Purpose: This study investigated the quality characteristics of traditional and commercial red pepper pastes (Gochujang) for development of traditional red pepper pastes. Methods: Proximate compositions, amino acid nitrogen contents, salt contents, color, viable bacteria, and inorganic substance contents of 19 traditional and three commercial Gochujang were investigated. Results: Analysis of proximate compositions showed huge differences between the samples. For moisture content, six kinds of traditional Gochujang did not meet the standard of the food codex. Contents of moisture, fat, and protein were significantly higher in traditional than commercial Gochujang. However, carbohydrate content was higher in commercial than traditional Gochujang, and thus calories were also higher. The amino nitrogen contents of the three kinds of traditional Gochujang did not meet the standard of the food codex. Salt contents and color were not significantly different between traditional and commercial Gochujang. In the microbiological analysis, total bacteria counts were higher in traditional than commercial Gochujang. The number of B. cereus did not exceed the standard of the food codex in all samples. S. aureus and fungi were not detected in any samples. In the inorganic substance analysis, inorganic substances contents in Gochujang were in the order of Na, K, Mg, and Ca. Overall, Na, K, Mg, and Ca were more abundant in traditional than commercial Gochujang. However, due to wide variations in inorganic substance contents of traditional Gochujang, there were no significant differences between traditional and commercial Gochujang. Conclusion: For the development of traditional Gochujang, quality standardization is considered necessary.
In urban areas, groundwater pollution is heavily affected by urbanization with land use types. This study aims to characterize groundwater quality and contamination in Sasang industrial area of Busan Metropolitan City where metalworking, machinery and footwear factories are located. Busan Metropolitan City is the highest in the utilization of groundwater resources among the metropolitan cities in Korea. $K^+,\;Na^+,\;Ca^{2+},\;Mg^{2+},\;Cl^-,\;SO_4^{2-}\;and\;HCO_3^-$ concentrations, and electrical conductivity (EC), total dissolved solids (TDS) and salinity are high in the areas near the Nakdong River. The results are attributed to the influence of salt water which intruded into the coastal sediments during sedimentation. In addition, the dominant chemical type of Ca-Cl indicates the influence of salt water in the geological formations as well as anthropogenic pollution. $SiO_2$ ion is interpreted to originate from both water-silicate mineral reactions and the decomposition of cement concretes. Trichloroethylene (TCE) was detected at 12 sites of total 18 sites. However, tetrachloroethylene (PCE) was detected at f sites and 1.1.1-trichloroethane (TCA) at 3 sites. According to the factor analysis, factor 1 was explained by 49.8%, factor 2 19.8%, and factor 3 11.0% with total 80.6% explanation. pH, TDS, salinity, $Ca^{2+},\;K^+,\;Mg^{2+},\;Na^+,\;Al^{3+},\;As^{3+},\;Cl^-\;and\;Fe^{2+}$ were positively highly loaded to factor 1. The chemical components loaded to factor 1 represent the chemical characteristics of both industrial pollution and influence by salt water. Based on the cluster analysis and distribution pattern of chemical components, the concentration of $Na^+,\;Ca^{2+},\;Cl^-,\;SO_4^{2-}\;K^+,\;and\;Mg^{2+}$ is high in the riverside area of the Nakdong River composed of coastal sediments that is influenced by salt water. The downstream area of the Hakjang Stream is judged to be affected by both salt water and artificial pollution. The other part of the study area is interpreted by anthropogenic pollution.

Saemangeum Lake is the largest artificial lake in Korea. The continuous deterioration of lake water quality necessitates the introduction of novel water quality management strategies. Therefore, this study aims to identify the spatiotemporal water quality characteristics of Saemangeum Lake using data from the National Water Quality Measurement Network and provide basic information for water quality management. In the water quality parameters of Saemangeum Lake, water temperature and total phosphorous content were correlated, and salt, total nitrogen content, pH, and chemical oxygen demand were significantly correlated. Other parameters showed a low correlation. The spatial principal component analysis of Saemangeum Lake showed the characteristics of its four zones. The mid-to-downstream section of the river affected by freshwater inflow showed a high nutrient salt concentration, and the deep-water section of the drainage gate and the lake affected by seawater showed a high salt concentration. Two types of water qualities were observed in the intermediate water area where river water and outer sea water were mixed: waters with relatively low salt and high chemical oxygen demand, and waters with relatively low salt and high pH concentration. In the principal component analysis by time, the water quality was divided into four groups based on the observation month. Group I occurred during May and June in late spring and early summer, Group II was in early spring (March-April) and late autumn (November-December), Group III was in winter (January-February), and Group IV was in summer (July-October) during high temperatures. The water quality characteristics of Saemangeum Lake were found to be affected by the inflow of the upper Mangyeong and Dongjin rivers, and the seawater through the Garuk and Shinshi gates installed in the Saemangeum Embankment. In order to achieve the target water quality of Saemangeum Lake, it is necessary to establish water quality management measures for Saemangeum Lake along with pollution source management measures in the upper basin.
Productivity of a rice plant is greatly influenced by salt stress. One of the ways to achieve tolerance to salinity is to transfer genes encoding protective enzymes from other organisms, such as microorganisms. The bacterial gene, mtlD, which encodes mannitol-1-phosphate dehydrogenase (Mtl-DH), was introduced to the cytosol of a rice plant by an imbibition technique to overproduce mannitol. The germination and survival rate of the imbibed rice seeds were markedly increased by transferring the mtlD gene when it was delivered in either a pBIN19 or pBmin binary vector. When a polymerase chain reaction was performed with the genomic DNAs of the imbibed rice leaves as a template and with mtlD-specific primers, several lines were shown to contain an exogenous mtlD DNA. However, a reverse transcription (RT)-PCR analysis revealed that not all of them showed an expression of this foreign gene. This paper demonstrates that the growth and germination of rice plants transiently transformed with the bacterial gene, mtlD, are enhanced and these enhancements may have resulted from the experssion of the mtlD gene. The imbibition method empolyed in this study fulfills the requirements for testing the function of such a putative gene in vivo prior to the production of a stable transgenic plant.
Phenotypic screening for bile salt hydrolase (BSH) activity was performed on Lactobacillus acidophilus PF01 isolated from piglet feces. A gene encoding BSH was identified and cloned from the genomic library of L. acidophilus PF01. The bsh gene and surrounding regions were characterized by nucleotide sequence analysis and were found to contain a single open reading frame (ORF) of 951 nucleotides encoding a 316 amino acid protein. The potential bsh promoter region was located upstream of the start codon. The protein deduced from the complete ORF had high similarity with other BSHs, and four amino acid motifs located around the active site, FGRNXD, AGLNF, VLTNXP, and GXGXGXXGXPGD, were highly conserved. The bsh gene was cloned into the pET21b expression vector and expressed in Escherichia coli BLR(DE3) by induction with 0.1mM of isopropylthiogalactopyranoside. The BSH enzyme was purified with apparent homogeneity using a $Ni^{2+}$-NTA agarose column and characterized. The overexpressed recombinant BSH enzyme of L. acidophilus PF01 exhibited hydrolase activity against tauroconjugated bile salts, but not glycoconjugated bile salts. It showed the highest activity against taurocholic acid. The maximum BSH activity occurred at approximately $40^{\circ}C$. The enzyme maintained approximately 70% of its maximum activity even at $60^{\circ}C$, whereas its activity rapidly decreased at below $37^{\circ}C$. The optimum pH was 6, and BSH activity was rapidly inactivated below pH 5 and above pH 7.
A fungal strain, capable of solubilizing insoluble phosphate under diverse temperature, pH and salt conditions was isolated from Waste Mushroom bed of Agaricus bisporus in South Korea. Based on 18S rRNA analysis, the strain was identified as Aspergillus awamori bxq33110. The strain showed maximum phosphate solubilization in AYG medium (525 ${\mu}g\;mL^{-1}$) followed by NBRIP medium (515 ${\mu}g\;mL^{-1}$). The strain solubilized $Ca_3(PO_4)_2$ to a greater extent and rock phosphate and $FePO_4$ to a certain extent. However $AlPO_4$ solubilizing ability of the strain was found to be very low. Glucose at the rate of 2% ($561{\mu}g\;mL^{-1}$) was found be the best carbon source for Aspergillus awamori bxq33110 to solubilize maximum amount of phosphate. However, no significant difference ($P{\leq}0.05$) in phosphorus solubilization was found between 1% and 2% glucose concentrations. $(NH_4)_2SO_4$ was the best nitrogen source for Aspergillus awamori bxq33110 followed by $NH_4Cl$ and $NH_4NO_3$. At pH 7, temperature $30^{\circ}C$ and 5% salt concentration (674 ${\mu}g\;mL^{-1}$) were found to be the optimal conditions for insoluble phosphate solubilization. However, strain Aspergillus awamori bxq33110 was shown to have the ability to solublize phosphate under different stress conditions at $30-40^{\circ}C$ temperature, pH 7-10 and 0-10% salt concentrations indicating it's potential to be used as bio-inoculants in different environmental conditions.
Background: Dental caries is one of several prevalent oral diseases caused by dental plaque biofilms. This study evaluated the anti-cariogenic effects of a bamboo salt (BS) and sodium fluoride (NaF) mixture on oral bacteria. Methods: The effects of several mixtures of NaF and BS on acid production, growth, and adhesion to glass beads of Streptococcus mutans, and their anti-cariogenic properties were investigated. The growth of S. mutans was measured according to optical density at 3, 6, 9, 12, 15, 18, and 24 hours after treatment using spectrophotometry at a wavelength of 600 nm, while pH was measured using a pH meter. Adhesion of S. mutans was measured according to the weight of glass beads from each group before and after incubation. Gene expression was measured using real-time polymerase chain reaction. Acid production and growth patterns of S. mutans were compared using repeated measures analysis of variance, followed by Scheffe's post-hoc test. The Kruskal-Wallis test was used to compare adhesion, followed by the Mann-Whitney test. Gene expression in the experimental and control samples was compared using the Student's t-test. Results: Growth, acid production, and adhesion of S. mutans were inhibited in all experimental groups. Expression of gft and fructosyltransferase in S. mutans was inhibited in all groups. A mixture of NaF and BS significantly reduced growth, acid production, adhesion, and gene expression of S. mutans compared with the other groups. Conclusion: Results of the present study demonstrated that a mixture of NaF and BS was useful as a mouth rinse in preventing dental caries.
This study was carried out to evaluate the effect of rice husk (RHB) and food waste biochar (FWB) on upland soil with sandy loam texture, in terms of physico-chemical analysis, lettuce seed germination test, and orgainc carbon leaching experiment. RHB and FWB had different physico-chemical properties each other. Carbon to nitrogen ratio (C/N ratio) of RHB was 32, showing two times higher than that of FWB. FWB had high salt and heavy metal content, compared to RHB. This is probably due to different ingredients and production processing between two biochars each other. Results of germination test with Lettuce showed lower germination rate when FWB was applied because of higher salt concentration compared to control and RHB. Organic carbon leaching test using saturated soil column (${\Phi}75{\times}h75mm$) with $10MT\;ha^{-1}$ biochar application rate, showed higher saturated hydraulic conductivity in rice husk biochar treatment column, compared to control and food waste biochar treatment. The highest total organic carbon concentration in column effluent was lower than those in both of rice husk biochar and food waste biochar, whereas the differences was negligible after 9 pore volumes of effluent. Consequently, biochars from byproducts such as rice husk and food waste in sandy loam textured upland soil could enhance a buffer function such as reduction of leaching from soil, but the harmful ingredient to crops such as high salt and heavy metals could limit the agricultural use of biochars.
To enrich the genetic resource of microbial xylanases with high activity and stability under alkaline conditions, a xylanase gene (xynSL4) was cloned from Planococcus sp. SL4, an alkaline xylanase-producing strain isolated from the sediment of soda lake Dabusu. Deduced XynSL4 consists of a putative signal peptide of 29 residues and a catalytic domain (30-380 residues) of glycosyl hydrolase family 10, and shares the highest identity of 77% with a hypothetical protein from Planomicrobium glaciei CHR43. Phylogenetic analysis indicated that deduced XynSL4 is closely related with thermophilic and alkaline xylanases from Geobacillus and Bacillus species. The gene xynSL4 was expressed heterologously in Escherichia coli and the recombinant enzyme showed some superior properties. Purified recombinant XynSL4 (rXynSL4) was highly active and stable over the neutral and alkaline pH range from 6 to 11, with maximum activity at pH 7 and more than 60% activity at pH 11. It had an apparent temperature optimum of 70℃ and retained stable at this temperature in the presence of substrate. rXynSL4 was highly halotolerant, retaining more than 55% activity with 0.25-3.0 M NaCl and was stable at the concentration of NaCl up to 4M. The enzyme activity was significantly enhanced by β-mercaptoethanol and Ca2+ but strongly inhibited by heavy-metal ions and SDS. This thermophilic and alkaline- and salt-tolerant enzyme has great potential for basic research and industrial applications.
The purpose of this study was to investigate dietary habits and food compliance in stroke patients. One-hundred sixty eight elderly stroke patients and 97 young patients with first -ever stroke admitted to Asan Medical Center between 1994 and 1998 were studied. Using a structured interview, we assessed food intake. food consumption frequency and compliance to low salt, low meat hight fish and high fruit and vegetable diets. These results were analyzed with X$^2$, t-tests, and analysis of variance (ANOVA) using the SAS package program. Salted food intake and cholesterol-containing food frequency were increased whereas frequency of fruits and vegetables intake was decreased in young stroke patients compared to the elderly. Meat intake and cholesterol-containing food frequency were increased in the males compared to the females in elderly stroke patients. and fish intake and cholesterol-containing food frequency were higher in the males than the females in the young. In patients with high economic status, frequency of fruits and vegetables was elevated. Also compliance the low meat and high fruit and vegetable diet in young patients was lower than that in the elderly. When the life-style risk factors influencing the food intake of frequency of fruits and vegetables was affected by education in young stroke patients. In elderly stroke patients, meat intake frequency of cholesterol-containing foods and fruits and vegetables were influenced by sex and /or income. Our results suggest that dietary intake of salt meat ,cholesterol-containing foods. fruits and vegetables in stroke patients may vary with age, sex the presence of risk factors or economic status therefore guidelines and nutrition education should by formulated to prevent stroke recurrence based on dietary habits and risk factors of individual patients.
