The present investigation was carried out to identify salivary components of mucosal pellicle and to explore the difference of mucosal pellicle components according to the location of oral mucosa. By using antisera and immunoblotting, high-(MG1) and low-(MG2) molecular-mass salivary mucins, amylase, IgA, proline-rich proteins(PRPs) were detected in mucosal pellicle in vivo. In addition, the data indicated that mucins, IgA and proline-rich proteins could be cleaved into lower-molecular-mass products, whereas the IgA, proline-rich proteins could also be cross-linked into higher-molecular-mass complexes. Mucosal pellicles from buccal, labial and palatal mucosa showed similar pattern in immunoblotting experiments using anti-MG2 and anti-PRPs antisera. The data from this study suggest that during mucosal pellicle formation multiple components of saliva adsorb to oral mucosal epithelial cell surfaces, and selected components can be proteolytically cleaved into smaller fragments and/or cross-linked into higher-molecular products.
Purpose: The reaction of cells to a titanium implant depends on the surface characteristics of the implant which are affected by decontamination. The aim of this study was to evaluate the cytocompatibility of titanium disks treated with various decontamination methods, using salivary bacterial contamination with dental pellicle formation as an in vitro model. Methods: Sand-blasted and acid-etched (SA) titanium disks were used. Three control groups (pristine SA disks [SA group]; salivary pellicle-coated SA disks [pellicle group]; and biofilm-coated, untreated SA disks [NT group]) were not subjected to any decontamination treatments. Decontamination of the biofilm-coated disks was performed by 14 methods, including ultrasonic instruments, rotating instruments, an air-powder abrasive system, a laser, and chemical agents. MG63 cells were cultured in the presence of the treated disks. Cell proliferation assays were performed on days 2 and 5 of cell culture, and cell morphology was analyzed by immunofluorescence and scanning electron microscopy (SEM). A vascular endothelial growth factor (VEGF) assay was performed on day 5 of culture. Results: The cell proliferation assay revealed that all decontaminated disks, except for the 2 groups treated using a plastic tip, showed significantly less cell proliferation than the SA group. The immunofluorescence and SEM analyses revealed that most groups showed comparable cell density, with the exception of the NT group, in which the cell density was lower and bacterial residue was observed. Furthermore, the cells grown with tetracycline-treated titanium disks showed significantly lower VEGF production than those in the SA group. Conclusions: None of the decontamination methods resulted in cytocompatibility similar to that of pristine SA titanium. However, many methods caused improvement in the biocompatibility of the titanium disks in comparison with the biofilm-coated, untreated titanium disks. This suggests that decontamination is indispensable for the treatment of peri-implantitis, even if the original biocompatibility cannot be restored.
The present study was performed to investigate the binding between salivary proteins(low-molecular-weight mucin;MG2, amylase, proline-rich proteins;PRPs) and oral pathogens(Streptococcus gordonii, Actinomyces viscosus, Staphylococcus aureus) by using solid-phase assay. In the case of transferring proteins to Immobilon-P, S. gordonii binds to MG2. A. viscosus binds to MG2, amylase, and PRPs, and S. aureus binds to MG2 and amylase. On nitrocellulose membrane, S, gordonii and A. viscosus bind to MG2, amylase, and PRPs. S. aureus binds to MG2 and PRPs. However, rabbit anti-A. viscosus antisera and rabbit anti-S. aureus antisera showed cross reactivity to PRPs adsorbed to only nitrocellulose membrane in negative control experiments, which were done without bacterial overlay. The results were different according to the membrane used as solid-phase, which reflected the assay-sensitive nature of binding experiment. PRPs and amylase are known to be components of tooth enamel pellicle. In addition, there was experimental evidence that PRPs and MG2 may covalently bind to oral mucosal epithelium. Considering above facts, the results of the present study can provide information on the interactions between salivary proteins and oral bacteria on tooth and oral mucosal surfaces.
The adherence of $^{3}H$-labeled oral streptococcal cells to protein-coated hydroxyapatite (HA) beads was studied by a standard adherence assay. The adherence equilibrium for S. mutans 10449 occured in about 2 hrs. The cell numbers adhering to SHA was 50% less than those on bare HA. Sailva from different subjects had varying effect on bacterial adherence. The use of saliva adsorbed with homologouis bacteria decreased S. mutans adherence by 38% ; this indicates the presence of salivary agglutinin in acquired pellicle formed on HA. Animal sera and BSA decreased S. sanguis adherence. BSA concentration as high as 10mg/ml caused up to 87% adherence inhibition. The desorption experiment of adhered bacteria confirmed the previous reports that the adhesive sites on HA beads for S. mutans were different from those for S. sanguis and that S. mutans could enhance the adherence of S. sanguis but not vice versa.
Silver diamine fluoride(SDF)는 우식의 정지를 위해 사용될 수 있는 효과적이고 경제적인 약제로, 협조가 힘든 환자나 의과적인 문제로 치료를 받기 힘든 경우 등에서 사용될 수 있다. 이 연구는 타액 생물막을 이용하여 SDF 의 항미생물효과를 평가하고자 시행되었다. 타액을 이용하여 획득피막 유사 구조를 형성한 후, 타액과 Streptococcus mutans를 첨가하여 우식원성의 타액 생물막을 형성하였다. 타액 생물막에 SDF 를 처리한 결과, 전체 미생물과 S. mutans의 수가 유의하게 감소하였다(p < 0.000). CLSM에서도 생물막 내 죽은 세포가 증가하였다. 유치 법랑질과 상아질 시편에 SDF를 처리한 후 우식원성의 생물막을 배양한 결과, SDF 를 처리할 경우 생물막 형성이 저해되는 것을 확인할 수 있었다(p = 0.029). 연구 결과 SDF의 뛰어난 항미생물효과를 확인할 수 있었으며, 이로 인한 임상에서의 항우식효과를 기대할 수 있다.
치과용 임플란트 실패의 주요 원인은 임플란트 표면에 부착되는 세균의 침착의 결과로 생기는 임플란트 주위염이다. 구강 내에서 세균성 치태의 침착은 치태가 부착하는 기질 표면의 물리적 성상과 타액성 피막의 성분에 영향을 받으며 형성된 피막의 유기질 성분의 차이가 치태의 성분과 병원성에 영향을 미친다. 최근 연구에 의하면 생체재료의 표면에 침착되는 치태세균은 사용되는 재료에 따라 특이한 세균 침착을 보이며 이는 초기 타액성 피막의 차이에 의한 것으로 알려져 있다. 이 연구의 목적은 플라즈마분사법으로 표면 처리된 타이태늄 임플란트에 흡착되는 타액성 단백질 피막의 특성을 정성적인 방법으로 분석하는 데 있었다. 법랑질 조각과 플라즈마분사법으로 표면 처리된 타이태늄 임플란트를 스프린트에 치실을 이용하여 연결한 장치를 구강 내 장착하여 2시간 동안 피막이 침착되게 한 후 피막을 분리 추출하여 냉동 건조시켰다. 재수화 과정을 거치고 나서 전기 영동법과 Western transfer 분석을 통해 단백질 성분에 관한 분석을 시행하였다. 사람의 총 타액과 이하선 타액 및 악하선-설하선 타액을 수집기를 이용하여 채취하고 같은 방법으로 처리한 후 성분분석을 실시하였다. 피막 흡착 전후의 표면변화를 주사전자현미경을 이용하여 관찰하였다. 실험결과 타이태늄 임플란트에 흡착된 피막은 법랑질 표면의 피막과는 다른 단백질 성분을 가지고 있었으며, 주로 악하선-설하선 타액에서 유래하였다. 임플란트와 법랑질 표면 모두에서 흡착된 피막에는 아밀라제, 분비성 면역 글로불린A 및 락토페린이 존재함을 알 수 있었으나 법랑질의 경우는 blotting이 약하게 나타났다. 주사전자현미경 관찰결과 시편의 표면에 균질한 피막이 덮고 있었으며 세균의 부착은 거의 관찰되지 않았다. 이상의 실험 결과들을 통하여 플라즈마분사법으로 표면 처리된 타이태늄 임플란트 표면에 부착된 타액성 단백질 성분은 법랑질과는 차이가 있음을 알 수 있었으며, 이러한 차이는 치태세균의 종류 및 병원성에 영향을 미칠 것으로 생각된다. 법랑질과 타이태늄 임플란트는 기질과 표면구조가 다르므로 표면에 형성 되는 치태성분도 다르다는 사실과 본 연구 결과를 종합하여 볼 때, 타이태늄 임플란트 표면에 흡착되는 초기 타액성 단백질의 성불이 타이태늄 표면에 침착되는 미생물 군의 조절에 중요한 역할을 가지고 있으며, 임플란트 치료 시에 올바른 치태 관리법의 교육을 통하여 환자 스스로 적절한 관리를 하도록 함으로써 임플란트 치료의 성공률을 높일 수 있을 것으로 생각된다.
Salivary proteins which are produced in the saliary acinar cells have been known to be involved in the Calcium and phosphate metabolism. The acquired pellicle resulting from such metabolism is considered as a secondary defence membrane against tooth caries. In this respect, some proteins included in saliva probably play an important role in the prevention of demineralization in enamel. On the other hand, fluoride has long been known to prevent the demineralization of enamel by the inhibition of the growth of Streptococcus mutans(S. mutans) and by the chemical reaction with calcium and phosphate, Therefore, I have examined the roles of amylase and albumin in the demineralization of enamel and compared these preteins with fluoride in terms of anticariogenic effect. 1. The demineralization caused by S. mutans occurred slowly and progressively for the first 60 min, then the rate of demineralization was accelerated afterwards. 2. pH decreased continuously during the entire period of each experiment. 3. The demineralization was significantly inhibited by the preteatment of amylase and fluoride but albumin had little effect on it. 4. An addition of 0.1 mM lactic acid (final concentration 0.1 ${\mu}M$) caused a rapid increase in calcium concentration reaching a maximum within 10 min. 5. pH decreased rapidly by the addition of 0.1 mM lactic acid and reached a minimum within a few seconds followed by an increase in pH. pH reaced a plateu with 10 min. 6. Fluoride, amylase and albumin played little role in the 0.1 mM lactic acid-induced demineralization. 7. A slow infusion of 0.1 M lactic acid at a rate of 5 ${\mu}l/min$ caused a slower increase in calcium concentration compared with the bolus addition of lactic acid. 8. Fluoride had an inhibitory effect on the calcium release caused by slow infusion of lactic acid while amylase and albumin had no effect on it. These results suggest that fluoride inhibits demineralization by protecting the HA from the acid attack whereas amylase has a direct effect on S. mutans to prevent demineralization.
치아우식증의 원인균으로 알려져 있는 Streptococcus mutans의 여러 균주 중 GS-5균주는 AgI/II 유전자 내에 생기는 nonsense mutation에 의하여 절편의 분비형 AgI/II 단백질을 발현한다. 이러한 사실은 S. nutans GS-5가 독특한 임상 기능을 가질 수 있음을 암시하며, 최근의 보고는 임상적으로 분리된 S. mutans 균주를 이용한 실험 결과에 근거하여 이러한 가능성을 지지한다. 본 연구는 이전의 실험을 통하여 S. mutans의 agI/II 유전자를 확보한 후 재조합 단백질인 N-terminal AgI/II 단백질을 생산하였다. 이 후 하이브리도마를 통하여 1C11A라 명명되는 세포막 단백질 AgI/II에 대한 단항체를 생산하였으며, Western blot과 ELISA를 통하여 이 항체가 AgI/II 단백질에 매우 높은 특이성을 나타냄을 알 수 있었다. 새로이 얻어진 단항체는 AgI/II와 관련된 질환의 기전을 규명하는데 기초 재료가 될 것이다.
치아 우식은 구강 내 미생물에 의해 치아 석회 조직의 일부가 용해되고 파괴되는 감염성 질환이다. 치아 우식의 원인균은 Streptococcus mutans와 같은 Mutans streptococci로 알려져 있고, 이 미생물이 치면에 접착하여 군집을 형성하는 능력이 균의 독성에 중요한 역할을 한다. S. mutans가 치면의 타액성 피막에 부착하는 데에는 AgI/II와 같은 세포표면의 섬유성 단백질을 매개로 한다. 그러므로, AgI/II는 S. mutans GS-5에 대한 백신 개발에 적절한 목표가 된다. 본 실험은 S. mutans GS-5로부터 AgI/II 유전자를 복제하고 염기서열분석을 하였다. 복제된 AgI/II와 앞서 보고된 S. mutans GS-5의 해당 부위의 280개의 핵산은 완벽하게 일치하였다. 복제된 유전자를 두 부위로 절단하여 형질전환을 통해 재조합 단백질인 AgI/IImr을 얻었고, 정제된 재조합 단백질을 쥐에게 주입하여 다클론 항체를 얻었다. 추출된 다클론 항체는 AgI/IImr항원단백질에 반응하였고, 대조군으로 쓰인 단백질에는 반응하지 않았다.
동물성 mucin은 인체 타액 mucin과 유사한 구조적 특성을 가지고 있으므로 효과적인 타액대체제의 개발에 적합한 성분으로 여겨져 왔다. 구강건조증 환자가 동물성 mucin 함유 타액대체제를 사용할 경우, 동물성 mucin과 인체 타액에 존재하는 항균 단백질이 용액상태인 전타액과 구강표면에 형성된 pellicle에 동시에 존재할 수 있으므로 이들 물질사이에 상호작용이 일어 날 수 있을 것이다. 본 연구의 목적은 용액과 hydroxyapatite(HA) 표면에서 동물성 mucin이 peroxidase 활성에 미치는 영향을 평가하기 위하여 시행되었다. 동물성 mucin이 peroxidase 활성에 미치는 영향은 돼지의 위장 mucin(porcine gastric mucin, PGM) 이나 소의 악하선 mucin(bovine submaxillary mucin, BSM)을 소의 lactoperoxidase(bovine lactoperoxidase, bLPO)나 타액검체와 incubation하는 방법을 사용하여 분석하였고, 표면상태에서의 연구를 위해 HA beads, HA disc, 소의 치아와 같은 3가지 종류의 HA 표면을 활용하였다. Peroxidase 활성은 NbsSCN 법을 이용하여 분석하였다. 1. 돼지위장 mucin은 용액상태에서 bLPO 활성을 증가시켰으나 타액검체의 peroxidase(peroxidase in saliva sample, POS) 활성에는 영향을 미치지 않았다. 2. 소 악하선 mucin은 용액상태에서 bLPO와 POS 활성에 영향을 미치지 않았다. 3. HA 표면에 부착된 돼지위장 mucin은 peroxidase의 부착과 활성을 증가시켰고 이러한 효과는 세 종류의 HA 표면 모두에서 일어났으며, POS의 활성증가는 HA beads와 소 치아 표면에서만 나타났다. 4. bLPO와 돼지위장 mucin의 혼합물을 HA 표면에 부착시킬 경우, HA beads와 HA disc 표면에서의 peroxidase 활성은 증가하였다. 5. bLPO의 돼지위장 mucin에 대한 부착친화도는 소 악하선 mucin에 비해 컸다. 이상의 결과를 종합해 볼 때 동물성 mucin은 용액상태와 HA 표면에서 peroxidase 활성에 영향을 미침을 알 수 있으며, 동물성 mucin을 포함하고 있는 타액대체제는 인체타액 및 타액대체제에 있는 peroxidase 활성에 영향을 미칠 수 있을 것이다.
