Effect of Phosphate Fretilization Levels on the Agronomic Characters of Soiling Cowpea ( Vigna sinensis Endlicher ) (인산시용량 차이가 청예동부의 형질변화에 미치는 영향)

  • This study was carried out to determine the optimum fertilizer level of phosphate for maximum yield of soiling cowpea(Vigna sinensis Endlicher) in Cheju. The results obtained are summarized as follows:1. Plant length, fresh yield, stem and leaf weight per plant, node number of main stem, primary branch number per plant, pod number per plant and pod weight per plant increased with increasing of phosphate level regardless of growth stage.2. Plant length and node number of main stem did not increase after August 25, 40 days after planting. Leaf weight and fresh yield were greatest on August 25, and then reduced. 3. Stem weight the number of leaves and pods and pod weight per plant increased until September 13, 80 days after planting. 4. Increased level of phosphate delayed leaf weight decrease after August 25. 5. Root length and root weight per plant increased with increasing level of phosphate regardless of growth stage, and they did not increase after Augest 25. 6. Nodule number and nodule weight per plant also increased as phosphate rate increased regardlessof growth stage. Nodule number and nodule weight were greatest on August 5, and then rapidly decreased.

Seasonal Changes of Nodule Activity, Carbohydrates and Nitrogen and their Inter-relationships in Alfalfa (알팔파근류의 질소고정활성과 체내탄수화물 및 질소함량의 연중변화와 이들의 상호관계)

  • Alfalfa field was established to investigate seasonal changes of nodule activity and contents of carbohydrates, and nitrogen, and also to examine their relationships in alfalfa (Medicago sativa L.). Periodical sampling of alfalfa in cutting and uncutting plots was collected to measure growth of plants, development and activity of nodule, and content of carbohydrates and nitrogen in the third year following year of establishment. Nodule activity of alfalfa root appeared to early April, increased to a maximum in beginning of June (flowering stage), and then decreased and generally remained low from late-July to mid-August, and again increased from early September, and then decreased and generally remained low for the rest of growing season. After flowering, nodule weight tended to decrease slowly throughout the growing season. Until flowering stage, increase of dry weight was closely related with that of nodule activity. But after flowering the curve pattern of dry weight did not fit to that of nodule activity due to decrease of supply of assimilate to nodule, drought, and high temperature. Total nonstructural carbohydrates in roots were closely correlated with nodule weight and nodule activity. While, nitrogen contents in leaves were closely correlated with nodule weight and nodule activity. Also cutting on July prevented unnessesary losses of respiration during summer to provide rapid recovery of nodule activity.

Studies on the Root Nodule Formation of Several Leading Soybean Varieties (주요대두품종의 근류형 성성에 관한 연구)

  • 15 soybean cultivars were tested with the sandculture to investigate the affinity of cultivars to the nodule bacteria, Rhyzobium Japonicum #23, and the effects of inoculation on the vegetative growth of cultivars. The amount of nodule formation until the flowering stage was significantly different among the cultivars. The cultivar Haman showed the best nodule formation of 122.2 nodules per plant, while the cultivar Busuk showed the least amount of nodule formation. The inoculation of the nodule bacteria resulted in an increased plant growth and especially the chlorophyll content was significantly improved by inoculation of the nodule bacteria.

Nodulation Experiment by Cross-Inoculation of Nitrogen-Fixing Bacteria Isolated from Root Nodules of Several Leguminous Plants

  • Root-nodule nitrogen-fixing bacteria are known for being specific to particular legumes. This study isolated the endophytic root-nodule bacteria from the nodules of legumes and examined them to determine whether they could be used to promote the formation of nodules in other legumes. Forty-six isolates were collected from five leguminous plants and screened for housekeeping (16S rRNA), nitrogen fixation (nifH), and nodulation (nodC) genes. Based on the 16S rRNA gene sequencing and phylogenetic analysis, the bacterial isolates WC15, WC16, WC24, and GM5 were identified as Rhizobium, Sphingomonas, Methylobacterium, and Bradyrhizobium, respectively. The four isolates were found to have the nifH gene, and the study confirmed that one isolate (GM5) had both the nifH and nodC genes. The Salkowski method was used to measure the isolated bacteria for their capacity to produce phytohormone indole acetic acid (IAA). Additional experiments were performed to examine the effect of the isolated bacteria on root morphology and nodulation. Among the four tested isolates, both WC24 and GM5 induced nodulation in Glycine max. The gene expression studies revealed that GM5 had a higher expression of the nifH gene. The existence and expression of the nitrogen-fixing genes implied that the tested strain had the ability to fix the atmospheric nitrogen. These findings demonstrated that a nitrogen-fixing bacterium, Methylobacterium (WC24), isolated from a Trifolium repens, induced the formation of root nodules in non-host leguminous plants (Glycine max). This suggested the potential application of these rhizobia as biofertilizer. Further studies are required to verify the N2-fixing efficiency of the isolates.

Screening of Rhizobium, Hairy Vetch Root Nodule Bacteria, with Promotion of Nodulation and Nitrogen Fixation (뿌리혹 형성능과 질소 고정능이 우수한 헤어리베치 유래 Rhizobium의 분리 및 선발)

  • This study was conducted to select rhizobia from hairy vetch (Vicia villosa Roth) with nodulation and excellent nitrogen-fixing ability. Hairy vetch root was collected from 7 of cultivation region of all over the country, rhizobia were isolated from the root nodules. Isolates were re-inoculated into a hairy vetch separately and studied nodulation and nitrogen-fixing ability. As a result, total of 52 Rhizobium isolates were isolated from the hairy vetch root nodules, among these, 16 isolates were Rhizobium which show good growth at more than 0.5% NaCl concentration. These 16 isolates were re-inoculated separately, 8 weeks after, good root nodule formation was observed from Rhizobium sp. RH1, RH3, RH81, RH82, RH84, and RH93 strain treated samples. Six isolates were positive for nitrogen fixing ability, the highest acetylene reduction activity was shown by Rhizobium sp. RH84. Results suggest that the Rhizobium sp. RH84 could be used as the possibility of its application as a green manure crop of hairy vetches in nonuniform salt distribution reclaimed land.

Nodulation and Early Growth of Supernodulating Mutants in Soybean (초다 근류착생 돌연변이체 콩의 뿌리혹 형성 및 초기생육)

  • Increase in nodulation and nitrogen fixation was achieved partly through the isolation of supernodulating soybean mutant plants. This experiment was conducted to compare nodulation, nitrogen fixation, and early growth characters of wild type 'Bragg' with those of its supernodulating soybean mutant, 'nts 382' and 'nts 246'. At 31 days after planting, nodule dry weight of nts mutants was 2.5 to 3.7 times greater than that of Bragg. Higher nodulation of nts mutants showed the reduced top growth, indicating that photosynthates might be translocated and used for nodule growth attached to the root system. Total acetylene reduction activity was higher in nts mutants than Bragg, whereas specific acetylene reduction activity of nts mutant was the half of that of Bragg. Mixture of nts mutants and Bragg did not affect nodulation characters each other, suggesting that factors affecting supernodulating characters exist inside rather than outside the root system.

Isolation and Characterization of Ilhizobium loti from Lotus corniculatus var. japonicus (벌노랑이로부터 Rhizobium loti 의 분리 동정 및 그 특성)

  • Five strains of the fast-growing endosymbionts were isolated from the nodules of Lotus eorniculatus var. japonicus inhabited in Taejeon. From morphological and physiOlogical characteristics and nodulation test, the isolated strains were identified as Rhizobium loti. Compared to the control plant, both Lotus cornieulatus var. japonieus and Lotus corniculatus seedlings inoculated with the isolated strains. grew normally due to effective root nodule. The reisolated endosymbiont from the induced root nodule was confirmed identical to those of the first isolates by investigating antibiotic resistance and morphological characteristics. Three strains among the isolates. R. loti TUS I. TUS5 and TUS6 produced a ca1cof1uorbinding exopolysaccharide.
