Intraspecific variation of gene structure in the mitochondrial large subunit ribosomal RNA and cytochrome c oxidase subunit 1 of Pyropia yezoensis (Bangiales, Rhodophyta)

  • Red algal mitochondrial genomes (mtDNAs) can provide useful information on species identification. mtDNAs of Pyropia / Porphyra (Bangiales, Rhodophyta) have shown diverse variation in their size and gene structure. In particular, the introns and intronic open reading frames found in the ribosomal RNA large subunit gene (rnl) and cytochrome c oxidase subunit 1 gene (cox1) significantly vary the mitochondrial genome size in Pyropia / Porphyra species. In this study, we examined the exon / intron structure of rnl and cox1 genes of Pyropia yezoensis at the intraspecific level. The combined data of rnl and cox1 genes exhibited 12 genotypes for 40 P. yezoensis strains, based on the existence of introns. These genotypes were more effective to identify P. yezoensis strains in comparison to the traditional DNA barcode cox1 marker (5 haplotypes). Therefore, the variation in gene structure of rnl and cox1 can be a novel molecular marker to discriminate the strains of Pyropia species.

The Effects of Extracts Mixture Drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on Hematopoietic Stem Cells and Lymphocyte Subset of Blood in Human (차가버섯, 상황버섯 및 영지버섯 복합추출물 복용이 인체의 혈중 조혈모세포와 면역세포에 미치는 영향)

  • This study was performed to investigate the effect of extract mixture(IPGE) drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on hematopoietic stem cells and lymphocyte subset[lymphocyte, $CD4^+T$ cell, $CD8^+T$ cell, Natural Killer(NK) cells] of blood in 37 participants who were healthy and about $40{\sim}70$ years old. They were divided into two groups; extract mixture drink administration group(n=27) and placebo administration group(n=12). They were given the test drink daily for 4 weeks. Blood was obtained from the subjects every two week in the beginning of administration day to evaluate the $CD34^+$ hematopoietic stem cells and immune cells. As results, $CD34^+$ hematopoietic stem cells were significantly increased after taking IPGE drink for 4 weeks compared to that before taking the drink (p<0.001). There was no significant changes in number of lymphocytes, $CD4^+T$ cells, $CD8^+T$ cells, NK cells and in the ratio of $CD4^+/CD8^+$ cell after taking the test drink. From these results, it was suggested that IPGE have a good health effect by promoting the proliferation of the hematopoietic stem cells.

Functional Metagenome Mining of Soil for a Novel Gentamicin Resistance Gene

  • Extensive use of antibiotics over recent decades has led to bacterial resistance against antibiotics, including gentamicin, one of the most effective aminoglycosides. The emergence of resistance is problematic for hospitals, since gentamicin is an important broad-spectrum antibiotic for the control of bacterial pathogens in the clinic. Previous study to identify gentamicin resistance genes from environmental samples have been conducted using culture-dependent screening methods. To overcome these limitations, we employed a metagenome-based culture-independent protocol to identify gentamicin resistance genes. Through functional screening of metagenome libraries derived from soil samples, a fosmid clone was selected as it conferred strong gentamicin resistance. To identify a specific functioning gene conferring gentamicin resistance from a selected fosmid clone (35-40 kb), a shot-gun library was constructed and four shot-gun clones (2-3 kb) were selected. Further characterization of these clones revealed that they contained sequences similar to that of the RNA ligase, T4 rnlA that is known as a toxin gene. The overexpression of the rnlA-like gene in Escherichia coli increased gentamicin resistance, indicating that this toxin gene modulates this trait. The results of our metagenome library analysis suggest that the rnlA-like gene may represent a new class of gentamicin resistance genes in pathogenic bacteria. In addition, we demonstrate that the soil metagenome can provide an important resource for the identification of antibiotic resistance genes, which are valuable molecular targets in efforts to overcome antibiotic resistance.

Studies on the Development of Rise with Plant Extracts for Enhancing Growth Rate (생약추출물을 이용한 키크는 쌀 개발에 관한 연구)

  • Various kinds of chinese herbs have been known as to promote growth rate of animals. We investigated the growth promoting effect of the selected plants by using fish and rats as experimental animals. In a preliminary test by using fish, Eleutherococcus senticosus and Artemisia capillasis were found to be effective. Based on this result, we manufactured the mixture of plant extracts and applied this mixture to rice, named as Kiwoomi/sup TM/. When we administered Kiwoomi/sup TM/ to rats, it was found to be effective in growth promotion. Kiwoomi/sup TM/ - administered group showed 49.3% (0.75g/kg B.W.) and 52.3% (1.59/kg.B.W.) growth promotion, respectively. On the other hand, Control group showed 46.2% growth promotion. This growth promotion was supported by increase of GH and IGF-1 level in the Kiwoomi/sup TM/- administered group. From these results, we suggest that this functional rice could be helpful for growing children.

New Isolation Technique and Culture System for Clinical Applications of Human Amniotic Epithelial Stem Cells (인간태반양막유래 상피줄기세포의 임상적용을 위한 새로운 세포분리 및 배양 기술)

  • Human placenta is abundant source of adult stem cells. Especially, amniotic epithelial cells have stem cell characteristics, expressing surface markers normally present on embryonic stem cells and germ cells. However, culturing and expanding amniotic epithelial cells in vitro without feeder cells are difficult due to endogenous characteristics of epithelial cells. In the present study, amniotic epithelial cells are isolated and proliferated in several passages by applying dithiothreitol and a Rho-associated kinase inhibitor in culture media. The cultured amniotic epithelial cells showed the epithelial and stem cell characteristics. In conclusion, human placenta-derived amniotic epithelial stem cells can be a major source of stem cells for medical treatment of various diseases without any controversial issues.

Studies on the anti-parasitic efficacy and safety of ivermectin and pyrantel pamoate compound against Dirofilaria immitis in dogs

  • Dirofilaria (D.) immitis is an important canine parasitic nematode in dogs. D. immitis parasitizes the right ventricle and pulmonary artery of dogs. An ivermectin and pyrantel pamoate compound (IPPC) was administered to dogs naturally infected with this parasite. IPPC is composed of 68.0, 136.0 and $272.0{\mu}g$ of ivermectin and 57.0, 114.0 and 227.0 mg pyrantel pamoate for small, middle, and large animals. Ivermectin has activity against nematodes and ectoparasites in dogs. Pyrantel pamoate is also effective against nematodes in dogs. Our results showed that this drug combination has good efficacy in D. immitis infected dogs.

Studies on a New Alimentotheraphy for Ddiabetic Patients (당뇨환자를 위한 새로운 식이요법제에 관한 연구)

  • Mulberry, Mori folium leaves (ML) and fruits (MF), Lycii fructus (LF), and Panax ginseng have been known to maintain normal blood sugar levels (BSL) in folk medicine. Here we investigated the effects of coated rice with these herb extract to diabetic patients. Alloxan-induced diabetes mellitus (DB) mice (80 mg/kg BW., i.v.) separated into 3 groups. One group received the extract in high dose (6.5 mg/kg B.W.), and another one group received in low dose (1.3 mg/kg B.W.) perorally for 30 days. (+) control received no extracts. (-) control received neither of them. BSL, volume of water consumed (VWC), body weights (BW) of each group were measured and compared. BSL in both of the extracts-received groups got close to normal levels on 4th week after starting the experiments. VWC also showed the same pattern. BSL in (+) control remained high through the experiment periods. BW in (+) control remained high, but those in the extracts-received groups recovered to normal levels. The clinical trials for both of healthy volunteers and DB patients, conducted in a University Hospital, also showed that the rice coated with the extracts could control the BSL. From these results, we suggest that the functional rice with herb extracts could be used as a new alimentotherapy for DB patients.

Studies on the Progesterone Level for Early Pregnency Diagnosis of Cheju Native Mares (혈중 Progesterone 수준 측정에 의한 제주재래마의 조기 임신진단 응용에 관한 연구)

  • Plasma progesterone concentrations were measured by using radjoimmunoassay for early diagnosis of pregnancy in Cheju-native mares. A total of 226 pony mares were examined for pregnancy during breeding and non breeding seasons. Plasma progesterone levels 20~23 days after the onset of oestrus were 4.67+O.67ng /rnl and O.55+O.O4ng /ml for mares becornrning pregnant and not pregnant after the estrus, respectively, and there was a significant differences (p<0.01) between the two groups. Progesterone concentration of pregnant mares gradually increased in 30 days andreached a peak (10.3ng /ml) during the third month of gestation. However, the concentration decresed to the base line (1.llng /rnl) at 7 months and gradually increased again as foaling approached (2.lng /ml). Early diagnosis for pregnancy of Cheju mares by progesterone level at 20~23 days after onset of oestrus was 88% accurate when 4.6ng /ml was used to classify mares as pregnancy and below 1.3ng /rnl was used to determine nonpregnant mares. However, the accuracy of the diagnosis was improved to 96% when a progesterone level of above 2ng /mi was used to classify mares for pregnancy. Diagnosis for pregnancy was 69.6% accurate when mares were classified as pregnancy by horse owners during breeding season. The progesterone levels of pregnant and non-pregnant mares during non-breeding season varied greatly between individual animals, Plasma progesterone levels of pregnant animals ranged from 3.5ng /mi to above 6.2ng /mi whereas similar values were observed in non-pregnant animals. Radioirnrnunoassay technicjues can be applied for the early pregnant diagnosis of Cheju native mares when progesterone levels are measured during the early gestation period (18~23 days after onset of oestrus). However, progesterone concentration of mares in non-breeding season is conisidered unsuitable as a indicator of pregnant diagnosis.

The Development of Functional Food with Plant Extracts for Enhancing Growth Rate (생약추출물을 이용한 키 성장 기능성 식품 개발)

  • It has been reported that various kinds of chinese herbs have an activity of promote growth rate in both animals and human. To investigate the growth promoting effect of the selected plants, fish and pigs were used as experimental animals. In fish, Eleutherococcus senticosus extract and Artemisia capillasis extract were found to be most effective. And these plant extracts were given to pigs. The result showed that plant extracts-fed pigs were significantly increased their body weight gain at 7.06% of average daily gain, as compared to control. The verify this results, secreted growth hormone (GH)levels and insulin-like growth factor-1 (ICF-1) levels into blood were measured. This result indicated that GH and IGF-1 levels in the blood in plant extracts-fed pigs were higher that those of control. To confirm growth promotion effect on human, we manufactured the mixture of these plant extracts, and coated this mixture onto rice, named as $Kiwoomi^{TM}$. When we administered $Kiwoomi^{TM}$ to elementary students, it was found to be effective in growth promotion. This result showed that $Kiwoomi^{TM}$-treated elementary students significantly increased their growth rate (about 2.14 times), as compared to untreated children. Taken together, it is suggested that this functional rice ($Kiwoomi^{TM}$) might be helpful for growing children without any side effects.
