활나물(Crotalaria sessiliflora L.)로부터 모상근의 유도 및 배양 (Induction and Culture of Hairy Roots of Crotalaria sessiliflora L.)

  • The hairy roots of Crotalaria sessiliflora were induced from the tissue segments infected with Agrobacterium rhizogenes ATCC 15834. The induced hairy roots were subjected to paper electrophoresis fro the detection of opine-positive clones which were considered to have been transformed. Mannopine and agropine were presented in hairy root clones while mannopine was presented in two hairy root clones. Eight hairy root clones were selected and cultured in MS, B5 and WP media. Each of hairy root clones was showed a difference in branch pattern and growth rate. The best culture medium and culture conditions of hairy roots were in $\frac{1}{2}$MS(3% sucrose, pH 5.7) liquid medium at 25$^\circ C$, 70 rpm under dark, the growth rate in $\frac{1}{2}$MS liquid medium was increased with 210-fold more than that of inoculated hairy roots and with 2-fold more than that in MS liquid medium. Also, the adequate condition for hairy root growth was such that concentration of KH$_2PO$_4 was 1.25mM and the ratio of NH${_4}{^+}$ : NO${_3}{^-}$ was 1 to 3 in MS medium. The presence of pyrrolizidine alkaloids, monocrotaline, in the hairy roots was detected by TLC.

오옥신 처리가 에키네시아의 부정근 및 생리활성물질 생산에 미치는 영향 (Auxin Affects on Production of Adventitious Roots and Secondary Metabolites in Echinacea angustifolia)

  • The production of adventitious roots derived from root explant of Echinacea angustifolia and its secondary metabolite content were assessed in different types and levels of auxin. The induction of adventitious roots from root explant cultured in Murashige and Skoog solid medium supplemented with 1.0 mg/L indole -3-butyric acid (IBA) attained highest as 20.87 mg fresh weight and 3.07 mg dry weight per culture but root suspension culture at the same concentration of IBA enhanced biomass production as 3.07 g fresh weight and 0.38 g per culture after 4 weeks in culture. 3.0 mg/L ${\alpha}$-naphthalene acetic acid (NAA) treatment had similar effect on root biomass production as 3.07 g fresh weight and 0.38 g per culture with liquid suspension culture, whereas adventitious roots exposed to over 3.0-5.0 mg/L IBA or 5.0 mg/L NAA were less responsive by reducing the number of adventitious roots and/or changing root morphology such as short and thick. The content of secondary metabolites such as phenolic, flavonoids and total caffeic acid in adventitious roots cultured on MS medium supplemented with 1.0 mg/L IBA were attained highest as 27.20, 9.60. 10.67 mg/g dry weight, respectively. Overall, the best production of root biomass and secondary metabolites were given by 1.0 mg/L IBA.

Resistance Induction by Salicylic Acid Formulation in Cassava Plant against Fusarium solani

  • Fusarium root rot caused by the soil-borne fungus Fusarium solani is one of the most important fungal diseases of cassava in Thailand, resulting in high yield losses of more than 80%. This study aimed to investigate if the exogenous application of salicylic acid formulations (Zacha) can induce resistance in cassava against Fusarium root rot and observe the biochemical changes in induced cassava leaf tissues through synchrotron radiation based on Fourier-transform infrared (SR-FTIR) microspectroscopy. We demonstrated that the application of Zacha11 prototype formulations could induce resistance against Fusarium root rot in cassava. The in vitro experimental results showed that Zacha11 prototype formulations inhibited the growth of F. solani at approximately 34.83%. Furthermore, a significant reduction in the disease severity of Fusarium root rot disease at 60 days after challenge inoculation was observed in cassava plants treated with Zacha11 at a concentration of 500 ppm (9.0%). Population densities of F. solani were determined at 7 days after inoculation. Treatment of the Zacha11 at a concentration of 500 ppm resulted in reduced populations compared with the distilled water control and differences among treatment means at each assay date. Moreover, the SR-FTIR spectral changes of Zacha11-treated epidermal tissues of leaves had higher integral areas of lipids, lignins, and pectins (1,770-1,700/cm), amide I (1,700-1,600/cm), amide II (1,600-1,500/cm), hemicellulose, lignin (1,300-1,200/cm), and cellulose (1,155/cm). Therefore, alteration in defensive carbohydrates, lipids, and proteins contributed to generate barriers against Fusarium invasion in cassava roots, leading to lower the root rot disease severity.

  • This paper presents a comprehensive study on the stability of several control schemes for the induction motor driven by current source inverters. A stator voltage-controlled current source inverter drive system without a speed sensor is investigated in order to find appropriate control schemes, which are primarily based on direct or, alternatively, indirect frequency control scheme. The overall control systems with either voltage control loop or current and voltage control loops provided in addition to each frequency control scheme are analyzed by utilizing the root locus method and simulated by computer to illustrate the validity of this analysis.

  • The phase-locked loop technique developed in the 1930's has many advantages when applied to speed control. The speed control and analysis of a three phase induction motor using the PLL are described in this paper. In this system, the phase frequency detector (PFD) compares the actual motor speed from the pulses received from a shaft encoder and desired speed, and the difference adjusts the frequency of the inverter that feeds the motor, and excellent speed regulation in the order of 0.035(%) has been-obtained. A linear continuous model of the drive is developed and system response is analysed using conventional root locus techniques. Various compensating filters and feedback signals are considered and the need for addition of derivative feedback is shown. A sampled data model is used to study the effects of discrete PFD output. Stability limitson speed are predicted. A drive was implimented and experimental results are presented to verify theoretical predictions.

Mass Production of Sand Dune Plant, Vitex rotundifolia via Micropropagation

  • The fruits of Vitex rotundifolia in Korea, known as 'Man Hyung Ja', occupy an important position as traditional oriental medicine in Asian countries. It is known that propagation of this plant by seed is difficult and time-consuming with little success. Attempts were made to develop a method by using nodal culture techniques. Explants of stem node without leaves cultured on Nitsch medium containing 1 ml/L BA, gave the best shoot induction ratio. Also, BA with IAA or TDZ treatment showed positive effect on shoot induction. Half-strength Nitsch medium was supplemented with 0.5 mg/L NAA produced better success than did the others on root formation. It showed that many of the regenerants grew successfully on growth chamber at $24^{\circ}c$.

농어촌 동력용 전동기구를 위한 단상/3상변환기의 개발에 관한 연구 (Study on single/three phase converter for motor applications of rural district)

  • This study is initiated to solve the problem that the development of an electric machine to drive heavy horse-power load required in the rural district, where only single phase supply is available, is very urgent. As a method for this purpose, the single/three phase converter by single phase induction machine with a tapped auxiliary winding, running unloaded mechanically under single-phase source and supplying three-phase output to a loaded 3-phase induction motor, is devised and the pilot machine is put into test. Analysis based on hybrid equivalent circuit for the phase converter and symmertical component theory for the 3-phase load motor and practical experiment result in that optimum auxiliary winding ratio is to be 1.25 rather than theoretical .root.3/2 in order to keep the voltage unbalance ratio of 3-phase output from the converter as low as possible in both cases of starting and running the load motor.

Sequential Induction of Ethylene, Lipoxygenase, and Ascorbate Peroxidase in Senescing Soybean Callus

  • Bursts of ethylene production occurred in twice at an early exponential (EEP) and prestationary (PSP) phases, respectively, during growth of callus tissue isolated from the root of soybean seedlings. The second burst of ethylene production at PSP was smaller in magnitude than the earlier one at EEP, but was followed by increases in both guaiacol peroxidase (GuPOX) and ascorbate peroxidase (AsPOX). The increase in AsPOX activity was also preceded by an increase in lipoxygenase (LOX) activity. Treatment of the tissue with the ethylene antagonist 2,5-norbonadiene (NBD) resulted in substantial reduction in LOX and AsPOX activities during this period. GuPOX activity was reduced only slightly, if any, by NBD. Role of ethylene in the sequential induction of LOX and AsPOX in senescing callus tissue is discussed.

  • Lycii Radicis Cortex, the root bark of Lycium chinense Miller (Solanaceae) is used in oriental medicine as a tonic and is reported to exhibit hypotensive, hypoglycemic, and antipyretic activity. Recently, we have isolated dihydro-N-caffeoyltyramine, a phenolic amide, from the Lycii Radicis Cortex. Treatment with dihydro-N-caffeoyltyramine significantly inhibited the proliferation of human leukemia cell lines HL-60 in a dose-dependent manner. We found also that the growth inhibition of HL-60 by dihydro-N-caffeoyltyramine is associated with induction of apoptosis of cells. (omitted)

Transcriptional Activation of CuIZn Superoxide Dismutase And Catalase Genes by Panaxadiol Ginsenosides Extracted From Panax ginseng

  • Superoxide dismutase (SOD) and catalase constitute the first coordinated unit of defense against reactive oxygen species. Here, we examined the effect of ginseng saponins on the induction of SOD and catalase gene expression. To explore this possibility, the upstream regulatory promoter region of Cu/Zn superoxide dismutase (SODI) and catalase genes were linked to the chloramphenicol acetyl-transferase (CATI structural gene and introduced into human hepatoma HepG2 cells. Total saponin and panaxatriol did not activate the transcription of SODI and catalase genes but panaxadiol increased the transcription of these genes about 2-3 fold. Among the Panaxadiol ginsenosides, the Rb2 subtraction appeared to is a major induce of SODI and catalase genes. Using the deletion analyses and mobility shift assays, we showed that the 5051 gene was greatly activated by ginsenoside Rba through transcription factor AP2 binding sites and its induction. We also examined the effect of the content ratio of panaxadiol extracted from various compartment of ginseng on the transcription of 5031 gene. Saponin extract that contains 2.6-fold more PD than PT from the fine root Increased the SODI induction about 3-fold. These results suggest that the panaxadiol fraction and its ginsenosides could induce the antioxidant enzymes, which are important for maintaining cell viability by lowering level of oxygen radical generated from intracellular metabolism.
