  • We have isolated a full-length cDNA clone, CaCDPK4 encoding a typical calcium-dependent protein kinase (CDPK) from hot pepper cDNA library. Genomic southern blot analysis showed that it belongs to a multigene family, but represents a single copy gone in hot pepper genome. RNA expression pattern of this gene revealed that it is induced by infiltration of Xanthomonas axonopodis pv. glycines Bra into hot pepper leaves but not by water deficit stress. However, high salt treatment of NaCl (0.4 M) solution to hot pepper plants strongly induced CaCDPK4 gene. In addition, this gene is weakly responsive to the exogenous application of salicylic acid or ethephon. Biochemical study of the GST-CaCDPK4 recominant protein showed that it autophosphorylates in vitro and the presence of EGTA, a calcium chelater, eliminates the kinase activity of the recombinant protein. As a way to identify the in vivo function of CaCDPK4 in plants, VIGS (Virus-Induced Gene Silencing) was employed. Agrobacterium-mediated TRV silencing construct containing the kinase and calmodulin domain of CaCDPK4 resulted in cell death of Nicotiana benthamiana plants. A highly homologous H benthamiana CDPK gene, NbCDPK5, to CaCDPK4 was cloned from N. benthamiana cDNA library. VIGS of NbCDPK5 also resulted in cell death. The molecular characterization of this cell death phenotype is being under investigation.

  • Concerns remain regarding the biocompatibility and adverse effects of dental casting alloys. The aim of this study was to understand the cytopathogenic effect of metal ions, which might be released from dental alloys, on oral squamous carcinoma(OSC) cells. The cellular morphology, viability, the type of cell death and molecular change in response to metal ion salt solutions including aluminum(Al), cobalt(Co), copper(Cu) and nickel(Ni) were examined. The $TC_{50}$ values for the metal ions with the exception of AI were estimated to be between 400 and $600{\mu}M$. The cells treated with the metal ions showed apoptotic change with the exception of Al ions. Metal ion-induced apoptosis was further confirmed using flow cytometric analysis. This study showed that the cytotoxicity and the mode of cell death by metal ions clearly depend on the cell type, the type of metal ion and the duration of exposure. The protein level of Rb, a tumor suppressor that affects apoptosis para-doxically, was higher in the cells treated with Co, Cu and Ni. It is believed that apoptosis and cell damage in the OSC cells treated with Co, Cu or Ni can be evoked by the regulation of Rb.

  • $NO^3$-N removal was examined from July 2002 to December 2002 of a surface-flow constructed treatment wetland cell, which was a part of a treatment wetland system composed of four wetland cells and one distribution pond. The system was established on rice paddy near the Kohung Estuarine Lake located at the southern part of the Korean Peninsula. The lake and the paddy were formed by a salt marsh reclamation project. Effluent from a secondary-level treatment plant was funneled into the system. The investigated cell was created in June 2002. Its dimensions were 87 m in length and 14 m in width. It had an open water zone at its center, which was equivalent to 10 percent of its total area. Reeds(Phragmites australis) were transplanted from natural wetlands into the cell and their stems were cut at about 40 cm height from their bottom ends. Average 25 $m^3$/day of effluent from the plant was funneled into the cell by gravity flow and average 24.2$m^3$/day of its treated effluent was discharged into the Sinyang Stream flowing into the lake. Its water depth was maintained about 0.2 m and its hydraulic detention time averaged 5.2 days. The average height of the reed stems was 45.2 cm in July 2002 and 80.5 cm in September 2002. The number of stems averaged 40.3 stems/$m^2$ in July 2002 and 74.5 stems/$m^2$ in September 2002. The reeds were established initially well. $NO_3$-N loading rate of influent and effluent averaged 173.7 and $93.5mg/m2{\cdot}day$, respectively. Removal of $NO_3$-N averaged $80.2mg/m2{\cdot}day$ and its removal rate by mass was about 50 %. Considering the initial operation of the cell and the inclusion of the cold months of November and December in the analysis period, the $NO_3$-N removal rate was good.

  • Seed viability testing provides valuable information for assessing seed lot germinability. However, most testing methods require destruction of seed prior to test. Because the dissected seeds for viability test cannot be used further evaluation, the nondestructively X-ray photography technique that can be applied for the evaluation of seed quality has been developed. In order to know the validity and accuracy of X-ray photography technique in seed evaluation test that conducted to remove the abnormal seed from a seed lot, we have compared the results from tetrazolium viability test, germination test and X-ray contrast method in cotton. Metallic salts treatment increased the efficiency of X-ray photographic method by enhancing the penetration of X-ray in abnormal or damaged seeds rather than normal seeds that have strong and well-organized tissues in seed. Cotton seeds presoaked for 16 hr in distilled water followed by soaking into metallic salt solution (5% NaI in water) for 60 min were easily classified seeds into dead seed and viable seed based on the radiography images obtained by X-ray radiation. We concluded that soft X-ray photography was reliable to find out the various defective characters due to heat and mechanical damage of seeds.

  • To shorten the production cycle of Zaodan, this study first pickled Zaodan by a novel technology - vacuum decompression technology. Vacuum decompression technology could reduce the pickling time of Zaodan from 20 wk to about 9 wk. The protein content, moisture and pH of the Zaodan egg white gradually decreased with a concomitant increase in salt during the pickling process. The total sulfhydryl group (SH) group content of the egg white proteins was increased to $2.43{\times}10^{-3}mol/L$ after being pickled for 30 d, whereas the content of disulphide bonds (SS) was reduced to $23.35{\times}10^{-3}mol/L$. The surface hydrophobicity was lowest after pickling for 30 d. In addition, great changes occurred in the secondary structure of the egg white proteins after pickling for 20 d. The disappearance of ovomucin was noticeable based on sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis.

  • To investigate the oil degrading properties of psychrotrophic bacterium Acinetobacter calcoaceticus Al-l the effects of environmental factors on this bacterium were studied. The optimal environmental conditions for cell growth rate and oil-emulsifying activity were as follows; temperature $15^{\circ}C$, pH 7.5, salt concentration 0- 3% and crude oil concentration 0.1%. Additionally the optimal concentration of Nand P source for cell growth rate and oil-emulsifying activity were 0.76 mM and 0.057 mM as $(NH_4)HS0_4$ and $K_2HP0_4$, respectively. Analysis of remnant oil by gas chromatography showed time dependent oil degradation pattern by A. calcoaceticus during cultivation; At lOoe and $15^{circ}$e, most alkane peaks were disappeared and it showed large quantities of crude oil were degraded. But at $25^{circ}$e alkane compounds in the crude oil were partially degraded even after 120 hours incubation.

  • A new tetracyclic triterpenoid [4,4,24-trimethylcholesta-${\Delta}^{8,9;14,5;24,28}$-trien-$3{\beta},11{\beta},12{\alpha}$-triol-12-acetate, 3-sulfate] sodium salt (1), together with eight known compounds including ergosterol $5{\alpha},8{\alpha}$-endoperoxide (2), 1,9-dihydroxy-3-methoxy-2-methylpterocarpan (3), 3-O-${\beta}$-D-2-acetyl-amino-2-deoxyglucopyranoxyloleanoic acid (4), hydnocarpin (5), derrone (6), isovitexin (7), erythrinin C (8), and 5,4'-dihydroxy-2"-hydroxyisopropyldihydrofurano [4,5:7,8]-isoflavone (9), were isolated from the EtOAc soluble fraction of the methanol extract of aerial part of Desmodium uncinatum collected in the western highland of Cameroon. The structures of these compounds were established by comprehensive interpretation of their spectral data mainly including 1D- ($^1H$ and $^{13}C$), 2D-NMR($^1H$-$^1H$ COSY, HMQC, HMBC) spectroscopic and ESI-TOF-MS mass spectrometric analysis. The isolation of an integracide-like compound from plant origin is a very unusual finding.

  • The potential antioxidant activities of different fractions from Prunella vulgaris var. lilacina were assayed in vitro. Among several fractions, n-BuOH fraction showed the highest 1,1-di[henyl-2-picrylhydrazyl (DPPH) free radical scavenging ($IC_{50}=0.50{\mu}g/mL$). The results of 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) assay showed the concentration dependency and n-BuOH fraction appeared a better result than the other fractions at the same concentrati on in this study. Moreover the total phenol and flavonoid contents of n-BuOH fraction contained the highest level. Additionally, correlation analysis indicated a high correlation between the antiradical activity and the total phenolic and flavonoid contents (p < 0.001). It suggests that n-BuOH fraction obtained from the 70% EtOH crude extract of Prunella vulgaris var. lilacina has wide potential for use as a source of antioxidant material.

  • The purpose of this study was to evaluate the salt water resistance of concrete depending on various types of cement. In this regard, 5 types of concrete were selected and their strength, void ratio and chloride ion diffusion characteristics were tested, and mutual correlation were analyzed. From the test results, the compressive strength and void ratio of concrete which using Type V cement was as good as Type I cement at long-term ages but the chloride diffusion coefficient of Type V cement was larger than Type I cement. And the concrete replacing some portion of the Type I cement with fly ash was superior in the cases of compressive strength, void ratio and the resistance of chloride ion permeation compared to the Type I cement with the lapse of ages. On the other hand, the compressive strength, the void ratio and the chloride diffusion coefficient of the concrete all indicated high levels of the correlation coefficient and the coefficient of determination regardless of the type of cement.
