First Report of Lasiodiplodia theobromae Causing Gummosis on Citrus grandis (L.) Osbeck in Vietnam

  • This study aims to isolate and identify the fungal pathogen responsible for gummosis disease affecting Thanh Tra pomelo in Vietnam. Through molecular identification utilizing primer pairs ITS5 and ITS4, the analysis pinpointed Lasiodiplodia theobromae as the specific fungal pathogen. Notably, the fungal colonies exhibited vigorous growth on potato dextrose agar. Initially, these colonies appeared whitish-grey, transforming into a black-grey hue within 5-7 days at a temperature of 30℃. According to previous reports, Phytophthora spp. was the most common pathogenic genus causing gummosis on Thanh Tra pomelo in Vietnam. To our knowledge, this is the first report on L. theobromae causing gummosis on Thanh Tra pomelo in Vietnam.

A 43 kD Protein Isolated from the Herb Cajanus indicus L Attenuates Sodium Fluoride-induced Hepatic and Renal Disorders in Vivo

  • The herb, Cajanus indicus L, is well known for its hepatoprotective action. A 43 kD protein has been isolated, purified and partially sequenced from the leaves of this herb. A number of in vivo and in vitro studies carried out in our laboratory suggest that this protein might be a major component responsible for the hepatoprotective action of the herb. Our successive studies have been designed to evaluate the potential efficacy of this protein in protecting the hepatic as well as renal tissues from the sodium fluoride (NaF) induced oxidative stress. The experimental groups of mice were exposed to NaF at a dose of 600 ppm through drinking water for one week. This exposure significantly altered the activities of the antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), glutathione reductase (GR) and the cellular metabolites such as reduced glutathione (GSH), oxidized glutathione (GSSG), total thiols, lipid peroxidation end products in liver and kidney compared to the normal mice. Intraperitoneal administration of the protein at a dose of 2 mg/kg body weight for seven days followed by NaF treatment (600 ppm for next seven days) normalized the activities of the hepato-renal antioxidant enzymes, the level of cellular metabolites and lipid peroxidation end products. Post treatment with the protein for four days showed that it could help recovering the damages after NaF administration. Time-course study suggests that the protein could stimulate the recovery of both the organs faster than natural process. Effects of a known antioxidant, vitamin E, and a non-relevant protein, bovine serum albumin (BSA) have been included in the study to validate the experimental data. Combining all, result suggests that NaF could induce severe oxidative stress both in the liver and kidney tissues in mice and the protein possessed the ability to attenuate that hepato-renal toxic effect of NaF probably via its antioxidant activity.

Effects of Starvation on Lipid Metabolism and Gluconeogenesis in Yak

  • This research was conducted to investigate the physiological consequences of undernourished yak. Twelve Maiwa yak ($110.3{\pm}5.85kg$) were randomly divided into two groups (baseline and starvation group). The yak of baseline group were slaughtered at day 0, while the other group of yak were kept in shed without feed but allowed free access to water, salt and free movement for 9 days. Blood samples of the starvation group were collected on day 0, 1, 2, 3, 5, 7, 9 and the starved yak were slaughtered after the final blood sample collection. The liver and muscle glycogen of the starvation group decreased (p<0.01), and the lipid content also decreased while the content of moisture and ash increased (p<0.05) both in Longissimus dorsi and liver compared with the baseline group. The plasma insulin and glucose of the starved yak decreased at first and then kept stable but at a relatively lower level during the following days (p<0.01). On the contrary, the non-esterified fatty acids was increased (p<0.01). Beyond our expectation, the ketone bodies of ${\beta}$-hydroxybutyric acid and acetoacetic acid decreased with prolonged starvation (p<0.01). Furthermore, the mRNA expression of lipogenetic enzyme fatty acid synthase and lipoprotein lipase in subcutaneous adipose tissue of starved yak were down-regulated (p<0.01), whereas the mRNA expression of lipolytic enzyme carnitine palmitoyltransferase-1 and hormone sensitive lipase were up-regulated (p<0.01) after 9 days of starvation. The phosphoenolpyruvate carboxykinase and pyruvate carboxylase, responsible for hepatic gluconeogenesis were up-regulated (p<0.01). It was concluded that yak derive energy by gluconeogenesis promotion and fat storage mobilization during starvation but without ketone body accumulation in the plasma.

Educational Levels and Delays in Start of Treatment for Head and Neck Cancers in North-East India

  • Background: There are various patient and professional factors responsible for the delay in start of treatment (SOT) for head and neck cancers (HNC). Materials and Methods: This retrospective study was conducted on data for HNC patients registered at the hospital cancer registry in North-East India. All cases diagnosed during the period of January 2010 to December 2012 were considered for the present analysis. Educational levels of all patients were clustered into 3 groups; illiterates (unable to read or write), qualified (school or high school level education), and highly qualified (college and above). Results: In the present analysis 1066 (34.6%) patients were illiterates, 1,869 (60.6%) patients were literates and 145 (4.7%) of all patients with HNC were highly qualified. The stage at diagnosis were stage I, seen in 62 (34.6%), stage II in 393 (12.8%), stage III in 1,371 (44.5%) and stage IV in 1,254 (40.7%). The median time (MT) to the SOT from date of attending cancer hospital (DOACH) was, in illiterate group MT was 18 days, whereas in the qualified group of patients it was 15 days and in the highly qualified group was 10 days. Analysis of variance showed there was a significant difference on the mean time for the delay in SOT from DOACH for different educational levels (F=9.923, p=0.000). Conclusions: Educational level is a patient related factor in the delays for the SOT in HNCs in our population.

고추의 생장에 미치는 Azospirillum brasilense 및 Methylobacterium oryzae 접종 효과 (Effect of Inoculation of Azospirillum brasilense and Methylobacterium oryzae on the Growth of Red Pepper Plant)

  • BACKGROUND: Rhizosphere bacteria may improve plant growth and productivity both by supply nutrients and hormonal stimulation. Although many experiments have shown improvements in plant growth with inoculation of bacterial cultures to the rhizosphere, the main obstacle in the applications of plant growth promoting rhizobacteria in a large scale is the inconsistency of the results. We tested the growth promoting effects of Azospirillum and Methylobacterium strains on red pepper plant. METHODS AND RESULTS: Red pepper seedlings were grown for 25 days in a growth media inoculated with A. brasilense CW903 or M. oryzae CBMB20. The seedlings were transplanted and grown for 45 days in pots with soil in a greenhouse, at half the recommended level of fertilizer. Bacterial culture, $4.0{\times}10^9$ for A. brasilense CW903 and $5.8{\times}10^8$ CFU for M. oryzae CBMB20, was applied in root zone soil periodically every 10 days during the experiment. Inoculation of M. oryzae CBMB20 significantly increased the red pepper plant growth in terms of leaf number, height and mass of shoot, or root mass compared to uninoculated control plants. Although beneficial effects of A. brasilense on plant growth of many crops were observed, the growthpromoting effect of A. brasilense CW903 on red pepper plant was not found in this study. CONCLUSION(s): The factors responsible for the irregularities in plant growth promoting of rhizobacteria are difficult to elucidate. Extensive inoculation experiments in the greenhouse and in the field should enable us to define the factors critical to obtain successful application of plant growth promoting rhizobacteria.

공장산 고추장의 저장기간중 이화학적 특성의 변화 (Changes in Physicochemical Properties of Industry-type Kochujang during Storage)

  • 국내에서 생산되고 있는 공장산 고추장의 제조후 경과기간에 따른 품질특성치의 변화를 구명하기 위하여 시제된 공장산 고추장을 $37^{\circ}C$에서 105일간 저장하면서 고추장의 품질변화를 측정한 결과는 다음과 같았다. 환원당은 저장 15일까지는 비교적 큰폭으로 감소한 다음 완만하게 감소하였다. 또한 유리당으로는 glucose, fructose, maltose가 검출되었으며 이중에서는 glucose가 가장 많았다. 유리아미노산 함량은 저장기간이 길어짐에 따라 감소하는 경향이었다. 즉, 저장초기 유리아미노산 총량은 2092.50mg%에서 저장 60일후 22.3% 정도 감소하였으며 90일 후에는 1360.57mg%로 저장초기에 비해 35% 정도가 감소한 것으로 나타났다. 저장기간중 전반적으로 효소활성은 유의적인 변화를 보이지 않았으나, 증성 protease의 활성이 미약하나마 완만하게 증가되는 경향을 보였다. 수분함량 및 수분활성도는 각각 저장기간의 증가에 비례하여 감소하는 추세를 보였으며, 양자간의 상관계수는 r=0.964였다. 저장기간의 경과에 따른 겉보기점도는 증가하였으며, 수분함량보다는 수분 활성도에 의해 더 큰 영향을 받는 것으로 판단되었다.

Methanol이 배양된 흰쥐 해마의 신경세포 및 신경교 세포의 성장에 미치는 영향 (Effect of Methanol on Cultured Neuronal and Glial Cells on Rat Hippocampus)

  • Methanol has been widely used as an industrial solvent and environmental exposure to methanol would be expected to be increasing. In humans, methanol causes metabolic acidosis and damage to ocular system, and can lead to death in severe and untreated case. Clinical symptoms are attributed to accumulation of forrnic acid which is a metabolic product of methanol. In humans and primates, formic acid is accumulated after methanol intake but not in rodents due to the rapid metabolism of methanol. Neverthless, the developmental and reproductive toxicity were reported in rodents. Previous reports showed that perinatal exposure to ethanol produces a variety of damage in human central nervous system by direct neurotoxicity. This suggests that the mechanism of toxic symptoms by methanol in rodents might mimic that of ethanol in human. In the present study I hypothesized that methanol can also induce toxicity in neuronal cells. For the study, primary culture of rat hippocampal neurons and glias were empolyed. Hippocampal cells were prepared from the embryonic day-17 fetuses and maintained up to 7 days. Effect of methanol (10, 100, 500 and 1000 mM) on neurite outgrowth and cell viability was investigated at 0, 18 and 24 hours following methanol treatment. To study the changes in proliferation of glial cells, protein content was measured at 7 days. Neuronal cell viability in culture was not altered during 0-24 hours after methanol treatment. 10 and 100 mM methanol treatment significantly enhanced neurite outgrowth between 18-24 hours. 7-day exposure to 10 or 100 mM methanol significantly increased protein contents but that to 1000 mM methanol decreased in culture. In conclusion, methanol may have a variety of effects on growing and differentiation of neurons and glial cells in hippocampus. Treatment with low concentration of methanol caused that neurite outgrowth was enhanced during 18-24 hours and the numbers of glial cell were increased for 7 days. High concentration of methanol brought about decreased protein contents. At present, the mechanism responsible for the methanol- induced enhancement of neurite outgrowth is not clear. Further studies are required to delineate the mechanism possibly by employing molecular biological techniques.

Penicillin G Potassium 주사액 조제 후 보관방법에 따른 안정성 (The Stability of Penicillin G Potassium Injection after Reconstitution in Various Storage Conditions)

  • The stability of penicillin G potassium injection after reconstitution was evaluated in two different diluents of sodium chloride $0.9\%$ and dextrose $5\%$ in water stored at room temperature or refrigerated condition. The concentrations of penicillin G, stored for 24 hours at room temperature or for 10 days at refrigerated condition, were determined by HPLC. Also the pHs of the reconstituted solutions were monitored. The concentrations and pHs of penicillin G potassium 500,000 U/ml injection after reconstitution gradually decreased in all conditions. Stored at room temperature after reconstitution, a new peak which suspected as degradation products of penicillin G was detected in 5 hours in sodium chloride $0.9\%$, 4 hours in dextrose $5\%$ in water. At refrigerated condition, the new peak was detected in 4 days in both sodium chloride $0.9\%$ and dextrose $5\%$ in water. The degradation products of penicillin G allergy have been thought to be one of the substances responsible for evoking allergic reactions. In conclusion, the penicillin G potassium 500,000 U/ml injection after reconstitution was stable for 4 hours in sodium chloride $0.9\%$ 3 hours in dextrose $5\%$ in water solution at room temperature. At refrigerated condition, both solutions were stable for 3 days after reconstitution.

Multifactor Dimensionality Reduction (MDR) Analysis to Detect Single Nucleotide Polymorphisms Associated with a Carcass Trait in a Hanwoo Population

  • Studies to detect genes responsible for economic traits in farm animals have been performed using parametric linear models. A non-parametric, model-free approach using the 'expanded multifactor-dimensionality reduction (MDR) method' considering high dimensionalities of interaction effects between multiple single nucleotide polymorphisms (SNPs), was applied to identify interaction effects of SNPs responsible for carcass traits in a Hanwoo beef cattle population. Data were obtained from the Hanwoo Improvement Center, National Agricultural Cooperation Federation, Korea, and comprised 299 steers from 16 paternal half-sib proven sires that were delivered in Namwon or Daegwanryong livestock testing stations between spring of 2002 and fall of 2003. For each steer at approximately 722 days of age, the Longssimus dorsi muscle area (LMA) was measured after slaughter. Three functional SNPs (19_1, 18_4, 28_2) near the microsatellite marker ILSTS035 on BTA6, around which the QTL for meat quality were previously detected, were assessed. Application of the expanded MDR method revealed the best model with an interaction effect between the SNPs 19_1 and 28_2, while only one main effect of SNP19_1 was statistically significant for LMA (p<0.01) under a general linear mixed model. Our results suggest that the expanded MDR method better identifies interaction effects between multiple genes that are related to polygenic traits, and that the method is an alternative to the current model choices to find associations of multiple functional SNPs and/or their interaction effects with economic traits in livestock populations.
