동작연기의 표현력 향상을 위한 한국춤의 호흡운용법 (Breathing of Korean Dance for Develop Methodology of Expression)

  • 공연예술에 있어서 무대 위에서 움직이는 연기자의 역할은 중요한 의미를 지니고 있다. 공연예술이 목표하는 바가 단순히 표피적 또는 상투적 즐거움의 생산과 표현에 있는 것이 아니라 인간의 근원적 삶에 대한 철학적 정신의 정화를 추구하며, 이러한 역할 표현의 중심이 바로 연기자의 몫으로 설정되기 때문이다. 공연예술은 단순히 기술적인 작업이 아니며, 한 나라의 정체성과 전통을 아우르는 문화적 창조 작업이어야 한다. 그러나 오늘날 우리의 공연예술의 표현방법은 다분히 서구적 공연예술 양식의 방법론을 따르고 있다. 이러한 서구 지향의 공연예술 방법론을 극복하고, 보다 주체적이고 한국적인 공연예술의 창작과 동작연기의 표현력 향상을 위한 새로운 방법론이 제시되어져야 할 것이다. 본 논문은 전통 한국춤에서 운용되어지고 있는 호흡법과 장단을 현대 공연예술의 시간성과 공간성을 연관시켰을 때 나타나는 동작연기의 시각적 효과와 표현력 향상에 관한 연구이다. 이를 위해 한국춤의 근간을 이루는 호흡운용에 관한 기본적 이해를 바탕으로 연기자의 동작연기의 표현력 향상을 위한 호흡운용 프로그램 모델을 제시하고 있다.

  • 본 연구는 웹이라는 환경에서의 감성에 대해 연구하고자 하였다. 즉, 이는 웹에서의 사용자 감성에 대한 객관적인 기준을 제시하고자 한 것이다. 본 연구는 크게 두 가지의 연구 주제를 가지며, 그 내용은 다음과 같다. 첫째, 웹에서 웹사이트가 가지고 있는 감성요인을 분석, 이를 바탕으로 한 감성구조모형을 개발하고자 하였다. 둘째, 위 연구를 바탕으로 웹 디자인에서의 감성요인과 조형요소간의 상관관계를 알아보고자 하였다. 감성요소와 조형요소간의 상관관계를 밝힘으로서, 웹 디자인에서의 성공적인 감성디자인을 구현하고자 하였다. 이는 감성을 위한 다양한 접근방식 중 하나를 제시하고자 한 것이며, 결국 이러한 연구는 감성적 디자인의 객관적 기준을 마련하는 과장이라 할 수 있다. 디자이너가 감성적 디자인을 함에 있어서 제작과 평가에 보다 이성적인 가이드라인을 제시, 성공적인 감성디자인 구현에 도움이 될 것으로 기대된다.

Expression of Human Cytomegalovirus Immediate Early US3 Gene in Human Fibroblast Cells

  • US3 gene is a member of the human cytomegalovirus (HCMV) immediate early gene. Although the precise functions of the US3 gene in HCMV replication and pathogenesis are not known, it has been reported to play a role in inhibiting major histocompatibility class I antigen presentation. For further knowledge of US3 gene expression, rabbit polyclonal antiserum of the US3 gene product was used for indirect immunofluorescence assay. In permissive human foreskin fibroblast (HFF) cells, US3 gene expression was detectable as crescent or half-moon shape in the perinuclear region at immediate early times after virus infection. HFF cells infected with mutant HCMV lacking US3 open reading frames were negative for US3 immunofluorescence assay. Double immunofluorescence assay using monoclonal antibody to gamma adaptin (specific for the Golgi complex) and rabbit anti-US3 antiserum revealed that US3 gene product could be localized to the Golgi complex. At later time after HCMV infection, US3 gene products were detected as globular aggregates in the cytosol. These aggregates were positive for gamma adaptin and stained with preimmune serum, suggesting a nonspecific reaction to the Golgi complex. Northern blot analysis revealed that transcription of US3 was observed only during immediate early times after virus infection (until 6 h postinfection). Therefore US3 gene expression appears to be confined to immediate early time and its gene products are localized to the Golgi complex as crescent shaped forms in the perinuclear cytoplasm.

Surfer's Myelopathy : Case Series and Literature Review

  • Three male patients diagnosed with surfer's myelopathy (19-30 years) were admitted to our hospital. All three patients were novice surfers showing a typical clinical course of rapid progression of paraplegia following the onset of back pain. Typical history and magnetic resonance imaging features indicated the diagnosis of surfer's myelopathy. Two patients received high-dose steroid therapy and the other was treated with induced hypertension. One patient treated with induced hypertension showed almost full recovery; however, two patients who received high-dose steroid therapy remained completely paraplegic and required catheterization for bladder and bowel dysfunction despite months of rehabilitation. Our case series demonstrates the potentially devastating neurological outcome of surfer's myelopathy; however, early recovery in the initial 24-72 hours of presentation can occur in some patients, which is in accordance with previous reports. Ischemic insult to the spinal cord is thought to play a crucial role in the pathophysiology of surfer's myelopathy. Treatment recommendations include hydration, induced hypertension, early spinal angiography with intra-arterial intervention, intravenous tissue plasminogen activator, and high-dose steroid therapy; however, there is no standardized treatment option available. Early recovery appears to be important for long-term neurological outcome. Induced hypertension for initial treatment can be helpful for improving spinal cord perfusion; therefore, it is important for early and long-term neurological recovery. Education and awareness are essential for preventing surfer's myelopathy and avoiding further deterioration of neurological function.

Clinicohematological parameters and outcomes in a cohort of chronic lymphocytic leukemia patients with Deletion 17p from Pakistan

  • Background Chronic lymphocytic leukemia (CLL) exhibits profound heterogeneity in its clinical course. Its clinicohematological and cytogenetic features play a significant role in determining the clinical course and in predicting the treatment response and prognosis. In this context, 17p deletion is known to predict a poor prognosis, as these cases are refractory to conventional therapy. This study aimed to evaluate the clinicohematological characteristics, outcomes, and prognostic factors among CLL patients with and without del 17p in Pakistan. Methods This prospective observational study was conducted at the Department of Haematology, Armed Forces Institute of Pathology (Rawalpindi, Pakistan) between January 2013 and December 2017. Patients were diagnosed based on the International Workshop on Chronic Lymphocytic Leukaemia IWCLL criteria, their clinicohematological parameters were recorded, and cytogenetic analyses were performed. The time from diagnosis to treatment and the 2-year overall survival rate were also evaluated. Results We evaluated 130 CLL cases, including 24 patients (18.5%) with del 17p, who included 18 men (75%) and 6 women (25%). The median age was 68 years. Binet stage C was detected at the presentation in 16 patients (67%). Treatment was administered to 14 patients (70%) at a median interval of 11 months (range, 0-28 mo) after diagnosis. The overall response rate was 64.3%, the median event-free survival was 9 months (range, 1-23 mo), and the 2-year overall survival rate was 65%. Conclusion Del 17p is relatively common in Pakistan, and patients harboring this deletion had poor treatment response and survival outcomes.

다중신속순차제시아래 자극의 명암대비 및 색상이 표적 탐지에 미치는 영향 (The Influence of Stimulus Contrast and Color on Target Detection under Multiple Rapid Serial Visual Presentation)

  • 본 연구는 일련의 문자열들 중 하나의 표적을 탐지하는 데 자극 명암대비와 색상이 미치는 영향을 관찰하였다. 실험 1의 각 시행에서는 네 문자로 구성된 RSVP가 제시되고 각 RSVP 화면상의 문자들이 서로 다른 위치를 점유하였다. RSVP 배열 내에서는 한 위치에 회색 표적 문자가, 나머지 위치에 세 방해자극들이 제시된 표적화면을 제외하고 나머지 RSVP 화면에서 네 흰색 방해자극 문자들이 상응하는 위치에 제시되었다. 고가시성 조건에서는 회색 배경화면 보다 표적 회색이 현저히 어두웠던 반면 저가시성 조건에서는 그 밝기 차이가 상대적으로 분명치 않았다. 참가자들은 사전에 지정된 표적들 중 각 시행에서 어느 것이 출현했는지를 가능한 빠르고 정확하게 탐지하도록 요구받았으며 그 결과 관찰된 탐지 수행은 고가시성 조건에서 더 정확했으며 신속했다. 실험 2에서는 고가시성 조건의 회색 표적을 고선명도 유채색 표적으로 대체한 것을 제외하고 실험 1과 동일한 RSVP 화면 및 과제가 사용되었다. 그 결과 참가자들은 고가시성 조건의 표적을 더 정확히 탐지했으나 반응 속도에 있어서는 두 가시성 조건 간 차이가 없었다. 두 실험의 결과는 시각적 부담이 높은 상황일지라도 현저한 명암대비 및 색상이 부여될 경우 자극에 대한 지각이 촉진됨을 시사하며 자극 색상에 비해 명암대비가 지각적 촉진에 더 중요한 역할을 수행할 가능성을 제안한다.

In Vitro 고삼투압이 정자 원형질막의 Protein Tyrosine Phosphorylation에 미치는 영향 (In vitro Effect of High Osmolality on Plasma Membrane Activities in the Spermatozoa)

  • 정자의 원형질막은 삼투압에 의해서 영향을 받는다고 보고되고 있다. 이중 세포막내 분자구조의 변화 특히 막지질 구조의 변화와 동반되는 이온채널의 변화 그리고 $Ca^{2+}$과 HCO$^{-}_{3}$의 유동성과도 밀접한 관련이 있는 것으로 보고되고 있다. 지금까지의 연구보고에 의하면, 정자의 첨체반웅 (acrosome reaction)이 일어날 경우 protein tyrosine phosphorylation이 증가되는데 이것은 cAMP, protein kinase A 둥을 통하여 작용되는 것으로 보고되고 있다. 막의 지질변화를 유도하는 물질로 일종의 sterol acceptor인 BSA가 알려져 있는 바, 실제로 BSA가 막지질 성분에 미치는 영향을 관찰한 결과 cholesterol이 유출되고 이온 둥의 유동성 변화가 일어나, 이 유동성 변화가 정자의 adenylyl cyclase를 활성화시켜 cAMP를 증가시키고, PKA가 활성화되어 결과적으로 protein tyrosine phosphorylation이 유도된다고 보는 것이다. 첨체반응과 protein tyrosine phosphorylation과는 밀접한 관계가 있는 것으로 사료되고 있다. 본 연구는 정자 원형질막에서 cholesterol이 유출되어 protein tyrosine phosphorylation이 유도될 때, BSA와 같은 sterol acceptor가 작용할 것이라는 전제하에, 고삼투압 하에서 탈수로 인해 원형질막이 위축되더라도 sterol acceptor가 존재한다면 막지질 성분의 구조적 변화가 억제될 수 있을 것이라는 가설을 설정하였다. 실험결과, 저온 및 고삼투압 하에서 정자운동은 감소되지만 원형질막의 구조적 변화는 없고, 삼투압에 대한 반응정도는 원형질막을 통한 수분이동과 세포공적 변화에 따라 비례적으로 일어난다고 하는 사실을 발견하였다. 이 결과는 정자보존에 있어서 저온변화에 영향을 미치는 여러 인자들 특히 protein tyrosine phosphorylation의 증가와 밀접한 관계가 있음을 시시해 준다. 또한 sterol acceptor로 알려진 BSA는 삼투압이 변화되더라도 역시 중요한 인자로 작용할 수 있음을 알 수 있었으며, 특히 고삼투압으로의 변화는 cAMP와 protein kinase A를 거치는 신호전달과정에 있어서 중요한 요인이라는 사실을 확인할 수 있었다.

Vimentin Is Involved in Peptidylarginine Deiminase 2-Induced Apoptosis of Activated Jurkat Cells

  • Peptidylarginine deiminase type 2 (PADI2) deiminates (or citrullinates) arginine residues in protein to citrulline residues in a $Ca^{2+}$-dependent manner, and is found in lymphocytes and macrophages. Vimentin is an intermediate filament protein and a well-known substrate of PADI2. Citrullinated vimentin is found in ionomycin-induced macrophage apoptosis. Citrullinated vimentin is the target of anti-Sa antibodies, which are specific to rheumatoid arthritis, and play a critical role in the pathogenesis of the disease. To investigate the role of PADI2 in apoptosis, we generated a Jurkat cell line that overexpressed the PADI2 transgene from a tetracycline-inducible promoter, and used a combination of 12-O-tetradecanoylphorbol-13-acetate and ionomycin to activate Jurkat cells. We found that PADI2 overexpression reduced the cell viability of activated Jurkat cells in1a dose- and time-dependent manner. The PADI2-overexpressed and -activated Jurkat cells presented typical manifestations of apoptosis, and exhibited greater levels of citrullinated proteins, including citrullinated vimentin. Vimentin overexpression rescued a portion of the cells from apoptosis. In conclusion, PADI2 overexpression induces apoptosis in activated Jurkat cells. Vimentin is involved in PADI2-induced apoptosis. Moreover, PADI2-overexpressed Jurkat cells secreted greater levels of vimentin after activation, and expressed more vimentin on their cell surfaces when undergoing apoptosis. Through artificially highlighting PADI2 and vimentin, we demonstrated that PADI2 and vimentin participate in the apoptotic mechanisms of activated T lymphocytes. The secretion and surface expression of vimentin are possible ways of autoantigen presentation to the immune system.

Chicken novel leukocyte immunoglobulin-like receptor subfamilies B1 and B3 are transcriptional regulators of major histocompatibility complex class I genes and signaling pathways

  • Objective: The inhibitory leukocyte immunoglobulin-like receptors (LILRBs) play an important role in innate immunity. The present study represents the first description of the cloning and structural and functional analysis of LILRB1 and LILRB3 isolated from two genetically disparate chicken lines. Methods: Chicken LILRB1-3 genes were identified by bioinformatics approach. Expression studies were performed by transfection, quantitative polymerase chain reaction. Signal transduction was analyzed by western blots, immunoprecipitation and flow cytometric. Cytokine levels were determined by enzyme-linked immunosorbent assay. Results: Amino acid homology and phylogenetic analyses showed that the homologies of LILRB1 and LILRB3 in the chicken line 6.3 to those proteins in the chicken line 7.2 ranged between 97%-99%, while homologies between chicken and mammal proteins ranged between 13%-19%, and 13%-69%, respectively. Our findings indicate that LILRB1 and LILRB3 subdivided into two groups based on the immunoreceptor tyrosine-based inhibitory motifs (ITIM) present in the transmembrane domain. Chicken line 6.3 has two ITIM motifs of the sequence LxYxxL and SxYxxV while line 7.2 has two ITIM motifs of the sequences LxYxxL and LxYxxV. These motifs bind to SHP-2 (protein tyrosine phosphatase, non-receptor type 11) that plays a regulatory role in immune functions. Moreover, our data indicate that LILRB1 and LILRB3 associated with and activated major histocompatibility complex (MHC) class I and ${\beta}2-microglobulin$ and induced the expression of transporters associated with antigen processing, which are essential for MHC class I antigen presentation. This suggests that LILRB1 and LILRB3 are transcriptional regulators, modulating the expression of components in the MHC class I pathway and thereby regulating immune responses. Furthermore, LILRB1 and LILRB3 activated Janus kinase2/tyrosine kinase 2 (JAK2/TYK2); signal transducer and activator of transcription1/3 (STAT1/3), and suppressor of cytokine signaling 1 genes expressed in Macrophage (HD11) cells, which induced Th1, Th2, and Th17 cytokines. Conclusion: These data indicate that LILRB1 and LILRB3 are innate immune receptors associated with SHP-2, MHC class I, ${\beta}2-microglobulin$, and they activate the Janus kinase/signal transducer and activator of transcription signaling pathway. Thus, our study provides novel insights into the regulation of immunity and immunopathology.

The Decision-Making Journey of Malaysian Women with Early Breast Cancer: A Qualitative Study

  • Background: The survival outcomes for women presenting with early breast cancer are influenced by treatment decisions. In Malaysia, survival outcome is generally poor due to late presentation. Of those who present early, many refuse treatment for complementary therapy. Objective: This study aimed to explore the decision making experiences of women with early breast cancer. Materials and Methods: A qualitative study using individual in-depth interviews was conducted to capture the decision making process of women with early breast cancer in Malaysia. We used purposive sampling to recruit women yet to undergo surgical treatment. A total of eight participants consented and were interviewed using a semi-structured interview guide. These women were recruited from a period of one week after they were informed of their diagnoses. A topic guide, based on the Ottawa decision support framework (ODSF), was used to facilitate the interviews, which were audio recorded, transcribed and analysed using a thematic approach. Results: We identified four phases in the decision-making process of women with early breast cancer: discovery (pre-diagnosis); confirmatory ('receiving bad news'); deliberation; and decision (making a decision). These phases ranged from when women first discovered abnormalities in their breasts to them making final surgical treatment decisions. Information was vital in guiding these women. Support from family members, friends, healthcare professionals as well as survivors also has an influencing role. However, the final say on treatment decision was from themselves. Conclusions: The treatment decision for women with early breast cancer in Malaysia is a result of information they gather on their decision making journey. This journey starts with diagnosis. The women's spouses, friends, family members and healthcare professionals play different roles as information providers and supporters at different stages of treatment decisions. However, the final treatment decision is influenced mainly by women's own experiences, knowledge and understanding.
