To study the effect of installation and the shapes of filling pieces used for windows on the infiltration rate, infiltration tests were performed with two types of sliding windows and two shapes for the filling pieces. Infiltration occurred most at the top and bottom parts of the center location where two windows overlap, indicating the necessity of a filling piece to be installed at these locations. Infiltration velocity is proportional to the square root of the pressure difference, complying well with the theoretical correlation. Infiltration is reduced about 50% by the filling piece installation. This study shows that filling pieces are essential and their shapes play significant roles in reducing the infiltration of windows.

The solubility and miscibility measurement apparatus was developed and used to obtain data for refrigerant/oil mixture. The solubility and miscibility data for R-410A/68 ISO VG polyol ester (POE) oil mixture are obtained over the temperature range from -20 to $60^{circ}C\;with\;10^{\circ}C$ intervals and the oil concentration range from 0 to 90 wt%. Using the experimental data, an empirical model was developed to predict the solubility relations for R-410A/POE oil mixture at equilibrium. In the R-410A/Solest 68 oil mixture, the average root-mean-square deviation between measured data and calculated results from the empirical model is 3.4% and in the R-4104/EMKARATE RL 68H oil mixture, that is 2.86%. For R-410A/68 ISO VG POE oil mixture immiscibility was usually observed at the low oil concentrations(5~30 wt%) over the all experimental temperature range($-20~60^{circ}C$) and at the high oil concentrations(50~90 wt%) over the low experimental temperature range($-2O~0^{circ}C$).

Evaporative heat transfer characteristics of carbon dioxide have been investi- gated by experiment. The experiments have been carried out for a seamless stainless steel tube of the outer diameter of 9.55 mm, the inner diameter of 7.75 mm and the length of 5.0 m. Direct heating method was used for supplying heat to the refrigerant where the test tube was uniformly heated by electric current which was applied to the tube wall. Experiments were conducted with$CO_2$of purity 99.99% at saturation temperatures of 0.0 to 10.5$^{\circ}C$, heat fluxes of 12 to 27kW/$m^2$s and mass fluxes of 212 to 530 kg/$m^2$s. The heat transfer coefficients of $CO_2$are decreased as the vapor quality increases and these phenomena are explained by dimensionless Weber and Bond numbers. The heat transfer coefficients of$CO_2$increase when the heat and mass fluxes increase, and the saturation temperature effects are minor in the test range of this study. The present experimental data are compared with six renowned correlations with root-mean-squared deviations ranging from 23.0 to 94.9% respectively.

The vapor-liquid equilibrium and miscibility measurement apparatus was developed and used to obtain data for refrigerant/oil mixture. The vapor-liquid equilibrium and miscibility data for R-410a/POE32 and R-410A/POE46 oil mixtures are obtained over the temperature range from -20 to $60^{\circ}C\;with\;10^{\circ}C$ intervals and the oil concentration range from 0 to 90 wt%. Using the experimental data, an empirical model is developed to predict the temperature-pressure-concentration relations for R-410A/POE oil mixtures at equilibrium. In the R-410A/POE32 oil mixture, the average root-mean-square deviation between measured data and calculated results from the empirical model is 2.00% and in the R-410a/POE46 oil mixture, that is 3.69%. Flory-Huggins theory is also used to predict refrigerant/oil mixture behavior. Miscibility for R-410A/POE32 oil mixture was observed all over the experimental conditions. Immiscibility for R-410A/POE46 oil mixture was observed at the low oil concentrations(10~30 wt%).

Effects of low temperature ($8^{\circ}C$) on the hydraulic conductivity of young roots of a chilling-sensitive (cucumber; Cucumis sativus L.) and a chilling-resistant (figleaf gourd; Cucurbita ficifolia Bouche) crop have been measured at the levels of whole root systems (root hydraulic conductivity, $Lp_r$) and of individual cortical cells (cell hydraulic conductivity, Lp). In figleaf gourd, there was a reduction only in hydrostatic $Lp_r$ but not in osmotic $Lp_r$ suggesting that the activity of water channels was not much affected by low root temperature (LRT)treatment in this species. Changes in cell Lp in response to chilling and recovery were similar asroot level, although they were more intense at the root level. Roots of figleaf gourd recovered better from LRT treatment than those of cucumber. In figleaf gourd, recovery (both at the root and cell level) often resulted in Lp and $Lp_r$ values which were even bigger than the original, i.e. there was an overshoot in hydraulic conductivity. These effects were larger forosmotic (representing the cell-to-cell passage of water) than for hydrostatic $Lp_r$. After a short term (1 d) exposure to $8\;^{\circ}C$ followed by 1 d at $20\;^{\circ}C$, hydrostatic $Lp_r$ of cucumber nearly recovered and that of figleaf gourd still remained higher due to the overshoot. On the contrary, osmotic $Lp_r$ and cell Lp in both species remained high by a factor of 3 as compared to the control, possibly due to an increased activity of water channels. After pre-conditioning of roots at LRT, increased hydraulic conductivitywas completely inhibited by $HgCl_2$ at both the root and cell levels. Different from figleaf gourd, recovery from chilling was not complete in cucumber after longer exposure to LRT. It is concluded that at LRT, both changes in the activity of aquaporins and alterations of root anatomy determine the water uptake in both species. To better understand the aquaporin function in plants under various stress conditions, we examined the transgenic Arabidopsisand tobacco plants that constitutively overexpress ArabidopsisPIP1;4 or PIP2;5 under various abiotic stress conditions. No significant differences in growth rates were found between the transgenic and wild-type plants under favorable growth conditions. By contrast, overexpression of PIP1;4 or PIP2;5 had a negative effect on seed germination and seedling growth under drought stress, whereas it had a positive effect under cold stress and no effect under salt stress. Measurement of water transport by cell pressure probe revealed that these observed phenotypes under different stress conditions were closely correlated with the ability of water transport by each aquaporin in the transgenic plants. Together, our results demonstrate that PIP-type aquaporins play roles in seed germination, seedling growth, and stress response of Arabidopsis and tobacco plants under various stress conditions, and emphasize the importance of a single aquaporin-mediated water transport in these cellular processes.
Thin layer drying tests of short gain rough rice were conducted in an experimental dryer equiped with air conditioning unit. The drying tests were performed in triplicate at three air temperatures of $35^circ$, $45^circ$, $55^circ$, and three relative humidities of 40%, 55%, 70%, respectively. Previously published thin layer equations were reviewed and four different models widely used as thin layer drying equations for cereal grains were selected. The selected four models were Pages, simplified diffusion, Lewis's and Thompson's models. Experimental data were fitted to these equations using stepwise multiple regression analysis. The experimental constants involved in tow equations were represented as a function of temperature and relative humidity of drying air. The results of comparing coefficients of determination and root mean square errors of miosture ratio for low equations showed that Page's and Thompsons models were found to fit adequately to all drying test data with coefficient of determination of 0.99 or better and root mean square error of moisture ratio of 0.025.
Melting curve analysis of fluorescently labeled DNA fragments is used extensively for genotyping single nucleotide polymorphism(SNP). Here, we evaluated a SNP genotyping method by melting curve analysis with the two probe chemistries in a 384-well plate format on a Roche LightCycler 480. The HybProbe chemistry is based on the fluorescence resonance energy transfer(FRET) and the SimpleProbe chemistry uses a terminal self-quenching fluorophore. We evaluated FRET HybProbes and SimpleProbes for two SNP sites closely linked to two quantitative trait loci(QTL) for southern root-knot nematode resistance. These probes were used to genotype the two parents and 94 $F_2$ plants from the cross of PI 96354$\times$Bossier. The SNP genotypes of all samples determined by the LightCycler software agreed with previously determined SSR genotypes and the SNP genotypes determined on a Luminex 100 flow cytometry instrument. Multiplexed HybProbes for the two SNPs showed a 98.4% success rate and 100% concordance between repeats two of the same 96 DNA samples. Also, we developed a HybProbe assay for the Rcs3 gene conditioning broad resistance to the frogeye leaf spot(FLS) disease. The LightCycler 480 provides rapid PCR on 384-well plate and allows simultaneous amplification and analysis in approximately 2 hours without any additional steps after amplification. This allowed for a reduction of the potential contamination of PCR products, simplicity, and enablement of a streamlined workflow. The melting curve analysis on the LightCycler 480 provided high-throughput and rapid SNP genotyping and appears highly effective for marker-assisted selection in soybean.
Desorption equilibrium moisture contents of oak mushroom were measured by the static method using salt solutions at flour temperature levels of 35$\^{C}$, 45$\^{C}$, 55$\^{C}$ and 6$\^{C}$ and five relative humidity levels in the range from 11.0% to 90.8%. EMC data were fitted to the modified Henderson, Chung-Pfost, modified Halsey and modified Oswin models using nonlinear regression analysis. Drying tests far oak mushroom were conducted in an experimental dryer equipped with air conditioning unit. The drying test were performed in triplicate at flour air temperatures of 35$\^{C}$, 45$\^{C}$, 55$\^{C}$ and 65$\^{C}$ and three relative humidities of 30%, 50% and 70% respectively. Measured moisture ratio data were fitted to the selected four drying models(Lewis, Page, simplified diffusion and Thompson models) using stepwise multiple regression analysis. The results of comparing root mean square errors for EMC models showed that modified Halsey was the best model, and modified Oswin models could be available far oak mushroom. The results of comparing coefficients of determination and root mean square errors of moisture ratio for four drying models showed that Page model were found to fit adequately to all drying test data with a coefficient of determination of 0.9990 and root mean square error of moisture ratio of 0.00739.
This research was conducted to investigate the influence of various organic substrates on growth and yield of ginseng seedling grown organically in the closed plastic house. The pH and EC of substrates used for organically ginseng cultivation ranged 5.93~6.78 and 0.03~0.15 dS/m respectively. The concentrations $NH_4$-N and $NO_3$-N respectively was 14.01~68.63 mg/L, 5.60~58.83 mg/L. The average quantum of the closed plastic house was range from 10 to 16% of natural light. In July and August, the maximum temperature of the closed plastic house did not exceed 30 and the average temperature was maintained within 25 lower than the field because air conditioning ran. The PPV-1 and PPV-2 bed soil substrates produced higher stem length, stem diameter, shoot fresh weight and leaf area than those of conventional culture. In PPV-2 bed soil substrates, root fresh weight and root diameter was the highest. The root fresh weight of PPV-2 bed soil substrates in closed plastic house was maximum 25% heavier than the conventional cultivation. The results of this experiment will be utilized for making new substrate application for organic ginseng culture in the plastic house.
Dentin hypersensitivity must be one of the most frequent postoperative complaints in periodontal patients. Obliterating the open dentinal tubules or decreasing the diameter of their orifices would, therefore, be an objective of treatment for hypersensitive teeth. The purpose of this study was to evaluate the effect of a pulsed Nd:YAG laser irradiation on obliteration of dentinal tubules and to determine any difference according to irradiation methods. The 45 posterior teeth that had been extracted due to periodontal disease were initially treated with tetracycline HCI(100 mg/ml, 4 min.) to remove the smear layer after root planing. The root surfaces were then irradiated by a pulsed Nd:YAG laser(EL.EN.EN060, Italy) by different laser beam spot size and different exposure condition: ${\cdot}$ group 1: irradiated group by small spot(beam diameter=1mm, lW, 2 sec) ${\cdot}$ group 2: irradiated group by large spot(beam diameter=10mm, 1W, 200 sec) ${\cdot}$ group 3: irradiated group by gradual increase of watt (from 0.3W to 1.0W), beam diameter=4mm ${\cdot}$ group 4: irradiated group by fixed watt(1.0 W), beam diameter=4mm ${\cdot}$ control group: no irradiation but root planing and tetracycline HCI conditioning only. Additionally, the specimens were retreated with tetracycline HCI(100mg/ml, 4min.) to evaluate the stability of obliteration effect by Nd:YAG laser. Specimens were examined under the scanning electron microscope(JEOL, JSM-840A, Japan). Photomicrographs were taken at ${\times}4,000$ magnification and were analyzed statistically. The results were as follows: l. Scanning electron micrographs of root surface treated by tetracycline HCI alone(control group) showed widened, funnel-shaped dentinal tubules, while those of the root surface irradiated by various methods showed partially or completely obliterated dentinal tubules and various surface alterations, eg, flat, multiple pitted, melted and resolidified surface at the same energy density. 2. There was no significant difference in the obliteration effect of dentinal tubules between group 1 and group 2, and between group 3 and group 4(p>0.05). 3. The obliteration effect of dentinal tubules by a Nd:YAG laser irradiation was relatively stable to tetracycline HCI. The results demonstrate that a pulsed Nd:YAG laser irradiation within 1.0W, regardless of irradiation methods, can obliterate dentinal tubules effectively.
