관개저수지의 적정 홍수조절용량 설정방법 (Optimal Flood Control Volume in the Irrigation Reservoir)

  • Water level of irrigation reservoir during the flood season could be kept to a certain level, so called, flood control level by releasing the flood inflow in advance in order to reduce the peak discharge of next coming flood and the damage of inundation. Concept of restriction intensity of water supply was introduced to evaluate the influence of flood control volume on the irrigation water supply. Restriction intensity can be calculated by multiplying the ratio of restriction to the days of restriction which are obtained from the operation rule curve and daily water level of irrigation reservoir and it has the dimension of % day. The method of restriction intensity was applied to the Yedang irrigation reservoir with the observed data of 30 years to review whether the present flood control volume is reasonable or not, and suggest the optimal flood control volume, if possible.

  • During the last decade enzyme reaction in organic solvent has been studied to show that specificity in buffer is different from that in organic solvent. The specificity of restriction enzyme was effected by various factors such as ionic strength, salt organic solvent and temperature. In this study, restriction enzyme PvuII which is used most frequently in genetic engineering and the substrate was vector pGEM3 whose sequence was already known were used. As a result the recognition sequence site was changed in the presence of organic solvents whose Log P are -1.5∼0. Their specificities were contrast with activities were contrasted. Specificities were not changed in organic solvent easily in inactivating enzyme. We think that the enzyme recognition site was not changed randomly but by preferential order. A recombinant vector which does not contain typical cleavage site CAG↓CTG was cleaved in 20% ethanol solution. This result might show that restriction enzyme could be used to cleave at unusual sites by changing the reaction conditions.

제주마의 mitochondrial DNA 다형(多型)의 분석(分析) (Mitochondrial DNA polymorphism in the Cheju horses)

  • As a result of the detection of mitochondrial DNA(mtDNA) polymorphism to Thoroughbred and Percheron using 14 restriction enzymes, mtDNA polymorphism of Cheju horse observed in the Bam HI and Sac I. Only in both restriction enzymes two types were classified as of A type, which is high expression frequency and B type, which is low expression frequency. In the other 12 restriction enzymes mtDNA polymorphism was not detected. On the basis of this information mtDNA polymorphism of Cheju horse was examined but was not observed the polymorphism and only A type was expressed both Bam HI and Sac I restriction enzymes. Through this study Cheju horse was demonstrated that lower genetic variation was expressed from the detection of mtDNA polymorphism.

  • This paper presents an of a brake pressure restriction valve to a motorcycle anti-lock brake system(ABS). In the conventional anti-lock brake system of automobiles, slip ratio as a control variable is actively controlled, which requires wheel speed sensors, ECU, and a pressure modulator. In the ABS valve that has been developed for use in motorcycles, however, the brake pressure that is close to the wheel locking pressure is preset by simple exercises and then the valve just allows to pass the wheel locking pressure and cutoff the remaining pressure. Simulation studies with a single wheel braking dynamics and lumped chassis model show that the pressure restriction valve has basic ABS functions as well as some robustness properties for the uncertain load and road conditions as well as various initial braking speeds. Field tests also show that the pressure restriction valve avoids the wheel locking effectively.

  • Genetic diversity among 27 isolates of Rhizoctonia solani, which were obtained from diseased crops in Korea and classified into 9 intraspecific groups by anastomosis test and cultural characteristics, was studied by PCR-RFLP. Gene regions of nuclear 17S ribosomal DNA and internal transcribed spacers including 5.8S rDNA of the isolates were amplified with polymerase chain reaction and digested with 12 restriction enzymes. Differences of restriction patterns were not shown among isolates within each intraspecific groups, however, each anastomosis group and culturala type sowed unique restriction fragment length polymorphisms by restriction patterns using HaeIII, Cfr13I and MspI. The results suggest that PCR-FRLP of rDNA using three restriction enzymes could be used to differentiate intraspecific groups of Rhizoctonia solani in Korea.

Dietary Protein Restriction Alters Lipid Metabolism and Insulin Sensitivity in Rats

  • Dietary protein restriction affects lipid metabolism in rats. This study was performed to determine the effect of a low protein diet on hepatic lipid metabolism and insulin sensitivity in growing male rats. Growing rats were fed either a control 20% protein diet or an 8% low protein diet. Feeding a low protein diet for four weeks from 8 weeks of age induced a fatty liver. Expression of acetyl-CoA carboxylase, a key lipogenic enzyme, was increased in rats fed a low protein diet. Feeding a low protein diet decreased very low density lipoprotein (VLDL) secretion without statistical significance. Feeding a low protein diet down-regulated protein expression of microsomal triglyceride transfer protein, an important enzyme of VLDL secretion. Feeding a low protein diet increased serum adiponectin levels. We performed glucose tolerance test (GTT) and insulin tolerance test (ITT). Both GTT and ITT were increased in protein-restricted growing rats. Our results demonstrate that dietary protein restriction increases insulin sensitivity and that this could be due to low-protein diet-mediated metabolic adaptation. In addition, increased adiponectin levels may influences insulin sensitivity. In conclusion, dietary protein restriction induces a fatty liver. Both increased lipogenesis and decreased VLDL secretion has contributed to this metabolic changes. In addition, insulin resistance was not associated with fatty liver induced by protein restriction.

분리 대두단백질 섭취와 염분 제한이 Streptozotocin으로 유도된 당뇨 횐쥐의 혈청 지질 수준 및 신장기능에 미치는 영향 (The Effects of Isolated Soyprotein and Salt Restriction on Serum Lipid and Kidney Function of Streptozotocin-Induced Diabetic Rats)

  • This study was performed to investigate the effects of isolated soyprotein and salt (NaCl) restriction on the serum lipid and the kidney functions of streptozotocin-induced diabetic rats. Sprague-Dawley males of normal and streptozotocin-induced diabetic rats were raised for 6 weeds divided into 4 groups each according to protein sources and salt levels. The sources of protein were isolated soyprotein and casein. Salt levels tested were 0.1% (normal) and 0.01% (low). The results are summarized as fellows: kidney weight, blood glucose, hemoglobinAlc, GFR and urinary protein of diabetic groups were higher than those of normal groups. Isolated soyprotein lowered total lipids, triglycerides, and total cholesterol in serum and plasma angiotensin II concentration as well as alleviated kidney enlargement and GFR in diabetic rats. Salt restriction didn\\`t affect serum lipid level but decreased GFR and increased angiotensin If concentration. In conclusion, isolated soyprotein decreased serum lipids, plasma angiotensin II concentration, sidney enlargement and GFR, while salt restriction increased plasma angiotensin II concentration. The results suggest that isolated soyprotein and salt restriction seem to cause different effects on plasma angiotensin II concentration and that isolated soyprotein might be of value in the prevention of diabetic artherosclerosis and diabetic hypertension.

만성 뇌졸중 환자의 참여제한에 활동과 가족지지가 미치는 영향 (The Effects of Activity and Family Support on the Participation Restriction of Chronic Stroke Patients)

  • The purpose of this study was to identify the factors determining the participation restriction of chronic stroke patients based on international classification of functioning, disability, and health (ICF) model. Sixty-eight stroke patients participated. The participants were assessed participation restriction using the Korean version of London handicap scale (K-LHS), modified Barthel index (K-MBI) to measure activities of daily living, Berg balance scale (K-BBS) to assess balance, and the center for epidemiologic studies depression (K-CES-D) to gauge depression. Also, 3 minutes walking test (3MWT), gait velocity, asymmetric posture, and family support were assessed. A stepwise multiple regression analysis was used to explore the factors determining participation restriction. There were no significant different in the K-LHS and K-MBI results by gender (p>.05). Correlations between the K-LHS and K-MBI (r=-.656), K-BBS (r=-.543), K-CES-D (r=.266), 3MWT (r=-.363), gait velocity (r=.348), and family support (r=-.389) were significant (p<.05). Also, the K-MBI and family support were the factors that determined participation restriction (p<.05) and that 40.2% of the variation in the K-LHS can be explained. Therefore, it is suggested that evaluation and intervention of patient's activity level and extent of family support is necessary to reduce participation restriction of chronic stroke patients.

열량제한(熱量制限) 및 무단백식이(無蛋白食餌)가 흰쥐의 체내대사(體內代謝)에 미치는 영향(影響) (Effects of Protein Depletion and Protein Calorie Restriction on Metabolic and Enzymatic Activities in Adult Rats)

  • Metabolic responses to the protein-free, high-carbohydrate diet and subsequent food restriction on the same diet at the level of 50% and 75% has been studied on the adult albino rats. The energy source was either corn starch or sugar. In experiment I, adult male rats weighing $509{\pm}8g$ were divided into two groups 10 rats each. Rats fed on the stock diet served as a control. Rats of restriction group received a protein free diet until they reduced their weight down to 400g and continue on a protein-calorie restriction diet until they reduced their weight down to 300g. In experiment II, 28 adult male rats and the same numbers of female rats weighing $329{\pm}5g$ and $223{\pm}4g$ respectively were divided into four groups, 7 males and females in each. Rats fed on a stock diet were sacrificed at the point when others started a protein free diet. These were served as the control. The protein free group received a protein free diet ad libitum for 4 weeks. The 50% restriction group and 75% restriction group were fed on a protein free diet coupled with food restriction at levels of 50% and 75% respectively for 3 weeks. In the result of this study: 1. The rate of body weight changes was similar between the males and the females. Feeding protein free diet ad lib. initiated a rapid weight lost of approximately 25% and protein free diet coupled with food restriction showed 37-43% reduction of their initial weight. 2. There was no significant differences in the value of the N concentration in liver, spleen, brain and muscle between controls and experimental groups. 3. Rats fed on protein free diet showed 1/10 value of the control in the nitrogen excretion in urine. However female showed less N excretion than male. 4. Observing blood picture, the effects of protein depletion and calorie restriction were not appeared any remarkable changes. 5. There was no sign of fatty liver which might result from protein depletion and calorie restriction. 6. Following semi-starvation, FAO and HMP-DH total enzyme activity was reduced, but activity per unit weight was relatively stable.

Effects of Water Restriction on the Growth Performance, Carcass Characteristics and Organ Weights of Naked Neck and Ovambo Chickens of Southern Africa

  • In semi-arid areas of Southern Africa, dehydration can compromise the performance and welfare of local chickens, particularly during the growing period when confinement is curtailed and birds are left to scavenge for feed and water. The effect of water restriction on the growth performance was compared in Naked Neck (NNK) and Ovambo (OVB) chickens that are predominant in Southern Africa. A total of 54 eight-wk-old pullets each of NNK and OVB chickens with an initial average weight of $641{\pm}10g/bird$ were randomly assigned to three water intake treatments, each having six birds for 8 wk. The water restriction treatments were ad libitum, 70% of ad libitum and 40% of ad libitum intake. Nine experimental pens with a floor space of $3.3m^2$ per strain were used. The pens were housed in an open-sided house with cement floor deep littered with a 20 cm layer of untreated wood shavings. Feed was provided ad libitum. Average daily water intake (ADWI), BW at 16 weeks of age (FBW), ADG, ADFI, feed conversion ratio (FCR) and water to feed ratios (WFR) were determined. Ovambo chickens had superior (p<0.05) FBW, ADG and ADWI than NNK chickens. Body weight of birds at 16 weeks of age, ADG, ADFI, ADWI, and WFR declined progressively (p<0.05) with increasing severity of water restriction while FCR values increased (p<0.05) as the severity of water restriction increased. Naked Neck chickens had better FCR at the 40% of ad libitum water intake level than Ovambo chickens. The dressing percentage per bird was higher in water restricted birds than those on ad libitum water consumption, irrespective of strain. Heart weight was significantly lower in birds on 40% of ad libitum water intake than those on ad libitum and 70% of ad libitum water intake, respectively. In conclusion, NNK chickens performed better than OVB chickens under conditions of water restriction and would be ideal to raise for meat and egg production in locations where water shortages are a major challenge.
