Detection of viability Change of Escherichia coli O157:H7 using Surface Plasmon Resonance

  • For the acute assessment on biological toxicity of wastewater, surface plasmon resonance(SPR) based cell viability detection was performed using gold surface-confined cell as a result of adhesion-modifying chemicals. Escherichia coli O157:H7 (E. coli O157:H7) was investigated after exposure to EDTA. Cells were immobilized on gold coated slide glass for SPR analysis by the method of cross-linking carboxyl group on the bacterial surface with amine group of poly-L-lysine that had been coupled to the gold surface modified by a self-assembled monolayer of 11-mercaptounde canoic acid (11-(MUA)). Reflective intensity of each flow step was changed with respect to confect of ethylenediaminetetraacetic acid (EDTA) disodium salt and phosphate-buffered saline (PBS) solution. The proposed detection technique can be used for biological toxicity test.

Response and transcriptional regulation of rice SUMOylation system during development and stress conditions

  • Modification of proteins by the reversible covalent addition of the small ubiquitin like modifier (SUMO) protein has important consequences affecting target protein stability, sub-cellular localization, and protein-protein interactions. SUMOylation involves a cascade of enzymatic reactions, which resembles the process of ubiquitination. In this study, we characterized the SUMOylation system from an important crop plant, rice, and show that it responds to cold, salt and ABA stress conditions on a protein level via the accumulation of SUMOylated proteins. We also characterized the transcriptional regulation of individual SUMOylation cascade components during stress and development. During stress conditions, majority of the SUMO cascade components are transcriptionally down regulated. SUMO conjugate proteins and SUMO cascade component transcripts accumulated differentially in various tissues during plant development with highest levels in reproductive tissues. Taken together, these data suggest a role for SUMOylation in rice development and stress responses.

  • To be dyed with cationic Berberine, the cotton fiber was modified with reactive anionic agent. The Berberine, a natural cationic colorant of pure yellow, is the major component of Amur Cork tree extract and also can be employed as a natural antimicrobial agent due to its characteristic of cationic quaternary ammonium salt. By LC/MS analysis, it became obvious that the Berberine was contained in Amur Cork tree extract as a major color component. The adsorption of the Berberine on the cotton fabrics pretreated with the anionic agent was greatly increased comparing to that of untreated fabric. Because the anionic agent was colorless, it did not cause unintended color change of the dyeings. The dyed fabric with the Berberine has strong antimicrobial activity showing 99.5% of reduction of bacteria against Staphylococcu aureus.

IMPROVEMENT OF FRESHENING PROCESS BY MEANS OF UNDERDRAINAGE CONDUIT

  • This paper is concerned with the actual comparison analysis for the freshening process in the two selected experimental reservoirs. At the deep freshening reservoir, salinity and depth of the freshwater layer were estimated by simulation technique using the quantitative equation for the two layered flow structures. First of all, it is shown that the effects of underdraiange conduit in the lower layer were reported more effective for the control of upper layer salinity comparing with the case of no underdraiange conduit. Further the results of computation were later compared with the real observed values and the relating parameters of the salt balance equation are conformed even though approximately. Finally it was represented that the salinity of upper layer is easily diluted not only by the tidal gate but also by the underdraiange conduit in the lower layer of the freshening reservoir.

  • Langmuir-Blodgett(LB) technique can speak the best candidate of the future molecular electronic devices. But, precursor as molecular ultrathin film devices require the bulk property that are influenced by the molecular orientation. So, this device is one of current interest in molecular electronic device development of new materials. In this study, quantitative evaluation of molecular orientation in LB films of polyamic acid alkylamine salt was performed analysis experiment comparing the absorption or transmission intensity of the FT-IR spectrometer and reflection or absorption spectra with UV-visible absorption spectra. It could find that the polar angle(${\theta}$) of the dipole moment appears in about $68^{\circ}$ and the tilting angle of the alkyl chain is about $11.5^{\circ}$.

  • The PTDRT model as prediction techniques for desalinization in reclaimed tidelands was developed and verified whether it is applicable to reclaimed tidelands at the beginning stage. The changes of salt concentration during desalinization, the water requirements and period required for desalinization, were simulated according to soil properties and desalinization methods by repression analyses. The program was also designed to systematize input data and analysis data associated with desalinization, and to confirm the results by a graphic form. All input data and the results can be printed after the form of a typical report.

  • While considering the current scenario, in this world power demand goes on increases day by day. Frequent power outages occur in high voltage transmission line due to the deprived performance of polluted insulators; this affects overall operation of power system and may indirectly impinge on the growth of production sector. Many researchers are keenly taking efforts to provide highly reliable and stable power to neediest. In this paper, A.C pollution flashover performance of disc type glass insulator and composite long rod insulators investigation under various artificial pollutions by varying Equivalent Salt Density Deposition (ESDD) levels. Here, we use different types of pollution methods like binding method, dipping method and spraying methods with different types of pollutants concentration. Based on dimensional analysis, four different Mathematical models have been developed to predict the A.C pollution Flashover Voltage (FOV) of insulators. Both the experimental and mathematically modeled results are compared; it's observed that mathematical model 3 yields better results.

A Study on Quality Characteristics and Establishment of Fermentation Process for Traditional Kyungsando Squid sikhe (경상도 전통 마른 오징어 식해의 제법조사 및 품질특성)

  • The method of the squid safe made in Kyungsando was examined, and fermentation precess was established. The chemical composition and functional effects of the squid sikhe were examined. In the sensory evaluation, the squid sikhe made from Gampo accepted to be best for traditional Kyungsangdo squid sikhe and it is called standard sikhe. The Gampo sguid sikhe showed higher values in total sugar, free reducing sugar and nitrogen compound analysis compared to the orthers. The sugar of standard sikhe consisted of four kinds including glucose. The contents of free amino acid was increased in the order of glutamic acid, alanine, and methionine. The composition of amino acid in water or salt soluble protein of squid sikhe contained 17 kinds, and the contents was increased in order of glut amine acid, aspartic acid, and proline.

  • This study was conducted to establish Korean food culture by analizing in sets of Jinyounigue (進宴儀軌), Jinchanuigue (進饌儀軌), and Jinjarkuigue (進酌儀軌) which were the records of royal party procedures in Yi-Dynasity. Korean desserts were 141 kinds and could be classified into 8 groups such as Yoomilkwa(油蜜菓)16, Gangjung(强精) 51, Dasikl(茶食) 13, Jungkwa(正菓) 22, Suksilkwa(熟實菓) 7, Byung(餠) 8, Dang(糖) 28, Junyak(煎藥) 1. Food materials were fruits, fruit vegetables, roots, cereals, wine, pepper, cinnamon, ginger powder, pine spike, maximowiczia chinensis, fruit of buckthorn, cape jasmine, japanese touchwood, green bean, sesame oil, honey, salt, sesame, rouge and so on.

  • Grown in the secondary broth of production media, the strain Streptomyces albulus has increased more the production of its metabolite ${\varepsilon}$-poly-L-lysine, one of poly(amino acid)s used as disinfecting food additives, than the strain in the primary culture of growth nutrients. Having the strain removed, the large concentrate obtained by ultrafiltrating the secondary culture broth. The concentrated production broth exchanged into followed by detecting in UV flowcell at 220nm the peptide bond of the components eluting the adsorbed proteins and polylysine with NaCl salt of gradient concentration, and has separated into five components. Among them the component in the fourth peak fraction has proved to be the pure ${\varepsilon}$-poly-L-lysine after the portion being hydrolyzed the fraction with HCl into amino acid followed by being the composing amino acid analysis.
