Production of Weak Acid by Anaerobic Fermentation of Soil and Antifungal Effect

  • Acetic acid and butyric acid were produced by the anaerobic fermentation of soil mixed with wheat or rice bran. The concentration of acetic acid produced in the wheat and rice bran-treated soil was 31.2mM and 8mM, respectively, whereas the concentration of butyric acid in the wheat and rice bran-treated soil was 25.0mM and 8mM, respectively. The minimal fungicidal concentration (MFC) for all the fungal strains was 40-60mM acetic acid, 20-40mM butyric acid, and 40-60mM mixture of acetic acid: butyric acid (1:1, v/v). Consequently, the efficacy of mixing wheat-bran with soil to control soil diseases was demonstrated.

Development of Protein Biomarkers for the Authentication of Organic Rice

  • The rice protein profiles of Oryza sativa L (Koshihikari) grown under organic and conventional cultivation regimes were compared on 2-D gels to develop diagnostic marker proteins for organic rice. The selected proteins, differentially expressed between organic and conventional rice, were compared with the differentially expressed proteins of another organic and conventional rice pairing, produced at a different location. In the first comparison among conventional, no-chemical, and organic rice grown in the same region, Korea, 13 proteins exhibiting differential expression in organic and conventionally grown plants were selected. Eight of the 13 proteins were down-regulated, and the 5 remaining proteins were up-regulated from conventional to organic rice. The second comparison pairing from Kyungju, revealed 12 differentially expressed proteins, with 8 down-regulated and 4 up-regulated proteins. Ten of the differentially expressed proteins that overlapped between the two comparison sets could not be clustered into any functional group using a functional annotation clustering tool. Further comparisons using another set of conventional and organic rice, belonging to a different variety of Oryza sativa L and produced in Sanchung, revealed 8 differentially expressed proteins, 5 of which were down-regulated and 3 of which were upregulated in the organic rice. Overall, 3 differentially expressed proteins were commonly found in all three organic rice crops. These 3 proteins, along with other overlapping differentially expressed proteins, can provide a good starting point for the development of signature proteins that can be used for the authentication of organic rice with a follow-up studies with more comparison sets.

Plastid Transformation in the Monocotyledonous Cereal Crop, Rice (Oryza sativa) and Transmission of Transgenes to Their Progeny

  • The plastid transformation approach offers a number of unique advantages, including high-level transgene expression, multi-gene engineering, transgene containment, and a lack of gene silencing and position effects. The extension of plastid transformation technology to monocotyledonous cereal crops, including rice, bears great promise for the improvement of agronomic traits, and the efficient production of pharmaceutical or nutritional enhancement. Here, we report a promising step towards stable plastid transformation in rice. We produced fertile transplastomic rice plants and demonstrated transmission of the plastidexpressed green fluorescent protein (GFP) and aminoglycoside 3′-adenylyltransferase genes to the progeny of these plants. Transgenic chloroplasts were determined to have stably expressed the GFP, which was confirmed by both confocal microscopy and Western blot analyses. Although the produced rice plastid transformants were found to be heteroplastomic, and the transformation efficiency requires further improvement, this study has established a variety of parameters for the use of plastid transformation technology in cereal crops.

알코올 발효조건 및 효모를 달리한 현미식초의 품질 특성 (Quality Characteristics of Brown Rice Vinegar by Different Yeasts and Fermentation Condition)

  • 알코올 발효 효모 종류를 달리하여 현미탁주를 제조하고 이를 이용한 현미식초(정치배양 및 교반배양)의 품질 특성을 조사하였다. 효모 종류에 따른 알코올 함량은 B가 14.3%로 가장 높았으며 적정산도는 모든 구간에서 0.6% 이하로 나타났다. 이를 이용한 교반 및 정치배양 현미식초의 품질특성을 비교한 결과, 교반배양구간에서는 초기산도 1.0%, 초기 pH는 3.9~4.0에서 발효 7일째 DV구간에서 산도가 6.05%로 가장 높았고 AV, CV 및 BV가 5.64, 5.55 및 5.32% 순으로 나타났다. 또한 정치배양구간에서도 산도는 발효 14일째까지 꾸준히 증가하여 5.01~5.31%로 나타났고 발효 16일째부터 조금씩 감소하는 경향이었다. 효모 종류에 따른 현미식초의 산도 및 pH 차이는 크게 나타나지 않았다. 현미식초의 유리아미노산 함량을 비교한 결과, 교반배양구간에서 총 유리아미노산 함량은 BV, DV, AV 및 CV 순으로 높게 나타났고 필수 유리아미노산 함량은 BV에서 100 mg% 이상으로 가장 높았다. 정치배양구간에서 유리아미노산 함량은 모든 구간에서 교반배양구간보다 높은 함량을 나타내었으며 serine, alanine, valine, isoleucine, leucine 및 $\gamma$-aminobutyric acid 등이 교반배양구간에 비하여 높았으며 특히 $\gamma$-aminobutyric acid는 45.69~52.27 mg%로 약 10배 이상 높게 나타났다. 현재까지의 결과는 현미식초의 품질특성은 효모 종류보다는 초산발효 방법에 따른 품질 차이가 더 큰 것으로 나타났다. 그러나 향후 숙성과정에서 변화를 거치면서 향기성분, 관능적 특성에 차이가 있을 것으로 예상되어 다양한 식초의 품질인자에 관한 연구가 요구된다.

The Fruiting Body Formation of Oudemansiella radicata in the Sawdust of Oak (Quercus variabilis) Mixed with Rice Bran

  • To screen additives and their mixed ratio suitable for the mycelial growth and fruiting body formation of Oudemansiella radicata in the oak sawdust, additives such as rice bran, fermented soybean powder and wheat bran were used. Generally, the mycelial growth of O. radicata has been stable on oak sawdust mixed with rice bran of $5{\sim}20%$. In case that O. radicata was cultured for about 30 days at $22{\pm}1^{\circ}C$ under the illumination (350 lux) of 12 hours and moisture condition of $90{\pm}5%$, the primordia have been formed gradually from red-brown crusts covering the surface of oak sawdust media. Based on the experimental results from 9 strains of O. radicata, fruiting bodies were produced widely on oak sawdust medium mixed with rice bran of 5 to 30%. Even though fruiting bodies of O. radicata have been produced well on oak sawdust media mixed with rice bran, fruiting bodies of O. radicata were produced intensively on oak sawdust media mixed with rice bran of 10%. Therefore, this result will provide a basic information for commercial production of fruiting body of wild O. radicata. This result is the first report associated with an artificial fruiting body formation of O. radicata in Korea.

Strength and Some Durability Properties of Concrete Containing Rice Husk Ash Produced in a Charcoal Incinerator at Low Specific Surface

  • Strength and some durability properties of concrete containing rice husk ash (RHA) predominantly composed of amorphous silica at a specific surface of 235 $m^2/kg$ produced using a charcoal incinerator were determined. The maximum ordinary Portland cement (OPC) replacement with the RHA increased with increase in water/binder (w/b) ratio of the concrete mixes. The results show that 15 % OPC could be substituted by the RHAwithout strength loss at w/b ratio of 0.50. The split tensile strength generally increased with increase in RHA content for the mixes.

  • One of effective utilization technique of rice husk is known to carbonize it for using as the culture materials. A series of study on the production of carbonized rice husk by a cyclone combustor shows that the carbonized rice husk produced have a strong alkalinity. Therefore, carbonized rice husk produced by a cyclone combustor is required to neutralize with proper normality. This work is the third part of a series on the production of carbonized rice husk by a cyclone combustor. In this work, the development of neutralization process was carried out in the range of experimental conditions recommended in the previous study. Those include the preheat temperature of combustion chamber of T1b=1273∼1373K, equivalence ratio =1.68∼2.17, auxiliary gas flow rates Qg=5.15∼6.43$\ell$/min. The injection technique of dilute acid was employed for neutralization. At the lower position of the outside of combustor, a dilute nitric acid selected as neutralization liquid was injected to the carbonized rice husk exhausted from the combustion chamber. The normalities of dilute nitric acid were varied to 0.01, 0.03 and 0.05N, respectively. The injection flow rates of the solution were changes from 1.7∼4$\ell$/min. The required carbonized and neutralized rice husk could be obtained at the dilute nitric acid with normality of 0.3N and flow rate of dilute nitric acid of 2∼3.5$\ell$/min. However, the carbonized and neutralized rice husks of about 10∼20% were destroyed by spray with high injection pressure.

고미(古米)의 품질변이 특성 (Variation of Rice Quality of Milled Rice according to Storage Period)

  • 본 연구는 쌀의 생산년도별(2005년, 2006년, 2009년) 품질변이 양상을 구명하기 위하여 남평벼와 동진벼에 대한 다양한 이화학적 특성 및 품질특성을 조사하였다. 저장기간이 길수록 단백질 함량은 증가하였으며, amylose 함량 및 Toyo 윤기치는 감소하는 경향을 보여주었다. 또한 지방산도(KOH mg/100 g)는 2006년산 남평벼와 동진벼가 각각 7.79와 4.79를 나타내었고 2005년산 남평벼와 동진벼가 각각 15.95와 15.68를 나타내어 저장기간이 길수록 2009년산에 비하여 지방산도가 월등히 증가되어 고미취 발생과 산패가 진전된 것으로 보여주었다. 또한 색도는 저장기간이 증가할수록 L값, a값, b값 모두에서 증가하는 경향을 보였다. 아밀로그래프에 의한 호화점도 측정 결과에서는 저장 기간이 증가함에 따라 최저점도, 최종점도 및 치반점도 값은 감소하는 경향을 보였지만 강하점도는 증가하는 경향을 보였다. 경도는 저장기간이 증가할수록 잘 부숴지는 경향을 보였고 식미관능평가 결과에서는 일반적으로 저장기간이 증가할수록 선호도가 매우 낮음을 알 수 있었다. 이상의 결과로부터 쌀은 수확 후 저장기간이 5년이상 경과되면 신선도와 품질이 저하되고 지방산도가 증가되어 고미취 및 이취가 발생하고 쌀자체의 호화 및 취반특성에 많은 변화가 나타나 밥맛과 관련하여 품질이 저하될 뿐만아니라 식품원료로서의 가치가 떨어지는 것으로 확인 할 수 있었다.

  • Twenty six isolates of Magnaporthe grisea originated from rice and other gramineous hosts Korea were tested for mating type with MAT1-1 and MAT1-2 standard isolates. Ninety three and 64% of rice and grass isolated mated and produced perithecia with standard isolates, respectively. Both mating types were found from rice and non-rice isolates, but MAT1-1 allele of M. grisea was predominant in Korea.
