Changes in Fatty Acid Compositions of Sesame Seed Lipid Fractions during Storage (저장중(貯藏中) 참깨의 결합형태별(結合形態別) 지질(脂質)의 지방산(脂肪酸) 조성변화(組成變化))

  • The changes in fatty acid compositions of the each fraction of oil, that is, steryl ester, triglyceride, diglyceride and free fatty acid, were analyzed during the storage of sesame seed for 4, 9, and 15 months at four different storage conditions, that is, storage under light with linen pouch(LA), storage in dark with linen pouch(DA), storage under light with poly ethylene pouch(LS), and storage in dark with poly ethylene pouch(DS). In the fatty acid composition of steryl ester fraction, palmitic and stearic acid were decreased but linoleic acid was increased during the storage in all the storage conditions. The ratio of $C_{18}$ to $C_{16}$ fatty acids in steryl ester from DA and LA was higher than that from DS and LS during storage until 15 months. The palmitic and stearic acid content in triglyceride were decreased until 9 months of storage, then slightly increased until 15 months of storage in all the storage conditions, but reverse tendency was observed in the lineoleic acid content. The ratio of $C_{18}$ to $C_{16}$ fatty acids and unstaturated to saturated fatty acids in DS were slightly higher than these in DA and LA. In the fatty acid composition of free fatty acid, the content of palmitic and stearic acid were decreased until 9 months of storage, then increased until 15 months, but reverse tendency was observed in the oleic and linoleic acid: the ratio of unsaturated to staturated fatty acids in DA and LA was higher than that in DS and LS during the storage for 15 months.

  • The electrical properties of polysilicon thin films implanted with $B_2H_6$ diluted in $H_2$ as dopant source using ion mass doping technique and the effect of radiation damage on the dopant activation behavior were investigated. Comparing the SIMS profiles of boron in polysilicon films with that obtained from computer simulation using TRIM92 the most probable ion species were $B_2H_x\;^+$(x=1, 2, 3‥‥) type molecular ions. As a result of the Implantation of energetic massive ions, a continuous amorphized layer was created in polysilicon films where the fraction of amorphized layer varied with doping time. This amorphization comes from the fact that mass separation of implanting species is not employed in this ion mass doping technique. In the dopant activation behavior, reverse annealing phenomenon appeared in the intermediate annealing temperature range for a severely damaged specimen. The experimental result showed that the off-state current of the p-channel polysilicon thin film transistor is dependent on the degree of radiation damage.

Reduction in Salmonella mutagenicity of mainstream cigarette smoke condensate by cation exchange chromatography

  • Mutagenicity of cigarette smoke is one of the major health concerns related to smoking. Reduction of the components comprising mutagenic activity in cigarette mainstream smoke can be expected to bring about reduced risk of smoking. The purpose of this study is to isolate mutagenic compounds and to investigate the relative contribution to allover mutagenicity of smoke to find clues for the effective elimination of the components. Cigarette smoke condensate (CSC) was obtained from total particulate matter (TPM) of mainstream smoke, and several fractions fractionated from CSC were made by combination of cation exchange chromatograph and reverse-phase chromatography. The mutagenic activity of these fractions was assessed using Salmonella mutagenicity assay with S. typimurium TA98 strain in the presence of metabolic activation system (S-9). The fractions isolated by cation exchange and reverse-phase column showed relatively high mutagenic activity. The basic and hydrophilic fraction 9 showed approximately 33% of mutagenic activity of CSC and its specific activity was 2,459 revertants/mg TPM. These results suggest that hydrophilic cation exchanger and/or other adsorbents possessing similar properties may be used to remove the mutagenic compounds from mainstream smoke.

Genotoxicity Study of Water Extract of Anemarrhena asphodeloides and Phellodendron amurense in Bacterial and Mammalian Cell Systems

  • In order to investigate the safety of a water extract (ADP) of 1 : 1 mixture of Anemarrhena rhizoma and Phellodendron cortex for alleviating benign prostate hyperplasia, genotoxicity studies in bacterial and mammalian cell assay systems, namely, the Ames bacterial reverse mutation and chromosomal aberration assays were performed. As shown by the results of the Ames bacterial reversion assay, ADP in the range of 625-5000 $\mu\textrm{g}$/plate did not induce mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535 and TA 1537 strains in the absence or in the presence of S9 (the microsomal fraction of rat liver homogenate) metabolic activation. The $IC_{50}$ (50% cell growth inhibition concentration) values of ADP for the chromosomal aberration assay were determined; these were 2425 $\mu\textrm{g}$/ml in the absence and 8126 $\mu\textrm{g}$/ml in the presence of S9 metabolic activation in Chinese hamster lung (CHL) fibroblast cell culture. No chromosomal aberration was observed in CHL cells treated with ADP at 2425, 1212.5 and 606.25 $\mu\textrm{g}$/ml in the absence, or at 8126, 4063 and 2031.5 $\mu\textrm{g}$/ml in the presence of S9 metabolic activation. These results show that under the conditions used, ADP does not harmfully affect the bacterial or mammalian cell system at the gene level.

Interpretation of Permeation Characteristics and Membrane Transport Models Through Polyamide Reverse Osmosis Membrane (Polyamide 역삼투막의 투과성능과 막 이동 모델의 해석)

  • In this study, we present a noble study far membrane transport models using chlorine resistance of polyamide RO membranes. Membrane transport mechanism is investigated by the comparison of membrane permeation performance under the continuous and Intermittent operation modes with mixed feed solution containing NaOCl and NaCl. Analysis of permeation performance indicates that solution-diffusion model and preferential adsorption-capillary flow model are relatively efficient according to operation mode. Under the continuous flow state, mass transfer depends on preferential adsorption-capillary flow model rather than solution-diffusion model. On the other hand, it prefers solution-diffusion model to preferential adsorption-capillary flow model under the stationary state. SEM images of NaOCl treated membrane surfaces strongly support these conclusions. These surface images reveal that NaOCl treated membrane in continuous operation mode exhibits ridge and valley structure in some fraction of the surface area, whereas that in intermittent operation mode shows surface degradation entirely.

Characterization of the Membrane-bound Adenosine Triphosphatase from Corn Roots (옥수수 뿌리로부터 분리한 Membrane-bound ATPase의 특성에 관한 연구)

  • The membrane-bound ATPases were separated on sucrose gradient from corn roots and characterized by pH optima, sensitivity to monovalent salt, Km and Vmax. The pH optima for the activity of all the ATPases associated with 13, 000g pellet and 13, 000~80, 000g pellet were 5 and 9, respectively. The ATPases in Fractions B and C of the 13, 000 g pellet were more active at pH 5 than pH 9. While, in the case of Fractions D, E and F, they were reverse. The activities of the ATPase in Fractions A and C of the 13, 000~80, 000 g pellet were greater at pH 5 than pH 9. On the other hand, the ATPases in Fractions B, D, E, and F were more active at pH 9 than pH 5. The optimum concentraction of ATP for the assay was about 3 to 5 mM. The Km's for the membrane-bound ATPases in 13, 000g pellet and in 13, 000~80, 000 g pellet were 0.25 mM. While Vmax values for 13, 000g pellet were from 8.0 to 12.5 $\mu$M Pi/mg protein/hr. according to pH values, those for 13, 000~80, 000 g pellet were from 35.7 to 55.6 $\mu$M Pi/mg protein/hr. Activities of the membrane-bound ATPases in both 13, 000 g pellet and 13, 000~80, 000 g pellet were stimulated with increasing the concentration of $K^+$.

Evaluation of Antioxidant Capacity and Antimutagen Activity of Bulbil Extracts of the Dioscorea japonica Decaisne and Dioscorea batatas Decaisne (단마와 장마 영여자의 항산화능 및 항돌연변이 활성 검정)

  • The bulbil of the $Dioscorea$ $species$ is produced, the amount of which is 2,000 tons annually, but it has been discarded without specific use. In this study the antioxidant and antimutagenicity of bulbil of the $Dioscorea$, which compared to bulbil of Danma($Dioscorea$ $japonica$ Decaisne) and Jangma($Dioscorea$ $batatas$ Decaisne), a major domestic cultivation species. The study was done by extracting bulbil of the Dioscorea methanol and the methanol extracts was re-extracted with chloroform, ethyl acetate, $n$-butanol and water. In methanol extract of Danma and Jangma, polyphenolic compounds contained 2.2 and 3.9 mg/g extract, respectively. The ethyl acetate fraction of Danma and Jangma had higher polyphenolic contents of 33.9 and 39.1 mg/g, whereas water fractions were much lower at 2.4 and 5.8 mg/g. Determination of antioxidant activity showed that the ethylacetate fraction strong DPPH radical scavenging activity and ABTS radical scavenging activity. The inhibitory effects of methanol extracts and chloroform, ethylacetate, $n$-butanol, water fraction from bulbil of Danma and Jangma on the mutagenicity in 1-NP, $AFB_1$, Trp-P-1 were investigated using $S.$ $typhimurium$ TA98. Danma and Jangma cultivars decreased the reverse mutation induced by 1-NP, $AFB_1$, Trp-P-1 in $S.$ $typhimurium$ TA98. The fraction of chloroform and ethylacetate showed strong inhibitory effects, in a dose dependant manner against the mutagenicities induced by 1-NP, $AFB_1$, Trp-P-1 in $S.$ $typhimurium$ TA98.

Isolation and Partial Physicochemical Characterization of Bile Acid-Binding Fraction from Rice Bran Protein Hydrolysates (미강 단백질 가수분해물에서 담즙산 결합 획분의 분리 및 특성구명)

  • Rice bran protein hydrolysates were prepared and some of their physicochemical properties were investigated to utilize rice bran as starting material for functional food ingredient. Rice bran proteins (RBP) were prepared from defatted rice bran by alkaline extraction and isoelectric precipitation. The enzyme for hydrolysis of RBP was selected through measuring relative activity by pH-drop method and comparing the degree of hydrolysis (DH) of hydrolysates. The enzymatic hydrolysates prepared by $Esperase^{\circledR}$ treatment were partitioned into two fractions by ultrafiltration(UF) with a 10 kDa molecular weight cut-off membrane. Each fraction was applied to a cholic acid-conjugated ${\omega}-aminohexyl$ Sepharose 4B column and the bile acid-binding components were obtained by eluting with deoxycholate. Gel permeation chromatography on a Sephadex G-50 column revealed that molecular weight of the bile acid-binding fraction of UF permeate was distributed in ranges of $2\;kDa{\sim}10\;kDa$ and $0.2\;kDa{\sim}0.6\;kDa$. Three peaks (R-1, R-2 and R-3) were obtained by prep-HPLC of bile acid-binding fraction of UF retentate and analyzed for total and free amino acid composition. The results showed that proline content of the bile-acid binding polypeptides and peptides was four times as much as that of rice bran protein and that the peak corresponding to higher average hydrophobicity had a higher free amino acid content. Average hydrophobicity slightly increased with enzymatic hydrolysis.

Comparison of anticancer activities of Korean Red Ginseng-derived fractions

  • Background: Korean Red Ginseng (KRG) is an ethnopharmacological plant that is traditionally used to improve the body's immune functions and ameliorate the symptoms of various diseases. However, the antitumorigenic effects of KRG and its underlying molecular and cellular mechanisms are not fully understood in terms of its individual components. In this study, in vitro and in vivo antitumorigenic activities of KRG were explored in water extract (WE), saponin fraction (SF), and nonsaponin fraction (NSF). Methods: In vitro antitumorigenic activities of WE, SF, and NSF of KRG were investigated in the C6 glioma cell line using cytotoxicity, migration, and proliferation assays. The underlying molecular mechanisms of KRG fractions were determined by examining the signaling cascades of apoptotic cell death by semiquantitative reverse transcriptase polymerase chain reaction and Western blot analysis. The in vivo antitumorigenic activities of WE, SF, and NSF were investigated in a xenograft mouse model. Results: SF induced apoptotic death of C6 glioma cells and suppressed migration and proliferation of C6 glioma cells, whereas WE and NSF neither induced apoptosis nor suppressed migration of C6 glioma cells. SF downregulated the expression of the anti-apoptotic gene B-cell lymphoma-2 (Bcl-2) and upregulated the expression of the pro-apoptotic gene Bcl-2-associated X protein (BAX) in C6 glioma cells but had no effect on the expression of the p53 tumor-suppressor gene. Moreover, SF treatment resulted in activation of caspase-3 as evidenced by increased levels of cleaved caspase-3. Finally, WE, SF, and NSF exhibited in vivo antitumorigenic activities in the xenograft mouse model by suppressing the growth of grafted CT-26 carcinoma cells without decreasing the animal body weight. Conclusion: These results suggest that WE, SF, and NSF of KRG are able to suppress tumor growth via different molecular and cellular mechanisms, including induction of apoptosis and activation of immune cells.

Total Phenolics of Dried Platycodon grandiflorum and Its Effect on Growth of Human Cancer Cell Lines (건조 도라지 추출물 및 분획물의 총 페놀계 화합물 함량 및 인체 암세포 증식 억제효과)

  • We investigated the effects of extracts from dried Platycodon grandiflorum on total phenolic content and growth of cancer cell lines (HT-29 and AGS). Total phenolic contents of acetone/methylene chloride (A+M) extract and methanol (MeOH) fraction were 4.53 and 27.22 TAE mg/100 g, respectively. Among the fractions, n-butanol (n-BuOH) fraction contained the highest phenolic content. Treatments of crude extracts and fractions significantly inhibited the growth of HT-29 and AGS cancer cell lines (p<0.05). Among the fractions, n-BuOH fraction exhibited the highest inhibitory effect and was then sub-fractionated by reverse phase flash column chromatography (rfc). The rfc 1-3 exhibited a higher inhibitory effect on proliferation of both cancer cells. The rfc 1 contained the highest phenol content. Our results showed that n-BuOH fraction possessed a potent inhibitory effect on proliferation of human cancer cells. We suggested that this anticancer activity was partially related to the content of phenolic compounds.
