Organic acids 의 첨가가 in vitro 반추위 발효성상과 메탄 생성에 미치는 영향 (Effects of Organic Acids on In Vitro Ruminal Fermentation Characteristics and Methane Emission)

  • 본 연구의 목적은 organic acids를 첨가하여 in vitro 상의 반추위 발효성상과 반추위 내 메탄 억제에 미치는 영향에 대한 효과를 알아보고자 하였다. 반추위액은 순천대학교 부속목장의 반추위 cannula가 시술된 Holstein에서 채취하였고, organic acids는 반추위액과 버퍼의 혼합액에 첨가하여 배양하였다. 그 결과 pH 값은 lactic acid, malic acid 및 succinic acid첨가구에서 6.69에서 6.16 정도로, 대조구와 다른 첨가구보다 낮았다. 총 가스 발생량은 배양 48시간에 aspartic acid, malic acid 및 succinic acid첨가구에서 유의적(p<0.05)으로 높았고, 메탄 발생량은 lactic acid 첨가구에서 대조구보다 낮았다. 총 VFA와 propionic acid의 농도는 배양 12시간에 모든 첨가구가 대조구에 비해 높았다. 반추위 미생물 측정 결과에서는 Fumaric acid와 malic acid의 bacteria수가 대조구에 비해 유의적으로 증가하였으며(p<0.05), protozoa수는 유의적(p<0.05)으로 감소되었다. 이상의 실험 결과를 종합해 보면, organic acids의 첨가는 반추위 내 pH를 감소시키고 가스 발생량, 반추위 미생물 성장량 및 propionic acid 모두 증가시켰으며, 특히 lactic acid는 메탄생성을 억제하였다. 앞으로 Organic acid와 다른 메탄억제 물질과 혼합하여 반추위 내 메탄생성 억제에 관한 구체적인 연구가 필요한 것으로 사료된다.

Digestion and Nitrogen Utilization by Sheep Fed Diets Supplemented with Processed Broiler Litter

  • In vivo digestion and metabolism trials were conducted with 10 wethers equipped with ruminal, abomasal, and ileal cannulae to evaluate digestion of ensiled broiler litter (EBL), deepstacked broiler litter (DBL), and composted broiler litter (CBL). Wethers were fed a low protein (6.3% CP) basal diet alone or supplemented to 10.3% CP with EBL, DBL, CBL or soybean meal (SBM). All diets were formulated to be isoenergetic (56% TDN, DM basis). Apparent digestibilities of DM, OM, and ADF were not affected (p<0.05) by diet, but digestibility of CP was improved (p<0.05) by N supplementation. Apparent digestibility of CP was lower (p<0.05) for diets supplemented with CBL and DBL than for diets supplemented with SBM and EBL. Ruminal $NH_3$ concentration was 20 to 24 mg/dl at 2 h after feeding litter-supplemented diets compared with 13 mg/dl for SBM. Abomasal N, $NH_3$ N, and nonammonia N flows were increased (p<0.05) by N supplementation, whereas microbial N flow was not influenced (p<0.05) by diet. Compared with SBM and EBL, undegraded dietary CP flow to the abomasum tended to be greater (p<0.1) when wethers were fed DBL and CBLsupplemented diets. Retention of N (g/d) also was greater (p<0.05) due to greater (p<0.05) N intake and lower (p<0.05) urinary N excretion when wethers were fed diets supplemented with litter (especially EBL) vs. SBM. Overall, characteristics of ruminal fermentation and digestion indicated that broiler litter N was utilized efficiently by wethers, but ensiling may be preferable to deepstacking or composting.

Effect of Feed Additives in Growing Lambs Fed Diets Containing Wet Brewers Grains

  • The study was conducted to evaluate and compare the effects of feed additives on ruminal fermentation, nutrient digestibility and performance of lambs fed diets containing 60% wet brewers grains (WBG). In Experiment 1, two simultaneous trials were conducted. Fifty intact ($20.2{\pm}0.8kg\;BW$) lambs were used in a feedlot trial and 10 (rumen cannulated; $32{\pm}1kg\;BW$) in a digestion trial. The pH, volatile fatty acids (VFA) and ammonia-N in lambs were also estimated. Lambs were randomly assigned to one of five diets: i) without additives (Con), ii) with 1% bicarbonate (Bic), iii) with 1% bentonite (Ben), iv) with 33 mg/kg monensin (Mon) and v) with 200 mg/kg fibrolityc enzymes (Enz). In Experiment 2, 120 RambouilletPelibuey intact male lambs ($19.5{\pm}1.5kg\;BW$) were used in a feedlot trial and randomly assigned to four diets: i) without additives (control), ii) with 1% Bic, iii) with 33 mg/kg Mon and iv) with 1% Bic and 33 mg/kg Mon. In Experiment 1, lambs fed diets containing Bic or Mon had significantly higher final weight, DMI, ADG than other lambs. However, apparent DM, OM, CP, NDF and ADF digestibilities and ruminal individual VFA content were similar (p>0.05) among treatments. Conversely, treatmentcollection period interaction was significant for ruminal pH and NH3. In Experiment 2, lambs fed diets containing a Bic and Mon combination had significantly higher final weight, DMI and ADG. It is concluded lambs fed Bic or Mon or Bic and Mon combination had better performance characteristics than lambs on Ben or Enz.

Effect of Concentrate Level on the Formation of Conjugated Linoleic Acid and Trans-octadecenoic Acid by Ruminal Bacteria when Incubated with Oilseeds In Vitro

  • An in vitro study was conducted to examine the effect of addition level of concentrate on fermentation characteristics and long-chain unsaturated fatty acids composition, especially conjugated linoleic acid (CLA) and trans-octadecenoic acid (t-FA) by mixed ruminal bacteria when incubated with linseed or rapeseed. Four levels (0.83, 1.25, 1.67 and 2.08%, w/v) of concentrate and ground oilseeds (linseed or rapeseed; 0.83%, w/v) were added to mixed solution of strained rumen fluid with artificial saliva (1:1, v/v) in the glass jar with a glass lid equipped with stirrer, and was incubated anaerobically for 24 h at $39^{\circ}C$. Addition level of concentrate slightly reflect on pH and ammonia concentration of the culture solution at the various incubation times when incubated with both linseed and rapeseed. Total VFA concentration slightly increased with incubation times and concentrate levels for incubations with oilseeds. While CLA composition had a clearly increasing trend with incubation time when incubated with linseed, percent CLA was relatively stable when incubated with rapeseed. Percent CLA, however, had a clearly decreasing trend with concentrate level throughout incubation times with significances at 3 h incubations when incubated with linseed (p<0.038) and rapeseed (p<0.0009). The differences in compositions of t-FA were relatively small among concentrate levels for both incubations with linseed and rapeseed. The ratios of t-FA to CLA were lower for linseed with increased proportion of CLA than for rapeseed.

Effect of feeding a by-product feed-based silage on nutrients intake, apparent digestibility, and nitrogen balance in sheep

  • Background: Literature is lacking on the effects of feeding by-product feed (BF)-based silage on rumen fermentation parameters, nutrient digestion and nitrogen (N) retention in sheep. Therefore, this study was conducted to determine the effect of replacing rye straw with BF-based silage as a roughage source on ruminal parameters, total-tract apparent nutrient digestibility, and N balance in sheep. Methods: The by-product feed silage was composed of spent mushroom substrate (SMS) (45 %), recycled poultry bedding (RPB) (21 %), rye straw (11 %), rice bran (10.8 %), corn taffy residue (10 %), protected fat (1.0 %), bentonite (0.6 %), and mixed microbial additive (0.6 %). Six sheep were assigned randomly to either the control (concentrate mix + rye straw) or a treatment diet (concentrate mix + BF-based silage). Results: Compared with the control diet, feeding a BF-based silage diet resulted in similar ruminal characteristics (pH, acetate, propionate, and butyrate concentrations, and acetate: propionate ratio), higher (p < 0.05) ruminal NH3-N, higher (p < 0.05) ether extract digestibility, similar crude protein digestibility, lower (p < 0.05) dry matter, fiber, and crude ash digestibilities, and higher (p < 0.05) N retention (g/d) Conclusion: The BF-based silage showed similar energy value, higher protein metabolism and utilization, and lower fiber digestion in sheep compared to the control diet containing rye straw.

Evaluation of Different Yeast Species for Improving In vitro Fermentation of Cereal Straws

  • Information on the effects of different yeast species on ruminal fermentation is limited. This experiment was conducted in a $3{\times}4$ factorial arrangement to explore and compare the effects of addition of three different live yeast species (Candida utilis 1314, Saccharomyces cerevisiae 1355, and Candida tropicalis 1254) at four doses (0, $0.25{\times}10^7$, $0.50{\times}10^7$, and $0.75{\times}10^7$ colony-forming unit [cfu]) on in vitro gas production kinetics, fiber degradation, methane production and ruminal fermentation characteristics of maize stover, and rice straw by mixed rumen microorganisms in dairy cows. The maximum gas production (Vf), dry matter disappearance (IVDMD), neutral detergent fiber disappearance (IVNDFD), and methane production in C. utilis group were less (p<0.01) than other two live yeast supplemented groups. The inclusion of S. cerevisiae reduced (p<0.01) the concentrations of ammonia nitrogen ($NH_3$-N), isobutyrate, and isovalerate compared to the other two yeast groups. C. tropicalis addition generally enhanced (p<0.05) IVDMD and IVNDFD. The $NH_3$-N concentration and $CH_4$ production were increased (p<0.05) by the addition of S. cerevisiae and C. tropicalis compared with the control. Supplementation of three yeast species decreased (p<0.05) or numerically decreased the ratio of acetate to propionate. The current results indicate that C. tropicalis is more preferred as yeast culture supplements, and its optimal dose should be $0.25{\times}10^7$ cfu/500 mg substrates in vitro.

Effects of Gelidium amansii extracts on in vitro ruminal fermentation characteristics, methanogenesis, and microbial populations

  • Objective: Gelidium amansii (Lamouroux) is a red alga belonging to the family Gelidaceae and is commonly found in the shallow coasts of many East Asian countries, including Korea, China, and Japan. G. amansii has traditionally been utilized as an edible alga, and has various biological activities. The objective of this study was to determine whether dietary supplementation of G. amansii could be useful for improving ruminal fermentation. Methods: As assessed by in vitro fermentation parameters such as pH, total gas, volatile fatty acid (VFA) production, gas profile (methane, carbon dioxide, hydrogen, and ammonia), and microbial growth rate was compared to a basal diet with timothy hay. Cannulated Holstein cows were used as rumen fluid donors and 15 mL rumen fluid: buffer (1:2) was incubated for up to 72 h with four treatments with three replicates. The treatments were: control (timothy only), basal diet with 1% G. amansii extract, basal diet with 3% G. amansii extract, and basal diet with 5% G. amansii extract. Results: Overall, the results of our study indicate that G. amansii supplementation is potentially useful for improving ruminant growth performance, via increased total gas and VFA production, but does come with some undesirable effects, such as increasing pH, ammonia concentration, and methane production. In particular, real-time polymerase chain reaction indicated that the methanogenic archaea and Fibrobacter succinogenes populations were significantly reduced, while the Ruminococcus flavefaciens populations were significantly increased at 24 h, when supplemented with G. amansii extracts as compared with controls. Conclusion: More research is required to elucidate what G. amansii supplementation can do to improve growth performance, and its effect on methane production in ruminants.

한약재 추출물 첨가가 in vitro 반추위 발효 시 메탄생성 및 발효성상에 미치는 영향 (Effect of Herbal Extracts Supplementation on Ruminal Methane Production and Fermentation Characteristics In vitro)

  • 본 연구는 반추동물에 있어서 결명자, 계피, 산초 및 감초 등의 한약재 추출물의 첨가가 메탄생성에 미치는 영향을 구명하고자 수행되었다. In vitro 반추위 발효기질로는 대두박을 사용하였으며, 한약재 추출물 첨가수준은 기질의 10%로 하여 시간대별(0, 3, 6, 9, 12, 24 hr)로 $39^{\circ}C$에서 발효시켰다. In vitro 반추위 배양액의 pH는 6.46~6.89 수준으로서 시간이 경과함에 따라 감소하는 경향이었다. 총 가스발생량은 감초, 산초, 계피, 결명자 처리구 순으로 감초 추출물 첨가구에서 가장 많았으며, 모든 한약재 추출물 첨가구가 대조구에 비해 유의적(p<0.05)으로 많았다. 메탄생성 비율은 3시간 발효 시, 감초 추출물 처리구가 타 처리구에 비해 낮았으나 시간이 경과함에 따라 비율이 오히려 높아졌으며, 한약재 추출물 첨가구가 대조구에 비해 유의적으로 높았다(p<0.05). 수소가스의 생성비율은 6시간 발효 시까지는 처리구간에 차이가 없었으나 12시간 발효 시에는 한약재 추출물 첨가구가 대조구에 비해 유의적(p<0.05)으로 높게 나타났다. 암모니아 생성량은 발효 3시간대에는 산초 추출물 첨가구와 감초추출물 첨가구가 대조구에 비해 높았으나, 발효 12시간대에는 감초 추출물 첨가구가 대조구에 비해 유의적으로 낮았다(p<0.05). 이상의 결과에서 본 시험에 사용된 한약재 추출물은 가스 생성량을 비롯한 메탄 생성량을 증가시키고, 메탄 발효를 촉진시키는 것으로 나타났다.

Characteristics of Wet Distillers Grains on In vitro Ruminal Fermentation and Its Effects on Performance and Carcass Characteristics of Finishing Hanwoo Steers

  • Two experiments were conducted to determine the nutrient composition, in vitro ruminal ammonia concentrations and pH of wet distillers grains (WDG, produced from tapioca 70% and rice 30%) and to evaluate dietary effects of fermented total mixed ration (TMR) using WDG on the performance, blood metabolites and carcass characteristics of Hanwoo steers from mid fattening to slaughter. In Exp. I, average dry matter (DM), crude protein, ether extract, crude fiber, ash, neutral detergent fiber, acid detergent fiber, and nitrogen free extract of seven WDG samples from an ethanol plant with different sampling dates were 19.9%, 24.8%, 3.8%, 21.8%, 8.87%, 60.3%, 34.5%, and 40.7% (DM basis), respectively. For in vitro ammonia concentrations and pH, each sample was assigned to 7 incubation times (0, 4, 8, 12, 24, 48, and 72 h). Linear increase was observed between 12 and 48 h for ammonia concentrations, but final ammonia concentrations (72 h) were not significantly different among WDG samples and fermentation patterns of WDG samples showed similar tendency. In vitro pH varied among treatments from 0 to 24 h, but were not different statistically after 48 h. In Exp. II, 45 Hanwoo steers of 23 months ($641{\pm}123kg$) from mid fattening period to slaughter (248 days) were randomly divided into three groups of 15 pens each (five repetitions/each treatment) and assigned to one of three dietary treatments; i) Control (TMR), ii) WDG 15 (TMR containing 15% of WDG, as fed basis) and iii) WDG 28 (TMR containing 28% of WDG, as fed basis). The body weight (BW), ADG, and feed conversion ratio (FCR) of control and WDG 15 and 28 during 248 days were 760.8, 740.1, and 765.5 kg, and 0.50, 0.50, and 0.52 kg/d, and 18.6, 17.6, and 17.1, respectively. The dry matter intake (DMI) (kg/d) of control (9.11) was higher (p<0.05) than WDG treatments (WDG 15%, 8.57; 28%, 8.70). Nevertheless, DMI did not affect BW, ADG, and FCR of Hanwoo finishing steers. Blood metabolites were in normal ranges and were not different among treatments except the albumin concentration. In carcass characteristics, WDG 15 (30%) showed higher frequency of A-carcass yield grade than WDG 28 (15%) and control (7%), and WDG 28 (61%) showed higher frequency of $1^{{+}{+}}$ and $1^+$-carcass quality grade than WDG 15 (40%) and control (60%). In conclusion, using WDG up to 28% in TMR did not show any negative effect on the performance and blood metabolites, and improved carcass quality of Hanwoo steers. Therefore, WDG can be a useful feed ingredient for Hanwoo steers in mid-fattening period to slaughter.

  • 본 실험은 alcohol 사료 처리에 따른 in vitro 발효 pattern의 변화를 검토하여 alcohol 사료 처리가 반추위내 발효 pattern에 미치는 영향을 규명코자 실시하였다. 시험구 처리는 배합사료를 첨가하는 대조구(control), 배합사료와 1, 3 및 5%의 alcohol 사료(Alcoholic feed; AF)를 첨가하는 처리구를 각각 AF-1구, AF-3구 및 AF-5구로 나누어 실시하였다. 배양 12시간 동안 ammonia 농도는 AF-1, AF-5구에서 대조구에 비해 낮았다(p<0.05). 배양액의 alcohol 농도는 alcohol 첨가 수준이 증가할수록 높았다(p<0.05). 배양 12시간에 TVFA 농도는 AF-1, AF-3 및 AF-5에서 대조구에 비해 현저히 높았다(p<0.05). 배양 8~12시간 사이에 acetate 비율은 대조구에서 빠른 속도로 감소하였으며, AF-1, AF-3 및 AF-5구에서는 일정한 수준을 유지하는 것으로 나타났다. Propionate 비율은 배양 8~12시간 사이에 AF-1, AF-3 및 AF-5에 비해 대조구에서 빠른 속도로 증가하였다(p<0.05). Butyrate 및 valerate 비율은 AF-1, AF-3 및 AF-5에서 대조구에 비해 높았다(p<0.05). Caproate 비율은 배양 8시간에 AF-1구, AF-3구 및 AF-5구에서 각각 0.05, 0.58 및 0.47M%로 나타났으나, 대조구에서는 검출되지 않았다. 본 실험의 결과에서 alcohol 사료의 처리는 in vitro 반추위액의 ammonia, alcohol 및 휘발성지방산 농도와 pH에 긍정적인 영향을 미치는 것으로 판단된다. 또한 본 실험의 결과에서는 5%의 alcohol 첨가 수준이 여타 alcohol 첨가 수준에 비해 반추위 발효에 보다 효과적인 것으로 사료된다.
