재조합 인간상피세포 성장인자(rhEGF, DWP401)의 배${\cdot}$태자발달 독성 연구 (Embryo and Fetal Developmental toxicity Study on Recombinant Human Epidermal Growth Factor (rhEGF) in Rats)

  • Effect of recombinant human epidermal growth factor (rhEGF, DWP401) on fetal external, visceral and skeletal malformation during organogenesis was examined. Pregnant Sprauge-Daw ley rats were administered with 0.2, 1 and 5mg/kg/day subcutaneously on gestation day 6 through 16. Dams were sacrified at 20th day of gestation. Materal body weight, food consumption and clinical observation were not changed. Significant dose-dependent increase of relative and absolute liver weight were observed in the treatment group, whereas other organ weights were not changed. Placental weight of 1 and 5mg/kg/day group and number of resorption in 5mg/kg/day treatment group were significantly increased. External and visceral malformation of fetuses were not observed with treatment. However, skeletal variations(increase of asymmetry sternebrae, decrease of dumb-bell and asymmetry sternbrae at 5mg/kg/day, and fused stemebrae at 5mg/kg/day) were observed. These results showed that rhEGF (DWP401) may not have embryo and/or fetal developmental toxicity effect in rats.

  • Multivesicular DepoFoam technology is best suited for the encapsulation and sustained release of water-soluble drugs. The purpose of the present study was to prepare multivesicular DepoFoam particles and investigated possibility of oral delivery of a peptide, human epidermal growth factor (rhEGF). The multivesicular DepoFoam particles containing rhEGF was prepared by a two step water-in-oil-in-water double emulsification process. (omitted)

Effect of Recombinant Human Epidermal Growth Factor Impregnated Chitosan Film on Hemostasis and Healing of Blood Vessels

  • Background Bleeding can be a problem in wound debridement. In search for an effective hemostatic agent, we experimented with a chitosan film combined with the recombinant human epidermal growth factor (rh-EGF), hypothesizing that it would achieve effective hemostasis and simultaneously enhance arterial healing. Methods Forty-eight Sprague-Dawley rats were used, and 96 puncture wounds were made. The wounds were divided into the following four groups: treated with sterile gauze, treated with gelatin sponge, treated with chitosan, and treated with chitosan combined with rh-EGF. Immediate hemostasis was evaluated, and arterial healing was observed histologically. Results Groups B, C, and D showed a significant rate of immediate hemostasis as compared to group A (P<0.05), but there were no significant differences among groups B, C, and D. Histologically, only group D showed good continuity of the vessel wall after 1 week. It was the only group to show smooth muscle cell nuclei of the vessel wall. Conclusions We observed that chitosan has an effective hemostatic potential and the mix of rh-EGF and chitosan does not interfere with chitosan's hemostatic capabilities. We also identified enhanced healing of vessel walls when rh-EGF was added to chitosan. Further research based on these positive findings is needed to evaluate the potential use of this combination on difficult wounds like chronic diabetic ulcerations.

  • Natural human epidermal growth factor (nhEGF) was purified from pregnant human urine by benzoic acid adsorption, DEAE-Sepharose ion exchange, and immunoaffinity chromatography. The purified nhEGF was further separated into four fractions using Bondapak C$_{18}$ HPLC system. Following characterization by Western blot analysis and double immu- nodiffusion, we found that each fraction corresponds to four derivatives of the nhEGF. For biological analysis of nhEGF, we optimized the labeling time and serum concentration for the incorporatioin of 5-bromo-2'-deoxy uridine (BrdU), a non-radioactive alternative for [$^{3}$H]-thymidine uptake, into NIH 3T3 cells. The DNA synthesis of NIH 3T3 cells was gradually increased at the nhEGF concentrations between 0.1 - 10 ng/ml in the Dulbecco's Modified Eagles Medium (DMEM) containing 0.2% Fetal calf serum (FCS). When we assayed the biological activity of four fractions, the activity of the second fraction was superior to that of the others. Based on the results from the HPLC analysis spiked with recombinant human epidermal growth factor (rhEGF) and amino acid sequencing, we concluded that the second fraction was nhEGF and the other three fractions were the derivatives of nhEGF. In addition, the proportion of nhEGF was approximately 46% is compared with that of the other three derivatives.

  • To develop the buccal delivery system of rhEGF for the treatment of buccal mucosal ulcer, polymer films and hydrogels were investigated. Methods: Hydrogels for thermosenstive sol/gel systems were prepared by the cold method (Schmolka, 1972). And mucoadhesive films were prepared by mixing sod. alginate/polycarbophil 974p. To find an optimum buccal mucosal adhesive gel or film, the gel strength of the poloxamer and sod. alginate/polycarbophil 974p hydrogels were determined by the Simple Rheology Method and their mucoadhesiveness were measured by the Instron (M 4400, Instron Co., U.S.A.) method. Optimization of growth inducing factors for colony forming and attachment of bone marrow-derived mesenchymal stem cells regarding bioengineering application

  • PURPOSE. These days, mesenchymal stem cells (MSCs) have received worldwide attention because of their potentiality in tissue engineering for implant dentistry. The purpose of this study was to evaluate various growth inducing factors in media for improvement of acquisition of bone marrow mesenchymal stem cells (BMMSCs) and colony forming unit-fibroblast (CFU-F). MATERIALS AND METHODS. The mouse BMMSCs were freshly obtained from female C3H mouse femur and tibia. The cells seeded at the density of $10^6$/dish in media supplemented with different density of fetal bovine serum (FBS), $1{\alpha}$, 25-dihydroxyvitamin (VD3) and recombinant human epidermal growth factor (rhEGF). After 14 days, CFU-F assay was conducted to analyze the cell attachment and proliferation, and moreover for VD3, the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was additionally conducted. RESULTS. The cell proliferation was increased with the increase of FBS concentration (P<.05). The cell proliferation was highest at the density of 20 ng/mL rhEGF compared with 0 ng/mL and 200 ng/mL rhEGF (P<.05). For VD3, although the colony number was increased with the increase of its concentration, the difference was not statistically significant (P>.05). CONCLUTION. FBS played the main role in cell attachment and growth, and the growth factor like rhEGF played the additional effect. However, VD3 did not have much efficacy compare with the other two factors. Improvement of the conditions could be adopted to acquire more functional MSCs to apply into bony defect around implants easily.

한국인 여드름 환자에서 표피성장인자가 함유된 외용제의 피부 적용에 대한 유효성 및 안전성 평가 (Efficacy and Safety of Topical Application of Epidermal Growth Factor (EGF) for Korean Acne Patient)

  • 여드름은 면포, 구진, 낭종, 결절, 색소 침착 등 다양한 피부병변으로 나타나는 모낭피지선의 만성 염증질환으로 사춘기부터 성인기까지 발생 연령대가 다양해지고 있다. 한편, 약물 부작용으로 여드름이 발생하기도 하는데, 표피성장인자(epidermal grouwth factor, EGF) 수용체 억제제 항암제를 사용할 경우 75 ~ 100%에서 여드름양 모낭염이 발생된다고 보고되고 있다. 여드름의 치료로 항생제, 레티노이드 경구 복용 및 외용 약제 도포 등 다양한 방법이 이용되고 있다. 그러나 최근 들어 레티노이드 기형 유발 가능성 및 Propionibacterium acne의 항생제 내성률 증가는 기존 치료의 한계로 여겨진다. 따라서 본 연구는 최근 여드름양 발진에 효과가 있다고 알려진 EGF를 함유한 외용제가 여드름 치료에 미치는 효과와 안정성을 평가하였다. 한국 성인 10 ~ 29세 23명을 대상으로 EGF 함유 제품(트러블컨트롤 EGF)과 3종 제품(트러블컨트롤 클래리파잉 클렌징폼, 트러블컨트롤 올-클리어 필링토너, 레드롤 카밍 모이스처)을 하루 두 번 사용하도록 하였다. 사용 후 영상 피지량, 경표피수분손실량, 피부 홍조 측정, 전문가 육안 평가, 사용 후 만족도 설문조사를 평가하였다. 최종 측정 시, 피부 피지량, 경피수분손실량, 피부 홍조가 통계학적으로 감소하였으며, 전문가 육안 평가에서 여드름 병변(면포, 구진)도 통계학적으로 감소하였다. 연구동안 심각한 부작용은 관찰되지 않았다. 표피성장인자 함유 외용제는 경도의 여드름에 안전하면서도 효과적으로 사용할 수 있을 것으로 생각된다.
