Characteristics of Endosperm Starch of the Rice Mutant Lines Induced by Sodium Azide

  • Rice consumption per capta, in South Korea, has been decreased dramatically, owing to the changes of living patterns. Because of not only the major energy source of Korean people but also major income source of Korean farmers, diversifying end-use-quality of rice has been demanded. To the context, 'Suweon 472', a high yielding and early mature japonica line and released as 'Namilbyeo' to framers in 2002, was treated with a chemical mutagen, Sodium Azide to find endosperm mutant types. A total of nine endosperm mutat lines, including five waxy, one dull, two floury, and one white core type, were identified from the 3,542 mutatagen treated lines. Amylose contents, iodine reaction, disintegration in alkali solution, gelatinization in urea solution and amylogram properties of those nine endosperm mutant lines were evaluated to address the possibility as new genetic materials for diversifying rice quality of Korean japonica cultivars. All embryo mutants were clearly differentiated from their wild type, 'Suweon 472', in terms of physic-chemical properties evaluated. The endosperm mutant lines would be very useful in expanding untiliztation of rice through opening new rice markets of processed foods from Korean japonica rice.

  • Physicochemical factors affecting cooking and eating quality of rice and their mechanisms were investigated. The stickiness of cooked rice was negatively correlated with amylose content(r=0.58, p<0.05) and protein content(r=-0.72, p<0.01), but not affected by crude fat content of rice. The ultrastructure of cooked rice grain showed the progressive gelatinization of starch from the periphery toward the center of the endosperm as water and heat energy diffused into. The rate of water diffusion appears to be dependent on the cell arrangement in the endosperm and the protein content of milled rice. Once water and heat reach the starch granules, the rate of in situ gelatinization of starches appears to be dependent on their own gelatinization temperature range and amylose content. Protein acts as a barrier for the swelling of starch and water diffusion in two ways : 1) by encasing starch granules in the starchy endosperm, and 2) by forming a barrier between the subaleurone layer and the starchy endosperm. Therefore, the separation and fragmentation of the outermost layers of the endosperm occurred more easily in the low-protein content rices, and was associated with increases of solids lost in cooking-water at 95$^{\circ}C$ and stickiness of cooked rice.

Quantitative comparison of acidic polysaccharides in the endosperm of two major varieties of rice

  • Rice endosperm, the portion that remains after milling, is the part of the rice seed that is primarily consumed as a source of nutrients. There have been many studies on polysaccharides, such as hemicellulose, cellulose, and pectins, derived from the cell walls of various plant groups. It has been reported that the acidic polysaccharide fractions, which contain water-soluble pectins that have been shown to have pharmacological effects in vivo and in vitro, have common chemical structures that include galacturonic acid polymers, rhamnose, arabinose, and galactose. However, few studies have been conducted on the acidic polysaccharides contained in the endosperm of rice. In this study, we quantitatively compared the differences in the acidic polysaccharide contents from samples from two of the main varieties of rice consumed as staple foods, japonica and indica, using a colorimetric method. Rice samples were collected from 39 different regions in Korea, China, Thailand and Vietnam. Acidic polysaccharide fractions were obtained by precipitation of the alcohol-insoluble residue (AIR) and enzyme treatment of each sample. The total amount of carbohydrates and uronic acid in each acidic polysaccharide fraction were measured using the phenol-sulfuric acid method and the carbazole-sulfuric acid method, respectively. The differences in the total polysaccharide contents in the acidic polysaccharide fractions were not statistically significant (p = 0.07), but the uronic acid contents were significantly different between the two groups (p = 0.04).

흑미의 도정분획 메탄올 추출물의 항산화 성분 및 항산화 효과 (Antioxidant Compounds and Antioxidant Activities of the Methanolic Extracts from Milling Fractions of Black Rice)

  • 본 연구에서는 흑미 두 품종(흑진주벼와 흑광벼)의 도정 분획별 메탄올 추출물의 항산화 성분과 항산화 활성을 비교 분석하고자 하였다. 흑미의 항산화 성분으로 polyphenolic, flavonoid, anthocyanin과 ${\gamma}$-oryzanol, tocopherol 및 tocotrienol 함량을 측정하였으며 항산화 활성은 DPPH와 ABTS radical 제거능을 측정하였다. 각 도정 분획별 메탄올 추출물에서 rice bran 층에서 약 $15.3{\sim}15.5%$의 가장 높은 수율을 보였으며 endosperm 메탄올 추출물에서 약 $0.9{\sim}1.1%$의 가장 낮은 수율을 보였다. 실험결과 rice bran의 추출물에서 endosperm, whole grain의 추출물보다 높은 함량의 항산화 성분이 분포함을 확인할 수 있었다. 또한 이러한 항산화 성분이 다량 분포하는 rice bran 추출물의 우수한 항산화력을 측정할 수 있었다. 품종 간에는 흑진주벼의 rice bran층의 항산화 성분이 흑광벼보다 높은 함량을 나타내었다. 본 연구결과 흑미의 rice bran은 기능성식품의 성분으로 활용가능성이 높을 것으로 예상된다.

Pre-harvest Sprouting Tolerance Test in Rice with Floury Endosperm

  • Pre-harvest sprouting(PHS) refers to germinating seeds in the mother plant before harvesting under low dormancy and humid climate, deteriorating grain quality, and rice yield. Rice varieties with floury endosperm(RFE) have been developed to boost domestic rice consumption by invigorating the processed rice industry, reducing milling and environmental cost. However, the PHS tolerance of RFE is relatively low in the rice varieties with transparent endosperm(RTE) since they soak moisture rapidly due to soft endosperm. In this study, Baromi2(BR2), floury endosperm, and Jomyeong1(JM1), PHS tolerance donor, were crossed to improve PHS tolerance. Major agronomic traits and PHS tolerance test of ten F7(BR2/JM1) lines were conducted in NICS, 2022. The evaluations of PHS were carried out according to the method of RDA(2012) with slight modifications. Briefly, three panicles were treated and incubated 25℃ in a growth chamber 35 days after the heading date. Ten PHS tolerance promising lines demonstrated floury endosperm. The heading date of BR2 and JM1 was 7/27 and 8/5, respectively. The heading date of promising lines was 7/23~8/10. The PHS rate of BR2 and JM1 exhibited 56.3% and 10.7%, respectively. However, the PHS rate often promising lines demonstrated 2.4%~52.4%, 3 lines significantly lower than BR2. Further studies such as ABA contents are necessary to elucidate the mechanism of PHS tolerance in BR2/JM1. These results may contribute to developing elite RFE lines with improved PHS tolerance.

Proteomic Analysis of Drought Stress-Responsive Proteins in Rice Endosperm Affecting Grain Quality

  • Drought stress is one of the major abiotic stresses in agriculture worldwide. We report here a proteomic approach to investigate the impact of post-fertilization drought on grain quality in rice seed endosperm (Oryza sativa cv. IR-64). Plants were stressed for 4 days at 3 days before heading. Total proteins of endosperm were extracted and separated by two-dimensional gel electrophoresis. Not many protein spots showed differential accumulation in drought-stressed samples. More than 400 protein spots were reproducibly detected, including three that were up-regulated and five down-regulated. Mass spectrometry analysis and database searching helped us to identify six spots representing different proteins. Functionally, the identified proteins were related to protein synthesis and carbohydrate metabolism, such as Granule-Bound Starch Synthase (GBSS, Wx protein), which is thought to play a very important role in starch biosynthesis and quality, a very crucial factor in determining rice grain quality.

남일벼 돌연변이 후대 계통 'Namil(SA)-flo1'의 분질배유 특성에 대한 유전분석 (Genetic Analysis on Floury Endosperm Characteristics of 'Namil(SA)-flo1', a Japonica Rice Mutant Line)

  • 쌀 가공산업을 활성화하고 소비를 촉진하여 국내 쌀 생산 기반을 유지하기 위해서는 쌀가루 제분적성을 보유한 가공용 벼 품종 개발이 시급하다. 농촌진흥청 국립식량과학원에서는 아지드화나트륨을 돌연변이원으로 활용하여 건식제분 적합성을 보유한 분질배유 돌연변이 후대계통인 'Namil(SA)-flo1'를 육성한 바 있다. 본 연구는 염색체 상에서 'Namil(SA)-flo1'의 분질배유 특성을 지배하는 유전자위를 탐색하고자 수행하였다. 주요 결과는 아래와 같다. 1. 'Namil(SA)-flo1' ${\times}$ '밀양23호'로부터 유래한 F2 94 개체로부터 종자 분질립 비율을 검정하고 54개 SSR 마커의 유전자형을 검정하여 연관성분석(association analysis)을 수행한 결과 목표 유전자위는 5번 염색체 중하단 부위로 추정되었다. 2. 목표 부위의 SSR 마커 밀도를 높여 추가 연관성분석을 실시하였고, F2:3 종자 분질립 변이의 79.7%가 5번 염색체 상의 RM164의 유전자형 변이에 의하여 설명된다는 것을 확인하였다. 3. 이를 통하여 분질배유 지배 유전자위를 5번 염색체 17.7~20.7 Mbp 부위로 추정하였으며, 추후 추가 분리집단을 이용하여 목표 유전자를 동정하고 쌀가루용 품종 개발에 활용할 수 있는 핵산정밀표지인자를 개발할 계획이다.

  • A strategy for development of a functional rice in proved with human lactoferrin and enhancement of nutrient compounds was planned. For the purposes we have cloned and characterized a human lactoferrin cDNA from human mammary gland cDNA library A endosperm storage vacuole targeting sequence and the cDNA fragment was linked to endosperm specific glutelin promoter. The fusion gene fragment was inserted into a binary vector containing MAR gene. In addition a new ${\beta}$-galactosidase gene from Bifidobacterium of human was used as a reporter gene in the vector system, Rice plants showing a high concentration of amino acids in the endosperm cells were developed by using a biochemical mutation and bred for the transformation with the binary vector system Finally we have established a transformation method for the rice endosperm cells.

Effect of Particle Size on the Solubility and Dispersibility of Endosperm, Bran, and Husk Powders of Rice

  • Size effects of rice product powders on physical properties including suspension stability were investigated in this study. Endosperm, bran, and husk powders of rice with different size particles were prepared using the pin crusher or the ultrafine air mill. The physical properties of the powders were examined using particle size analysis, scanning electron microscopy, and spectrophotometry. The peak of the volume-weighted particle distribution of ultrafine endosperm particles was at $5.4\;{\mu}m$ whereas those of the bran and the husk appeared at 65 and $35\;{\mu}m$, respectively. Ultrafine particles of the endosperm and the husks dispersed better than larger-sized particles. As time elapsed, the dispersibility decreased, but the ultrafine particles were precipitated at the slowest rate. Our results suggest that ultrafine particles, including future nanosized particles, provide improved solubility and dispersibility resulting in better stability in the food colloidal suspension.

제분방법을 달리하여 제조한 8품종 변이체벼의 쌀빵가공성 비교 (Comparison of Some Characteristics Relevant to Rice Bread made from Eight Varieties of Endosperm Mutants between Dry and Wet Milling Process)

  • 아밀로오스함량, 당함량, 전분입자의 형태 등에 차이를 보이는 8품종 변이체 벼들의 쌀빵가공성에 대한 제분방법(습식 및 건식)의 차이에 대해서 검토하였다. 이들 8품종의 최대수분흡수량은 품종에 따라 상당히 차이가 있어 shr.은 61.5%, 분질미는 48.4%였고, 화청벼 남풍벼 및 그들의 변이체들은 아밀로오스 함량이 낮은 품종의 쌀일수록 최대 수분흡수량은 증가하고 있었다. 건식제분에 의해서 제조한 쌀빵이 성형성, 기계적인 물성(경도, 탄력성, 응집성. 씹힘성) 및 관능검사에 의한 질감의 면에서 습식제분의 경우보다 우수하였다.
