Salinity Tolerance of Progenies between Korean Cultivars and IRRI's New Plant Type Lines in Rice

  • To select new germplasm for salinity tolerance from new plant type (NPT) breeding lines, the sixty F$_4$ lines selected from the crosses between Korean cultivars and IRRI's NPT lines were evaluated for salinity tolerance at the seedling stage with salinized culture solution (EC=12 dS/m) in the controlled conditions. Two NTP lines derived from a cross between 'Ilmibyeo' and 'IR66152-AC5-1', 'HR15258-7-1' and 'HR15258-27-1', were found to have good tolerance. The salinity tolerance of the lines was compared to their parents and the sensitive ('IR29') and tolerant ('Pokkali') checks in three salinity levels, no salinity (control) and an EC of 12 and 16 dS/m. Visual salinity score, shoot Na+ and Na-K ratio in two NPT lines was significantly low compared with the parents and IR29. Indicating that salinity tolerance of the lines might be derived from a transgressive segregation. The relative water content of the lines was higher than Pokkali, and the dry weight of shoot and root was proportionally decreased to salinity score and salinizing concentration. The visual salinity scores were significantly correlated with shoot Na concentration, Na-K ratio, relative water content, and reduction of dry weight (P<0.01). Their tolerance was attributed to root and shoot characteristics that led to high shoot water content, thus diluting the toxic effect of salts.

Evaluation and classification of selected rice varieties for salinity tolerance at seedling stage

  • To identify the new source of breeding materials for rice salt tolerance, the salinity tolerance of thirty-four varieties was evaluated under 0.5% saline condition at seedling stage. The salinity score showed highly significant correlations to dry weight and dead leaf ratio. The tested varieties were classified into three groups by visual score, reduction ratio of dry weight, and dead leaf ratio. Eighteen varieties were classified as the highly tolerant group (salinity scores of 1.3-3.7), seven varieties were fallen into the tolerant group (salinity scores of 4.2-5.8), and others were susceptible (salinity scores of 6.7-9.0). In highly tolerant group, most indica varieties including Getu, Dikwee and Kuatic Putic, didn't exsert a panicle under the Korean climate. But six varieties, Xiangcho V, Annapuruna, HP 3319-2wx-6-3-1, Giza 175, and GZ 2447-S-17, GZ 4255-6-3 were suitable to the Korean climate, and their heading date (6-16, August) and culm length (65-78㎝) were similar to the Korean varieties. Accordingly, these varieties can be utilized as crossing materials for the salt tolerance in japonica rice.

  • Abiotic stress is one of the major serious limiting factors in rice (Oryza sativa) and caused rice production losses. It is important to precisely screen valuable genetic resources for improving stress tolerance and understanding tolerance mechanism to abiotic stresses. Because there are differences of experiment designs for screening of tolerant plant in several studies related to abiotic stress, this study has performed to provide the rapid and efficiency screening method for selection of tolerance rice to drought and salinity stresses. Two week-old rice seedlings that reached about three leaf stage were treated with drought and salinity stresses and examined tolerant levels with tolerant and susceptible control varieties, and transgenic plants. To determine the optimum concentration for the selection of drought and salinity condition, tolerant, susceptible and wild-type plants were grown under three soil moisture contents (5, 10 and 20% water contents) and three NaCl concentrations (100, 200 and 250 mM) for 10 days at seedling stage. 200 mM NaCl concentration and 5% moisture content soil were determined as the optimum conditions, respectively. The described methodologies in this study are simple and efficiency and might help the selection of drought and salinity tolerance plants at the 3,4-leaf-seedling stage.

  • Salinity is one of the major abiotic stresses that severely affect crop production throughout the world; especially rice plant which is generally categorized as a typical glycophyte as it cannot grow in the presence of salinity. Phenotypic resistance of salinity is expressed as the ability to survive and grow in a salinity condition. Salinity resistance has, at least implicitly, been treated as a single trait. Physiological studies of rice suggest that a range of characteristics (such as low shoot sodium concentration, compartmentation of salt in older rather than younger leaves, high potassium concentration, high $K^+/Na^+$ ratio, high biomass and plant vigour) would increase the ability of the plant to cope with salinity. Criteria for evaluating and screening salinity tolerance in crop plants vary depending on the level and duration of salt stress and the plant developmental stage. Plant growth responses to salinity vary with plant life cycle; critical stages sensitive to salinity are germination, seedling establishment and flowering. We have established a standard protocol to evaluate large rice germplasms for overall performance based on specific physiological traits for salt tolerance at seedling stage. This protocol will help in identifying germplasms which can perform better in the presence of different salinity treatments based on single trait and also combination of different physiological traits. The salt tolerant germplasm can be taken forward into developing better varieties by conventional breeding and exploring genes for salt tolerance.

Varietal Difference in Salinity Tolerance during Germination Stage of Rice

  • This study was conducted to find out a desirable screening condition for the salinity tolerance in germination of rice. Seeds of 33 rice varieties were tested in NaCl solutions with various concentration levels. The germination percentage had a decreasing tendency with increasing NaCl concentration and inhibition concentration of 50% germination was 320mM. Standard deviation of germination percentage was highest (28.6) under 300mM NaCl. There was a highly significant correlation between the 50% germination concentration and the germination percentage at 20th day after seeding in 300mM NaCl. Also in 300mM NaCl, the germination percentage at 20th day after seeding was significantly correlated with the germination percentage at the 6th day after seeding. The salinity tolerance on the basis of germination percentage at 6th day after seeding in 300mM NaCl, was strong in 'Hyangnambyeo', 'Ilmibyeo', 'Kancheogbyeo', and 'Namwonbyeo', while weak in 'Ansanbyeo', 'Odaebyeo', 'Nonganbyeo', 'Dasanbyeo', and 'Namcheonbyeo'.

Soybean Improvement for Drought, Salt and Flooding Tolerance

  • Drought, salinity and flooding are three important abiotic factors limiting soybean production worldwide. Irrigation, soil reclamation, and drainage systems are not generally available or economically feasible for soybean production. Therefore, productive soybean varieties with tolerance are a cost effective means for reducing yield losses due to these factors. Genetic variability for higher tolerance to drought, salt and flooding is important. However, only a small portion of nearly 200,000 world soybean accessions have been screened to find genotypes with tolerance for use in breeding programs. Evaluation for tolerance to drought, salinity and flooding is difficult due to lack of faster, cost effective, repeatable screening methods. Soybean strains with higher tolerance to the above stresses have been identified. Crosses with lines with drought, salt and flooding tolerance through conventional breeding has made a significant contribution to improving tolerance to abiotic stress in soybean. Molecular markers associated with tolerance to drought, salt and flooding will allow faster, reliable screening for these traits. Germplasm resources, genome sequence information and various genomic tools are available for soybean. Integration of genomic tools coupled with well-designed breeding strategies and effective uses of these resources will help to develop soybean varieties with higher tolerance to drought, salt and flooding.

Induced Tolerance to Salinity Stress by Halotolerant Bacteria Bacillus aryabhattai H19-1 and B. mesonae H20-5 in Tomato Plants

  • Salinity is one of the major abiotic stresses that cause reduction of plant growth and crop productivity. It has been reported that plant growth-promoting bacteria (PGPB) could confer abiotic stress tolerance to plants. In a previous study, we screened bacterial strains capable of enhancing plant health under abiotic stresses and identified these strains based on 16s rRNA sequencing analysis. In this study, we investigated the effects of two selected strains, Bacillus aryabhattai H19-1 and B. mesonae H20-5, on responses of tomato plants against salinity stress. As a result, they alleviated decrease in plant growth and chlorophyll content; only strain H19-1 increased carotenoid content compared to that in untreated plants under salinity stress. Strains H19-1 and H20-5 significantly decreased electrolyte leakage, whereas they increased $Ca^{2+}$ content compared to that in the untreated control. Our results also indicated that H20-5-treated plants accumulated significantly higher levels of proline, abscisic acid (ABA), and antioxidant enzyme activities compared to untreated and H19-1-treated plants during salinity stress. Moreover, strain H20-5 upregulated 9-cisepoxycarotenoid dioxygenase 1 (NCED1) and abscisic acid-response element-binding proteins 1 (AREB1) genes, otherwise strain H19-1 downregulated AREB1 in tomato plants after the salinity challenge. These findings demonstrated that strains H19-1 and H20-5 induced ABA-independent and -dependent salinity tolerance, respectively, in tomato plants, therefore these strains can be used as effective bio-fertilizers for sustainable agriculture.

급격한 저염분 노출에 따른 참돔 Pagrus major 치어의 단기 행동반응 및 내성 한계에 관한 연구 (Short-term behavioral responses and tolerance limits of red seabream Pagrus major fingerlings following sudden low salinity exposure)

  • In this study, using a continuous behavior measurement technique, the short-term behavioral responses and tolerance limits of red seabream Pagrus major fingerlings to sudden exposure to low salinity in a controlled environment were observed. The activity of the fingerlings suddenly exposed to 21.4, 17.3, and 9.8 psu increased temporarily at the initial exposure to show irregular swimming behavior, but then recovered a stable activity pattern through rapid salinity adaptation. However, the organisms suddenly exposed to 7.3 and 4.3 psu could not withstand the salinity stress, and their swimming behavior was severely disturbed and all individuals died within 48 hours. The findings suggest that red seabream underwent a temporary salinity stress process at the beginning of the exposure to concentrations of 10.0 psu or higher. At these concentrations, osmotic control was possible within at least 11 hours, so stable metabolic activity was also possible. However, organisms suddenly exposed to concentrations below 5.0 psu exceeded the tolerance to low salinity and the sublethal limit. In red seabream exposed to this concentration range, severe behavioral and metabolic disturbances were observed, and death was observed due to osmotic control failure. In conclusion, a salinity range of 5.0 to 10.0 psu can be predicted to correspond to a concentration range in which the osmotic control ability of the red seabream fingerlings is lost, and sub-lethal reactions occur.

Theoretical Effects of Altered Biological and Chemical Properties on Salinity Tolerance of Acacia seeds

  • Multiple regression equations (Rehman et al. 2000) have been developed to predict the salinity tolerance of Acacia seeds, expressed as the I$\sub$50/ (the concentration of NaCl required to reduce final germination to 50% of the control value in DW). Accurate predictions can be made using one or more chemical and biological seed parameters. In this study the theoretical effect of varying final germination percentage in distilled water, germination rate in distilled water. (Rate), Ca$\^$2+/ or K$\^$+/ contents and their ratios, as independent factors or related factors, on the predicted salinity tolerance (I$\sub$50/) of Acacia species was investigated. Simulation of the effects of changing final germination, rate, calcium and potassium suggest the possibility of practical application of these results to modify the salinity tolerance of seeds. The predicted I$\sub$50/ increased with increasing final germination. Similarly, the higher the rate of germination, the higher the predicted salt tolerance of Acacia species. The Ca$\^$2+/ content of seeds was found to be positively correlated with I$\sub$50/. Species with higher Ca$\^$2+/ contents had a higher I$\sub$50/. This suggests that I$\sub$50/ might be increased by increasing the Ca$\^$2+/ contents of seeds by pretreatment with calcium salts or by supplying these to the mother plants.

대게, Chionoecetes opilio의 수온 및 염분변화에 따른 생존내성과 Haemolymph의 삼투질과 이온 농도변화 (Temperature and Salinity Tolerance and Haemolymph Osmotic and ionic Concentration of the Snow Crab, Chionoecetes opilio)

  • 수온(5~2$0^{\circ}C$)과 염분(12~32$\textperthousand$)의 변화에 따른 대게 암수의 생존내성과, 삼투질, 이온 및 글루코스 농도 변화 등에 대한 생리적 반응을 연구하였다. 14, 16, 18$^{\circ}C$에서 50% 폐사시간은 암컷의 경우 각각 24.3일(수컷 11.3일), 10.7일(수컷 3.3일), 1.8일(수컷 0.6일)로 나타나, 수온의 증가에 따라 50% 폐사시간이 빨라지는 경향을 나타냈으며, 암수 각각 16$^{\circ}C$와 14$^{\circ}C$를 기점으로 폐사시간이 급격히 빨라졌다. 염분의 변화에 대해 암수 모두 24$\textperthousand$이상에서는 실험기간동안 50% 이하의 폐사율을 나타냈으며, 21$\textperthousand$에서 50% 폐사시간이 암컷 20.3시간(수컷 17시간), 12$\textperthousand$에서는 암컷 2.2시간(수컷 3시간)으로 나타났다 수온에 따른 삼투질 및 이온 농도의 변화에서, 수컷은 14$^{\circ}C$에서 급격한 감소경향 을 나타냈으나, 암컷은 뚜렷한 변화경향이 없었다. 염분의 변화에 대해, 암컷은 삼투질 및 이온 농도가 순응하는 경향을 나타냈으나, 수컷은 다소 불안정한 경향을 나타냈다. 또, 수온의 증가와 염분의 저하에 따라, 암수 모두 글루코스 농도가 증가하는 경향을 나타냈다.
