시마연어 0+기 스몰트에 대한 유전율 추정 (Estimation of Heritability for Underyearing Smolt of Masu Salmon (Oncorhpchus masou))

  • Heritability estimations of masu salmon smelt at underyearing of age were described. Masu salmon was taken from aquaculture station of Mori in Japan. After fertilization, the offsprings in each family were reared in separate egg trays and fingerling tanks. The percentage of fish in each tank which smoltified at 11-month old was recorded, and environmental and genetic factors influencing the percentage smoltification were also studied. Percentage of smoltification was not significantly affected by the number of fish held in a tank, or fish density (correlation coefficient 0.10). However, there was a high correlation ($0.821\~0.565$) between smelt percentage and growth related traits of fish in a family. Heritability estimated from total data was $0.47\pm0.13$, when effects of tanks were ignored.

  • The pathogenicity of viral hemorrhagic septicemia virus (VHSV) from olive flounder Paralichthys olivaceus was investigated with masu salmon Oncorhynchus masou fry. The cumulative mortality of fish challenged with FYeosu05 isolate at $10^{6.5}$ $TCID_{50}$/fish was 60%. No mortality was observed in fish challenged with the isolates at $10^{5.5}$ $TCID_{50}$/fish and in mock-challenged fish. The affected fish showed darkening of the body, expanded abdomen, pale gills and enlarged spleen. VHSV from $10^{6.3}$ to $10^{7.8}$ $TCID_{50}$/g-tissue was re-isolated from the dead fish. These results suggest that the VHSV from olive flounder is pathogenic to masu salmon fry, although with low virulence.

  • A genetic analysis of reproductive characters of masu salmon at three year classes was described. the reproductive performance of masu salmon spawning at 2 years of age was analyzed using data number fertility hatchability and growth traits to the juvenile stage. The phenotypic correlations among the traits were also estimated. it was determined that egg volume was the principle deter-minant of egg number and that the relationship of number to size was negative. It is recommended that selection for egg size be included in all selection programs and egg number be ignored in any welection program designed to increase body size. Phenotypic correlations between body size of parents and egg traits as well as between body size of offspring and egg traits were not significantly positive or negative magnitude at three year classes.

Effects of oyster mushroom as a feed addictive in juvenile cherry salmon

  • Our research on juvenile Oncorhynchus masou masou in oyster mushroom supplemented diet was studied to investigate the effect of feeding. Mixing of feed ingredients for dried oyster mushrooms, 3.5, 7.0, 10.5, 14.0 (%) was added to the amount of oyster mushroom dietary beta-glucan content of the more abundant. After the weight of feed given to salmon survey oyster mushrooms diet for 3.5 to 7.0% was similar to the formula feed and the weight of the fish ate oyster mushroom feed over 10.5% were reduced. The oyster mushroom of the experimental diets containing 3.5 to 7.0 percent hepatosomatic index and feed coefficient figure was similar to those of the formulated diets. Therefore we have juvenile cherry salmon fed diets containing 3.5 to 7.0% was considered good to eat and additional research on the immune response will be carried out was necessary.

  • 한국산 연어속에 속하는 연어, 산천어 및 무지개송어 3종의 형태학적 비교 연구를 하였다. 연어속 어류인 연어, 산천어, 무지개송어의 계측, 계수형질을 비교한 결과 체형에선 암컷이 두장을 비롯한 13개, 수컷이 11개 형질에서 차이가 있었고, 계수 형질에선 유문수와 새파수에서 가장 뚜렷한 차이를 나타내었다. 이들 세 종은 암, 수컷 모두에서 표준체장에 대한 체고, 미병고, 등지느러미 길이, 뒷지느러미의 길이와 두장에 대한 주둥이 길이, 안경, 위턱, 뺨의 비에서 차이를 보였다. 이중 두장에 대한 안경은 암수 모두에서 산천어 무지개송어, 연어 순으로 크게 나타나 3종을 분류할 수 있는 새로운 형질로 나타났다. 유문수 수는 연어가 103~236개(평균 151개)로 무지개송어 37~84개(평균 56개), 산천어 23~63개(평균 41개)로 종간에 좋은 분류 형질로 나타났다. 본 연구에 나타난 체형 및 계수 형질 분석 결과 산천어는 연어와 무지개송어의 중간 형질을 가지고 있는 종으로 나타나, Smith and Stealy(1989)의 계통수와 잘 일치하였다.

  • This study was conducted to improve yield and functional properties of autoclave-treated salmon frame extracts (SFETA) using commercial enzymes (Alcalase 2.4 L FG, Flavourzyme 500 MG, Neutrase 0.8 L and Protamex 1.5 MG). Yield and angiotensin I converting enzyme (ACE) inhibitory activity of all enzymatic hydrolysates improved compared to those of control (undigested extracts), which were the highest in hydrolysates incubated with Protamex 1.5 MG for 4 hrs (P4-treated hydrolysates) and 2 hrs (P2-treated hydrolysates), respectively. However, antioxidant activities of all enzymatic hydrolysates showed less than 29%. According to the trichloroacetic acid soluble-N, volatile component intensity and sensory evaluation, when compared to control, taste of P4-treated hydrolysates improved, while its fish odor strongly smelt. Therefore, for efficient use of P4-hydrolysates, the fish odor should be improved by Maillard reaction of extracts or pre-treatment of salmon frame.

  • 본 연구는 PCR을 이용하여 brook trout의 성장호르몬 cDNA를 증폭시켰다. 증폭된 brook trout의 성장호르몬 cDNA의 전 염기서열은 1,120bp로 13bp의 5'uncoding region과 477bp의 3'uncoding region 그리고 630bp로 coding되는 210 아미노산 잔기가 open reading frame(ORF)을 구성하고 있음을 알았다, 또한, ORF의 아미노산 배열로부터 22개의 아미노산으로 이루어진 signal peptide 그리고 4개의 cysteine 잔기로부터 2개의 disulfide bond 결합을 하고있었으며, 이는 성장호르몬 단백질이 종을 초월하여 2개의 disulfide bond 결합으로 이루어진 고치구조를 형성하고 있는 것이 시사되었다. 그리고 Atlantic salmon과 97.1%, chum salmon과 94.8%, rainbow trout와 94.3%, coho salmon과 91.9%, tuna와 66.2%, tilapia와 63.5%, yellow tail과 62.9%, carp와 62.3%, flounder와 53.8%, eel과 48.1%의 아미노산 상동성을 나타냈다.

Nutritional Value and Bioactive Properties of Enzymatic Hydrolysates prepared from the Livers of Oncorhynchus keta and Oncorhynchus gorbuscha (Pacific Salmon)

  • Calculated chemical scores (computed in relation to the FAO/WHO reference protein) for salmon liver protein hydrolysates indicated that all amino acids (other than methionine and threonine) were present in adequate or excess quantities; thus, the raw liver material is a good source of essential amino acids. The hydrophobic amino acids contents in hydrolysates prepared from Oncorhynchus keta and O. gorbuscha were 38.4 and 39.1%, respectively. The proportion of released peptides exceeding 500 kDa was reduced when hydrolysates were treated with the commercial enzyme Alcalase, although proportions in the following MW ranges were elevated: 100-500 kDa and <50 kDa. The optimal conditions for enzymatic hydrolysis were as follows: pH 7.0, $50^{\circ}C$, and a reaction time of 1 h. Of the different proteases tested, Alcalase was the most efficient for production of salmon liver hydrolysate with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity. The hydrolysates prepared from salmon liver had a balanced amino acid composition. The liver protein hydrolysates contained low molecular weight peptides, some of which may be bio-active; this bio-active potential should be investigated. Inhibition of the DPPH radical increased with increased degree of hydrolysis (DH), regardless of protease type. DPPH radical scavenging abilities, antithrombotic effects and ${\alpha}$-glucosidase enzyme inhibition effects of O. keta liver hydrolysate increased in a dose-dependent manner. Thus, salmon liver hydrolysate may be useful in functional food applications and as a source of novel products.

Prevalence and Characterization of Typical Aeromonas salmonicida Chum Salmon Isolates in Korea

  • Aeromonas salmonicida is an important fish pathogen commonly associated with furunculosis in salmonids. Typical A. salmonicida strains have the surface virulence A-layer protein, a major virulence determinant encoded by the vapA gene. In this study, 880 chum salmon Oncorhynchus keta were collected from the east coast of Korea during 2006-2011, including 560 wild adults and 320 artificially hatched fry pools, and the presence of typical A. salmonicida was examined by PCR using the typical A. salmonicida-specific vapA gene primers. The results demonstrated that 34.5% of the samples (304/880 samples) were PCR positive, implying that a typical A. salmonicida infection is highly prevalent among chum salmon in Korea. Twenty typical A. salmonicida isolates were recovered based on their brown pigmentation on Trypticase Soy Agar (TSA) plates, which indicates the existence of the A-layer protein. Further biochemical analyses with the four randomly selected typical A. salmonicida isolates revealed some variations in their amino acid decarboxylation and carbohydrate fermentation activity. A phylogenetic analysis based on the entire vapA gene sequence suggested that the A. salmonicida isolates from chum salmon were clustered with those isolated from Atlantic salmon in Europe. Further study is needed to resolve such an interesting relationship in detail.

급격한 염분변화에 따른 담수어 3종의 프로락틴 및 성장호르몬 유전자 발현변화 (Changes in Prolactin and Growth Hormone Gene Expression in Three Freshwater Teleosts with Rapid Changes in Salinity)

  • The changes in osmolality and the gene expression profiles of prolactin (PRL) and growth hormone (GH) with rapid changes in salinity were compared in the eel (Anguilla japonica), crucian carp (Carassius carassius), and masu salmon (Oncorhynchus masou). Fish stocked in freshwater (FW) were abruptly transferred to experimental tanks containing FW, 50% seawater (50% SW), or 100% SW (SW). Blood samples and pituitary glands were collected 2 and 24 hrs after the exposure. No mortality was observed in SW eel (n=6), whereas all of the crucian carp (n=6) and two masu salmon (n=6) exposed to SW died after land 24 hrs, respectively. The PRL mRNA levels of the eel and masu salmon decreased in 50% SW and SW compared to those of the fish kept in FW after 24 hrs, whereas the PRL levels of crucian carp were higher in 50% SW than in FW. Unlike the PRL mRNA levels, the GH mRNA levels of the eel did not differ significantly among three different salinities, while the GH mRNA levels of crucian carp and masu salmon increased significantly in 50% SW and SW after 24 hrs. The serum osmolalities increased marginally in the eel and masu salmon in 50% SW at 24 hrs (19% and 9%, respectively), whereas those of crucian carp increased abruptly in 50% SW (50% increase). These results suggest that the synthesis of PRL and GH is important in relation to the osmoregulatory system with environmental changes in salinity.
