화장품에서 금 콜로이드 입자에 의한 사프라닌 염료의 분해 연구 (Photodegradation of Safranin-O Dye by Au Metal Colloid in Cosmetics)

  • 본 연구에서는 금 나노입자 콜로이드를 이용하여 safranin-O의 광촉매적 분해를 관찰하였다. 금 나노입자는 용액상에서 safranin-O의 분해 속도를 빠르게 하기 위해서 사용되었다. 금 나노입자 콜로이드는 수용액상에서 $Na_2CO_3$와 PVP 고분자(poly(vinyl pyrrolidone))를 이용하는 환원방법에 의하여 제조하였다. Safranin-O의 분해현상은 자외선(UV light)와 과산화수소($H_2O_2$)의 존재 하에서 금 나노입자 콜로이드와 염화금의 농도, 반응계의 산도(pH), 반응시간과 같은 실험조건들의 조절을 통해 연구되었다. 분해반응에 사용된 금 나노입자 콜로이드의 농도가 증가함에 따라서 염료가 분해되는 속도가 증가하였다. Safranin-O의 광산화 반응은 광학적으로 측정되었고, 금 나노입자의 기본적인 물성과 촉매 특성은 UV-Vis 광학계를 이용하여 측정되었다.

Disparity between MR Imaging and Histochemical Grading in Human Intervertebral Disc Degeneration

  • Objective : In order to establish the index of degeneration, the authors performed a histochemical study with Safranin-O staining and investigated the occurrence of apoptosis in the human intervertebral disc. Methods : Eighteen intervertebral disc specimens surgically extracted from the patients and two additional specimens from the autopsied cases were stained with Safranin-O for proteoglycan according to a standard protocol. Terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate- biotin nick end labeling[TUNEL] was used to detect the fragmented DNA known to be associated with apoptotic cell death and classification scheme was formulated for categorization of the degree of Safranin-O staining [normal, moderate reduction, faint] by modification of Makin's histological-histochemical grading. The Kruskal-Wallis H test and Chi-square test were used for statistical analysis. Results : The statistical results showed a significant difference in the mean age between "normal" Safranin-O staining group and the others [19.3 versus 55, 43.4, p=0021]. However, there was no statistically significant correlation between Safranin-O staining and MR grading of disc degeneration. Only six of eighteen surgical specimens and none in autopsies showed positive apoptotic cells in TUNEL staining. Conclusion : The determination of the degree of degeneration in surgically obtained disc tissue per se by histochemical staining or by the degree of apoptosis that corresponds to its morphologic change was not feasible.

입상 활성탄에 의한 Safranin의 흡착에 관한 평형, 동력학 및 열역학에 관한 연구 (Equilibrium, Kinetics and Thermodynamics Studies about Adsorption of Safranin by Granular Activated Carbon)

  • 입상 활성탄을 사용하여 수용액으로부터 Safranin의 흡착에 대해 조사하였다. 회분식 실험은 흡착제의 양, 초기농도와 접촉시간과 온도를 흡착변수로 사용하여 수행하였다. 흡착평형자료는 Langmuir, Freundlich 및 Dubinin-Radushkevich식을 사용하여 해석한 결과 Freundlich식이 가장 좋은 일치도를 나타냈다. 평가된 Langmuir 분리계수, $R_L=0.183{\sim}0.254$와 Freundlich 분리계수, 1/n = 0.518~0.547로부터 이 흡착공정이 적절한 처리방법이 될 수 있음을 알았다. 흡착속도실험자료를 유사1차 및 유사2차 반응속도식에 적용해 본 결과는 유사2차반응속도식에 잘 맞는 것으로 나타났다. 음수값의 Gibbs 자유에너지(${\Delta}G=-3.688{\sim}-7.220kJ/mol$)와 양수값의 엔탈피(${\Delta}H=33.126kJ/mol$)는 흡착이 자발적이고 흡열공정으로 진행된다는 것을 나타냈다.

Removal of safranin from aqueous solution through liquid emulsion membrane

  • One of the real issues of the recent years is water contamination because of harmful synthetic dyes. Liquid Membranes (LM) resemble a promising alternative to the current separation processes, demonstrating various points of interest as far as effectiveness, selectivity, and operational expenses. The improvement of various Liquid Membranes designs has been a matter of examination by few researchers, particularly for the expulsion of dyes from aqueous solutions. The choice of organic surfactants plays an essential role in the efficiency of the dye removal. In LM design, the most significant step towards productivity is the decision of the surfactant type and its concentration. Liquid emulsion membrane (LEM) was used to remove safranin from aqueous solutions in which the emulsion was made with the help of D2EHPA as carrier, kerosene was used as a diluent and Span 80 (Sorbiton monooleate) was used as an emulsifying agent or surfactant. Various sorts of internal stages were utilized, to be specific sulphuric acid and sodium hydroxide. The impact of parameters influencing extraction efficiency such as pH of feed solution, concentrations of surfactant and emulsifying agent in membrane phase, volume ratio of internal phase to membrane phase, internal phase concentration, agitation speed and time of extraction were analyzed.

수종(數種) 수복물(修復物)의 색소침투(色素浸透)에 관(關)한 실험적(實驗的) 연구(硏究) (AN EXPERIMENTAL STUDY ON PENETRATION OF DYE IN FILLING MATERIALS)

  • The purpose of this study was to measure penetration of dye stuff(5% Methylene blue, Hematoxylin, Crystal violet and Safranin-O) on silicate cement, Adaptic, Hi-pol and unfilled resin. Each filling material was mixed on the mixing pad and the mixed material was inserted with condensation force of 500gr, 1000gr and 2000gr and without condensation force into preformed glass tube (10mm in diameter and 10mm in height). The specimen was stored in the air for 24 hours, then specimen was immersed in various dye solution (5% methylene blue, hematoxin, crystal violet and safranin-O) for different period of time (l hour and 24 hours). These dye-treated specimen was cut horizontally at the middle portion and the dye penetration in cut surface was measured. Following results were obtained. 1. Pentration of various dye was excessive in silicate cement with and without Condensation force. 2. There has been no evidence of dye penetration in unfilled resin. 3. Dye penetration occurced with in 1 hour period and the extending time didn't affect the dye penetration.

전방 십자 인대 파열시 손상된 대퇴골 외과 연골의 병리조직학적 연구 (Histopathological Study of Cartilage in the Bone Bruise of the Lateral Femoral Condyle Associated with Anterior Cruciate Ligament Rupture)

  • 목적: 급성 전방십자인대 파열 시 동반된 제 3형의 골멍의 조직학적 소견을 분석하였다. 대상 및 방법: 25례의 급성 전방십자인대 파열 환자에서 MRI상 대퇴골 외과에 제 3형의 골멍이 동반된 14례와 골멍이 동반되지 않은 11례를 대상으로 하였다. 남자가 22례, 여자가 3례였으며 평균 나이는 25세였다. 수상 후부터 MRI 검사까지의 기간은 최소 1일에서 최대 30일이였고 수상 후부터 전방십자인대 재건술까지의 기간은 평균 45일이었다. 전방십자인대 재건술 시 연골과 연골하 골에 trephine biopsy를 시행하여 조직학적 및 면역조직학적 소견을 비교 분석하였다. 결과: hematoxylin-eosin 염색시 세포에 대한 변화는 발견할 수 없었고, Masson's trichrome 염색에서 콜라겐의 분포차이는 보이지 않았다. 또한 면역화학염색으로 관찰된 제 1형 콜라겐과 제 2형 콜라겐의 분포에서 아무런 차이를 발견할 수 없었다(p>0.05). 그러나 Safranin-O를 이용한 글리코스아미노글리칸 염색에서 골멍군의 표층과 중간층의 글리코스아미노글리칸 염색 분포가 정상 대조군보다 떨어졌다(p<0.05). 또한 골멍군에서 석회화층에 지방세포들이 같이 발견되는 빈도가 높았다. 결론: 전방십자인대 파열시 동반되는 제 3형의 골임은 슬관절의 불안정성과 함께 추후 관절 연골의 손상을 유발하는 중요한 요인이 될 것으로 사료된다.

류마티스 관절염에서 우슬과 전침자극의 항관절염 효과 (Effects of Achyranthes Radix and Electroacupuncture on Type II Collagen-Induced Arthritis)

  • The purpose of this study was to observe the effects of Achyranthes Radix(AR) and electroacupuncture(EA) in rats with rheumatoid arthritis induced by type II collagen for 28 days. Control group was daily administered 0.9% NaCl 0.5 $m{\ell}$, Group I was daily administered 0.9% NaCl 0.5 $m{\ell}$ to arthritic rats, Group II was orally administered with Achyranthes Radix 500 mg/kg 0.5 $m{\ell}$ to arthritic rats. Group III was given 2 Hz EA of chok samni acupoint(ST36) in the test group for 30 min/days to arthritic rats. Group IV was daily orally administered with Achyranthes Radix 500 mg/kg 0.5 $m{\ell}$ and 2 Hz EA of chok samni acupoint(ST36) in the test group for 30 min/days to arthritic rats. This studies have been designed to evaluate the hind paw edema, assessment of arthritis indices, analgetic effects by analysis of blood chemistry(WBC, CRP, ALP, AST). In each group, histologic observations, Safranin O-fast green stain were observed and analyzed. The following results were obtained. Group II, III, IV were significantly decreased arthritis indices and the rate of paw edema compared with Group I . Especially group IV was the most significantly decreased. The WBC, CRP, AST, ALT was that Group II, III, IV were significantly decreased compared with Group I . In conclusion, Achyranthes Radix and Ea contribute to the improvement of blood chemistry and change in safranin O-fast green by knee joint of arthritic rats.

전기자극이 흰쥐의 피부 비만세포에 미치는 영향 (Effects of Electrical Stimulation on the Mast Cell of Skin in Rats)

  • 전기자극이 흰쥐 피부의 비만세포 수 및 탈과림에 미치는 영향을 규명할 목적으로 본 실험을 시행하였다. Sprague-Dawley계 수컷 흰쥐 12마리를 6마리씩 무작위로 전기자극군과 대조군에 배치하고 전기자극군 흰쥐의 등에 양극전극을 부착한 후 맥동빈도 100pps, 맥동기간 $140{\mu}s$, 전류량 $500{\mu}A$로 60분간 전기작극하였으며 대조근은 전극만 부착하고 전류를 통전시키지 않았다. 전자자극 후 등의 피부를 적출하여 포르말린에 고정한 후 파리핀 포매 절편을 제작하여 toluidine blue 및 alcian blue 염색을 하였다. 이 표본을 고배율 현미경하에서 관찰하여 현미경에 부착된 CCD 카메라를 통해 개인용 컴퓨터로 보낸 다음 영상분석 프로그램을 이용하여 비만세포 수, 탈과립 비만세포 수, 비만세포의 아종 수, 부위별 비반세포를 계측한 후 비만세포의 밀도, 탈과립 비만세포 비율, 비만세포 종류의 비율, 분포 비율을 산출하여 분석하였다. 실험 결과 전기자극군의 비만세포 밀도 및 탈과립 비만세포 비율이 대조군보다 통계학적으로 유의하게 높게 나타났으며, 결합조직 비만세포의 비율이 유의하게 낮았다. 또한 비만세포는 표피에는 존재하지 않고 진피에 분포하며 진피 심부에 유의하게 많이 분포하고 있었다. 이러한 결과는 흰쥐 피부의 양극 전기자극이 비만세포의 수를 증가시키고 비만세포의 탈과립을 유발시키고 있음을 시사하고 있다. 향후 비만세포와 관련이 있는 피부질환 및 손상을 대상으로 한 연구가 기대된다.

류마티스 관절염 흰쥐에서 수중운동과 우슬추출물이 염증 및 통증 반응에 미치는 영향 (Effect of Swimming Exercise and Achyranthes Radix Extracts on Inflammatory and Pain Responses in Rheumatoid Arthritis Rats)

  • This study was performed to investigate the effects in inflammatory and pain status on rheumatoid arthritis(RA) induced rats by swimming exercise and Achyranthes Radix(AR) extracts ointment according to the application methods with the change of motor-behavioral and histochemistry study through the change of safranin o-fast green stain in the knee joint and prostaglandin $E_2(PGE_2)$ concentration production in serum for 28 days. They were randomly divided into four groups; Group I: RA induction, Group II: application of only swimming exercise after RA induction, Group III: application of only AR extracts ointment after RA induction, Group IV: application of both AR extracts ointment and swimming exercise after RA induction. The following results were obtained. Volume change of hind paw edema and arthritis indices test and arthritic dorsal flexion & plantar flexion pain test, group II, III, IV were showed that significantly decrease to each scores compared with group I (p<.01). Safranin o-fast green stain were showed histological indices, group II, III, IV were showed that significantly decrease the scores of cellular infiltration and synovial hyperplasia(p<.05), pannus formation and cartilage destruction(p<.01) compared with group I. Group IV were the most decreased compared with group II, III. The $PGE_2$ concentration of the group II, III, IV were decreased compared to the group I (p<.01) In conclusion, swimming exercise and AR extracts ointment acts were the most therapeutic intervention in inflammatory and pain control of RA induced rats.

생강감초탕(生薑甘草湯)이 골관절염 유발 Rat에 미치는 영향 (Effects of Saengkanggamchotang (SKT) on MIA-Induced Osteoarthritis in Rats)

  • Objectives This study was performed to investigate the effects of Saengkanggamchotang (SKT) on the monosodium iodoacetate (MIA) induced osteoarthritis in rats. Methods Osteoarthritis was induced by injection of MIA (50 ul, 60 mg/ml) into knee joints of rats. Rats are divided into a total of 4 groups (normal, control, positive comparison group, SKT treated group, each n=6). Normal group are not treated at all without inducing osteoarthritis whereas control group were induced for osteoarthritis by MIA and oral medicated with 20 ml of distilled water per day. Positive comparison group was injected with MIA and after 7 days, that was taken indomethacin (30 mg/kg/mouse). SKT treated group was injected with MIA and after 7 days that was taken SKT (30 mg/kg/mouse). Positive comparison group and SKT treated group were oral medicated for each substance a total of 4 weeks with one time per day. After experiments (from 1 week after injection of papain to 4 weeks elapsed), the functions of liver and kidney, Prostaglandin E2, inflammatory cytokine (IL-$1{\beta}$, IL-6, TNF-${\alpha}$), osteocalcin, TIMP-1, MMP-9 within serum. Knee joint structures were observed by H&E, safranin-O staining method, and amount of cartilage were measured by ${\mu}CT$-arthrography. Results 1) Hind paw weight bearing ability was significantly improved. 2) Functions of liver and kidney were not affected. 3) Prostaglandin E2, osteocalcin, TIMP-1, MMP-9 in serum were significantly decreased. 4) Inflammatory cytokine IL-$1{\beta}$ was significantly decreased, and IL-6, TNF-${\alpha}$ were decreased but had not significant. 5) In terms of histopathology, significantly reduced subsidence of cartilage and bone in H&E staining. And in Safranin O staining, proteoglycan content in synovial membrane was significantly increased compared with control group. 6) Destruction of cartilage on ${\mu}CT$-arthrography was significantly reduced. Conclusions Based on all results mentioned above, Saengkanggamchotang (SKT) is believed to be meaningful for suppressing the progress of osteoarthritis and its treatments.
