Effect of pH, Temperature, and added Sucrose on the Production of Vitamin $B_{12}$ and Riboflavin by Bacillus megaterium and Enterobacter aerogenes (온도, pH 및 첨가된 Sucrose가 Bacillus megaterium 과 Enterobacter aerogenes 에 의한 비타민 $B_{12}$ 와 Riboflavin 생산에 미치는 영향)

  • Optimal pH temperature and sucrose content for the production of vitamin B$_{12}$ and riboflavin by Bacillus megaterium and Enterobacter aerogenes was studied by microbiological analysis. Optimal pH for the production of B$_{12}$ was 6.0 by B. megaterium while the pH for E. aerogenes was 5.0. However, upon the addition of sucrose the optimal pH for B. megaterium shifted to 7.5 but E. aerogenes remained at pH 5.0. In the absence of sucrose, pH did not influence the yields of riboflavin produced by either bacterium. Addition of sucrose stimulated synthesis of riboflavin by both bacteria. Temperature had little effect on the production of vitamins by either bacterium.

  • For the production of riboflavin, strain development of Ashbya gossypii NRRL Y-1056 was attempted by NTG(N-methyl-N'-nitro-N-nitrosoguanidne) treatment. The optimum composition of culture medium and other culture conditions for the production of riboflavin by selected mutant Ashbya gossypii JAG-13 were determined. The optimum composition of medium was 9% of corn oil, 3% of gellatone, 4% of CSL, 0.3% of glycine, 0.2% of S770. The optimum culture temperature and initial pH of medium was $28^{\circ}C$ and 6.5, respectively. oxygen was essential for the production of riboflavin, but excess oxygen inhibit the production of riboflavin. When Ashbya gossypii JAG-13 was cultured under above conditions for 12 days with a bioreactor, 6.9 mg/ml of riboflavin was produced.

  • In this paper, the microbial production of riboflavin is reviewed and includes descriptions of riboflavin overproducers, and the biosynthesis and details of the key-enzyme genes related to riboflavin. There kinds of riboflavin overproducers are known; Bacillus subtilis and Candida famate utilize glucose as a carbon source, but the fungus Ashbya gossypii requires plant oil as its sole carbon source. The starting material in ribofalvin biosynthesis is guanosine triphospate (GTP), which is converted to riboflavin through six enzymatic reactions. Though Bacillus subtilis, Candida famate, and Ashbya gossypii operate via different pathways until GTP, they follow the same pathway from GTP to riboflavin. From the metabolic viewpoint, with respect to improved riboflavin production, the supplementation of GTP, aprocess-limiting precursor must be considered. The GTP fluxes originate from three sources, serine, threonine and glyoxylate cycles. The development of pathways to strengthen GTP supplementation using biotechnological techniques remains an issue fro future research.

Media Optimization and Fed-Batch Fermentation for Riboflavin Overproduction by Ashbya gossypii (Ashbya gossypii로부터 riboflavin 대량생산을 위한 배지 최적화와 유가식 배양)

  • In order to maximize the riboflavin production by a mutant strain Ashbya gosspyii, the optimization of medium and fed-batch fermentation were performed. As carbon sources, glucose and soybean oil were necessary for the riboflavin overproduction. Optimal concentrations of glucose and soybean oil in the flask cultures were found to be 3.0% and 0.5%, respectively, in a complex medium containing corn steep liquor(CLS) 1%. Among the various organic nitrogen sources tested, CSL was the most effective one both for the cell growth and riboflavin overproduction.

Physio-Morphological Changes in a Riboflavin Producer Eremothecium ashbyii DT1 and UV Mutants in Submerged Fermentation

  • By UV-irradiation of Eremothecium ashbyii DTl, a higWy flavinogenic mutant (UV-18-57) and a nonflavinogenic mutant (UV -85) were obtained. The physio-morphological characteristics of these three strains were studied on glucose medium in submerged fermentation. Glucose utilization and mycelial growth occurred in 0 - 2 days of fermentation. By the third day, the biomass had declined. Extracellular riboflavin excretion was distinct from the second day, reaching a maximum rate by the fourth day. The hyphae of the highly flavinogenic mutant UV-18-57 were broader than DTl, while the nonflavinogenic UV-85 hyphae were very thin. Riboflavin accumulation was high in UV-18-57 (extracellular riboflavin,$825\mu\textrm{g}/ml$ , and intracellular, $490\mu\textrm{g}/ml$) and caused the mycelia to swell into bulbous forms. Riboflavin accumulation was less in DTl ($108\mu\textrm{g}/ml$ extracellular and $24\mu\textrm{g}/ml$ intracellular) and correspondingly its hyphae were thinner than those of UV-18-57 and swollen bulbous mycelia were not prominent. UV-85 was nonflavinogenic and, accordingly, its mOlphological characteristics included long thin filaments with no intracellular riboflavin accumulation. A large number of greenish fluorescence spores were seen in UV-18-57, whereas DTI had less spores and UV-85 was nonsporulating. Sporulation is correlated with riboflavin production. UV-18-57 had better mycelial integrity and lysis started only by the seventh day, whereas DTI and UV -85 started to lyze earlier by 4 -5 days. By the late stage of fermentation (eighth day), DTl had a few long, thin filaments indicating some secondary growth, whereas UV -85 showed a compact pellet form of mycelia. Most mycelia of UV-18-57 still appeared intact.

  • Cultural conditions for $\alpha$-D-Glucosidase Inhibitor production from Streptomyces sp. YS-221-B isolated from soil arid identified as Streptomyces flauovirens or a subspecies of it were investigated. When the strain was cultured in a flask containing 2% glucose, 0.3% asparagine, 0.0002% riboflavin, 0.05% $K_2$HPO$_4$, 0.1% MgSO$_4$.7$H_2O$, 0.05% NaCl, pH 8.0 at 3$0^{\circ}C$, maximum production of the inhibitor was obtained after 8-9 days of cultivation. Sugar alcohols such as mannitol, i-inositol, erythritol as carbon sources, asparagine and beef extract as nitrogen sources were favorable for inhibitor production. Among vitamins, riboflavin, p-aminobenzoic acid, pyridoxamine and folic acid promoted the production of inhibitor, but depressed by the addition of hesperidine, and also depressed by cobalt, lithium and ferrous salts.

  • Ulcerative colitis (UC) is a chronic inflammatory intestinal disease characterized by an imbalance in immune function and the overexpression of inflammatory cytokines and mediators. Vitamin B2, also known as riboflavin (Libof), is an essential water-soluble vitamin with numerous beneficial properties, including antioxidant, anti-aging, anti-inflammatory, anti-nociceptive, and anti-cancer effects. In this study, we aimed to investigate the protective effects of Libof on dextran sulfate sodium (DSS)-induced experimental colitis. The C57BL/6 mice were used as the in vivo model of chronic colitis to investigate the anti-inflammatory effects of Libof. RAW 264.7 cells were used for the in vitro investigation of the molecular mechanisms underlying these effects. In vivo, Libof alleviated the DSS-induced disease activity index (DAI), colon length shortening, and colonic pathological damage. In vitro, Libof inhibited lipopolysaccharide (LPS)-induced tumor necrosis factor (TNF)-alpha and interleukin (IL)-6 production in RAW 264.7 cells. Moreover, Libof inhibited LPS-induced nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression in RAW 264.7 cells. In conclusion, these findings indicate that Libof shows potential as an agent for the treatment of UC.

Lactic acid bacterial inoculant effects on the vitamin content of alfalfa and Chinese leymus silage

  • Objective: Information regarding the vitamin content of silage is limited. This study investigated the changes in the vitamin content of alfalfa and Chinese leymus silages with or without a lactic acid bacterial inoculant. Methods: Alfalfa at the early flowering stage and Chinese leymus at the full-bloom stage were harvested. The treatments for each forage type were control (deionized water only) and $1{\times}10^6$ colony-forming units Lactobacillus plantarum (LP)/g fresh matter. After 45 days of ensiling, all silages were sampled for evaluating the vitamin content, fermentation quality and chemical composition. Results: The LP inoculant decreased the pH value and ammonia nitrogen content of the alfalfa and Chinese leymus silages and significantly (p<0.05) increased the lactic acid, acetic acid concentrations and Flieg's points. Prior to ensiling, the levels of five B-group vitamins (thiamin, riboflavin, niacin, pantothenic acid, and pyridoxine) and ${\alpha}$-tocopherol in alfalfa were significantly (p<0.01) higher than those in Chinese leymus. Ensiling decreased the levels of the five B-group vitamins in both alfalfa and Chinese leymus while increasing the ${\alpha}$-tocopherol content of Chinese leymus. The thiamin, riboflavin, niacin and pantothenic acid levels in the LP-treated silage were significantly (p<0.05) lower than those in the untreated silage for the alfalfa and Chinese leymus. The ${\alpha}$-tocopherol content in the LP-treated alfalfa silage was significantly (p<0.05) higher than that in the untreated alfalfa silage. There was no significant (p>0.05) difference in pyridoxine content between the untreated and LP-treated silages for both forages. Conclusion: With or without LP inoculation, the levels of the five B-group vitamins (thiamin, riboflavin, niacin, pantothenic acid, and pyridoxine) in alfalfa and Chinese leymus decreased after 45 days of ensiling, while the ${\alpha}$-tocopherol content of Chinese leymus increased. The LP inoculant improved the fermentation quality of both the alfalfa and Chinese leymus silages but increased the thiamin, riboflavin, niacin, and pantothenic acid loss in the two forages after fermentation.

Cultural Conditions of Lactococcus sp. 1112-1 for Production of Bacteriocin-like Substance (Bacteriocin을 생산하는 Lactococcus sp. 1112-1의 배양조건)

  • Cultural conditions of Lactococcus sp. 1112-1, a bacteriocin producing strain, were studied for enhancing its production with regard to environmental and nutritional factors. Optimal compositions of culture medium for bacteriocin production were glucose 20 g/l as carbon source, casein acid hydrolyzate 15 g/l as nitrogen source, and sodium acetate 3 g/l, ammonium citrate 2 g/l as morganic salt with other basal components. The optimal pH of medium and fermentation temperature were 6.2 and $35^{\circ}C$, respectively. This strain required exclusively riboflavin and pantothenic acid for growth and bacteriocin production. In a 1l batch culture, stationary phase emerged after 8.5 hours of fermentation when 1.81 g/l of biomass was accumulated. The maximum antimicrobial activity was 3,894 IU/ml after 12 hours.

  • The optimum concentrations of pH, sucrose and vitamins for the growth and tropane alkaloid production of hairy root clone DTLA9 (best growth line) were investigated. The optimum pH in growth and tropane alkaliod production of DTLA9 clone in SH (Schenk and Hildebrandt, 1972) basal medium without growth regulator were pH 6.3 and 6.5, respectively. Also, the optimum sucrose concentration in growth and tropane alkaliod production in the same medium were 3.0 and 2.8%, respectively. The optimum concentrations of ascorbic acid, D-pantothenate, nicotinic acid, pyridoxine, riboflavin, and thiamine on the growth of DTLA9 clone in SH basal medium without vitamins were 0.1 mM, 0.003 mM, 0.07 mM, 0.002 mM, 0.025 mM, and 0.01 mM, respectively. In particular, supplement of 0.1 mM ascorbic acid to SH basal medium without vitamins stimulated the tropane alkaloid production.
