Damping-off Disease in Mulberry Seedlings and Its Management

  • During the routine survey, the mortality of mulberry seedlings was noticed due to damping-off disease. The disease recognized by rotting of emerged seedlings near the soil line (just below the soil level) resulting in collapse of the seedlings. Two fungi were isolated from affected samples and identified as Alternaria alternata (Fr.) Keissler and Fusarium solani (Mart.) Sacc. Both the fungi were found to be responsible in causing pre and post emergence damping-off of seedlings in mulberry. For management of the disease, an experiment was conducted using fungicides. These fungicides were applied as seed treatment; soil drenching and foliar spray alone and in combination. Among the different treatments, integration of seed treatment and soil application of Dithane M-45 (Mancozeb 75% WP) + Bavistin (Carbendazim 50% WP) followed by foliar spray of these fungicides (after 35 days of sowing) resulted in better survivability of seedlings (93.3 %) on $90^th$ day and controlled the pre and post emergence damping off by 100 and 89.5%, respectively over the check.

Cutaneous Toxicity of Xylene Application to Rat Skin

  • To investigate the skin toxicity of xylene, xylene (25 mg/$\textrm{cm}^2$) has been sequentially applied to the rat skin for four days. On the light microscopic examination, epithelium was left out with infiltration of inflammatory cells in border with dermis, and formation of new epithelial layer was shown under the inflammatory zone. Application of xylene to the rat skin showed the marked rise of cutaneous xanthine oxidase activity whereas, He activities of oxygen free radical scavenging enzymes, superoxide dismutase and glutathione S-transferase, were significantly declined. Furthermore, the content of cutaneous glutathione was more and less decreased in rat skin applied with xylene. In conclusion, these results suggest that a part of oxygen free radical may be responsible for morphological changes in skin by applying xylene to the rat skin.

  • The gene responsible for dechlorination of 4-chlorobenzoate (4CBA) was cloned in E. coli XL1-Blue from Pseudomonas sp. DJ-12. The cloned cell of E. coli Cjl had the hybrid pBluescript SK(+) plasmid, into which about 9.5 kb genomic DNA fragment of PseudOmonas sp. DJ-12 was inserted. The subclone of pCJlOl was constructed by inserting the 3.4 kb EcoRI-HindIII fragment of pCJl into the vector. Those cloned cells could be simply selected by halo formation around the colonies which was the precipitate of AgCl produced by reaction of AgNO$_{3}$ and chloride ion liberated by bacterial dechlorination of 4CBA- Such a plate assay method was standardized by the procedure that the colonies grown for 2 days on the Cl$^{-}$-free plate medium containing 1 mM 4CBA were flooded with 0.1 M AgNO$_{3}$ solution.

Apoptotic Cell Death Following Traumatic Injury to the Central Nervous System

  • Apoptotic cell death is a fundamental and highly regulated biological process in which a cell is instructed to actively participate in its own demise. This process of cellular suicide is activated by developmental and environmental cues and normally plays an essential role in eliminating superfluous, damaged, and senescent cells of many tissue types. In recent years, a number of experimental studies have provided evidence of widespread neuronal and glial apoptosis following injury to the central nervous system (CNS). These studies indicate that injury-induced apoptosis can be detected from hours to days following injury and may contribute to neurological dysfunction. Given these findings, understanding the biochemical signaling events controlling apoptosis is a first step towards developing therapeutic agents that target this cell death process. This review will focus on molecular cell death pathways that are responsible for generating the apoptotic phenotype. It will also summarize what is currently known about the apoptotic signals that are activated in the injured CNS, and what potential strategies might be pursued to reduce this cell death process as a means to promote functional recovery.

Acid-base Balance and Metabolic Acidosis in Neonates (신생아의 산-염기 균형과 대사성 산증)

  • Metabolic acidosis is commonly encountered issues in the management of critically ill neonates and especially of preterm infants during early neonatal days. In extremely premature infants, low glomerular filtration rate and immaturity of renal tubules to produce new bicarbonate causes renal bicarbonate loss. Higher intake of amino acids, relatively greater contribution of protein to the energy metabolism and mineralization process in growing bones are also responsible for higher acid load in premature infant than in adult. Despite widespread use of sodium bicarbonate in the management of severe metabolic acidosis, use of sodium bicarbonate in premature infants should be restricted to a reasonable but unproven exception such as ongoing renal loss. Despite concern about the low pH value (<7.2) which can compromise cellular metabolic function, no treatment guideline has been established regarding the management of metabolic acidosis in premature infants. Appropriately powered randomized controlled trials of base therapy to treat metabolic acidosis in critically ill newborn infants are demanding.

Postinfarction Left Ventricular Free Wall Rupture (급성 심근경색후 좌심실벽 파열 -2례보고-)

  • Left ventricular free wall rupture following acute myocardial infarction (AMI) is the second most common cause of death and has been reported to be responsible for 4 to 24% of all infarction deaths. The rupture occurs anywhere from a few hours to several days after AMI. The common findings of ventricular rupture are persistent chest pain bradycardia and shock. This may be often mistaken for the ruptured dissection of the ascending aorta. The different points from dissection are 1) persistent chest pain 2)persistent ST segment elevation and 3) only intramural hematoma in ascending aorta. We have sucessfully managed two patients with postinfarction myocardial rupture. Surgical management consisted of infarctectomy repairi of the ventricular rupture and coronay artery bypass grafting. We conclude that successful surgical management of ventricular free wall rupture should require prompt diagnosis and emergency operation.

Effect of Cyclohexane Application to Rat Skin on the Skin Toxicity (흰쥐의 피부조직에 있어서 Cyclohexane의 독성)

  • To evaluate the skin toxicity of topical cyclohexane application (25mg/$\textrm{cm}^2$) was sequentially applied to the rat skin for four days. On the histopathological findings in the light micrographs, neutrophils and engulfed neutrophils are seen, and many cytoplasmic processes were appeared in proliferated layer whereas in the dermis area, increased numbers of fibroblast, accumulation of neutrophil and lipid droplets are demonstrated. On the other hand, applying the cyclohexane to the rat skin led to the remarkable rise of cutaneous xanthine oxidase activity and similar activities of superoxide dismutase and glutathione peroxidase and glutathione content and declined activity of glutathione S-transferase compared with control group. Especially the remarkably decreased activity of aniline hydroxylase (AH) was appeared in skin as little as scarcely determined. Furthermore, the applying the cyclohexane to skin led to the significantly increased activity of hepatic AH and alcohol dehydrogenase. These results indicate that oxygen free radical and intermediate metabolite of cyclohexane may be responsible for structural changes in skin by cyclohexane application to rat skin.

  • This study was undertaken to investigate to know whether spleen and lymph node cells from immunized mice can transfer systemically the delayed-type hypersensitivity(DTH) reaction to Salmonella typhimurium and to characterize the lymphoid cells using glass, nylon wool and rabbit anti-mouse thymocyte serum. Mice, C57BL/6 or ICR, were immunized subcutaneously at 11, 8 and 2 days before adoptive systemic transfer with $100{\mu}g$ of protein antigen from S. typhimurium in complete Freund adjuvant. It was found that DTH reaction to S. typhimurium could be transferred to normal recipient systemically by both spleen and lymph node cells($10^8\;cells$, respectively) from immunized mice. The cells responsible for this transfer of DTH reaction were glass nonadherent T lymphocytes.

Clinical Study of Neonatal Gastric Perforation (신생아 위 천공의 임상적 고찰)

  • Gastric perforation of the newborn is a rare and life threatening problem. The pathogenesis of gastric perforation is not clear. Since ischemia is responsible for intestinal perforation, a similar mechanism may result in gastric perforation. Twelve patients with neonatal gastric perforation who were treated at the Department of Pediatric Surgery, Hanyang University Hospital from 1987 to 2002 were reviewed. Eight patients were male and four female. The age of perforation was 1 day to 8 days of life. Ten patients were operated upon and 2 patients were treated nonoperatively. The perforation site was located on the anterior wall along the greater curvature of the stomach in 8 patients and along the lessor curvature of the stomach in 2. The precipitating factors were prematurity, gastroschisis, mechanical ventilation, intestinal obstruction, cyanotic heart disease and indomethacine medication. In 5 cases the cause of perforation was not identified. The mortality rate was 25 % (3 of 12). Earlier recognition and treatment were thought to be crucial prognostic factors.

Assessment of the effect of sulfate attack on cement stabilized montmorillonite

  • In this study, aiming to investigate the effects of sulfate attack on cement stabilized highly plastic clay; an experimental study was carried out considering the effects of cement type, sulfate type and its concentration, cement content and curing period. Unconfined compressive strength and chloride-ion penetration tests were performed to obtain strength and permeability characteristics of specimens cured under different conditions. Test results were evaluated along with microstructural investigations including SEM and EDS analyses. Results revealed that use of sulfate resistance cement instead of normal portland cement is more plausible for soils under the threat of sulfate attack. Besides, it was verified that sulfate concentration is responsible for strength loss and permeability increase in cement stabilized montmorillonite. Finally, empirical equations were proposed to estimate the unconfined compressive strength of cement stabilized montmorillonite, which was exposed to sulfate attack for 28 days.
