Streptococcus gordonii, Fusobacterium nucleatum 및 Porphyromonas gingivalis의 상호작용이 성장에 미치는 영향 (The Interactive Effect of These Bacterial Substrates on the Growth of Streptococcus gordonii, Fusobacterium nucleatum and Porphyromonas gingivalis)

  • 치주질환과 관련된 세균인 S. gordonii, F. nucleatum 및 P. gingivalis의 상호작용이 군집 형성에 미치는 영향을 알아보고자 trypticase soy hemin menadione broth에 단독 및 열처리한 사균과 혼합 분주하여 혐기성 균배양조를 통해 $37^{\circ}C$ $CO_2$ 배양기에서 anearobic gas pack 하에 7일간 배양하였다. 군집 형성 정도를 확인하기 위해 흡광도를 측정하였으며, 군집 구조 및 형태를 확인하기 위해 주사전자현미경으로 관찰하였다. P. gingivalis의 병원성에 미치는 영향을 확인하기 위해 real-time RT-PCR를 통해 gingipain인 HRgpA를 생성하는 rgpA 유전자에 대한 발현 분석을 시행하여 다음과 같은 결론을 얻었다. S. gordonii와 P. gingivalis의 군집 형성은 다른 사균들에 의해 증가하였다. F. nucleatum의 경우 P. gingivalis 사균에 의해 증가하는 양상을 보였으나 S. gordonii 사균에 의해서는 군집 형성이 감소되었다. 따라서 본 실험에 사용된 균주는 군집 형성 시 상호작용 인자뿐 아니라 세균 입자 그 자체 등을 통해서도 서로 영향을 주는 것으로 생각된다.

쥐의 초기 난포 발달에 관여하는 Cell Size Growth 및 CCN Family 유전자에 관한 연구 (Characterization of Genes Related to the Cell Size Growth and CCN Family According to the Early Folliculogenesis in the Mouse)

  • Objectives: Previously, we sought to compile a list of genes expressed during early folliculogenesis by using cDNA microarray to investigate follicular gene expression and changes during primordialprimary follicle transition and development of secondary follicles (Yoon et al., 2005). Among those genes, a group of genes related to the cell size growth was characterized during the ovarian development in the present study. Methods: We determined ovarian expression pattern of six genes related to the cell size growth (cyr61, emp1, fhl1, socs2, wig1 and wisp1) and extended into CCN family (${\underline{c}}onnective$ tissue growth factor/${\underline{c}}ysteine$-rich 61/${\underline{n}}ephroblastoma$-overexpressed), ctgf, nov, wisp2, wisp3, including cyr61 and wisp1 genes. Expression of mRNA and protein according to the ovarian developmental stage was evaluated by in situ hybridization, and/or semiquantitative reverse transcriptase polymerase chain reaction (RT-PCR), and immunohistochemistry, respectively. Results: Among 6 genes related to the cell size growth, cyr61 and wisp1 mRNA was detected only in oocytes in the postnatal day5 mouse ovaries. cyr61 mRNA expression was limited to the nucleolus of oocytes, while wisp1 was expressed in the cytoplasm and nucleolus of oocytes, except nucleus. cyr61 mRNA expression, however, was found in granulosa cells from secondary follicles. The rest 4 genes in the cell size growth group were detected in oocytes, granulosa and theca cells. Cyr61 and Wisp1 proteins were expressed in the oocyte cytoplasm from primordial follicle stage. Especially, Cyr61 protein was detected in pre-granulosa cells, Wisp1 protein was not. By using RT-PCR, we evaluated and decided that Cyr61 protein is produced by their own mRNA in pre-granulosa cells that was not detected by in situ hybridization. cyr61 and wisp1 genes are happen to be the CCN family members. The other members of CCN family were also studied, but their expression was detected in oocytes, granulose and theca cells. Conclusions: We firstly characterized the ovarian expression of genes related to the cell size growth and CCN family according to the early folliculogenesis. Cyr61 protein expression in the pre-granulosa cells is profound in meaning. Further functional analysis for cyr61 in early folliculogenesis is under investigation.

Immunosuppression-enhancing effect of the administration of allogeneic canine adipose-derived mesenchymal stem cells (cA-MSCs) compared with autologous cA-MSCs in vitro

  • Background: Recently, mesenchymal stem cells therapy has been performed in dogs, although the outcome is not always favorable. Objectives: To investigate the therapeutic efficacy of mesenchymal stem cells (MSCs) using dog leukocyte antigen (DLA) matching between the donor and recipient in vitro. Methods: Canine adipose-derived MSCs (cA-MSCs) isolated from the subcutaneous tissue of Dog 1 underwent characterization. For major DLA genotyping (DQA1, DQB1, and DRB1), peripheral blood mononuclear cells (PBMCs) from two dogs (Dogs 1 and 2) were analyzed by direct sequencing of polymerase chain reaction (PCR) products. The cA-MSCs were co-cultured at a 1:10 ratio with activated PBMCs (DLA matching or mismatching) for 3 days and analyzed for immunosuppressive (IDO, PTGS2, and PTGES), inflammatory (IL6 and IL10), and apoptotic genes (CASP8, BAX, TP53, and BCL2) by quantitative real-time reverse transcriptase-PCR. Results: cA-MSCs were expressed cell surface markers such as CD90+/44+/29+/45- and differentiated into osteocytes, chondrocytes, and adipocytes in vitro. According to the Immuno Polymorphism Database, DLA genotyping comparisons of Dogs 1 and 2 revealed complete differences in genes DQA1, DQB1, and DRB1. In the co-culturing of cA-MSCs and PBMCs, DLA mismatch between the two cell types induced a significant increase in the expression of immunosuppressive (IDO/PTGS2) and apoptotic (CASP8/BAX) genes. Conclusions: The administration of cA-MSCs matching the recipient DLA type can alleviate the need to regulate excessive immunosuppressive responses associated with genes, such as IDO and PTGES. Furthermore, easy and reliable DLA genotyping technology is required because of the high degree of genetic polymorphisms of DQA1, DQB1, and DRB1 and the low readability of DLA 88.

Exosomal miR-181b-5p Downregulation in Ascites Serves as a Potential Diagnostic Biomarker for Gastric Cancer-associated Malignant Ascites

  • Purpose: Peritoneal carcinomatosis in gastric cancer (GC) patients results in extremely poor prognosis. Malignant ascites samples are the most appropriate biological material to use to evaluate biomarkers for peritoneal carcinomatosis. This study identified exosomal MicroRNAs (miRNAs) differently expressed between benign liver cirrhosis-associated ascites (LC-ascites) and malignant gastric cancer-associated ascites (GC-ascites), and validated their role as diagnostic biomarkers for GC-ascites. Materials and Methods: Total RNA was extracted from exosomes isolated from 165 ascites samples (73 LC-ascites and 92 GC-ascites). Initially, microarrays were used to screen the expression levels of 2,006 miRNAs in the discovery cohort (n=22). Subsequently, quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) analyses were performed to validate the expression levels of selected exosomal miRNAs in the training (n=70) and validation (n=73) cohorts. Furthermore, carcinoembryonic antigen (CEA) levels were determined in ascites samples. Results: The miR-574-3p, miR-181b-5p, miR-4481, and miR-181d were significantly downregulated in the GC-ascites samples compared to the LC-ascites samples, and miR-181b-5p showed the best diagnostic performance for GC-ascites (area under the curve [AUC]=0.798 and 0.846 for the training and validation cohorts, respectively). The diagnostic performance of CEA for GC-ascites was improved by the combined analysis of miR-181b-5p and CEA (AUC=0.981 and 0.946 for the training and validation cohorts, respectively). Conclusions: We identified exosomal miRNAs capable of distinguishing between non-malignant and GC-ascites, showing that the combined use of miR-181b-5p and CEA could improve diagnosis.

국내 자연산 명태(Gadus chalcogrammus) 집단의 바이러스 모니터링 (Monitoring of viruses in wild walleye pollock (Gadus chalcogrammus) population in Korea)

  • 2015년 2월부터 2018년 8월까지 총 1,253 마리의 자연산 명태 (Gadus chalcogrammus)를 강원도 고성 아야진항 근해에서 정치망을 사용하여 포획한 후, 바이러스 (viral hemorrhagic septicemia virus, VHSV; nervous necrosis virus, NNV; marine birnavirus, MABV) 모니터링을 RT-PCR법으로 수행하였다. One-step PCR법으로 비장시료 및 뇌시료에서는 대상 바이러스가 모두 검출되지 않았으며, 일부 시료를 two-step PCR법으로 검사한 결과 VHSV는 19.7% (36/183)의 비장시료에서 검출되었다. 또한, NNV는 4.4% (8/183)의 비장시료, 1.2% (3/259)의 뇌시료에서 검출되었다. 검출된 바이러스의 계통분석 결과, 기존의 국내에서 분리되는 바이러스의 유전형에 각각 속하는 것으로 나타났다 (Genotype IVa, RGNNV genotype). 바이러스의 분리를 시도하지 않아 검출된 바이러스의 활성은 알 수 없지만, 모든 양성 시료가 two-step PCR법으로 검출되었으므로 매우 낮을 것으로 추측된다.

천궁다조산가미방(川芎茶調散加味方) 주정추출물이 산화적 손상 및 염증에 미치는 영향 (Effect of Cheongungdajosan-gamibang on oxidative damage and inflammation)

  • Objectives : The purpose of this study was verification of the anti-inflammation and anti-oxidant effect of Cheongungdajosan-gamibang extract (CG) in mouse macrophage, RAW 264.7 cells. Methods : We have basically using LPS-stimulated RAW 264.7 cells. The cell toxicity was determined by MTT assay. To evaluate the anti-inflammatory effect of Cheongungdajosan-gamibang, amount of nitric oxide(NO) was measured using the NO detection kit and the IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ expression was measured by reverse transcriptase polymerase chain reaction (RT-PCR). Also, free radical scavenging assay has tested for DPPH and ABTS radical activity as well as the contents of total polyphenol. Results : In this study, 96.6% or higher cell viability was observed in all tested groups from 1, 10, $100{\mu}/m{\ell}$ in RAW 264.7 cells. The RAW 264.7 cells were induced by lipopolysaccharide (LPS) and CG 1, 10, $100{\mu}/m{\ell}$. The CG decreased nitric oxide (NO) production activity dose dependently, especially at $100{\mu}/m{\ell}$ of 55%. The production of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ were decreased by 51%, 78% and 35% in CG treated $100{\mu}g/m{\ell}$. CG showed dose-dependent suppression activity of reactive oxygen species (ROS) production, especially at $100{\mu}g/m{\ell}$ of 37%. DPPH radical scavenging activity and ABTS cation decolorization were activated over 86% and 88% in CG at $1,000{\mu}g/m{\ell}$ concentration. Conclusions : According to the results, we thought that CG showed anti-inflammatory and antioxidant activities on the RAW 264.7 cells in mouse macrophage. Therefore, this research is expected to provide the fundamental data about the natural material analysis of relating to the anti-inflammation and antioxidant.

산마늘로부터 단리한 kaempferol과 quercetin의 콜레스테롤 저하 활성 (Cholesterol inhibitory activities of kaempferol and quercetin isolated from Allium victorialis var. platyphyllum)

  • 식용 임산자원인 산채류를 기능성 식품으로 개발하고자 산마늘을 비롯한 총 13종의 에탄올 조추출물에 대한 콜레스테롤 저하활성을 검정하였다. 즉, 콜레스테롤 생합성 과정 초기에 관여하는 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMG-CoA reductase)와 후기에 관여하는 squalene synthase의 효소 활성을 조사한 결과 산마늘 잎 에탄올 추출물이 두 효소의 활성을 공히 70% 이상 저해하여 활성이 가장 우수한 것으로 나타났다. 그리고 이러한 콜레스테롤 저하활성과 관련이 있는 물질을 탐색하고자 산마늘로부터 물질 단리를 시도하여 디클로로메탄 가용부로부터 kaempferol과 quercetin을 단리하였다. 또한 유전자 레벨에서의 콜레스테롤 저하 활성을 조사하기 위해 단리물질과 분획물을 C100세포(햄스터 유래 HMG-CoA reductase 고발현 세포주)에 각각 5 ㎍/㎖과 10 ㎍/㎖로 24시간 처리하여 HMG-CoA reductase와 squalene synthase의 mRNA 발현 정도를 조사하였다. 그 결과 10 ㎍/㎖로 kaempferol과 quercetin을 처리한 경우 두 효소의 mRNA가 전혀 발현하지 않는 것으로 나타나 유전자 레벨에서의 콜레스테롤 생합성 저해 효과를 확인할 수 있었다. 이상의 결과 산마늘 잎 에탄올 추출물은 콜레스테롤 생합성에 관여하는 HMG-CoA reductase와 squalene synthase의 활성을 저해하며 이러한 활성 저해 효과는 kaempferol과 quercetin에 기인하는 것으로 사료되었다. 특히 kaempferol과 quercetin은 여러 식물의 성분으로서 이미 알려진 화합물이지만 콜레스테롤 저하활성이 있는 것으로 밝혀진 것은 이번이 처음으로 금후 이들 물질과 이들 물질을 함유하고 있는 식물 활용에 필요한 자료를 제공하였다고 사료된다.

Noni Fruit Juice의 3T3-L1 지방전구세포 분화 억제 및 인터루킨-1β 유도 Inducible Nitric Oxide Synthase 염증유전자 발현 감소 효과 (Suppressive Effects on Lipid Accumulation and Expression of Interleukin-1β-Mediated Inducible Nitric Oxide Synthase in 3T3-L1 Preadipocytes by a Standardized Commercial Noni Fruit Juice)

  • Objectives: Noni fruit juice (NFJ) is liquor extracted from Morinda citrifolia (noni) fruit and has been used as an herbal remedy in many countries. However, the NFJ's anti-adipogenic and anti-inflammatory effects on adipocytes are poorly understood. The purpose of this study was to explore the commercially standardized NFJ effects on lipid accumulation throughout 3T3-L1 preadipocytes differentiation and interleukin-1β (IL-1β)-mediated inducible nitric oxide synthase (iNOS) expression in 3T3-L1 preadipocytes. Methods: Cellular lipid accumulation and triglyceride (TG) content in differentiating 3T3-L1 preadipocytes were assessed subsequently via the Oil Red O staining and AdipoRed assay. MTS assay was used to examine NFJ cytotoxicity in (differentiating) 3T3-L1 preadipocytes. Immunoblotting and reverse transcriptase polymerase chain reaction analysis were used to measure the expression levels of target protein and mRNA in (differentiating) 3T3-L1 preadipocytes, respectively. Results: NFJ treatment at 150 μL/mL led to a substantial reduction of fat accumulation and TG content during 3T3-L1 adipogenesis with no discernable impact on the cell viability. Of note, while NFJ treatment (150 μL/mL) largely inhibited the CCAAT/enhancer-binding protein-α (C/EBP-α) and peroxisome proliferator-activated receptor-β (PPAR-β) protein expressions, it did not influence PPAR-γ in differentiating 3T3-L1 preadipocytes. Of interest, treatment with IL-1β at 20 ng/mL for 4 hours elicited in firm induction of iNOS mRNA expression in 3T3-L1 preadipocytes. However, NFJ treatment at 100 or 200 μL/mL greatly attenuated the IL-1β-induced iNOS mRNA expression in 3T3-L1 preadipocytes. Conclusions: NFJ has anti-adipogenic and anti-inflammatory effects on (differentiating) 3T3-L1 preadipocytes which are in part intervened via control of the expression of C/EBP-α, PPAR-β, and iNOS.

미세먼지 유발 피부노화에 대한 쌍별귀뚜라미의 예방 효과 (Anti-skinaging effects of Gryllus bimaculatus on ERM-CZ100-exposed human diploid fibroblasts)

  • 국내 미세먼지의 증가는 만성 호흡기 질환과 피부 염증 및 노화를 유발하여 국민 전체의 건강을 위협하는 심각한 문제로 대두되었다. 본 연구는 미세먼지 유발 피부 염증과 노화에 대해 식용곤충인 쌍별귀뚜라미 70% 에탄올 추출물 (AE-GBE)의 미세먼지에 대한 활성산소 소거능, 세포내 β-galactosidase 효소 활성, MMP-1 발현, 콜라겐 생성, 그리고 염증성 반응에 대해 알아보았다. AE-GBE는 HDF 세포에서 ERM-CZ100에 의해 유도될 수 있는 활성산소종, DNA 단편화 수준 및 β-galactosidase 활성을 유의하게 감소시켰다. 또한 IL-1β, IL-6, TNF-α와 같은 전염증성 사이토카인의 생성과 이들 사이토카인에 의해 발현되는 것으로 알려진 COX2 단백질의 발현을 현저히 감소시켰으며, MMP-1을 억제하여 콜라겐 분해를 막았다. 따라서 본 연구결과는 쌍별귀뚜라미 추출물이 미세먼지 유발 피부 염증에 대한 잠재적인 치료 표적이 될 수 있으며 더 나아가 피부 노화를 늦추는 데 긍정적인 효과를 가질 수 있음을 시사한다.

Dietary Diversity during Early Infancy Increases Microbial Diversity and Prevents Egg Allergy in High-Risk Infants

  • We aimed to investigate associations of dietary diversity (DD) with gut microbial diversity and the development of hen's egg allergy (HEA) in infants. We enrolled 68 infants in a high-risk group and 32 infants in a control group based on a family history of allergic diseases. All infants were followed from birth until 12 months of age. We collected infant feeding data, and DD was defined using 3 measures: the World Health Organization definition of minimum DD, food group diversity, and food allergen diversity. Gut microbiome profiles and expression of cytokines were evaluated by bacterial 16S rRNA sequencing and real-time reverse transcriptase-polymerase chain reaction. High DD scores at 3 and 4 months were associated with a lower risk of developing HEA in the high-risk group, but not in the control group. In the high-risk group, high DD scores at 3, 4, and 5 months of age were associated with an increase in Chao1 index at 6 months. We found that the gene expression of IL-4, IL-5, IL-6, and IL-8 were higher among infants who had lower DD scores compared to those who had higher DD scores in high-risk infants. Additionally, high-risk infants with a higher FAD score at 5 months of age showed a reduced gene expression of IL-13. Increasing DD within 6 months of life may increase gut microbial diversity, and thus reduce the development of HEA in infants with a family history of allergic diseases.
