Effect of Garlic Oil (diallyl disulfide)/ Vitamin A( retinol acetate on Heat Shock Protein Induction in Cadmium Treated Rats. (마늘유(diallyl disulfide)와 비타민 A(retinol acetate)가 카드뮴 투여 랫드에 미치는영향)

  • Garlic occupies a special position among the many foods of vegetable origin because it is the sole food for Koreans during the their lives. And vitamin A has been ingested by forms of food or additives. Cadmium has been described as one of the most dangerous trace elements in the food and environment of man and livestocks. Since the de novo synthesis of stress proteins can be detected early after exposure to some agents, analysis of cadmium-induced changes in gene expression , ie. alterations in patterns of protein synthesis, may be useful to develop as biomarkers of exposure and damage for food hygiene. He acute and chronic combine effects of cadmium (Cd, CdCl2 20mg/kg), garlic oil(Dds: diallyl disulfide 50mg/kg, 3 times a week) and vitamin A(Ra: retinol acetate 50,000 IU/kg, 3 times a week) on Wistar male rats were evaluated concerning cadmium contents, tissues enzyme activity, HSP expression histopathological and electron microscopical examinations. The results of the study are as follows ; 1. Less cadmium was absorbed through the digestive tracts, but the ratio of contents in tissue were not changed by the simultaneous adminstration of diallyl disufide or retinol acetate. 2. ALT(alanine aminotransferase) , AST(aspartate aminotransferase), glucose, BUN (blood urea nitrogen), creatinine, the key indices of the clinical changes in hepatic and renal function were significantly hanged by the cadmium treatment after 1 week in liver, after 4 weeks in kidney. 3. Histopathological changes in cadmium treated rats were appeared at 8 weeks age treatment in kidneys. Homogenous eosinophilic material was accumulated in cortical and collecting tubular lumens at 16 weeks. Degenerated or necrotized tubular cells were observed in cortex and medulla. Degenerated seminiferous tubules and homogeneous eosinophilic material was seen in interstitial tissue of rat treated with cadmium for 16 weeks. Calcium deposits were seen in degenerated seminiferous tubules and the tubules showed severe calcification of rat treated with cadmium for 16 weeks. Electron microscope changes in kidney were observed in rats treated with CdCl2 20 mg/kg. Proximal convoluted tubule cells showed selling of cytoplasm and narrow lumen. Capillary endothelial cells showed cytoplasmic vacuoles and swelling. Degenerated epithelial cells were accumulated in tubular lumen of kidney. 4. Enhanced synthesis of 70 KDa relateve molecular mass proteins were detected in 2 hours after cadmium, exposure, with maximum activity occurring at 8~48 hours. Induction of HSP 70 was evident at proximal tubules and glomeruli in kidney. Testicular cells produced enough HSP to be detected normally. From the above results, it could be concluded that HSP70 induction by the cadmium treatment was a rapid reaction to indicated the exposure of xenobiotics, and retinol acetate reduced the cadmium induced nephrotoxicity.

Effect of Dietary Supplementation of Vitamin A or β-Carotene on Oxidative Damage Induced by Acute Ethanol Administration in Rats (비타민 A와 β-Carotene의 급여가 에탄올의 급성 투여에 의한 흰쥐의 산화적 손상에 미치는 영향)

  • The present study was conducted to investigate the effect of dietary supplementation of vitamin A or $\beta$-carotene on oxidative damage induced by acute ethanol administration. Sprague-Dawley rats were fed on the experimental diets supplemented with retinyl acetate (2.86 mg/kg diet) or $\beta$-carotene (15.2 mg/kg diet) for 5 weeks. After fed the diet, rats were administered 20% ethanol solution (3g/kg B.W.) acutely. Lipid peroxide values in hepatic tissue, hepatic antioxidative enzyme activities and contents of antioxidative nutrient such as vitamins A and E in serum and hepatic tissue were measured. Hepatic level of malondialdehyde decreased in $\beta$-carotene group compared to the control group. However, there was no significant difference between retinal acetate and $\beta$-carotene groups. Superoxide dismutase activity was higher in retinal acetate group than in the control group. Hepatic glutathione-S-transferase activity of retinal acetate and $\beta$-carotene groups significantly decreased as compared with that of control group. The hepatic content of retinol increased in retinal acetate and $\beta$-carotene groups, especially, in retinyl acetate group. But there was no significant difference in serum content of retinol among the groups. Hepatic content of $\alpha$-tocopherol was significantly increased in retinyl acetate and $\beta$-carotene groups. In conclusion, acute ethanol administration might induce lipid peroxidation, and the dietary supplementation of retinyl acetate or $\beta$-carotene improve partly the antioxidative system through activation of superoxide dismutase and retention of hepatic $\alpha$-tocopherol in ethanol-treated rats.

Screening of Antioxidation-related Functional Components in Brans of the Pigmented Rices (유색미 겨에 함유된 항산화 효과 관련 기능성 성분의 검정)

  • Lipid-soluble fractions extracted with methylene chloride and ethyl acetate as well as 70% ethanolic extract were made from 5 cultivars of the pigmented rice brans (DZ 78, Elwee, LK1A-2-12-1-1, LK1-3-6-12-1-1 and Killimhukmi) to determine their antioxidative activities and to measure the contents of phenolic compounds, phytic acid, together with unsaponificated compounds such as retinol, tocopherol and stigmasterol. We also attempted to characterize candidate components in charge of antioxidation and measure their relative contents in each methylene chloride and ethyl acetate fraction by using GC-MS techniques. As the results, we found a 30% higher antioxidative activity with 26-fold greater phenolic compound content of the pigmented rices as compared to those of the normal rice. Especially, in Killimhukmi, the contents of the unsaponificated compounds such as retinol, tocopherol and stigmasterol were also found to be more than 2.3-fold greater than those of normal rice. However, we could not find the overall correlations between antioxidativity of rices and their color values, or unsaponificated compounds contents. Methylene chloride fraction from Killimhukmi and LK1A-2-12-1-1, both showing strong antioxidativity, contained relatively high contents of (Z)-9-hexadecenyl ester-9-octadecanoic acid and 2-Hydroxyl-1-1(hydroxymethyl) ethyl-9-octadecanoic acid, respectively. In ethyl acetate fractions, on the contrary, 1H indene, methyl ester-tetradecanoic acid and methyl ester-hexadecanoic acid were detected as the commonly-occurring substances in the 5 cultivars of pigmented rices, implying a possible role in antioxidative action.

Effect of Dietary Supplementation of Vitamin A and Chronic Consumption of Ethanol on Oxidative Damage and Antioxidant System in Rats (비타민 A 보충 식이 및 에탄올의 만성적 급여가 흰쥐의 체내 산화적 손상과 항산화체계에 미치는 영향)

  • Alcohol is well known agent which can damage the human tissues such as liver via stimulating lipid peroxidation. On the other hand, carotenoids in addition to vitamins A, C and I play important roles in protecting these oxidative damages as well as preventing the production of free radicals. This study was carried out to investigate the effect of dietary vitamin A on lipid peroxidation and antioxidants status in ethanol-treated rats. In the experiment, male Sprague-Dawley rats weighing 160~180 g were given a liquid diet containing 36% of total calories as ethanol for 7 weeks. The pair-fed control rats received an isocaloric amount of diet containing sucrose instead of ethanol on the following day Additionally, the liquid diet contained adequate amount of $\beta$-carotene, retinyl acetate or 13-sis-reinoic acid except vitamin A-deficient diet. The results obtained are as follows. The levels of plasma and hepatic lipid peroxide were increased after chronic ethanol feeding in rats. Retinyl acetate supplementation significantly reduced lipid peroxidation induced by ethanol feeding Glucose 6-phosphatase activity was significantly reduced in rats fed vitamin A-deficient diet with ethanol and alkaline phosphatase activity was significantly induced in rats fed 13-cis-reinoic acid diet with ethanol. Catalase and alcohol dehydrogenase activities did not show a consistent tendency in experiment groups. The hepatic antioxidant enzyme activities did not significantly changed by chronic ethanol feeding groups. The striking decrease in conversion of $\beta$-carotene to retinol was observed in rats fed a $\beta$-carotene diet with ethanol feeding The level of retinol and retinoic acid in plasma and liver was decreased after chronic ethanol administration Based on this result, these data suggest that ethanol feeding enhances oxidative stress especially in those fed a vitamin A-deficient diet, and vitamin A supplementation, especially, retinyl acetate intake can prevent enhanced lipid peroxidation and related damage to some extent.

STUDY ON STABILITY, EFFICACY, AND EFFECT OF A CREAM CONTAINING 5% OF RETINYL PALMITATE

  • Retinlyl Palmitate, the skin normalizerm is useful to promote greater skin elasticity, to diminish lipid peroxidation and skin roughness following UV exposure, and promote a youthfulgeneral skin appearance. In manufacturing creams, Retinyl Palmitate, which is a derivative of retinol, is used since reionol is easily oxidized by heat and light. However, only a small mount of retinol, is used since using a large amount of it may be harmful to its stability. In this study, thermal stability and UV stability of W/O-, W/S-, O/W-, and MLV-type creams containing 5% of retinyl palmitate and 10% of tocopheryl acetate are measured by Chroma Meters, and the content of RP is quantitatively analyzed by HPLC at 25 $^{\circ}C$ and 45 $^{\circ}C$. Also, how RP has been changed by heat, light, etc. is measured by HPLC, and toxicity of the changed substance is studied. Particle size of each type of the cream if measured, cellular renewal is measured by using DHP and Chroma Meters in order to study their efficacy and effect, moisture content is measured by using Corneometer and Tewameter, and how much wrinkles are improved is studied by using Image Analyzer. Development of MLV-type cream containing 5% of RP and 10% of TA, and satisfying conditions for better creams has been successful.

Effect of Vitamin E Supplementation on Plasma Antioxidant Vitamins and Immunity Status of Crossbred Cows

  • Twenty crossbred (HF${\times}$Tharparkar) dry pregnant cows were divided into four equal groups. They were supplemented with 1,000 I.U. $\alpha$-tocopheryl acetate from 0 (group I), 15 (group II), 30 (group III) and 60 (group IV) days before parturition to 1month of lactation. All the cows were kept under similar feeding and management conditions. Blood plasma samples collected on specific days were analyzed for $\alpha$-tocopherol, retinol, total antioxidant activity (FRAP), immunoglobulin and calcium. Plasma $\alpha$-tocopherol concentration at 30 days prepartum averaged 3.5, 4.1, 4.4 and $3.9{\mu}g/ml$ and decreased by 50.0, 41.4, 34.1 and 33.3 percent on the day of parturition in the four respective groups. After calving, plasma vitamin E started to recover earlier in groups II, III and IV as compared to group I. Mean plasma $\alpha$-tocopherol concentration at 21 days postpartum was significantly higher in groups II, III and IV (2.9, 3.5 and $3.1{\mu}g/ml$) compared to group I ($1.9{\mu}g/ml$) cows. Plasma retinol concentration also showed a substantial decrease in all the groups on the day of calving but recovered to its normal value at 3 weeks postpartum. Plasma total antioxidant activity averaged 901, 895, 859 and $875{\mu}mol/l$ in the four respective groups on 30 days prepartum and decreased on the day of calving in all the groups, but the decrease was less in groups III and IV. Plasma immunoglobulin concentration was higher in group IV, followed by groups III, II and I, respectively, showing better immune status of vitamin E supplemented cows due to less oxidative stress. Supplementation of vitamin E resulted in higher plasma calcium concentration. The data showed that vitamin E supplementation should be started at least 30 days prepartum to reduce oxidative stress in periparturient cows.

  • In this study, poly(DL-lactide-co-glycolide) nanoparticles loaded with water-insoluble vitamins such as vitamin A (Retinol) and vitamin E acetate were prepared by the emulsification diffusion method. Polymer solution was prepared by the two water-miscible organic solvent, such as ethanol and acetone. Because of its biocompatible property, polyethyleneglycol-polypropyleneglycol diblock copolymer was used as surfactant and stabilizer. The influence of some preparative variables on the nanoparticle formation and on the loading efficiency of active agents, such as the type and concentration of stabilizing agent, the stirring methods, the water/oil phase ratio and the polymer concentration were investigated in order to control and optimize the process. After preparation of nanoparticles loaded with active agent, particle size and distribution were evaluated by the light scattering particle analyzer. The loading efficiency of active agents was evaluated by the UV-visible spectroscopy. As the results, particle size were 50-200 nm and dispersibility was monodisperse. The optimum loading efficiency of active agents was observed 50-60%. It was found that the appropriate of selections of binary solvent mixtures and polymeric concentrations in both organic and aqueous phases could provide good yield and favorable physical properties of PLGA nanoparticles.

  • Retinyl Palmitate, the skin normalzer, is useful to promote greater skin elasticity, to diminish lipid peroxidation and skin roughness following UV exposure, and promote a youthfull general skin appearance. We knew that the reduction of retinyl palmitate in W/O, W/S, O/W, MLV cream was caused by variable compound factors. Among the retinoids, we chose retinyl palmitate and studied the stability behavior of retinyl palmitate is liposomed. Furthermore, HPLC, CHROMA METERS, LASER SIGHT SCATTERING SYSTEM and FREEZE FRACTURE SCANNING ELECTROM MICROSCOPY was used to analyzing the stability and efficacy of UV and heat.

Effects of Vitamin A on the Antioxidant Systems of the Growing Chicken

  • The present study was conducted to evaluate effects of the increased dietary vitamin A supplementation on the vitamin A, vitamin E and ascorbic acid concentrations in the plasma and liver and activities of some enzymes in the liver of the growing chicken. One hundred and twenty female chickens at 4 weeks of age were divided in 6 equal groups in accordance with their body weight. They were housed in cages and fed on standard wheat-barley-based broiler diet balanced in the major nutrients. Vitamin A was supplemented in the form of retinyl acetate. Control diet was supplemented with 10 IU/g and experimental feeds were supplemented with 50, 100, 500, 1000 and 2000 IU/g. At days 42 and 56 of the development 8 chickens from each group were killed, plasma and liver were collected for vitamin and enzyme analyses. The increased vitamin A supplementation was associated with its increased accumulation in the liver and with a reduction of ${\alpha}-tocopherol$ concentrations in the plasma and liver. The blood plasma was more resistant to vitamin A concentration changes and the retinol level was elevated only when the vitamin A dose exceeded 100 IU/g feed. Ascorbic acid concentration in the liver was elevated when moderately high vitamin A supplementation was used but significantly decreased at the highest vitamin A dose. Similar changes were observed with glycogen concentration in the liver. Activities of hexokinase, glucose-6-phosphatase and lactate dehydrogenase in the chicken liver were also dependent on vitamin A supplementation, decreasing with highest vitamin A doses. Therefore the observations showed that the vitamin A excess compromises antioxidant system of the growing chickens suggesting that prooxidant activity may be responsible for at least part of the toxicity of vitamin A.

Development of Alternative Testing Methods without Hazardous Reagents used in Korean Pharmaceutical Codex (고시의약품 시험에 사용되는 유해시약 대체 시험법 개발)

  • Development of alternative testing methods for the replacement of hazardous reagents with less hazardous ones is strongly enforced because exposure of human and environment to hazardous reagents are restricted and hazardous reagents are gradually prohibited from using in various testing methods. Thus, in this study, we developed 8 monographs from the Korean Pharmaceutical Codex by substituting the use of the hazardous reagents including ICH class 1 such as benzene, chloroform and dioxane to the use of less toxic ones like ICH class 2 or 3 reagents. We also improved their qualification and quantification performance. Among 8 monographs, the 6 newly developed TLC methods for the identification of nifedipine, oxolamine citrate, ketoprofen lysinate, chlorquinaldol, retinol acetate, and riboflavin showed a clear spot of corresponding material without any interference in spite of the replacement with ICH class 2 or 3 reagents. For the quantification of domperidone and trimebutine, HPLC methods were developed for the substitution of UV/VIS spectrometry and titrimetry, respectively. These HPLC methods were validated for the linearity, recovery, reproducibility, and inter-laboratory variations. In conclusion, the newly developed methods could be expected to become valuable tools for revising the Korean Pharmaceutical Codex.
