뉴스페이스(new space) 시대가 도래함에 따라 국내 KOMPSAT-3·3A 위성영상과 해외 위성영상과의 글로벌 융합활용 기술확보가 대두되고 있다. 일반적으로 다중센서 위성영상은 취득 당시의 다양한 외부요소로 인해 영상 간 상대적인 기하오차(relative geometric error)가 발생하며, 이로 인해 위성영상 산출물의 품질이 저하된다. 따라서 본 연구에서는 KOMPSAT-3·3A 위성영상과 해외 위성영상 간 존재하는 상대기하오차를 최소화하기 위한 정밀영상정합(fine-image registration) 방법론을 제안한다. KOMPSAT-3·3A 위성영상과 해외 위성영상 간 중첩영역을 선정한 후 두 영상 간 공간해상도를 통일한다. 이어서, 특징 및 영역 기반 정합기법을 결합한 형태의 하이브리드(hybrid) 정합기법을 이용하여 정합점(tie-point)을 추출한다. 그리고 피라미드(pyramid) 영상 기반의 반복적 정합을 수행하여 정밀영상정합을 수행한다. KOMPSAT-3·3A 위성영상과 Sentinel-2A 및 PlanetScope 영상을 이용하여 제안기법의 정확도 및 성능을 평가하였다. 그 결과, Sentienl-2A 영상 기준 평균 Root Mean Square Error (RMSE) 1.2 pixels, PlanetScope 영상 기준 평균 RMSE 3.59 pixels의 정확도가 도출되었다. 이를 통해 제안기법을 이용하여 효과적으로 정밀영상정합을 수행할 수 있을 것으로 사료된다.
Glycolysis is responsible for the conversion of glucose into pyruvate and for supplying reducing power and several metabolites. Fructose-1,6-bisphosphate aldolase (AtFBA1), a central enzyme in the glycolysis pathway, was isolated by functional complementation of the salt-sensitive phenotype of a calcineurin (CaN)-deficient yeast mutant. Under high salinity conditions, aldolase activity and the concentration of NADH were compromised. However, expression of AtFBA1 maintained aldolase activity and the NADH level in yeast cells. AtFBA1 shares a high degree of sequence identity with known class I type aldolases, and its expression was negatively regulated by stress conditions including NaCl. The fusion protein GFP-AtFBA1 was localized in the cytosol of Arabidopsis protoplasts. The seed germination and root elongation of AtFBA1 knock-out plants exhibited sensitivity to ABA and salt stress. These results indicate that AtFBA1 expression and aldolase activity is important for stress tolerance in yeast and plants.
To analyze temporal and spatial changes in vegetation, it is necessary to determine the associated continuous distribution and conduct growth observations using time series data. For this purpose, the normalized difference vegetation index, which is calculated from optical images, is employed. However, acquiring images under cloud cover and rainfall conditions is challenging; therefore, time series data may often be unavailable. To address this issue, La et al. (2015) developed a multilinear simulation method to generate missing images on the target date using the obtained images. This method was applied to a small simulation area, and it employed a simple analysis of variables with lower constraints on the simulation conditions (where the environmental characteristics at the moment of image capture are considered as the variables). In contrast, the present study employs variables that reflect the growth characteristics of vegetation in a greater simulation area, and the results are compared with those of the existing simulation method. By applying the accumulated temperature, the average coefficient of determination (R2) and RMSE (Root Mean-Squared Error) increased and decreased by 0.0850 and 0.0249, respectively. Moreover, when data were unavailable for the same season, R2 and RMSE increased and decreased by 0.2421 and 0.1289, respectively.
For geometric correction of high-resolution images, the authors matched corresponding objects between a large-scale digital map and a QuickBird image to obtain the coefficients of the first order polynomial. Proximity corrections were performed, using the Boolean operation, to perform automated matching accurately. The modified iterative closest point (ICP) algorithm was used between the point data of the surface linear objects and the point data of the edge objects of the image to determine accurate transformation coefficients. As a result of the automated geometric correction for the study site, an accuracy of 1.207 root mean square error (RMSE) per pixel was obtained.
A partial cDNA encoding a Korean radish isoperoxidase was obtained from a cDNA library prepared from 9 day old radish root. In order to obtain Korean radish isoperoxidase cDNA, 5' RACE (rapid amplification cDNA end) PCR was performed and a cDNA (prxK1) encoding a complete structural protein was obtained by RT (reverse transcription)-PCR. Sequence analysis revealed that the length of the cDNA was 945 base pairs, and that of the mRNA transcript was ca. 1.6 kb. The deduced amino acid of the protein were composed of 315 amino acid residues and the protein was 92% homologous to turnip peroxidase, and 46% to 50% homologous to other known peroxidases. The 945 bp cDNA encoding Korean radish isoperoxidase was overexpressed in Escherichia coli up to approximately 9% of total cellular protein. The recombinant fusion protein exhibited 43 kDa on SDS-PAGE analysis and the activity level of the recombinant nonglycosylated protein was two fold higher in IPTG induced cell extracts than that of uninduced ones.

Narrow gap welding has many advantages over conventional V-grooved butt welding such as high productivity, small deformation and improved mechanical property of joints. With narrower groove gap, less arc heat input is expected will all the other advantages of narrow gap welding. The main defects of narrow gap welding include the lack of root fusion, convex bead surface and irregular surface, all of which have negative effects on the next welding pass. This paper suggests an up-and-down torch oscillation for ultra narrow gap welding with gap size of 5mm and investigates the proper welding conditions to fulfill the reliable and high welding quality. First, GMA welding model was suggested for ultra narrow gap welding system with Halmoy's model referenced for wire melting modeling. And the arc length in ultra narrow gap was defined. Secondly, based on the experimental results of up-and-down torch oscillation welding, phase shift of current and wire extension length were simulated for varying oscillation frequency to show that weld the bead shape in ultra narrow gap welding can be predicted. As the result, it was confirmed that reliable weld quality in ultra narrow gap welding can be achieved with up-and-down torch oscillation above 15Hz due to its ability to provide uniform heat input along the sidewall of gap.
In this study, API-X100 steel pipes were welded with various kinds of welding wires in the laser-arc hybrid welding process. 10kW fiber laser source was combined to MIG arc welding process. API X-100 steel of base metal was of 16.9mm thickness, and butt welding applied. After welding, full penetration weld was acquired by 1-pass welding. A root porosity and the lack of fusion was observed in some welding conditions. By the mixing the melted wire, acicular ferrite, polygonal ferrite, pro-eutectoid, aligned side plate, and bainite structures were observed at the weld metal. From the observation of hybrid weld, unmixed zone had more Ni and Cr. The unmixed zone was a 1/3 area of the weld metal. As the mechanical test of the hybrid welding, tensile test and impact test applied. From the tensile test, all of the welding except SM70S was fractured at the base metal. The result of the impact test at -30 degree C led 60J~320J of the absorbed energy. The result of the low-absorbed energy might be from the coarse equiaxed structures of the weld metal.
Allium hookeri L. (Alliaceae family) is an important ethnomedicinal plant native to the Himalayan region of Asia. The aim of this research was to establish a high-frequency plant regeneration system for in vitro propagation of A. hookeri. Among the tissue types examined, receptacle explants derived from immature flower buds showed the highest regeneration rate of shoots ($93.33{\pm}4.63%$), roots ($76.67{\pm}7.85%$), and calli ($80.00{\pm}7.43%$) when cultured on Gamborg B5 (B5) medium containing $10{\mu}M$ 6-benzylaminopurine (BA) + $1{\mu}M$ naphthalene acetic acid (NAA), $0.5{\mu}M$ BA + $5{\mu}M$ NAA, and $1{\mu}M$ BA + $10{\mu}M$ NAA, respectively. Shoot multiplication was superior when cultured in liquid rather than on solid medium and relatively high concentrations of BA, ranging from 5 to $10{\mu}M$. Efficient bulblet formation following root induction from shoot clumps was achieved with culture in liquid B5 medium containing 7% (w/v) sucrose. Regenerated bulblets were successfully acclimatized to ex vitro conditions with a greater than 95% survival rate. By this method, a maximum of 62 plantlets per receptacle could be propagated within 9 weeks of initial culture. The in vitro propagation system established in this study will promote A. hookeri biotechnology, including large-scale production of healthy and aseptic clones, preserving parental genotypes with desirable traits, and genetic manipulation to enhance medicinal value.
To determine the patterns and the levels of expression of the cauliflower mosaic virus (CaMV 35S) promoter and the rice actin 1 (Act1) promoter in rice, transgenic rice plants containing CaMV 35S-$\beta$-glucuronidase (GUS) and Act1-GUS constructs were generated and examined by fluorometric and histochemical analyses. The fluorometric analysis of stably transformed calluses showed that the activity of the rice Act1 promoter was stronger than that of the CaMV 35S promoter in rice cells. In a histochemcial study of the transgenic rices, it was shown that the GUS activity directed by the CaMV 35S promoter was localized mainly in parenchymal cells of vascular tissues of leaves and roots and mesophyll cells of leaves. These results are similar to those of potato, a dicot plant. In contrast, rice plant transformed with Act1-GUS fusion construct revealed strong GUS activity in parenchymal cells of vascular tissue, mesophyll cells, epidermal cells, bulliform cells, guard subsidiary cells of leaves and most cells of the root, suggesting that the rice Act1 promoter is more constitutive than the CaMV 35S promoter. It was also confirmed that in both types of transgenic rice little or no staining was localized in metaxylen tracheary elements of vascular tissue from leaves or roots. These results indicate that the rice Act1 promoter can be utilized more successfully for expression of a variety of foreign gene in rice than the CaMV 35S promoter.
