Resistant cell lines to azetidine-2-carboxylic acid (AZCA) were selected through rice embryo culture after mutagenic treatment of callus irradiated with 30,50,70,90 and 120 Gy. The optimum AZCA concentration for the selection of resistant cell lines was 3 or 4 mM AZCA considering $LD_{50}$ and the fresh weight of callus. Survival rate of the AZCA resistant callus showed remarkable increase in the callus irradiated with 50 and 70 Gy. Regeneration rate of the AZCA resistant callus was much lower on the whole. Ninety and 120 Gy increased the regeneration rate for calli selected from 3 and 4 mM AZCA, respectively. Based on fresh weight, survival rate and regeneration for selection of the AZCA resistant cell line, 50-90 Gy was considered as the optimum range of gamma irradiation. Irradiated calli selected from AZCA were more tolerant to NaCl than those from non-irradiated calli. It suggests that elevated resistance to osmotic stress resulted from mutagenic treatment. The level of free proline content in the AZCA resistant cell line was increased up to 3.5 times compared with that in the control. Proline content in the regenerant derived from the AZCA resistant cell line also increased to 1.7 times that from the control plants regenerated from callus grown in AZCA free medium. Selection of proline overproducing cell lines by in vitro mutagenesis was successful and seems to be useful for improvement of stress tolerance in this crop.
From the sample seeds of rice cultivar Palkoeng with the brown spot symptom, Helminthosporium oryzae was detected in $86\~92\%$ of hulls including empty glumes, lemma, palea, and rachilla in five to seven days of incubation. When the seeds were dehulled, the percentage detection decreased to $74\~83\%$ of pericarps. The fungus sporulated on whole surface of the seed and was detected within about ten cell layers deep of endosperm but not detected in embryo. Fusarium moniliforme was found in embryo as well as pericarp and endosperm. The frequency of F. roseum, Trichoconis padwickii, Curvularia spp., Atternaria tenuis, Cladosporium cladosporoides and Phoma sp. was less than three percent of hulls and pericarps. In seven days of incubation, $60\~87\%$ of the germinating seeds showed the symptom of H. oryzae while $13\~40\%$ were apparently healthy. This fungus in hilum of infected rice transmitted through pericarp to plumule shoot and radicle of the germinating seed.
An ultimate purpose of this study was to develop an automatic system for brown rice quality inspection using image processing technique. In this study emphasis was put on developing an algorithm for discriminating the brown rice kernels depending on their external quality with a color image processing system equipped with an adaptor magnifying the input image and optical fiber for oblique lightening. Primarily, geometical and optical features of images were analyzed with paddy and the various brown rice kernel samples such as a sound, cracked, peen-transparent, green-opaque, colored, white-opaque and brokens. Secondary, geometrical and optical parameters significant for identifying each rice kernels were screened by a statistical analysis(STEPWISE and DISCRIM procedure, SAS wer. 6) and an algorithm fur on- line discrimination of the rice kernels in static state were developed, and finally its performance was evaluated. The results are summarized as follows. 1) It was ascertained that the cracked kernels can be detected when e incident angle of the oblique light is less than 2$0^{\circ}C$ but detectivity was significantly affected by the angle between the direction of the oblique light and the longitudinal axis of the rice kernel and also by the location of the embryo with respect to the oblique light. 2) The most significant Parameters which can discriminate brown rice kernels are area, length and R, B and r values among the several geometrical and optical parameters. 3) Discrimination accuracies of the algorithm were ranged from 90% to 96% for a sound, cracked, colored, broken and unhulled, about 81 % for green-transparent and white-opaque and 75 % for green-opaque, respectively.

Zinc finger proteins constitute a large family which has been studied to have various functions in different organisms. Tandem CCCH zinc finger proteins (TZFs), members of the zinc finger protein family, are known to participate as post-transcriptional regulators of gene expression in eukaryotes. Here, we showed that the OsZn15, a gene for tandem CCCH zinc finger protein, is induced by abiotic stress and its overexpression in transgenic rice plants (PGD1:OsZn15) gains higher drought tolerance. Gene expression analysis of promoter:GFP plants revealed that OsZn15 is specifically expressed in anther and embryo, but not in vegetative organs. In-field evaluation, grain yield was higher in the PGD1:OsZn15 than nontransgenic plants under drought conditions. Interestingly, OsZn15 is shown to not only localize at nucleus but also co-localize with both processing bodies (PB) and stress granules (SG), two messenger ribo-nucleoprotein complexes which are known to activate by forming cytoplasmic foci under stress conditions. In sum, these results suggest that OsZn15 increases drought stress tolerance of rice probably by participating in RNA turnover in PB and SG.
In vitro culture of panicle has been the method to accumulate starch and protein in immature grains by providing nutrients after florets crossed between remote genotypes artificially. Grain filling means embryo development and sucrose translocation from photosynthetic source, and starch manufacture in endosperm. The concentrations of sucrose used to culture immature rice panicle were 5, 10, 15, 20% and glutamine was 20 mM. When immature rice panicles at 5 days after flowering were cultured in distilled water with different concentrations of sucrose, glutamine 20 mM and MS medium with different concentrations of sucrose, glutamine 20 mM for 30 days the later was effective on grain filling. The optimal concentration of sucrose on grain filling in vitro culture of rice panicle was 10% and the weight of grain cultured was 10.14 mg that was corresponded to 57% of intact plant. In the method of treating plant growth regulators, the culture of immature rice panicle adding in MS medium with Kinetin, IAA, $\textrm{GA}_3$ were effective on grain filling than the culturing of immature rice panicle after submerging in solutions of Kinetin, IAA, $\textrm{GA}_3$ for 1day. When immature rice panicle was cultured in MS medium with sucrose 10% and Kinetin 46.47 $\mu$M it was effective on grain filling, respectively. The weight of grain cultured was 13.1mg that was corresponded to 75% of intact and germination rate was 51 %. When immature rice panicles were cultured in medium with different concentrations combined with Kinetin 4.65, 46.47, 464.7 $\mu\textrm{M}$, IAA 5.71, 57.08, 570.80 $\mu\textrm{M}$ for 30 days and in medium with IAA 5.71, 57.08, 570.80 $\mu\textrm{M}$ for 15 days after culturing in medium with Kinetin 4.65, 46.47, 464.70 $\mu\textrm{M}$ for 15 days the effect on grain filling was similar.

Fusarium graminearum causes the devastating plant disease Fusarium head blight and produces mycotoxins on small cultivated grains. To investigate the timeframe of F. graminearum infection during rice cultivation, a spore suspension of F. graminearum was applied to the rice cultivars Dongjin 1 and Nampyeongbyeo before and after the heading stage. The disease incidence rate was the highest (50%) directly after heading, when the greatest number of flowers were present, while only 10% of the rice infected 30 days after heading showed symptoms. To understand the mechanism of infection, an F. graminearum strain expressing green fluorescent protein (GFP) was inoculated, and the resulting infections were visually examined. Spores were found in all areas between the glume and inner seed, with the largest amount of GFP detected in the aleurone layer. When the inner part of the rice seed was infected, the pathogen was mainly observed in the embryo. These results suggest that F. graminearum migrates from the anthers to the ovaries and into the seeds during the flowering stage of rice. This study will contribute to uncovering the infection process of this pathogen in rice.

The objective of this study was to determine antioxidant compounds in specialty rice including milled rice, brown rice, red rice, giant embryonic rice, black rice, green rice, and Goami consumed in Korea. The concentrations of total polyphenols (insoluble and soluble polyphenols), phytic acid, and anthocyanin in the samples were measured using spectrophotometric methods and vitamin I analysis was carried out by HPLC. The contents of the total polyphenolic compounds were 565mg/100g for black rice, 405mg/100g for red rice, 140mg/100g for giant embryo rice, 138mg/100g for Goami, 133mg/100g for brown rice, 127mg/100g for green rice, and 66mg/100g for white rice. The black and red rices were significantly high in polyphenolic contents compared with the other rices, apparently due to their intense red-purple color. Black rice, red rice, and Goami showed significantly higher vitamin E and phytic acid contents compared with other rices. Anthocyanins were determined in only black rice (302mg/100g) due to the detection limits of spectrophotometric assay. hlthough vitamin I and anthocyanin contents were relatively lower than polyphenolics among the samples, the specialty rice may contribute to the significant supply of antioxidant compounds to prevent oxidative stress due to the fact that rice is used as a staple food and consumed in large amounts in our diets. The results can be used to increase rice consumption by enhancing consumer awareness on health benefits of the rice.
Recently, increasing interests in local foods have been highlighted along with active efforts and activities from autonomy organizations to develop local cultures and promote local economies by generating value added products and profits through the development of local foods. Subsequently, useful effects might result by making local foods attractive to tourists, such as by using indigenous Gimpo products to make menu items that incorporate the social, economic, and cultural aspects of the Gimpo area, and contributing to its food culture, the development and use of its indigenous products, and promoting local restaurant businesses, etc. The items of the survey used to select the local food menu items to be developed were limited to ideas offered by expert groups, and were implemented by a new menu development process. In order to derive the menu items to develop, the following method was applied: a brainstorming session with experts to generate and draft ideas, a questionnaire to chefs and cooks in special grade hotels is Seoul to select the menu items to develop from those drafted, and sensory evaluations by experts to evaluate the developed items. Frequency analysis and technical statistical analysis were performed using the SPSS 12.0 program package, where 13 kinds of local foods were ultimately developed using indigenous products of Gimpo. The developed local food menu items were rice & grape sujaebi, grape sikhae, fried rice embryo bud, grape yanggaeng, rice pancakes, rice spaghetti, grape seed oil dressing, grape sauce, rice pizza, grape pie, rice & grape ice cream, grape chocolate, and rice roll cakes.

Plants have evolved physiological, biochemical and metabolic mechanisms to increase their survival under the adverse conditions. This present study has been performed to select salt-tolerant rice mutant lines through in vivo and in vitro mutagenesis with gamma-rays. For the selection of the salt-tolerant rice mutants, we conducted three times of selection procedure using 1,500 gamma ray mutant lines resulted from an embryo culture of the original rice cv. Dongan (wild-type, WT): first, selection in the a nutrient solution with 171 mM NaCl; second, selection under in vitro condition with 171 mM NaCl; and third, selection in a reclaimed saline land. Based on a growth comparison of the entries, out of the mutant lines, two putative 2 salt tolerant (ST) rice mutant lines, ST-87 and ST-301, were finally selected. The survival rate of the WT, ST-87 and ST-301 were 36.6%, 60% and 66.3% after 7 days in 171 mM NaCl treatment, respectively. The WT and two salt tolerant mutant lines were used to analyze their genetic variations. A total of 21 EcoRI and Msel primer combinations were used to analyze the genetic relationship of among the two salt-tolerant lines and the WT using the ABI3130 capillary electrophoresis system. In the AFLP analysis, a total of 1469 bands were produced by the 21 primer combinations, and 700 (47.6%) of them were identified as having polymorphism. The genetic similarity coefficients were ranged from 0.52 between the ST-87 and WT to 0.24 between the ST-301 and the WT. These rice mutant lines will be used as a control plot for physiological analysis and genetic research on salt tolerance.
