Hypericum source of natural antimicrobials

  • Plants of the genus Hypericum (Family - Hypericaceae) are herbs, shrubs or small trees and are distributed chiefly in the temperate regions of the world. About 400 different species of Hypericum are available throughout the globe and 20 species occur in India, including a few cultivated in gardens. Almost all plants of the genus Hypericum are widely used in folk medicine. Several potent phytoconstituents from different Hypericum species have led to the isolation of antibacterial, antifungal and cytotoxic compounds. With the development of resistance and cross resistance with different microorganisms and the evolution of so many deadly diseases the screening and evaluation of the phytoconstituents so much so the development of varied phytoconstituents for the drug development for these deadly diseases is utmost essential in every aspects. The present review on the antimicrobial use of different Hypericum reports the findings from and extensive literature search on the Hypericum species around the globe that have been assessed for antimicrobial and antiviral activity. An attempt has been made through this review to summarize the information in this aspect in order to highlight the promising species of this genus which are worthy for further investigation as leads for drug development. Over 31 different Hypericum species have been reported to possess such activities with their varied number of phytoconstituents. Sixteen different constituents of six different classes of phytoconstituents have been reported to be present in different varieties of Hypericum, which may be considered responsible for this activity.

Characteristics of Transmissible CTX-M- and CMY-Type β-Lactamase-Producing Escherichia coli Isolates Collected from Pig and Chicken Farms in South Korea

  • The rapid dissemination of extended-spectrum ${\beta}$-lactamase (ESBL)-producing Escherichia coli has significantly contributed to public health hazard globally. A total of 281 E. coli strains recovered from pigs and chickens between 2009 and 2015 in South Korea were analyzed for ESBL production. ESBL phenotypes were recognized in 14 E. coli isolates; ten and three ESBL-producing isolates carried only $bla_{CTX-M}$ and $bla_{CMY}$ genes, respectively, and one isolate harbored both genes. The predominant CTX-M and CMY types were CTX-M-15 (n = 8) and CMY-2 (n = 3). We also detected ESBL-producing isolates harboring $bla_{CTX-M-65}$, $bla_{CTX-M-14}$, $bla_{CMY-6}$, $bla_{DHA-1}$, and $bla_{TEM-1}$ genes. All ESBL-producing isolates showed resistance to the extent of the fourth generation cephalosporins, along with multidrug resistance. CTX-M-15-producing isolates showed higher MIC values than CTX-M-14- and CTX-M-65-producing isolates. The $bla_{CTX-M}$ and $bla_{CMY}$ genes have the potential to be transferable. The spreading of $bla_{CMY}$ and $bla_{CTX-M}$ genes was arbitrated mainly via Frep and IncI1 plasmids. Our isolates showed clonal diversity in PFGE analysis. This is the first report of E. coli isolates carrying $bla_{CMY-6}$ in chicken from South Korea. The emergence of CMY-6 ESBLs in a population of poultry suggests that extensive screening with long-term surveillance is necessary to prevent the dissemination of ESBL from chicken to human.

Pathotype Classification of Plasmodiophora brassicae Isolates Using Clubroot-Resistant Cultivars of Chinese Cabbage

  • Clubroot disease caused by Plasmodiophora brassicae is one of the most serious diseases in Brassica crops worldwide. In this study, the pathotypes of 12 Korean P. brassicae field isolates were determined using various Chinese cabbage including 22 commercial cultivars from Korea, China, and Japan, and 15 inbred lines. All P. brassicae isolates exhibited the typical clubroot disease on non-clubroot resistant cultivar, indicating that the isolates were highly pathogenic. According to the reactions on the Williams' hosts, the 12 field isolates were initially classified into five races. However, when these isolates were inoculated onto clubroot-resistant (CR) cultivars of Chinese cabbage, several isolates led to different disease responses even though the isolates have been assigned to the same race by the Williams' host responses. Based on the pathogenicity results, the 12 field isolates were reclassified into four different groups: pathotype 1 (GN1, GN2, GS, JS, and HS), 2 (DJ and KS), 3 (HN1, PC, and YC), and 4 (HN2 and SS). In addition, the CR cultivars from Korea, China, and Japan exhibited distinguishable disease responses to the P. brassicae isolates, suggesting that the 22 cultivars used in this study, including the non-CR cultivars, are classified into four different host groups based on their disease resistance. Combining these findings, the four differential hosts of Chinese cabbage and four pathotype groups of P. brassicae might provide an efficient screening system for resistant cultivars and a new foundation of breeding strategies for CR Chinese cabbage.

다제내성 아시네토박터 바우마니의 에센셜 오일에 대한 항균효과 (Antimicrobial Effects of Essential Oils for Multidrug-Resistant Acinetobacter baumanii)

  • Acinetobacter baumannii는 광범위한 항생제에 대한 저항성으로 인해 감염된 환자의 사망률이 높아지는 적색 경보 병원체로 분류됩니다. 이 연구에서 다제 내성 A. baumannii(MRAB)의 18가지 임상 분리 균주에 대해 일부 에센셜 오일(티트리, 로즈마리, 라벤더 오일)의 항균 활성을 평가하고자 하였다. Carbapenemase 선별을 위한 Hodge 시험법은 A. baumannii의 20 가지 균주가 모두 imipenem에 내성이 있음을 보여주었습니다. 다제 내성 미생물의 확인은 VITEK 시스템을 통해 수행하였다. 에센셜 오일의 항균 활성은 MRAB에 대한 디스크 확산 방법으로 평가하였다. 디스크 확산 방법에서 tee tree는 라벤더 오일에 비해 억제 크기가 가장 크게 증가했으며, 로즈마리는 항균 효과가 없었다. 티 트리 오일은 가장 일반적인 인간 병원균 및 MRAB 감염의 치료 및 예방을 위한 대체 천연 제품으로 유용할 것으로 보인다. 따라서 이 연구의 결과는 다제 내성 A. baumannii의 항균 효과를 입증했으며, 미래에 천연 에센셜 오일을 사용하는 손 소독제와 같은 항균제로 사용될 것으로 예상됩니다.

Selection of indigenous starter culture for safety and its effect on reduction of biogenic amine content in Moo som

  • Objective: The aims of this study were to select one strain of Lactobacillus plantarum (L. plantarum) for a potential indigenous safe starter culture with low level antibiotic resistant and low biogenic amine production and evaluate its effect on biogenic amines reduction in Moo som. Methods: Three strains of indigenous L. plantarum starter culture (KL101, KL102, and KL103) were selected based on their safety including antibiotic resistance and decarboxylase activity, and fermentation property as compared with a commercial starter culture (L. plantarum TISIR543). Subsequently, the effect of the selected indigenous safe starter culture on biogenic amines formation during Moo som fermentation was studied. Results: KL102 and TISIR 543 were susceptible to penicillin G, tetracycline, chloramphenicol, erythromycin, gentamycin, streptomycin, vancomycin, ciprofloxacin and trimethoprim (MIC90 ranging from 0.25 to $4{\mu}g/mL$). All strains were negative amino acid-decarboxylase for lysis of biogenic amines in screening medium. For fermentation in Moo som broth, a relatively high maximum growth rate of KL102 and TISIR543 resulted in a generation time than in the other strains (p<0.05). These strain counts were constant during the end of fermentation. Similarly, KL102 or TISIR543 addition supported increases of lactic acid bacterial count and total acidity in Moo som fermentation. For biogenic amine reduction, tyramine, putrescine, histamine and spermine contents in Moo som decreased significantly by the addition KL102 during 1 d of fermentation (p<0.05). In final product, histamine, spermine and tryptamine contents in Moo som inoculated with KL102 were lower amount those with TISIR543 (p<0.05). Conclusion: KL102 was a suitable starter culture to reduce the biogenic amine formation in Moo som.

Serum alanine aminotransferase levels are closely associated with metabolic disturbances in apparently healthy young adolescents independent of obesity

  • Purpose: Liver metabolism plays a pivotal role in the development of metabolic disorders. We aimed to investigate the clinical and laboratory risk factors associated with alanine aminotransferase (ALT) levels in young adolescents from an urban population in Korea. Methods: A population of 120 apparently healthy adolescents aged 12-13 years was included in the cross-sectional design study; 58 were overweight or obese and 62 were of normal weight. We estimated anthropometric and laboratory measurements, including waist-to-height ratio, blood pressure, insulin sensitivity, aspartate aminotransferases (AST), ALT, and lipid profiles. Results: The mean ages of the overweight or obese and normal weight participants were $12.9{\pm}0.3$ and $13.0{\pm}0.3years$, respectively. Height, weight, body mass index, waist circumference, waist-to-height ratio, systolic and diastolic blood pressure, AST, ALT, total cholesterol, low-density lipoprotein-cholesterol, triglyceride, insulin, and the homeostatic model assessment of insulin resistance (HOMA-IR) score were significantly higher and the high-density lipoprotein-cholesterol and quantitative insulin-sensitivity check index were significantly lower in the overweight/obese participants in comparison to the normal-weight participants (all P<0.05). In multivariate linear regression analysis, waist-to-height ratio, systolic blood pressure, and HOMA-IR score were independently and positively associated with serum ALT levels. Conclusion: Screening for ALT levels in adolescents may help to differentiate those at risk of metabolic abnormalities and thus prevent disease progression at an early age.

Development of transgenic disease-resistance root stock for growth of watermelon.(oral)

  • To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. Gene Expression Analysis in Cucumber Leaves Primed by Root Colonization of Pseudomonas chlororaphis O6 upon Challenge-inoculation with Corynespora cassiicola.

  • Colonization of Pseudomonas chlororaphis O6, a nonpathogenic rhizobacterium, on the roots induced systemic resistance in cucumber plants against tai-get leaf spot, a foliar disease caused by Corynespora cassiicola. A cDNA library was constructed using mRNA extracted from the cucumber leaves 12 h after inoculation with C. cassiicola, which roots had been previously treated with O6. To identify the genes involved in the O6-mediated induced systemic resistance (ISR), we employed a subtractive hybridization method using mRNAs extracted from C cassiicola-inoculated cucumber leaves with and without previous O6 treatment on the plant roots. Differential screening of the cDNA library led to the isolation of 5 distinct genesencoding a GTP-binding protein, a putative senescence-associated protein, a galactinol synthase, a hypersensitive-induced reaction protein, and a putative aquaporin. Expressions of these genes are not induced by O6 colonization alone. Before challenge inoculation, no increase in the gene transcriptions could be detected in previously O6-treated and untreated plants but, upon subsequent inoculation with the pathogenic fungus, transcription levels in O6-treated plants rose significantly faster and stronger than in untreated plants. Therefore, the O6-mediated ISR may be associated with an enhanced capacity for the rapid and effective activation of cellular defense responses which becomes apparent only after challenge inoculation on the distal, untreated plant parts, as suggested by Conrath et al. (2002). Alternaria brassicicola에 의한 무 검은무늬병에 대한 효율적인 저항성 검정법 개발 (Development of an Effective Method for Testing Resistance to Black Spot of Radish Caused by Alternaria brassicicola)

  • 본 연구는 Alternaria brassicicola에 의해 발생하는 무 검은무늬병에 대한 효율적인 저항성 검정법을 확립하기 위하여 수행되었다. A. brassicicola 7균주의 포자 형성량과 병원성을 검정하였다. 실험한 균주 중, A. brassicicola KACC 40036과 43923 균주는 V-8 juice agar 배지에서 가장 많은 포자를 생산하였다. 그리고 실험한 균주 모두는 무 유묘에 대하여 강한 병원력을 나타냈다. 시판 중인 무 61개 품종의 A. brassicicola KACC 40036 균주에 대한 저항성 스크리닝을 수행하였다. 실험한 품종 중 높은 저항성을 보이는 품종은 없었으나 '금봉'과 '새롬'같이 약한 저항성을 나타내는 품종이 발견되었다. 이들 결과로부터 추후 실험을 위해 저항성 반응에 차이를 보이는 4개 품종을 선발하였다. 이들 4개 품종의 접종하는 무의 생육 시기, 접종원 농도 그리고 접종 후의 재배 온도에 따른 검은무늬병 발생을 조사하였다. 실험한 모든 품종은 어린 유묘일수록 검은무늬병에 대한 감수성이 증가하였으며, 더 많은 A. brassicicola 포자를 접종할수록 검은무늬병이 더 많이 발생하였다. 그리고 접종한 유묘를 $25^{\circ}C$의 습실상에서 배양하였을 때에는 $20^{\circ}C$$30^{\circ}C$에서 배양하였을 때보다 검은무늬병 발생이 증가하였다. 이들 실험의 결과로부터 무 품종의 검은무늬병에 대한 저항성을 검정하기 위해서는 무 종자를 파종하고 온실($25{\pm}5^{\circ}C$)에서 16일 동안 재배하고, 이 유묘에 A. brassicicola 균주의 포자현탁액($2.0{\times}10^5spores{\cdot}mL^{-1}$)을 분무하여 접종하고, 접종한 식물은 $25^{\circ}C$ 습실상에 24시간 동안 배양한 후에 $25^{\circ}C$, 상대습도 80%의 생육상에서 재배하고, 접종 2일 후에 병반면적율을 조사하는 것을 제안한다.

무 품종의 세균성 무름병 저항성 생물검정법 평가 (Evaluation of Bioassay Methods to Assess Bacterial Soft Rot Resistance in Radish Cultivars)

  • 세균성 무름병 균[Pectobacterium carotovorum subsp. carotovorum (Pcc)]에 의해서 일어나는 무름병은 아시아 국가에서 재배되는 무에 있어서 심각한 질병 중의 하나로 알려져 있다. 이번 연구의 목적은 상업적으로 시판되는 무 품종의 세균성 무름병의 저항성에 대한 효율적인 생물검정법을 확립하고자 하였다. 첫째, 무 품종에 대해 세균성 무름병의 효율적인 생물검정 방법을 조사하였다. 6개의 무 품종을 다양한 조건[두 가지 온도(25℃와 30℃), 3가지 접종방법(관주, 분무, 침지), 두 발생단계(2와 4잎 단계)]으로 조사하였다. 연구 결과는 무름병균 1×106 cfu/ml 농도를 분무한 4잎 단계와 30℃에서 배양한 생물검정 방법이 무 품종에 대해 가장 효율적인 방법임을 알 수 있었다. 둘째, 41개의 무 품종에 5가지 세균(KACC 10225, KACC 10343, KACC 10421, KACC 10458, KACC 13953)을 접종하여 저항성을 조사하였다. KACC 10421가 세균성 무름병의 감수성 및 저항성 질병 정도를 가장 잘 나타냈다. 41개의 무 품종 중 13개는 무름병 균에 대해 중도 저항성을 나타냈고 28개는 감수성을 나타냈다. 이 연구에서 중도저항성 무 품종은 세균성 무름병 저항성 육종을 위한 저항성 자원으로 활용 가능하고 육종가, 농민, 연구자, 최종 소비자에 의해서 다양한 목적을 위해 사용 가능할 것으로 사료된다.
