Hepatoprotective Activity of Salviae miltiorrhizae Radix Extract (단삼(丹蔘) 엑기스의 간보호작용)

  • Salviae miltiorrhizae(SM) Radix extract increased $[^3H]-thymidine$ incorporation into rat hepatocytes at the concentration ranging from $5{\times}10^{-5}\;to\;5{\times}10^{-1}mg/ml$. It decreased the activities of s-GOT and s-GPT in cirrhotic rats induced by $CCl_4$, TAA and D-GalN, respectively and reduced the sleeping time induced by hexobarbital in $CCl_4$,TAA and D-GalN intoxicated mice, respectively. SM extract shortened the half-life of sulfobromophthalein in $CCl_4$ intoxicated rats.

  • The methanol extracts of 162 herbal medicines were screened for the inhibitory activity against VHR dualspecificity protein tyrosine phosphatase (DS-PTPase). Seventeen medicinal plants, Scutellaria baicalensis, Cuscuta chinensis, Caesalpinia sappan, Arecae pericarpium, Rubus coreanus, Machilus thunbergii, Amsonia elliptica Cinnamomum cassia, Arisaema erubescens, Pueraria thunbergiana, Dendrobium moniliforme, Mentha arvensis, Peucedanum japonicum, Salvia miltiorrhiza, Leonurus sibiricus, Siegesbeckia orientalis, Prunella vulgaris showed potent VHR DS-PTPase inhibitory activity.

GC-MS Analysis of Diterpene Quinone Constituents of Salviae Miltiorrhizae Radix and Biological Activity

  • The ether extract of Salviae miltiorrhizae Radix(SMR) was fractioned to give five subfractions, so that two subfractions of them were recrystallized to yield each pure diterpene quinone pigment. On the basis of spectral evidence, these two compounds were identified as tanshinone II and crytotanshinone. Cryptotanshinone exhibited both of a potent platelet anti-aggregating activity in vitro and a potent antimicrobial activity. GC-MS analysis of the other extract showed that tanshinone II was contained in the largest proportion of all the diterpene quinones. In addition, GC-MS analysis gave other valuable analytical informations.

  • We observed the APD of rat ventricle myocyte and the effects of Lithospermic acid that was separated at Salvia miltiorrhiza having used in Oriental medicine by using classical whole cell patch clamp technique. We classified APD into APD30mV, APD0mV, APD-50mV, APD-60mV by cell membrane potential and the mean of cell resting membrane potential was -69.44${\pm}$1.72 mV.(omitted)

  • To evaluate anti-aggregatory activity of traditional prescriptions and medicines for treatment of Ohyul symptom, 70% methanol extracts were examined using collagen stimulated in vitro platelet aggregation by impedance method in rat whole blood. The crude extracts from DoHaekSeungKiTang, BoYangHwanOhTang, Caesalpinia sappan, Rhus verniciflua, Rheum palmatum, Polygonum cuspidatum, Salvia miltiorrhiza were found to inhibit platelet aggregation. The effective crude extracts of traditional medicine were fractionated to dichloromethane, ethyl acetate, butanol and aqueous layer. Polygonum cuspidatum, Caesalpinia sappan aud Rhus verniciflua ethyl acetate fractions concentration-dependently $(250-50{\mu}m/ml)$ inhibited the aggregation of platelet in whole blood induced by collagen. These results suggested that ethyl acetate fractions of Polygonum cuspidatum, Caesalpinia sappan and Rhus verniciflua have potent anti-aggregatory activity.

  • We have tested 101 species traditional medicinal plants on Acetylcholinester ase inhibition. Among them. Caesalpina sappan. Cnidium offcinale, Coptis japonica, Ephedra sinica, Eribotrya japonica, Gardenia jasminoides, Liriope platyphylla, Lonicera japonica, Paeonia suffruticosa, Phyllostachys nigra, Polygonum multiflorum, Prunella vulgaris, Rheum undulatum, Salvia miltiorrhiza and Uncaria rhynchophylla showed strong inhibition potency over 88%. In addition, Aconitum carmichaeli, Caesalphina sappan, Cinnamomun japonica, Citrus unshiu, Hordeum vulgare, Glycine max and Rehmannia glutinosa showed relatively good inhibition potency over 80%.

Suppression of AP-1 Activity by Tanshinone and Cancer Cell Growth Inhibition

  • The process of transcription is the major point at which gene expression is regulated. The jun and fos families of eukaryotic transcription factor heterodimerize to form complexes capable of binding 5'-TGAGTCA-3'DNA elements (AP-1 binding site). To search for the inhibitors of the jun-fos-DNA complex formation, several natural products extracts were screened and methanol extract of tanshen (the dried roots of Salvia miltiorrhiza Bunge) showed remarkable inhibitory activity. The active compounds of the extracts were purified using re-peated column chromatography and recrystallization. Their structures were identified as tanshinone I and tanshinone IIA. Through the electrophoresis mobility shift assay and cell cytotoxicity test, tanshinone I and tanshinone IIA were identified as inhibitors that suppress not only AP-1 function but also the cell proliferation. Tanshinone I also suppressed the jun-fos-DNA complex formation in TPA-induced NIH 3T3 cells.

  • The purpose of this study is to investigate the effect of Sungkangwon (Astragalus membranaceus Bunge + Pueraria lobata Ohwi + Salvia miltiorrhiza Bunge) on fatty liver induced by alcohol diet. Rats were either fed control diet or alcohol diet. On 6 weeks, alcohol group were randomly assigned to groups as follows; supplemented without SKW and with SKW (.033g/kg, 1g/kg, and 3g/kg). all of five groups were fed the chow diet for 4 weeks. After 6 weeks. the blood and liver concentration of TG and total-C is comparably high in alcohol group. (omitted)
