In this experiment, three kinds of mutations(ge, re, and eml )relating to the size of embryos were used to study their generation, genetic mechanism and developmental characteristics, and the interactions between embryo and endosperm were also examined. Giant embryo mutation comprises 7 kinds including the already isolated ge, and ge-2, which share an identical gene site. The SAM and the size of radicule for the ge showed little difference compared to a normal type. The number of embryo cells did not increased as much as it would affect the size of embryo. Therefore, the enlargement of embryo was due to the enlargement of scutellum that originated from the corpulence of each cell. Both F$_1$' s of re ]and odm 49 formed reduce embryos, and other combinations of hybridization showed all wild type of embryo sizes. Accordingly, the odm 49 must have an identical gene site of re 1, while odm 48 and odm 62 have different gene sites. Their shoots and radicules also shrank by the same ratio, however no sign of physical change was noticed. The size of embryo cell showed no change, while the number of cells was the half of that of wild types. The three gene sites of re represent all of them control the size of the entire embryo forming organs. The eml 1 was defined to have temperature sensibilities that the generation of endosperms was active at a high temperature while that was hampered at a low temperature.
An experiment was carried out to find out the changes in morphology, sugars, and $\alpha$-amylase activity during the priming of rice seeds (Oryza sativa L. cv. 'Ilpumbyeo'). For priming, seeds were soaked in -0.6 MPa PEG solution at 15$^{\circ}C$ for 4 days (properly primed) and at $25^{\circ}C$ for 4 and 10 days (over-primed) and dried at room temperature. The size of coleoptile and differentiated leaves of properly primed seeds were bigger and coleoptile was separated from the other part of embryo compared with non-primed and over-primed seeds. As priming of seeds advanced, compound starch grains in the endosperm disintegrated into tiny starch granules, and small holes were found in the tiny starch granules and a cavities developed between embryo and endosperm. The radicle and plumule of properly primed germinating seeds developed faster than non-primed and overprimed germinating seeds. Sucrose, maltose, and raffinose contents of properly primed seeds decreased, while content of glucose and fructose and $\alpha$-amylase activity increased. However, sugar content and $\alpha$-amylase activity of over-primed seeds were lower compared with non-primed seeds or properly primed seeds.
A positive selectable marker system was adapted for transformation of mature embryo-derived calli of Indica rice (Oryza sativa L.) utilizing the PMI gene encoding for phosphomannose-isomerase that converts mannose-6-phosphate to fructose-6-phosphate. The transformed cells grew on medium supplemented with 3% mannose as carbon source and calli were selected on media containing various concentrations of mannose. Molecular analyses showed that the transformed plants contained the PMI gene. The results indicate that the mannose selection system can be used for Agrobacterium-mediated transformation of mature embryo in rice to substitute the use of conventional selectable markers in genetic transformation.

The purpose of this study was to investigate the effect of Monacolin-inoculated rice embryo on the body fat and serum lipid profiles in 61 obese elementary school students. The subjects divided into two groups 31 obese children had the experimental diet (rice embryo inoculated Monacolin) and 30 obese children had the control diet three times a day after meals. The mean age, height, weight, and BMI of 61 subjects were $10.5\pm0.5$ years, $143.6\pm6.8cm,\;55.0\pm8.9kg,\;and\;26.1 \pm3.9$, respectively. The changes of body fat mass (kg) in experimental group and control group after 6 weeks were -0.60kg and -0.03kg, respectively. The changes of body fat $(\%)$ in experimental group and control group after 6weeks were $-1.44\%\;and\;-0.25\%$, respectively. These changes of body fat in experimental group were significantly higher than in control group (p<0.05). The change of total-cholesterol in experimental group and control group during 6weeks were -17.52mg/dL and -1.70mg/dL, respectively. The change of LDL-cholesterol in experimental group and control group during 6 weeks were -17.06mg/dL and - 2.80mg/dL, respectively. The change of triglyceride in experimental group and control group after 6 weeks were -9.58mg/dL and 11.67mg/dL. Total-cholesterol, LDL- cholesterol and triglyceride of experimental group after 6 weeks significantly decreased compared to control group (p<0.05). After experimental diet (6 weeks), total-cholesterol was negatively correlated with body water contents, soft lean mass and fat free mass (p<0.05). Triglyceride showed a significantly positive correlation with body weight and body fat mass, however, it was negatively correlated with fat free mass (p<0.05). HDL-cholesterol showed a significantly positive correlation with fat free mass (p<0.05). These results show that Monacolin-inoculated rice embryo is effective in decreasing body fat and blood lipid in obese children.
Recent identification and characterization of plant homeobox genes suggest that they play important roles in morphogenetic events. OSH1, one of the rice homeobox genes, is thought to be related to organ development since the changes of OSH1 gene expression cause morphological abnormalities of leaves by the ectopic expression and is expressed during early embryogenesis. In this experiment, the expression pattern of OSH1 was analyzedinmutants by in situ hybridization, and OSH1's potential as a molecular marker was explored. Region-specific expression of OSH1 during early embryogenesis shows that OSH1 could be used as a molecular marker for characterizing embryo mutants. Although several organless and shootless mutants showed normal expression of OSM1, some mutants exhibited abnormal expression patterns. In a minute organless cle1-1 embryo whose epidermis resembled morphologically the epithelium of scutellum, OSH1 expression was limited to a small basal region. This expression pattern suggests the gross deletion of the basal part. In a radicleless mutant, odm115, OSH1 expression was detected in a basal region instead of subcentral region of the ventral side. Together with other characteristics (short embryo and normal adventitious roots), odm115 was estimated to be derived from the deletion of basal region. Among five shootless mutants, three showed normal expression of OSH1. In the shl2 embryo, no expression of OSH1 was observed. In the shl1 embryo, however, OSH1 expression was extended to a dorsal side, indicating that SHL2 might be related to dorsoventral patterning. The above results of in situ hybrydization clearly indicate that OSH1 can be utilized as a marker for characterizing gene functions of embryo mutants.

The introduction of doubled haploid (DH) approach into breeding programs has reduced the times and population sizes required for the production of pure lines. We carried out the experiment for development on effective method of producing haploid in wheat. Emasculated spikelets of wheat were pollinated with maize pollen and cultured in the solution containing 40 g/L sucrose and 2,4-D, NAA, 2,4,5-T and dicamba 24 h after pollination, and then incubated until embryo rescue. Fourteen to sixteen days after pollination, the embryos are excised and cultured in half-strength MS basal medium supplemented with 20 g/L sucrose and 1 mg/L NAA. The type of plant growth regulators was found to be most significant in production of haploid plants. The application of synthetic auxins to pollinated florets, stimulates haploid embryo development to a stage where the embryos can be rescued onto nutrient media. The percentage of seeded florets was significantly affected by 100 mg/L 2,4-D, 150 mg/L 2,4,5-T and 50 mg/L dicamba. The percentage of embryos formed was significantly increased by treatment with 2,4-D and 2,4,5-T at 100 mg/L, and dicamba at 50 mg/L, but the treatments with 150 mg/L 2,4-D inhibited embryo development and plant regeneration. The optimum application time of plant growth regulators was 24 hrs after pollination.

Several types of endosperm and embryo mutants were induced by the treatment of MNU (N -methly- N -nitrosourea) on fertilized egg cell of rice plant. These mutants were named as dull. waxy, white core, floury, sugary, shrunken, colored seed coat and giant embryo according to their appearence, micro-scopic feature on SEM and amylose content. White cored mutant was the most frequent one among them. All of the mutants were segregated as controlled by a single recessive gene except 47320 (dull). Futher studies on the genetics and physico-chemical properities of the mutants are ongoing.
Recently developed black waxy rice with a giant embryo ('Nunkeunheukchal', BGE) was selected and processed to produce high quality nutritional food. BGE contains high levels of several phytochemicals with antioxidant activities, as well as other reported health beneficial properties. In addition, the giant embryo has high protein, lipid, and amino acids contents. Within the free amino acids, ${\gamma}$-aminobutyric acid (GABA), a major inhibitory neurotransmitter, has long been used for treating the aftereffects of brain injuries and stroke. A method for manufacturing pop-rice and black rice tea by popping process in BGE is provided to increase a taste, nutrition and functionality. The produced 'pop-rice' showed increased protein (11.3%) and lipid (3.7%) contents compared with control variety, IB ('Ilmibyeo'). In addition, melanoidin related products, polyphenol and functional amino acid contents were increased by the popping process. Pop-rice tea made of BGE showed the highest extraction of total sugar, glucose, raffinose and sucrose (4 times higher than brown rice) by hot water. Scavenging activity ($SC_{50}$) of processed BGE rice powder showed strong antioxidative activity of 0.24 mg/ml using DPPH and 1.82 mg/ml using ABTs method. Thereafter, these results suggested that the popping processed rice of BGE could be one of the promising materials for healthy food development.
This study was conducted to determine physiobiochemical basis of heterosis using rice hybrids such as Shanyou 63 (Zhenshan 97As Minhui 63) and Teyou 63 (Longtepu A Minhui 63) as compared with inbred rice like Milyang 23. Seed protein patterns of rice hybrid showed complementary genetic characteristics inherited from their parents. Hybrid rice had larger embryo and higher $\alpha$-amylase activity than those of inbred rice. The larger embryo of hybrid was significantly correlated with tillering ability and high number of low node tillers jplant increased by 60~70% in Shanyou 63, leading to higher productive tillers/plant which directly influenced on grain yield of hybrid rice. These characters were further supported by high chlorophyll content in hybrids. Exogenous application of GA$_3$ (0.02 ppm) on inbred rice like Milyang 23, increased significantly $\alpha$-amylase activity, but no effect of GA$_3$ on hybrid rice was observed, indicating that sufficient amount of GA$_3$ is endogenously present in hybrid rice, showing 1 to 3.5 fold higher activity of $\alpha$-amylase in hybrid rice, which trigger heterosis from the germinating stage. Further, activity of cytochrome c oxidase was 2.66 to 5.52 fold higher in hybrid rice than that of inbred rice, indicating that rice hybrids have very active metabolism reflecting vigorous growth starting from the germinating stage, in turn leading to higher tillering ability.
