생존율은 염분과 관계없이 8℃, 10℃일 때는 98.7~100%였으나, 4℃일 때는 생존율이 25~55%로 나타났다. 혈림프액 내 SOD와 glutathione 농도와 같은 스트레스 지표는 모든 염분구에서 4℃, 6℃일 때 높게 나타났다. THC, hemocyte population, apoptotic cell및 necrotic cell의 비율은 염분 별로는 26 psu구에서 가장 높게 나타났고, 4~6℃일 때 다른 실험구에 비해 유의하게 높았다. 이러한 결과들은 26 psu의 염분과 6℃ 이하의 수온에서는 참전복의 스트레스 반응이 증가하였으나, 30, 34 psu의 염분과 8, 10℃의 수온에서는 대조구와 차이가 없어, 참전복의 수송을 위한 적정 수온 및 염분 조건은 8~10℃, 30~34 psu인 것으로 나타난다.
Gene-expression analysis is increasingly important in biological research, with real-time reverse PCR (RTPCR) becoming the method of choice for high-throughput and accurate expression profiling of selected genes. However, this technique requires important preliminary work for standardizing and optimizing the many parameters involved in the analysis. Plant stress studies are more and more based on gene expression. The analysis of gene expression requires sensitive and reproducible measurements for specific mRNA sequence. Several genes are regulated in response to abitoic stresses, such as salinity, and their gene products function in stress response and tolerance. The design of the primers and TaqMan probes for real-time PCR assays were carried out using the Primer $Express^{TM}$ software 3.0. The PCR efficiency was estimated through the linear regression of the dilution curve. To understand the expression pattern of various genes under salt stressed condition, we have developed a unique public resource of 9 stress-related genes in poplar. In this study, real-time RT-PCR was used to quantify the transcript level of 10 genes (9 stress-related genes and 1 house keeping gene) that could play a role in adaptation of Populus davidiana. Real-time RT-PCR analyses exhibited different expression ratios of related genes. The data obtained showed that determination of mRNA levels could constitute a new approach to study the stress response of P. davidiana after adaptation during growth in salinity condition.
국내 일부 시설재배지는 장기간 과도한 양분 투입 등에 의한 염류 집적 현상이 문제가 되어왔으며, 최근 이상기온에 따른 온도장해에 의한 피해도 발생하고 있다. 이러한 현상에 대해 친환경적으로 대처하기 위하여 고염류와 온도 스트레스에 대해 작물에 내성을 증강시키는 미생물을 선발하였다. 국내 토양에서 분리한 1,944균주중 고염류 또는 온도 스트레스 조건에서 세균의 생장과 식물생장촉진 관련 특성(IAA 생성, ACC deaminase 활성, 인산가용화능)을 고려하여 20균주를 1차 선발(전체 균주의 1.03%)하였다. 1차 선발한 20균주 중 토마토 식물검정을 통해 고염류 또는 온도스트레스에 대한 내성을 유도하는 7세균(1차 선발균주의 35%, 전체 균주의 0.36%)을 단계적으로 선발할 수 있었다. 선발된 세균은 16S rRNA 유전자의 염기서열 분석을 통해 모두 Bacillus 속에 속하는 것으로 확인되었다. 이러한 결과로 선발된 7균주는 토마토의 고염류 또는 온도 스트레스에 대한 효과적인 미생물 제제로 활용이 가능한 것을 확인할 수 있었다.

Intertidal macroalgae are regularly exposed to hypo- or hypersaline conditions which are stressful. However, red algae in New Zealand are generally poorly studied in terms of salinity tolerance. Consequently, two native (Bostrychia arbuscula W. H. Harvey [Ceramiales], Champia novae-zelandiae [J. D. Hooker & Harvey] Harvey [Rhodymeniales]) and one introduced red algal taxon (Schizymenia spp. J. Agardh [Nemastomatales]) were exposed for 5 days in a controlled salt stress experiment to investigate photosynthetic activity and osmotic acclimation. The photosynthetic activity of B. arbuscula was not affected by salinity, as reflected in an almost unchanged maximum quantum yield (Fv/Fm). In contrast, the Fv/Fm of C. novae-zelandiae and Schizymenia spp. strongly decreased under hypo- and hypersaline conditions. Treatment with different salinities led to an increase of the total organic osmolyte concentrations with rising salt stress in B. arbuscula and Schizymenia spp. In C. novae-zelandiae the highest organic osmolyte concentrations were recorded at SA 38, followed by declining amounts with further hypersaline exposure. In B. arbuscula, sorbitol was the main organic osmolyte, while the other taxa contained floridoside. The data presented indicate that all three red algal species conspicuously differ in their salt tolerance. The upper intertidal B. arbuscula exhibited a wide salinity tolerance as reflected by unaffected photosynthetic parameters and strong sorbitol accumulation under increasing salinities, and hence can be characterized as euryhaline. In contrast, the introduced Schizymenia spp. and native C. novae-zelandiae, which preferentially occur in the mid-intertidal, showed a narrower salinity tolerance. The species-specific responses reflect their respective vertical positions in the intertidal zone.
Abiotic stress is one of the most serious problems in plant productivity because it dramatically delays plant growth and development. One of the abiotic stresses, soil salinity, has an adverse effect on plant growth, particularly in areas where irrigation is necessary like semiarid Asia and Africa. Among several physiological parameters, anthocyanin accumulation is a valuable indicator of the condition of the plant, and it tends to increase under salt stress conditions because of its protective role in such an environment. Consequently, it may be important to search for well adapted genotypes for upcoming climate changes. Anthocyanins are known to have important roles in defense against biotic and abiotic stresses, providing important functions for protecting plant cells from reactive oxygen species. In this study, we investigated the anthocyanin accumulation between two Korean sorghum genotypes, Sodamchal and Nampungchal. The two genotypes were subjected to a regulated salinity condition, and the anthocyanin contents were evaluated in both. In Nampungchal, the anthocyanin content increased with 150 mM NaCl treatment during the time course of the experiment. However, the anthocyanin content of Sodamchal decreased in the same condition. The measured values of the anthocyanin content should be useful to identify the intensity of the salt tolerance in Sorghum bicolor L. Furthermore, we studied gene expression profiling of salt stress related genes with qRT-PCR. These results suggest that Nampungchal is a more tolerant genotype to salt stress compared to Sodamchal. This information should be useful for breeding salt-resistant cultivars in sorghum.
Many studies have been conducted to understand plant stress responses to salinity because irrigation-dependent salt accumulation compromises crop productivity and also to understand the mechanism through which some plants thrive under saline conditions. As mechanistic understanding has increased during the last decades, discovery-oriented approaches have begun to identify genetic determinants of salt tolerance. In addition to osmolytes, osmoprotectants, radical detoxification, ion transport systems, and changes in hormone levels and hormone-guided communications, the Salt Overly Sensitive (SOS) pathway has emerged to be a major defense mechanism. However, the mechanism by which the components of the SOS pathway are integrated to ultimately orchestrate plant-wide tolerance to salinity stress remains unclear. A higher-level control mechanism has recently emerged as a result of recognizing the involvement of GIGANTEA (GI), a protein involved in maintaining the plant circadian clock and control switch in flowering. The loss of GI function confers high tolerance to salt stress via its interaction with the components of the SOS pathway. The mechanism underlying this observation indicates the association between GI and the SOS pathway and thus, given the key influence of the circadian clock and the pathway on photoperiodic flowering, the association between GI and SOS can regulate growth and stress tolerance. In this review, we will analyze the components of the SOS pathways, with emphasis on the integration of components recognized as hallmarks of a halophytic lifestyle.
사육 염분의 급성교환 실험에서, 해수의 담수화가 1시간 이내에 이루어졌을 경우 농어, 유어들은 다소의 스트레스 반응과 이온의 변화가 있었으며. 24시간 뒤에도 정상 회복이 더딘 것으로 나타났다. 그러나 5시간에 걸친 담수화 과정 중에 스트레스 반응은 담수화 직후 높게 나타났지만, 24시간 후 빠르게 정상으로 회복되는 것을 알 수 있다. 장기간 염분별 사육실험 결과, 20 ppt가 가장 높은 사료섭식량을 나타내었고, 2 ppt구가 가장 낮았다. 사료효율 또한 2 ppt구이 가장 낮게 나타났고, 10 ppt구가 가장 높았으며, 실험구별 유어의 성장은 실험 종료시 10, 20 및 30 ppt구가 2 ppt보다 유의하게 높은 길이 성장을 보였다. 무게 성장은 실험 종료시 20 ppt가 가장 빠른 무게 성장을 나타내었고, 다음으로 30 ppt, 10 ppt. 2 ppt순으로 나타났다. 염분별 사육유어의 혈액 성상은 2 ppt~30 ppt 사육수에 있어 농어 유어들의 혈액 조성은 큰 차이가 없는 것으로 나타났다.
Zeaxanthin is an important pigment in the photo-protection mechanism of microalgae. However, zeaxanthin epoxidase, an enzyme involved in the accumulation and conversion of zeaxanthin, has not been extensively studied in microalgae. In this work, we report the expression pattern of zeaxanthin epoxidase in Dunaliella tertiolecta (DtZEP) at different light and diverse salinity conditions. To confirm the responsiveness to light conditions, the ZEP expression pattern was investigated in photoperiodic (16 h of light and 8 h of dark) and continuous (24 h of light and 0 h of dark) light conditions. mRNA expression levels in photoperiodic conditions fluctuated along with the light/dark cycle, whereas those in continuous light remained unchanged. In varying salinity conditions, the highest mRNA and protein levels were detected in cells cultured in 1.5 M NaCl, and ZEP expression levels in cells shifted from 0.6 M NaCl to 1.5 M NaCl increased gradually. These results show that mRNA expression of DtZEP responds rapidly to the light/dark cycle or increased salinity, whereas changes in protein synthesis do not occur within a short period. Taken together, we show that DtZEP gene expression responds rapidly to light irradiation and hyperosmotic stress. In addition, ZEP expression patterns in light or salinity conditions are similar to those of higher plants, even though the habitat of D. tertiolecta is different.
간척조성지 밭작물재배기술개발의 일환으로 점적관개가 간척지토양의 염농도관리와 배추 및 양배추의 생육에 미치는 영향을 구명하기 위하여 영산강간척지, 이원간척지 및 화옹간척지에 시험지를 조성하여 시험재배를 하였다. 점적관개는 표토의 염농도를 비교적 효과적으로 염스트레스 기준이하로 제어할 수 있었지만 토양의 염농도에 대한 공간변이성을 완전히 해소시키기에는 다소 한계가 있었고 이 공간변이성이 생산물의 상품화율을 낮추는 원인중의 하나로 생각되었다. 표토의 염농도가 높을수록 유거수의 염농도가 직선적으로 증가함으로 고농도의 유거수는 간척지의 염농도관리에 매우 중요한 변수로 작용할 수 있다고 판단되었다. 적기 이식재배를 한 이원간척지에서는 평균 수량이 배추가 약 6-7톤/10a, 양배추가 5톤 정도로 비교적 높은 수량을 보였고 상품화율도 배추 51.2%, 양배추 59.4%로 어느 정도 경제성이 있는 것으로 조사되었지만 적기에 비하여 이식이 늦은 화옹간척지와 영산강 간척지는 수량도 매우 낮았고 상품화율도 거의 없거나 매우 낮아 적기를 맞추지 못한 것이 저수의 가장 큰 변수였다. 배추와 양배추의 결구비대는 토양염농도가 증가할수록 로그함수적으로 감소하였고, 결구배추가 최고 구중의 50% 이하로 감소되는 토양EC는 적기에 이식한 이원간척지의 경우 배추 $6.1dS\;m^{-1}$ 양배추 $5.7dS\;m^{-1}$이었다.
집약적인 시설재배 농업에서 화학비료와 퇴비의 연용과 과다한 사용은 용탈현상이 제한되어 있는 시설재배 토양의 염류집적현상을 쉽게 유발시키고 있다. 토양중의 염류집적에 따른 수분 흡수의 어려움을 극복하기 위하여 식물은 여러 가지의 무기 및 유기용질을 체내에 축적하는데, 특히 토양 중에 그 함량이 높아 쉽게 이용할 수 있는 $NO_3$를 삼투압 조절물질로 흡수 축적하는 것으로 결론지을 수 있다. 안전농산물의 생산 측면에서 특히 시설재배 신선야채류의 $NO_3$ 축적을 줄이기 위한 노력이 요구되고 있으며, 이를 이해서는 과다한 시비의 지양과 광조건의 개선이 필요하며 이에 더하여 토양 염류집적을 줄일 수 있는 토양 관리 및 시비 방법들이 심각하게 고려되어야 할 것이다.
