Review of Disease Incidence of Major Crops of the South Korea in 2004 (2004년 주요 농작물 병해 발생개황)

  • In 2004, average temperature was higher, and rainfall was less than those of previous year, The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Seedling diseases, leaf blast, sheath blight and bacterial blight of rice, phytophthova blight, virus diseases and anthracnose of pepper, and sudden wilt syndrome and powdery mildew of chinese melon grown in plastic greenhouse were severe. Especially, sheath blight and bacterial blight of rice occurred two times higher than those of previous year, Panicle blight of rice decreased about 4 times, compared with the previous year, presumed that the lower rainy days, rainfall and RH suppressed spread of the fungal pathogens to panicles of rice. Lower rainfall during mid- and late Aug caused three-times less occurrence of phytophtora blight of red-pepper than that of the previous year, Another diseases surveyed occurred similar or less than those of the previous year.

Discussions on the Efficacy, Phytotoxicity and Residue of Organo-Arsenic Fungicides (유기비소제의 약효 및 약해와 잔류에 관한 종합적 고찰)

  • Since rice high yielding practices with blast resistant varieties induced severe and earlier infestation of sheath blight recently, it is strongly suggested to spray at least 10 days earlier than the present recommendation and more than 2 sprays from early July to early August seems to be essential for the efficient control of sheath blight in the future. Among organo-arsenic fungicides, MAFe (Neo Asozin) is known as the most effective and economic as well as with the advantages of free phytotoxicity and safe residure on rice, and applications of the mixed formulation with other fungicide and insecticide is suitable for the one-hit control of rice diseases and insect pests in connection with cooperative mass control system. Present level of arsenic $(As_2O_3)$ residue in .ice is summarized as 0.2ppm to 0.65 ppm in highest average, and there are more possibilities of extensive MAFe fungicide uses for the efficient and economic control of this disease. During the past few years, the acreage of sheath blight control has been extended several times while the increase of the fungicides used for sheath blight showing about 10 times in quantity and mort than 15 times in amount, and also the fungicide cost estimated for the control is differ from 1 to 12 billion 1on by the use of MAFe or antibiotic fungicide in a year.

Control Efficacy of Phloretin Isolated from Apple Fruits Against Several Plant Diseases

  • In the course of a searching natural antifungal compounds from plant sources, we found that the methanol extract ($3,000\;{\mu}g/ml$) of Malus domestica fruits had potential of control against rice blast (Magnaporthe grisea) and tomato late blight (Phytophthora infestans). Under bioassay-guided purification, we isolated phloretin, a phenolic compound, with in vivo antifungal activity against M. grisea. By 1-day protective application of phloretin ($500\;{\mu}g/ml$), the compound strongly inhibited the disease development of M. grisea and P. infestans on rice and tomato seedlings, respectively. And red pepper anthracnose caused by Colletotrichum coccodes also was moderately suppressed. However, rice sheath blight (Rhizoctonia solani AG1), and barley powdery mildew (Blumeria graminis f. sp. hordei) were hardly controlled. In addition, the compound showed in vitro antifungal activity against some plant pathogenic fungi including Phytophthora capsici, Alternaria panax, Sclerotinia sclerotiorum, R. solani AG4, and M. grisea. This is the first report on the antifungal activity of phloretin against plant pathogenic fungi.

  • In Korea the most important plant industry is the rice-growing but she has not yet attained self-sufficiency with regard to rice which is the basic food of Korean. In the past, various measures of increasing rice production have been tried with some success but the increasing rate of did not show the sufficient result. Judging from experiences, disease control may be the most important factor which has influence on increasing of rice production in Korea. During the past 60 years three periods divided by the transition of rice diseases were noted, and especially the recent transition attracted much attention . That is, sheath blight and stripe which were minor diseases in the past have promptly spread all over the country and caused heavy losses to rice plant, and also local outbreak of bacterial leaf blight and dwarf were noticed. Various environmental factors are related to such transition of rice disease. Above all, cultivated varieties susceptible to these diseases, increase of application of nitrogenous fertilizer, earlier transplanting and density of transplanting are considered to be lucre closely related to the development of these diseases and successive development of blast. On the other hand. application of forage fungicides such as organic mercury compounds specific to blast contributed to reduce prevalent area if blast in spite of wide spread of susceptible varieties and increase of application of nitrogenous fertilizer. These facts were confirmed by Korean investigators At the present which various diseases to control coexist in the same region, no one variety resistant to all of these diseases is provided and cultural practices for the control of these diseases often do not produce sufficient yield of rice. it is emphasized that a role of pesticides is very great.

Purification and Properties of Trehalase from Rhizoctonia solani (Rhizoctonia solani가 생산하는 Trehalase의 정제 및 특성)

  • Nonregulatory trehalase has been purified from mycelia of Rhztoctonia solani. a pathogen of rice sheath blight. Purification procedures involved sonification, gel filtration and fast protein liquid chromatography. Purity was confirmed by isoelectric focusing with silver staining. The purified trehalase was estimated to have a molecular weight of 54,000 and pI point of 5.1. Maximal activity was observed at pH 5.4 and temperature $45^{\circ}C$. The purified trehalase exhibited on apparent Km for trehalose of 3.11 mM and a Vmax of 105.3 $\mu mol min^{-1}\times mg^{-1}$. Validamycin, a commercial antibiotics of rice sheath blight, was a non-competitive inhibitor of Rhzzoctoniu solani trehalase.

Inhibitory Effects of Resveratrol and Piceid against Pathogens of Rice Plant, and Disease Resistance Assay of Transgenic Rice Plant Transformed with Stilbene Synthase Gene

  • Resvestrol has been known to inhibit bacterial and fungal growth in vitro, and can be accumulated in plant to concentrations necessary to inhibit microbial pathogens. Hence, stilbene synthase gene has been used to transform to synthesize resveratrol in heterologous plant species to enhance resistance against pathogens. In the present study, we investigated the antimicrobial activities of resveratrol and piceid to bacterial and fungal pathogens, which causing severe damages to rice plants. In addition, disease resistance was compared between transgenic rice varieties, Iksan 515 and Iksan 526 transformed with stlibene synthase gene and non-transgenic rice varieties, Dongjin and Nampyeong. Minimum inhibitory concentration of resveratrol for Burkolderia glumae was 437.5 ${\mu}M$, and the mycelial growth of Biplaris oryzae was slightly inhibited at concentration of 10 ${\mu}M$. However, other bacterial and fungal pathogens are not inhibited by resveratrol and piceid. The expression of the stilbene synthase gene in Iksan 515 and Iksan 526 did not significantly enhanced resistance against bacterial grain rot, bacterial leaf blight, sheath blight, and leaf blight. This study is the first report on the effect of resveratrol and piceid against pathogens of rice plant, and changes of disease resistance of transgenic rice plants transformed with stilbene synthase gene.

Isolation and Identification of Myxobacteria KR025 and Searching of Their Bioactive Compounds (점액세균 KR025의 분리 동정 및 생리활성물질의 탐색)

  • Fifty isolates of myxobacteria were isolated from soils from several areas in Korea during 1996-1997 and bioactivity against plant pathogenic fungi of these isolates was examined. A myxobacterial isolate KR025 showed good antifungal activities against Pyricularia oryzae, Cryphonectria parasitica, Colletotrichum lagenarium, and C. gloeosporioides but did not against Rhizoctonia solani, Fusarium oxysporum and Pythium ultimum. The bacterium was identified as Myxococcus fulvus based on morphological and physiological characteristics. Antifungal substances were extracted from culture broth and bacterial cell of Myxococcus fulvus KR025 by ethyl acetate. Antifungal substance of Myxothiazole (100 ${\mu}{\textrm}{m}$/ml) produced by Myxococcus fulvus KR 025 controlled 97.0% rice blast, tomato late blight, wheat leaf rust, and barley powdery mildew and showed 45.0 and 82.6% disease control of rice sheath blight and cucumber gray model, respectively.

Evaluation of Disease Resistance of a Leaffolder-resistant (Cry1Ac1) Rice Event and Gene Transfer to Plant Pathogens (혹명나방 저항성벼(Cry1Ac1)의 병해 저항성 및 병원균으로의 유전자 전이)

  • The genetically modified leaffolder-resistant (Cry1Ac1) rice plant was evaluated for the changes of resistance by comparing the occurrence of major diseases with a japonica type Korean rice variety, Nakdong which was the mother plant of the transgenic rice event, in greenhouse and field conditions. There was no difference in the occurrence of sheath blight and Helminthosporium blight between the two varieties in the fields. We couldn't find any difference of resistance for fungal blast and bacterial leaf blight by artificial inoculation in greenhouse. There was also no difference in the susceptibility to sheath blight in artificial inoculation tests confirming the results in the fields. The possibility of gene transfer of Bar and Cry1Ac1 from the genetically modified rice plant to naturally infected pathogens such as Fusarium moniliforme and Pyricularia oryzae in the field conditions was tested by PCR. And the possible transfer of those genes by continuous inoculation of Xanthomonas oryzae pv. oryzae and Rhizoctonia solani was also tested. However, we couldn't find any possibility of transfer of the genes in natural and artificial conditions.

Ecological Studies on Rice Sheath Blight Caused by Rhizoctonia solani IV. Inoculation Time of Sclerotium and Disease Development (벼 잎집무늬마름병의 생태학적 연구 IV. 균핵의 접종시기와 발병)

  • Natural sclerotia of Rhizoctonia solani causing rice sheath blight were inoculated at 10 day intervals from June 15 to July 15 in paddy field, Icheon, Korea. Percentage of infected stems, top lesion height and percentage of. lesion height vs. plant height were higher in the early inoculated plots than in the late inoculated ones. However, rio significant differences among inoculation dates of sclerotia were found on the basis of degree of damage at maturing stage and rice yield. These results suggest that the time of initial symptom appearance under the same inoculum potential may not affect the damage of rice plants by the fungus.
