enhanced antioxidant activities (catalase, superoxide dismutase). Moreover, the protein expression of keratin markers (K16 and K17) and phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt) signaling proteins (pAkt/Akt and pPI3K/PI3K) were significantly downregulated after administration with SAB and MTX as compared with IMQ induced mice. Taking together, SAB and MTX significantly ameliorate psoriatic changes by inhibiting psoriatic inflammatory and keratin markers through abolishing PI3K/Akt signaling pathway. However, further studies (clinical trials) are needed to confirm the anti-psoriatic property of SAB before recommending to psoriasis patients.

Salvianolic Acid B Inhibits Hand-Foot-Mouth Disease Enterovirus 71 Replication through Enhancement of AKT Signaling Pathway

  • Hand, foot, and mouth disease (HFMD) is caused by enterovirus 71 (EV71) in infants and children under six years of age. HFMD is characterized by fever, mouth ulcers, and vesicular rashes on the palms and feet. EV71 also causes severe neurological manifestations, such as brainstem encephalitis and aseptic meningitis. Recently, frequent outbreaks of EV71 have occurred in the Asia-Pacific region, but currently, no effective antiviral drugs have been developed to treat the disease. In this study, we investigated the antiviral effect of salvianolic acid B (SalB) on EV71. SalB is a major component of the Salvia miltiorrhiza root and has been shown to be an effective treatment for subarachnoid hemorrhages and myocardial infarctions. HeLa cells were cultured in 12-well plates and treated with SalB (100 or 10 ㎍/ml) and 106 PFU/ml of EV71. SalB treatment (100 ㎍/ml) significantly decreased the cleavage of the eukaryotic eIF4G1 protein and reduced the expression of the EV71 capsid protein VP1. In addition, SalB treatment showed a dramatic decrease in viral infection, measured by immunofluorescence staining. The Akt signaling pathway, a key component of cell survival and proliferation, was significantly increased in EV71-infected HeLa cells treated with 100 ㎍/ml SalB. RT-PCR results showed that the mRNA for anti-apoptotic protein Bcl-2 and the cell cycle regulator Cyclin-D1 were significantly increased by SalB treatment. These results indicate that SalB activates Akt/PKB signaling and inhibits apoptosis in infected HeLa cells. Taken together, these results suggest that SalB could be used to develop a new therapeutic drug for EV71-induced HFMD.

재배지역별 단삼의 성분 분석 및 생리활성 비교 (Component Analysis and Comparison of Biological Activities of Salvia miltiorrhiza Bunge from Different Cultivation Regions)

  • 재배지역에 따른 단삼의 성분 및 생리활성 비교를 위하여 고창, 영양, 장흥 지역의 국내산 단삼 3종과 중국산 단삼에 대한 일반성분, 총 폴리페놀 및 플라보노이드 함량을 분석하고, 열수 및 75% 에탄올 추출을 시행하여 단삼 추출물의 항산화, 항균, 항염증 활성 평가와 salvianolic acid B 함량을 측정하였다. 단삼의 일반성분은 재배지역에 따라 상당한 차이를 보였으며, 총 폴리페놀 함량은 영양 단삼이 가장 높고, 총 플라보노이드 함량은 고창 단삼에서 가장 높은 결과를 보였다. 재배지역별 단삼의 열수 및 75% 에탄올 추출물을 이용하여 DPPH와 ABTS 라디칼 소거능을 측정한 결과 영양 단삼의 항산화 활성이 가장 우수하였으며, 75% 에탄올 추출물에 비해 열수 추출물의 항산화 활성이 더 우수하였다. B. cereus에 대한 지역별 단삼 추출물의 항균 활성은 영양 단삼이 가장 높았으며, 열수에 비해 75% 에탄올 추출물의 항균 활성이 더 우수하였다. LPS로 염증을 유도한 RAW 264.7 세포에 지역별 단삼 추출물을 처리하여 NO 저해율을 비교한 결과 열수 추출물에서는 중국산 단삼이 34% 저해율로 가장 높았으나, 75% 에탄올 추출물에서는 고창 단삼이 68% 저해율로 가장 우수한 결과를 나타내었다. 재배지역별 단삼 추출물에서 주요 성분인 salvianolic acid 함량을 비교한 결과 영양 단삼 추출물의 함량이 가장 높았으며, 열수에 비해 75% 에탄올 추출물의 함량이 좀 더 높은 결과를 보였다.
