[ $^{14}CO_2$ ] Assimilation and Metabolism of $^{14}C-$Assimilates in Whole Plants of Spring Barley In Relation to Adult-Plant Resistance to Powdery Mildew (흰가루병에 대해 성체식물 저항성을 지닌 봄보리에서 $^{14}CO_2$ 동화와 $^{14}C-$동화산물의 대사)

  • The effect of powdery mildew infection on the $^{14}CO_2$ assimilation and metabolism of $^{14}C-$assimilates was studied with spring barley cultivars, susceptible Peruvian and adult-plant resistant Asse at the four-leaf stage. No consistent differences between Peruvian and Asse were revealed in $^{14}CO_2$ assimilation and metabolism of $^{14}C-$assimilates in healthy whole plants. In the two cultivars, $^{14}CO_2$ assimilation and translocation of assimilates decreased as the number of infected leaves increased. Despite the same infection intensity, $^{14}CO_2$ assimilation was less inhibited in Asse than Peruvian. Infection reduced the fixation of $^{14}CO_2$ in noninfected fourth leaves of Peruvian more severely than that of Asse. Infection of the lower 3 leaves also inhibited the incorporation of 14 C into carbohydrates such as fructose and glucose in noninfected fourth leaves and their translocation into leaf sheathes, the inhibitions being greater in Peruvian than Asse. In the infected third leaves, there was a reduction of 14 C-activity in carbohydrates, more $^{14}C-$labeled fructose and glucose being retained in Peruvian. The stimulation of $^{14}C-$organic acid synthesis in all plant organs was more pronounced in Peruvian than Asse. Powdery mildew markedly increased the incorporation of $^{14}C$ into amino acids in infected third and noninfected fourth leaves, but reduced their translocation to the leaf sheathes. A greater rise of $^{14}C-$ activity in some amino acids in the two leaves was found in Peruvian than Asse.

  • Ethanol-soluble amino acids in healthy and powdery mildew-infected leaves of the susceptible cultivar Peruvian and the adult-plant-resistant cultivar Asse of spring barley were quantitatively analysed. At I day after inoculation, the levels of amino acids in the infected first leaves of the two cultivars were similar to those of comparable healthy controls. During sporulation, increases in amino acids were more pronounced in Peruvian than those in Asse. The changes in amino acid content in the infected first and fifth leaves were closely related to the number of colonies per leaf. The susceptible cultivar Peruvian showed higher amounts of amino acids in infected first and fifth leaves at all infection intensitives than did Asse.

Breeding of Black Tomato 'Hei' for Protected Cultivation (시설재배용 흑색토마토 신품종 '헤이' 육성)

  • 'Hei' is a new cultivar of red with black flesh; originated from the cross of two inbred lines, mother plant, TKUI separated from 'Kame' in 2007 and father plant, TLB separated from tomato collected in Europe by pedigree breeding method. 'Hei' was tested for specific character and productivity of lines of tomatoes in spring for 2 years, 2008 to 2009. This variety had resistance to powdery mildew. 'Hei' is an indeterminate type. The fruit shape is round and the skin color is blackish red. The average fruit weight of 'Hei' is about 127.9 g and marketable yield is $5,715kg{\cdot}10a^{-1}$. Lycopene contents of 'Hei' are about $18.5mg{\cdot}100g^{-1}$, high compared with other tomato ($3.1mg{\cdot}100g^{-1}$). 'Hei' is suitable for environment-friendly protected cultivation because of resistance to powdery mildew.

Selection of Desirable Cultivar for Organic Cultivation of Carrot (당근 유기재배를 위한 병해충 저항성 품종 선발)

  • This research was carried out to selection of desirable cultivar for organic cultivation of carrot in Korea. The assay of disease resistance, insect tolerance and rate of bolting were investigated on 32 cultivars of genetic resources of carrot. This experiment was carried out at experimental field and greenhouse of Gangneung-Wonju National University. 'PI 223360' was resistant against powdery mildew. Ten cultivars including 'Oxheart Carrot Heirloom' were moderately resistant, 22 cultivars including 'Long Impeator #58' were susceptible to powdery mildew. Although there was no resistant cultivar against black rot and leaf blight, our results showed that several cultivars have moderate resistance. Thirteen cultivars including 'Oxheart Carrot Heirloom' were having insect (Erythroplusia pyropia) tolerance and 'SA 102' and 'Scarlet Keeper Carrot Rare' were susceptible. According to the investigation of bolting, 6 cultivars including 'Hongsim Ouchon Carrot' were early bolting and 4 cultivars including 'Japanese Imperial Long Carrot' were moderate bolting. Other cultivars were identified as late bolting. From above results, we confirmed that 'Oxheart Carrot Heirloom' was suitable cultivar for organic cultivation among 32 genetic resources of carrot.

A New Cultivar of Gypsophila paniculata with Powdery Mildew Resistance and Double Florets of Pink Color, 'Dream Pink' (흰가루병에 강한 분홍색 절화용 안개꽃 '드림핑크' 육성)

  • 'Dream Pink', a cultivar of Gypsophila paniculata was newly developed for cut flowers by Jeollabuk-do Agricultural Research & Extension Services. It has morphological characteristics of the intermediate plant type, double florets with many petal and two pistils, emarginate petal tip and campanulate calyx shape. Also 'Dream Pink' had several other traits such as strong flower stalk, deep pink flower color, resistance to powdery mildew and pest injury. Furthermore, 'Dream Pink' produced higher number of nodes, primary branches, longer flower stalk length with long intermodal length. In subalpine area, 'Dream Pink' cultivar showed narrow branching angle as compare to 'My Pink', during the summer cultivation. Blooming in 'Dream Pink' was delayed by 5 days, its florets were broader with more number of petals as compare to 'My Pink'.

  • Starch contents in healthy and powdery mildew-infected spring barley leaves at first and fifth leaf stages were measured at different intervals after inoculation. Different patterns of starch accumulation in the susceptible cultivar Peruvian and the adult plant resistant cultivar Asse were found in infected first and fifth leaves. At the early phase of infection, the amount of starch in infected first leaves was slightly changed in both cultivars. During colony development and sporulation, the decrease in starch content was .more marked in the susceptible Peruvian, whereas the amounts of starch in the adult plant resistant Asse increased during colony development and then decreased sharply before and after sporulation. In heavily infected first leaves, the susceptible cultivar Peruvian showed a drastic decrease in starch content. In the adult plant resistant cultivar Asse, the higher amount of starch retained in these infected leaves, regardless of infection intensity. The regulation of starch accumulation in mildewed barley plants at different leaf stages was discussed in relation to adult plant resistance of spring barley.

A New Rose Variety, "Waltz" with Resistance to the Powdery Mildew (흰가루병에 강한 장미 신품종 "왈츠" 육성)

  • A new rose variety, was selected from the progenies of a cross between "Cosai" and "Tokimeky" in 2006 after evaluation trials for three years (2004-2006). "Waltz" was developed because of many petals and powdery mildew resistance. A standard type with large sized flower, "Waltz" shows in 11.0 cm flower diameter. "Waltz" produces 130 stems?m-2 in a year. It has red-white color flower and 49.6 petals per flower. The stem length of cut flower was long with 69.3 cm. Vase life of the variety could be as long as 9.6 days.

  • Systemic effect of two plant growth-promoting rhizobacterial (PGPR) strains, viz., Pseudomonas fluorescens (Pf4) and P. aeruginosa (Pag), was evaluated on pea (Pisum sativum) against the powdery mildew pathogen Erysiphe pisi. Foliar spray of the two PGPR strains was done on specific nodal leaves of pea and conidial germination of E. pisi was observed on other nodal leaves, distal to the treated ones. Conidial germination was reduced on distant leaves and at the same time, specific as well as total phenolic compounds increased in the leaves distal to those applied with PGPR strains, thereby indicating a positive correlation. The strains induced accumulation of phenolic compounds in pea leaves and the amount increased when such leaves were get inoculated with E. pisi conidia. Between the two strains, Pag was found to be more effective than Pf4 as its effect was more persistent in pea leaves. Foliar application of PGPR strains for the control of powdery mildew of pea is demonstrated in vitro while correlating it with the increased accumulation of plant phenolics.

Also 'Double Shine' has several traits such as strong flower stalk, bright colored flower, narrow branching angle and more or less resistance to powdery mildew and pest injury. Furthermore, 'Double Shine' produced higher primary branches, and longer flower stalk length with internodal length and than did a check variety 'Bristol Fairy' during summer cultivation in subalpine areas. Blooming of 'Double Shine' was delayed by 16 days, its floret had broader width, compared to 'Bristol Fairy'. Moreover, 'Double Shine' produced normal flowers with two pistils and 8.5 stamens, with little production of malformed flowers in the high temperature condition.

Examination of Correlations Between Several Biochemical Components and Powdery Mildew Resistance of Flax Cultivars

  • A field trial was conducted in 2009/2010 and 2010/2011 growing seasons at Giza Agricultural Research Station to examine correlations between some biochemical componets and powdery mildews ($PM_s$) resistance in flax cultivars. Nine flax cultivars could be divided into five distinct groups, i.e., highly susceptible (Cortland and C.I. 2008), moderately susceptible (Giza 7, and Marshall), moderately resistant (Cass), resistant (Koto, Dakota and Wilden), and highly resistant (Ottowa 770B). The cultivars showed considerable variation in PM severity ranged from 8.05 on Ottowa 770B to 97.02% on Cortland. Total soluble proteins, total phenols, antioxidant enzymes (peroxidase and polyphenoloxidase), ascorbic acid, tocopherol, and malondialdehyde (MDA), were determined in uninfected leaves of the tested cultivars. Pearson's correlation coefficient was calculated to measure the degree of association between PM severity and each component. All components showed significant (P < 0.05) or highly significant (P < 0.01) negative correlation with PM severity except MDA, which showed positive correlation (P < 0.01). Linear regression analysis was used to evaluate the causal relationship between the biochemical components (independent variables) and PM severity (dependent variable). Coefficient of determination ($R^2$) values of the generated models ranged from 48.76 to 77.15%. Tocopherol, MDA, and proteins were the most important contributors to the total variation in PM severity as the $R^2$ values of their models were 71.78, 75.28, and 77.15%, respectively. The results of the present study suggest that tocopherol, MDA, and proteins in uninfected leaves can be used as biochemical markers to predict PM resistance in flax.
