Partial Purification and Characterization of Halotolerant Alkaline Protease from Halomonas marisflava KCCM 10457 Isolated from Salt-fermented Food

  • Halotolerant protease produced by Halomonas marisflava KCCM 10457 was partially purified through ammonium sulfate precipitation and Sephacryl S-200HR gel permeation chromatography. Optimal pH and temperature of protease were 11.0 and $45^{\circ}C$, respectively. Enzyme activity was inhibited by $Cu^{2+}$, $Hg^{2+}$, $Fe^{2+}$, and $Fe^{3+}$, and selectively inhibited by p-chloromercuribenzoic acid (PCMB), suggesting this enzyme is cysteine protease. The enzyme is halotolerant, because it retained 77% of original activity in presence of 3.33 M NaCl. The protease showed broad substrate specificity to various natural proteins; BSA, casein, egg albumin, gelatin, and hemoglobin.

Removal of Cupper(II), Zinc(II) in Marine Environment by Heavy Metal Resistant Desulfovibrio desulfuricans (중금속 내성이 있는 Desulfovibrio desulfuricans를 이용한 해양 환경에서의 Cu(II), Zn(II) 제거)

  • Microorganisms play a significant role in bioremediation of heavy metal contaminated seawater. In this study, we reported an effective removal of Cu and Zn in marine envionment by using Desulfovibrio desulfuricans (D. desulfuricans) which belong to sulfate reducing bacteria. D. desulfuricans showed stable growth characteristics in high salt concentration and had resistance to heavy metals. Cu and Zn was removed not only by physical adsorption on the surface of bacteria but also by precipitation reaction of microbial metabolism by D. desulfuricans in seawater. In case of different heavy metal concentration, Cu was effectively removed 85% at 25 ppm and 60% at 50 ppm and Zn was effectively removed 54% at 50 ppm and 46% at 200 ppm, respectively.

Chemical Properties of Precipitation in Related to Wind Direction in Busan, Korea, 2009 (풍향에 따른 2009년 부산지역 강수의 화학적 특성)

  • The variation of acidity, conductivity, and ion components in precipitation depending on the dominant wind direction was investigated from January, 2009 to December 2009 in Busan, Korea. Both southwesterly and northeasterly winds were dominant in Busan area. The volume-weighted mean acidity showed pH 7, and the high conductivity indicated 200 ${\mu}scm^{-1}$ in westerly wind. The volume-weighted mean equivalent concentration showed higher value of $K^+$ and $Cl^-$ in all wind directions. The composition ratio of $NO{_3}^-/SO{_4}^{2-}$ showed over 3 in northerly wind. The neutralization factors have been found to have higher value for potassium ion in northeasterly, easterly, southwesterly, and westerly winds compared with different wind directions, which indicated significant neutralization of acidic components over the region by potassium. Also, the concentration of sea salt has been found over 800 ${\mu}sm^{-3}$ in northeasterly and southwesterly winds. Air masses passing through Manchuria, Inner Mongolia plateau, China, and Russia in spring, autumn, and winter covered Busan, Korea in northerly, westerly, and northwesterly winds. However, air masses passing through the ocean in summer covered Busan, Korea in easterly, northeasterly, and southwesterly winds. Therefore, the variation of acidity, conductivity, and ionic components contained in precipitation shows each seasonal characteristics with prevailing wind systems between the continental and coastal area in Busan, Korea.

Enhanced Dissolution and Duodenal Permeation of Atorvastatin Calcium Using Bile Salt and 2-Hydroxypropyl-${\beta}$-Cyclodextrin (담즙산염과 2-히드록시프로필-${\beta}$-시클로덱스트린을 이용한 아토르바스타틴칼슘의 용출 및 십이지장 점막 투과 증진)

  • This study was aimed to increase the solubility, dissolution and permeation rates of atorvastatin calcium (ATC) using bile salt and/or 2-hydroxypropyl-${\beta}$-cyclodextrin ($HP{\beta}CD$). From solubility studies, sodium deoxycholate (SDC) among bile salts studied was found to have the highest solubilizing effect on ATC ($4.4{\pm}0.4$ mg/ml), and the order of increasing solubility was SDC>sod. cholate>sod. glycocholate>sod. taurodeoxycholate>sod. taurocholate>conjugated bile acid. ATC solid dispersions were prepared at various ratios of drug to SDC and/or $HP{\beta}CD$, and evaluated by differential scanning calorimetry (DSC), dissolution studies and dissolution-permeation studies. DSC curves showed amorphous state of ATC in the physical mixture and solid dispersion. Dissolution rates of ATC-SDC solid dispersions and physical mixture were markedly increased at pH 6.8, but decreased at pH 1.2 with greater proportions of SDC due to the precipitation of SDC, compared with that of drug alone. On the other hand, dissolution rates of ATC-$HP{\beta}CD$ solid dispersion and physical mixture at pH 1.2 were varied with the ratio of drug to carriers. From duodenal permeation studies, it was found that fluxes of ATC (donor dose: 0.5 mg/3.5 ml) in the presence of 25 mM sodium glycocholate, SDC, sod. cholate and sod. taurocholate $(5.7{\pm}0.9$, $5.6{\pm}0.9$, $4.8{\pm}0.7$ and $4.6{\pm}0.9\;{\mu}g/cm^2/hr$, respectively) were enhanced, compared with drug alone ($3.4{\pm}0.9\;{\mu}g/cm^2/hr$). In the dissolution-permeation studies, 1 : 9 : 10 (w/w) ATC-SDC-$HP{\beta}CD$ solid dispersion increased the flux 2.2 times, compared with 1 : 5 : 4 (w/w) ATC-lactose-corn starch mixture as control. In conclusion, solid dispersions with bile salt and $HP{\beta}CD$ were found to be an effective means for increasing the dissolution and permeation rates of ATC.

Nutrient Recovery from Sludge Fermentation Effluent in Upflow Phosphate Crystallization Process (상향류 인 결정화공정을 이용한 슬러지 발효 유출수로 부터의 영양소 회수)

  • The nutrient recovery in phosphate crystallization process was investigated by using laboratory scale uptlow reactors, adopting sequencing batch type configuration. The industrial waste lime was used as potential cation source with magnesium salt($MgCl_2$) as control. The research was focused on its successful application in a novel integrated sludge treatment process, which is comprised of a high performance fermenter followed by a crystallization reactor. In the struvite precipitation test using synthetic wastewater first, which has the similar characteristics with the real fermentation effluent, the considerable nutrient removal(about 60%) in both ammonia and phosphate was observed within $0.5{\sim}1$ hr of retention time. The results also revealed that a minor amount(<5%) of ammonia stripping naturally occurred due to the alkaline(pH 9) characteristic in feed substrate. Stripping of $CO_2$ by air did not increase the struvite precipitation rate but it led to increased ammonia removal. In the second experiment using the fermentation effluent, the optimal dosage of magnesium salt for struvite precipitation was 0.86 g Mg $g^{-1}$ P, similar to the mass ratio of the struvite. The optimal dosage of waste lime was 0.3 g $L^{-1}$, resulting in 80% of $NH_4-N$ and 41% of $PO_4-P$ removal, at about 3 hrs of retention time. In the microscopic analysis, amorphous crystals were mainly observed in the settled solids with waste lime but prism-like crystals were observed with magnesium salt. Based on mass balance analysis for an integrated sludge treatment process(fermenter followed by crystallization reactor) for full-scale application(treatment capacity Q=158,880 $m^3\;d^{-1}$), nutrient recycle loading from the crystallization reactor effluent to the main liquid stream would be significantly reduced(0.13 g N and 0.19 g P per $m^3$ of wastewater, respectively). The results of the experiment reveal therefore that the reuse of waste lime, already an industrial waste, in a nutrient recovery system has various advantages such as higher economical benefits and sustainable treatment of the industrial waste.

Effect of Na Salt on the Formation of MgO Obtained from Mg(OH)2 by Precipitation Method (침전법으로 제조된 Mg(OH)2의 잔류 Na염이 MgO 입자 형성에 미치는 영향)

  • The particle size of MgO was examined as a function of the Na content in $Mg(OH)_2$ powders and the calcination temperature. $Mg(OH)_2$ suspension was obtained by dropwise precipitation of $Mg(NO_3)_2{\cdot}6H_2O$ and NaOH solutions. The suspension was diluted by varying the dilution volume ratio of distilled water to $Mg(OH)_2$ suspension to change the Na salt concentration in the suspension. $Mg(OH)_2$ slurry was filtered and dried at $60^{\circ}C$ under vacuum, and then its $Mg(OH)_2$ powder was calcined to produce MgO with different amount of Na content at $500\sim900^{\circ}C$ under air. Investigation of the physical and chemical properties of the various MgO powders with dilution ratio and calcination temperature variation was done by X-ray diffraction, transmission electron microscopy, BET specific surface area and thermal gravimetric analysis. It was observed that MgO particle size could depend on the condition of calcination temperature and dilution ratio of the $Mg(OH)_2$ suspension. The particle size of the MgO depends on the Na content remaining in the $Mg(OH)_2$ powder, which powder was prepared by changing the dilution ratio of the $Mg(OH)_2$ suspension. This change increased as the calcination temperature increased and decreased as the dilution ratio increased. The growth of MgO particle size according to the increase of temperature was more effective when there was a relatively high content of Na. The increase of Na content lowered the temperature at which decomposition of $Mg(OH)_2$ to MgO took place, thereby promoting the crystal growth of MgO.

A Study of Upgrading Real Biogas via CO2 Precipitation Route Under Indian Scenario

  • Our study focuses on upgrading real biogas obtained under Indian scenario using carbon capture and utilization (CCU) technology to remove carbon dioxide ($CO_2$) and utilize it by forming metal carbonate. Amines such as monoethanolamine (MEA), diethanolamine (DEA), and sodium hydroxide (NaOH) were used to rapidly convert gaseous $CO_2$ to aqueous $CO_2$, and $BaCl_2$ was used as an additive to react with the aqueous $CO_2$ and rapidly precipitating the aqueous $CO_2$. All experiments were conducted at $25^{\circ}C$ and 1 atm. We analyzed the characteristics of the $BaCO_3$ precipitates using X-ray diffractometry (XRD), scanning electron microscopy - Energy dispersive spectroscopy (SEM-EDS) and Fourier-transform infrared spectroscopy (FT-IR) analyses. The precipitates exhibited witherite morphology confirmed by the XRD results, and FT-IR confirmed that the metal salt formed was $BaCO_3$, and EDS showed that there were no traces of impurities present in it. The quantity of the $BaCO_3$ was larger when formed with DEA. Also, a comparison was done with a previous study of ours conducted in Korean conditions. Finally, we observed that the carbonate obtained using real biogas showed similar properties to carbonates available in the market. An economic analysis was done to show the cost effectiveness of the method employed by us.

A Study on Acid Recovering Process by Neutralization and Water-Splitting Electrodialysis (WSED) (중화법과 전기투석에 의한 산회수 공정연구)

  • Recently the treatment of industrial wastes by membrane processes has drawn much attention due to increasing demands for clean technology. In the process investigated in this study, metal species in the acidic wastes are precipitated as metal hydroxide forms in a neutralization tank, and acid and base solutions are regenerated by water-splitting electrodialysis(WSED) to be reused in the process. Material balances of the processes for treating pickle liquor and mixed wastewater were calculated to explain conceptual design of the process. Experiments for neutralization precipitation with KOH and NaOH for mixed wastewater were carried out to precipitate metal hydroxide and to recover salt solution as supernatant. Also WSED of the salt solutions producing acid and base was tested in 2 or 3 compartment stacks using KCl and NaCl to investigate the effects of stack configurations on the WSED performance.

Effects of Fat Reduction on the Stability, Microstructure, Rheological and Color Characteristics of White-Brined Cheese Emulsion with Different Emulsifying Salt Amounts

  • Cheese emulsion is an intermediate product for the production of cheese powder and needs to be stable, homogeneous and pumpable characteristics to convey to the spray drier. This study was conducted to evaluate the effects of fat reduction and emulsifying salt (ES) amount in cheese emulsion systems on the physicochemical characteristics. Reduced-fat (RF) and full-fat (FF) white-brined cheese emulsions were produced with different dry matters (DM; 15%-25% excluding ES) and ES concentrations (0%-3% based on cheese weight). Stable cheese emulsion was obtained at lower DM in RF cheese emulsion than that of FF cheese emulsion. Reduction in the amount of ES resulted in instability of both emulsions. Apparent viscosity with pseudoplastic flow behavior significantly increased with the decrease of fat content in stable cheese emulsions. Microstructure of emulsions appeared to be related to the fat content, stability and degree of emulsification. Reduction of fat content caused to get less lightness and more greenness in color, whereas yellowness was significantly decreased by increase in the amount of ES. In conclusion, fat reduction resulted in higher viscosities of cheese emulsion due to inducing the increment of protein, and the addition amount of ES considered as very important factor to produce stable cheese emulsion without protein precipitation or cream separation. Therefore, for preparation of RF cheese emulsion using a variety of white-brined cheese, lower amounts of DM would be suggested in this study to obtain homogenous droplets in the atomizing process of spray drying.

A Modified Methodology of Salt Removal through Flooding and Drainage in a Plastic Film House Soil (시설재배지에서 토양 담수 및 배수에 의한 염류집적 경감 방안)

  • One of the disadvantages of flooding treatment for desalting from soils is that salts move to deep soils after flooding and at the end reaccumulate at the soil surface through capillary movements. This study was carried out to remove salts from soils in plastic film houses by a modified flooding method, drainage after flooding. The method successfully removed salts at the soil surface and salts did not move to the deep soil. Drained water containing N, P and K could be reused as fertilizer. By applying small amount of MgO, turbidity of water flooded decreased in 30 min by 95%. Struvite should be formed since the flooded water contain ammonia and phosphorous and their concentrations were decreased. This could be utilized as fertilizer which provides a slow-release source of phosphorus, magnesium and nitrogen that features low inherent water solubility.
