Enzymatic saccharification of rice straw, a lignocellulosic biomass by the extracellular enzyme from KMU001 (KMU001 조효소에 의한 목질계 바이오매스, 볏짚의 효소당화성에 관한 연구)

  • This study shows that lignocellulosic biomass saccharification work has been carried out with rice-straw by the extracellular enzyme from KMU001, and the enzymes produced in 5%(w/v) wood biomass were characterized by protein and various enzyme activity measurements. Several cellulases such as Endoglucanase(EG), $\beta$-D-1,4-Glucosidase(BGL), Cellobiohydrolase(CBH), and $\beta$-D-1,4-Xylanase (BXL) were detected. Saccharification of rice-straw by the enzyme yielded about 233mg/g of glucose after 48hrs.

Degradation of Rice Straw by Rumen Fungi and Cellulolytic Bacteria through Mono-, Co- or Sequential- Cultures

  • Two strains of rumen fungi (Piromyces rhizinflata B157, Orpinomyces joyonii SG4) and three strains of rumen cellulolytic bacteria (Ruminococcus albus B199, Ruminococcus flavefaciens FD1 and Fibrobacter succinogenes S85) were used as mono-cultures or combinationally arranged as co- and sequential-cultures to assess the relative contributions and interactions between rumen fungi and cellulolytic bacteria on rice straw degradation. The rates of dry matter degradation of co-cultures were similar to those of corresponding bacterial mono-cultures. Compared to corresponding sequential-cultures, the degradation of rice straw was reduced in all co-cultures (P<0.01). Regardless of the microbial species, the cellulolytic bacteria seemed to inhibit the degradation of rice straw by rumen fungi. The high efficiency of fungal cellulolysis seems to affect bacterial degradation rates.

  • This study is performed to examine the physical and mechanical properties of Eco-concrete using rice straw ash for planting. The tests for void ratio, compressive and bending strength with neutralization treatment point, curing condition and coarse aggregate size are performed. The test result shows that the void ratio is decreased with increasing content of rice straw ash. But, the compressive and bending strength are increased with increasing content of rice straw ash. The greatest strength is appeared when neutralization is treated in curing age of 6 days. These Eco-concrete is very useful for planting.

  • Rice straw treated with anhydrous ammonia and/or sulphur dioxide was incubated with rumen liquor for 24 hours and 48 hours to investigate the changes in cell wall structure caused by the treatments and bacterial degradation using scanning electron microscopy (SEM). A less significant tissue loss of untreated rice straw was inspected after incubated for 24 hours and 48 hours. Sulphuration decreased the thickness of sclerenchyma and apparently removed parenchyma tissues. Ammoniation degraded the phloem, and the lignified inner portion of the cell wall was completely, however, little collapsed epidemis and vascular bundles. Ammonia and Sulphur dioxide combined treatment removed the inner layer from outer layer. The extent of apparent degradability following combination treatment was the largest due to the enhanced microbial degradation of sclerenchyma and parenchyma cells.

A Studyd on Adsorptive Properties of Activated Carbons Produced from Rice-Straw

  • Activated carbons from rice-straw can be used as an adsorbents for the purification of water were prepared and evaluated. The adsorptive capacities of activated carbons were measured by iodine, potassium permangante, phenol and metals. It was observed by electron microscope (SEM) and IR spectrum that organic components in the rice-straw and its carbonization product were disappeared. Slit-shaped and porousstructures were formed by activation. There was no relationship between temperature and adsorption of iodine but adsorption of potassium permanganate increased as temperature rose. The adsorption of the phenol was greater than 99%. The adsorption data of phenol at $25^\circ{C}$ obeyed the Freundlich's isotherm. Various metals except sodium were not removed by activated carbon.

  • This study was performed to develop the plantable concrete block using rice straw ash and apply for inclined plane. For the planting, plantable concrete block needs infiltration of water and air through void of block. The materials used for plantable concrete block are cement, rice straw ash, and coarse aggregate (5-10, 10-20 mm). Plantable block size is $23\times23\times4$ cm, and species of planting are Tall fescue, Lespedeza cyrtobotrya and Lespedeza cuneata. At the 6 months after seeding, germination ratio and grown-up length of Tall fescue, Lespedeza cyrtobotrya and Lespedeza cuneata are shown in 90%, 60%, 50%, and 40~50 cm, 90~120 cm, 60~75 cm, respectively.

Comparison of Dry-Seeding Methods for Improving Rice Seedling Stand on Reclaimed Saline Soil (간석지 벼 입모율 향상을 위한 건답직파 방법 비교)

  • This study was conducted to find out the feasibility in direct seeded rice cultivation on the reclaimed saline soil. Seedling emergence was tested under the different application rates of rice straw and seeding depth, and also under combined treatments soil moisture regimes and seeding depth on saline soils in a greenhouse. The comparison of seedling stand and yield performance of rice in rotary till after broadcast seeding and in non-plow after broadcast seeding were tested on a field that reclaimed saline soil. Seedling emergence ratio in application of rice straw(4 and 6 tons /ha) was higher than that in non-application of rice straw. Seedling emergence and plant height were remarkably increased with the shallow seeding depth in the application of rice straw(4 tons /ha). The seedling emergence under proper soil moisture condition(25%) was higher than that under excess soil moisture condition(35%). Under proper soil moisture condition, the plant height was increased with the shallowed seeding depth. The number of seedling stand per m$^2$ in non-plow after broadcast seeding was larger than that in rotary till after broadcast seeding. Panicle number per m$^2$ in non-plow after broadcast seeding was much larger than that in rotary till after broadcast seeding and the yield showed the same trends as panicle number.

Furrow Covering Effects with Rice Straw on Nutrient Discharge from Upland Soil Used for Red Pepper Cultivation (고추밭 고랑 볏짚피복에 의한 양분유출 특성)

  • Excessive application of nutrient supplement on the upland soil may increase the amount of discharge to surrounding water systems. The chemical fertilizer (CF), cow manure compost (CMC), and pig manure compost (PMC) are used as a nutrient supplement for cultivation of red pepper. Rice straws are widely used as a soil covering material in order to reduce weed occurrence, to protect soil moisture, and to supply organic matter in upland soil. This study was conducted to evaluate the furrow covering effect with rice straw on nutrient discharge in upland soil used for red pepper cultivation. The experimental plots of nutrient supplement were consisted of CF, CMC, and PMC and the amount of nutrient application were as recommended amount after soil test for red pepper cultivation. Each nutrient supplement treatment plot has no furrow covering (CFC) as a control and furrow covering with rice straw (FCS), respectively. Furrow covering with rice straw (FCS) of CF treatment and CMC treatment reduced the amount of T-N(total nitrogen) discharge by $1.4kg\;ha^{-1}$, $2.1kg\;ha^{-1}$, respectively, compared to control. While the amount of T-P(total phosphorus) discharge of the furrow covering with rice straw of CF, CMC, and PMC increased by $2.1kg\;ha^{-1}$, $2.1kg\;ha^{-1}$, and $0.2kg\;ha^{-1}$, respectively, compared to control. The phosphorus and nitrogen content of straw were 0.4 % and 0.3 % respectively. In addition, in three week the phosphorus was eluted from the straw which soaked in distilled water. Thus, it was assumed that T-P discharging originated from rice straw which applied as a furrow covering material. The furrow covering with rice straw reduced weed occurrence compared to control. But production of fresh red pepper was not influenced significantly by furrow covering with rice straw. In conclusion, excessive furrow covering with rice straw could induce T-P discharge from upland soil used for red pepper cultivation. Further studies are needed to evaluate the appropriate amount of rice straw as a furrow covering material.

The Effect of Replacing Grass with Urea Treated Fresh Rice Straw in Dairy Cow Diet

  • Nine tons of fresh rice straw from early-maturing varieties was ensiled with 50 g urea $kg^{-1}$ DM straw in plastic bags immediately after threshing. Five months after storage, preserved straw was used to partially replace elephant grass (Pennisetum purpureum, Schumach) for lactating cows. Eight crossbred Holstein lactating cows (75% of Holstein blood) in their second to fourth lactation and in mid-lactation were arranged in a balanced design with two squares consisting of $4\;periods{\times}4\;treatments$ (100% grass ad lib. as a control; 75% grass+urea treated fresh rice straw (UTrFRS) ad lib.; 50% grass+UTrFRS ad lib.; 25% grass+UTrFRS ad libitum) in one square. A concentrate supplement was given at a rate of 400 g per day per kg of milk produced. Samples of fresh straw taken in the field and UTrFRS and elephant grass taken at feeding time were evaluated in a degradation trial with 3 fistulated heifers (undefined blood ratio of crossbred of Sindhi and local yellow cattle). Straw preserved for 5-9 months was in nearly all cases of good quality. Crude protein (CP) content was increased 2.1 fold and 48 h dry matter loss (DML) was 20% higher compared to dry straw. Elephant grass cultivated intensively was low in DM content and 10% higher in 48 h DML compared to UTrFRS. Dry matter intake (DMI) was higher for the mixture of UtrFRS and Elephant grass, and highest when one-third of the roughage was UTrFRS. Higher DMI of mixed roughage diets was probably due to the low DM content of elephant grass in the sole grass roughage diet. Increasing substitution of elephant grass with UTrFRS up to 75% of the roughage component increased milk fat content and had no effect on milk yield and other milk composition parameters. Feeding UTrFRS, partially replacing elephant grass in the diets of lactating cows in the dry season can reduce the cost of roughage.
