Factors Affecting Adhesion of Lactic Acid Bacteria to Caco-2 Cells and Inhibitory Effect on Infection of Salmonella Typhimurium

  • In this study, seven strains isolated from mustard leaf kimchi were screened for their tolerance to simulated gastric and bile juices, the adhesive properties to Caco-2 cells, and the inhibition ability of Salmonella Typhimurium ATCC 29631 adhesion. Lactobacillus acidophilus GK20, Lactobacillus paracasei GK74, and Lactobacillus plantarum GK81, which were resistant to bile as well as gastric juices, possessed high bile-salt hydrolase (BSH) activity towards both sodium glycocholate and sodium taurocholate. The strongest in vitro adherence of $53.96{\pm}4.49%$ was exhibited by L. plantarum GK81 followed by L. acidophilus GK20 with adhesion levels of $40.72{\pm}9.46%$. The adhesion of these strains was significantly (p < 0.05) reduced after exposure to pepsin and heating for 30 min at $80^{\circ}C$. Addition of $Ca^{2+}$ led to a significant (p < 0.05) increase of the adhesion of L. acidophilus GK20, but the adhesion ability of L. plantarum GK81 was not different from the control by the addition of calcium. In the competition and exclusion experiment, the adhesion inhibition of S. Typhimurium by L. plantarum GK81 strain was much higher than the other strains. Moreover, the exclusion inhibition of S. Typhimurium by L. acidophilus GK20 was considerably high, although the inhibition activity of this strain was lower than L. plantarum GK81.

Burden of Noncommunicable Diseases and National Strategies to Control Them in Korea

  • Noncommunicable diseases (NCDs) are the most important causes of premature mortality and disability-adjusted life years in Korea. NCDs are also the main contributor to socioeconomic inequalities in mortality and life expectancy. Reduction of NCDs and NCD inequalities would result in significant improvement in healthy life expectancy and health equity in Korea. Major NCD risk factors such as dietary risks (including salt intake), alcohol consumption, cigarette smoking, and high blood pressure were found to be the leading modifiable risk factors of disability-adjusted life years in Korea, based on the 2010 Global Burden of Disease Study. Several Korean studies have shown that these risk factors play an important role in creating socioeconomic inequalities in NCD mortality and total mortality. Current international discussions on NCD policies in the United Nations and the World Health Organization would provide better opportunities for developing aggressive population-wide policy measures in Korea. Considering the paucity of population-wide policies to control major NCD risk factors in Korea, rigorous population approaches such as taxation and regulation of unhealthy commodities as well as public education and mass campaigns should be further developed in Korea.

Isolation of new microorganisms which degrades the protein of a food garbage efficiently and its application (음식물 쓰레기중의 단백질을 효과적으로 분해하는 신규 미생물의 분리 및 응용)

  • In this study, novel strains showing better protein degradation activity were isolated for the production of effective compost from garbages. Well growing bacteria with clear zone on the skim milk agar media were isolated . The strain was identified as Bacillus subtilis PNV-1 through various biochemical tests, Bergy's manual of determinative bacteriology and 165 rDNA partial sequence. The extracellular protease of the strain PNV-1 has its activity at broad pH and the optimal temperature was $50^{\circ}C$. Also, the strain PNV-1 was highly tolerant to high concentration of salt, red/black pepper and mustard.

  • Recent trend in petroleum/gas exploration is an application of 3D seismic exploration technique. Unlike the conventional 2D seismic data processing, 3D seismic data processing is considered as the one which requires expensive commercial software systems and high performance computer. This paper propose a practical 3D seismic processing methodology on a personal computer using public domain software such as SU, SEPlib, and SEPlib3D. The applicability of the proposed method has been demonstrated by successful application to a well known realistic 3D synthetic data, SEG/EAGE 3D salt model data.

  • Generally, atmospheric corrosion is the electrochemical degradation of metal that can be caused by various corrosion factors of atmospheric components and weather, as well as air pollutants. Specifically, moisture and particles of sea salt and sulfur dioxide are major factors in atmospheric corrosion. Using galvanized steel is one of the most efficient ways to protect iron from corrosion by zinc plating on the surface of the iron. Galvanized steel is widely used in automobiles, building structures, roofing, and other industrial structures due to their high corrosion resistance relative to iron. The atmospheric corrosion of galvanized steel shows complex corrosion behavior, depending on the plating, coating thickness, atmospheric environment, and air pollutants. In addition, corrosion products are produced in different types of environments. The lifespans of galvanized steels may vary depending on the use environment. Therefore, this study investigated the corrosion behavior of galvannealed steel under atmospheric corrosion in two locations in Korea, and the lifespan prediction of galvannealed steel in rural and coastal environments was conducted by means of the potentiostatic dissolution test and the chemical cyclic corrosion test.

Liquid-Liquid Equilibrium and Physical Properties of Aqueous Mixtures of Poly (Ethylene Glycol) 3000 with Tri-Potassium Citrate at Different pH: Experiment, Correlation and Thermodynamic Modeling

  • The new experimental data of liquid-liquid equilibrium (LLE) of aqueous two-phase system (ATPS) consisting of poly(ethylene glycol) 3000 + tri-potassium citrate at different pH were presented. It was found that an increase in pH resulted in the expansion of the two-phase region. The TLL and STL increased with increasing the pH values. The Merchuk equation can be appropriately employed to correlate the binodal curves and also the tie-line compositions were adjusted to both the Othmer-Tobias and Bancroft equations. In order to calculate the compositions of the phase and the ends of the tie-lines, density and refractive indices as two physical properties were used. Finally, the extended UNIQUAC, UNIFAC, Virial-(Mobalegholeslam & Bakhshi) and modified UNIQUAC-FV were used to measure the phase equilibria at different pH. The results of the models suggested that it can be used quite well to correlate the LLE in an aqueous solution of polymer-salt.

Plasmonic effects and size relation of gold-platinum alloy nanoparticles

  • Plasmonic effects of gold and platinum alloy nanoparticles (Au-Pt NPs) and their comparison to size was studied. Various factors including ratios of gold and platinum salt, temperature, pH and time of addition of reducing agent were studied for their effect on particle size. The size of gold and platinum alloy nanoparticles increases with increasing concentration of Pt NPs. Temperature dependent synthesis of gold and platinum alloy nanoparticles shows decrease in size at higher temperature while at lower temperature agglomeration occurs. For pH dependent synthesis of Au-Pt nanoparticles, size was found to be increased by increase in pH from 4 to 10. Increasing the time of addition of reducing agent for synthesis of pure and gold-platinum alloy nanoparticles shows gradual increase in size as well as increase in heterogeneity of nanoparticles. The size and elemental analysis of Au-Pt nanoparticles were characterized by UV-Vis spectroscopy, XRD, SEM and EDX techniques.

Complete genome sequence of Bacillus licheniformis strain 0DA23-1, a potential starter culture candidate for soybean fermentation (콩발효 종균후보 Bacillus licheniformis 0DA23-1의 유전체 염기서열)

  • Bacillus licheniformis strain 0DA23-1, a potential fermentation starter candidate, was isolated from doenjang, a Korean high-salt-fermented soybean food. Strain 0DA23-1 contains a single circular 4,405,373-bp chromosome with a G + C content of 45.96%. The complete genome of strain 0DA23-1 does not include any of the virulence factors found in the well-known pathogens Bacillus cereus and Staphylococcus aureus. Additionally, no genes associated with resistance to eight antibiotics (chloramphenicol, clindamycin, erythromycin, gentamicin, kanamycin, streptomycin, tetracycline, and vancomycin), hemolysis, or biofilm formation were identified.

Validation of an HPLC/UV-based method for Salicornia herbacea-derived isorhamnetin-3-O-glucoside and quercetin-3-O-glucoside quantification

  • Salicornia herbacea is a type of salt marsh plant that has been used in traditional medicine to treat several diseases. Isorhamnetin-3-O-glucoside (I3G) and quercetin-3-O-glucoside (Q3G) are major flavonoids in S. herbacea that are known to exert various pharmacological activities. Therefore, our study sought to validate and optimize an HPLC/UV-based analytical method for I3G and Q3G yield quantification, as well as to determine its limit of detection, limit of quantification, linearity, precision, and accuracy. Upon testing a concentration range of 31.5-1.9 ㎍/mL the results exhibited good linearity (r2 ≥0.9996 and r2 ≥0.9999 for I3G and Q3G, respectively), and the procedure was deemed precise (relative standard deviation of ≤3.19 and ≤3.85%, respectively), and accurate (102.6-105.0 and 92.9-95.2%, respectively). The results showed that our proposed method could be used for rapid I3G and Q3G evaluation in S. herbacea.

Surface Tentiometric Studies on the Interaction of Anionic Polyelectrolytes with Cationic Surfactants

  • The interaction of cationic surfactants, n-alkyltrimethylammonium bromide ($C_nTAB$; n = 12, 14, 16) with anionic polyelectrolyte, poly(styrenesulfonate) (PSS) has been studied by surface tension measurement. In the absence of added salt, the cationic surfactants bind to PSS quantitatively up to ca. 60% coverage of anionic sites of the polyanion and the complexes were surface inactive. Further binding of the surfactant cations on PSS caused a sharp conformational transition of the surfactant/ PSS complexes to surface active complexes and accompanied precipitation. The binding showed a biphasic behavior in the presence of NaCl and cooperativity of the binding became less as the concentration of NaCl increased. Binding of the cationic surfactants on poly(vinylsulfonate) also showed the biphasic behavior and the cooperativity of the binding was much less even in the absence of NaCl. The binding of surfactant to PSS provided hydrophobic environment to solubilized pyrene and reduced the viscosity of the solution greatly even at surfactant concentrations well below cmc. This study indicated that the surfactant bound to PSS up to $60{\%}$ coverage of PSS sites are present as surfactant aggregates which are wrapped up with PSS chains, and hydrophobic interaction is an important factor in the binding of the surfactants to PSS.
