USP14 inhibition regulates tumorigenesis by inducing apoptosis in gastric cancer

  • Deubiquitinases (DUBs) are an essential component of the ubiquitin-proteasome system (UPS). They trim ubiquitin from substrate proteins, thereby preventing them from degradation, and modulate different cellular processes. Ubiquitin-specific protease 14 (USP14) is a DUB that has mainly been studied for its role in tumorigenesis in several cancers. In the present study, we found that the protein levels of USP14 were remarkably higher in gastric cancer tissues than in the adjacent normal tissues. We also demonstrated that the inhibition of USP14 activity using IU1 (an USP14 inhibitor) or the inhibition of USP14 expression using USP14-specific siRNA markedly reduced the viability of gastric cancer cells and suppressed their migratory and invasive abilities. The reduction in gastric cancer cell proliferation due to the inhibition of USP14 activity was a result of the increase in the degree of apoptosis, as evidenced by the increased expression levels of cleaved caspase-3 and cleaved PARP. Furthermore, an experiment using the USP14 inhibitor IU1 revealed that the inhibition of USP14 activity suppressed 5-fluorouracil (5-FU) resistance in GC cells. Collectively, these findings indicate that USP14 plays critical roles in gastric cancer progression and suggest its potential to serve as a novel therapeutic target for gastric cancer treatment.

Antimicrobial resistance of Staphylococcus aureus isolated from domestic and imported raw meat in Korea

  • The rapid evolution of antibiotic resistance in Staphylococcus (S.) aureus is of considerable concern. Methicillin-resistant S. aureus (MRSA) strains are especially one of the greatest public concerns since the treatment of infections is more difficult when encountering resistance. In this study, we conducted a nationwide survey on the antimicrobial resistance of S. aureus isolated from raw meat samples collected from 16 countries, including Korea, and investigated the prevalence of MRSA as a possible source of human infection. Of 1,984 meat samples, S. aureus was isolated from 218 (11.0%) samples consisting of 23 (12.1%) from domestic meat and 195 (10.9%) from imported meat. The isolation rates of poultry meat, pork and beef were 12.8%, 7.0% and 10.0%, respectively. With regard to imported meat, the incidence varied from 4.8% to 16.6% from 13 countries, with the exception of Austria and Poland. In a resistance test to 20 antimicrobial agents, one hundred and eighty-four isolates (84.4%) were resistant to one or more antimicrobial agents tested. Especially, 17 (7.8%), 124 (56.9%) and 28 (12.8%) isolates showed a resistance to 3, 2 and 1 drugs, respectively. One isolate originating from domestic beef was resistant to 7 drugs. Another isolate originating from imported poultry meat showed resistance to oxacillin and methicillin by the disk diffusion test and minimal inhibition concentration methods, but showed negative for detection of the mecA gene.

  • The present study was conducted to investigate the biochemical characteristics and anti-biotic resistance of Escherichia coli(E. coli) isolated from piglets with diarrhea in Kyongbuk province during the Period from February to November 1991. 368 E. coli strains were isolated from 382 piglets with diarrhea and the biochemical and cultural reaction were compared with the classification criteria of Edwards and Ewing. Tetracycline and sulfadimethoxine were found to be highly ineffective at in vitro inhibition of the E. coli of piglets origin. The majority of E. coli were susceptible to amikacin, chloramphenicol and gentamicine. 89 (89.0%) of 100 strains of E. coil were resistant to one or more drugs. The organisms resistant to 20 or 3 drugs were 54(60.6%) of 89 strains, whereas 16(17.9%) strains were found to be resistant to one drug. 55(61.8%) out of 89 drug resistance strains carried R factors($R^+$) which were transfer-able to the recipients by conjugation.

  • In this present study, the corrosion mitigation effect of conifer cone extract (CC) was examined in the cement mortar to improve the steel rebar (SR) corrosion resistance. The corrosion inhibition properties of the SR embedded in cement mortar (CM) admixed with different percentage (0, 0.5, 1.0, 1.5, 2.0 %) of CC was studied by open circuit potential (OCP) and electrochemical impedance spectroscopy (EIS) tests. This result confirms that the CM with 0.5% of CC added has better corrosion resistance than the blank specimen (0 % of CC). Although, the percentage of CC increase above 0.5%, the CC could yield a negative impact on CM properties in terms of reducing the corrosion resistance due to the reduction of cement hydration reaction. It was highlighted that the SR embedded in CM containing 0.5% of CC had increased corrosion resistance.

Some Probiotic Properties of Some Lactic Acid Bacteria and Yeasts Isolated from Jeot-gal. (젓갈 프로바이오틱 생균의 내산성 및 내담즙 특성)

  • In order to select probiotics with a high survival rate in gut and the growth inhibition of virulent pathogens to human beings or animals, we have screened lactic acid bacteria and yeasts from Jeot-gal to assess resistance against the artificial gastric acid and bile juice. Lactic acid bacteria and yeasts isolated were incubated for 24 h in artificial bile juice after incubation for 2 h in artificial gastric acid. Especially, strain HW 161 and strain NK 181 showed the higher survival for 2 h incubation in artificial gastric acid. All of 3 strains of lactic acid bacteria and 2 strains of yeast were showed higher growth rate than the control in artificial bile. The antimicrobial activity of lactic acid bacteria and yeasts was also investigated to prove efficacy as probiotic organisms. Lactic acid bacteria were shown the inhibition of Gram positive and negative bacteria, but yeasts narrow inhibition.

  • Due to consumer reluctance to take synthetic drugs for nematode infections and the appearance of resistance to anthelminthic drugs, new drugs from natural products must be developed. Caenorhabditis elegans is one of the freely living nematodes and serves as a good model system for screening anthelminthic drugs. In this study, thirteen flavonoid derivatives were tested for anthelminthic activity and the relationships between their activities and structures were investigated. The structural information combined with the data for the larval growth inhibition of C. elegans provided meaningful structural insights in the search for new anthelminthic drugs.

  • Because of its inexpensive cost, mild steel is frequently employed as a construction material in different industries. Unfortunately, because of its limited resistance to corrosion, a protective layer must be applied to keep it from decaying in acidic or basic environments. The presence of heteroatoms, such as nitrogen, oxygen, and pi-electrons in the Schiff base could cause effective adsorption on the mild steel surface, preventing corrosion. The weight loss method and scanning electron microscopy (SEM) were used to investigate the inhibitory effects of APIDO on mild steel in a 1 M hydrochloric acid environment. The efficiency of inhibition increased as the inhibitor concentration increased and decreased as the temperature increased. The SEM analysis confirmed that the corrosion inhibition of APIDO proceeded by the formation of an organic protective layer over the mild steel surface by the adsorption process. Simulations based on the density functional theory are used to associate inhibitory efficacy with basic molecular characteristics. The findings acquired were compatible with the experimental information provided in the research.

  • In this study, the optimum amount of chloride ions is used to collaborate with hybrid corrosion inhibitor for carbon steel rebar treatment in simulated pore concrete (SCP) solution is discovered. The corrosion inhibition performance of hybrid inhibitors is carried on by open circuit potential (OCP), electrochemical impedance spectroscopy (EIS), and potentiodynamic polarization (PP). The highest corrosion inhibition resistance is found in case of LP-C2 after 240 h exposure. Surface studies including scanning electron microscopy (SEM), and atomic force microscopy (AFM) were used to figure out the surface morphology of the steel rebar treated with hybrid inhibitors in order to collaborate well with electrochemical studies. Anodic type inhibition action was confirmed by potentiodynamic polarization study.

Development of Effective Screening Method for Resistance of oyster mushroom to Trichoderma disease in vitro (느타리버섯 푸른곰팡이병 저항성 실내검정 방법의 개발)

  • Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.

Characterization of S. aureus Showing MLS Resistance with Foggy D Shape (fMLS) (안개 D형 저지원을 갖는 MLS 내성 황색 포도상 구균의 분포와 내성 기전)

  • A new type of macrolides, lincosamides and streptogramin B antibiotics (MLS) resistance, showing the characteristic phenotype growing within the inhibition zone area around the clindamycin disk, was identified among clinically isolated Staphylococcus aureus. We named the phenotype as MLS resistance with foggy-D shape (fMLS). The average frequency of MLS isolates was 9%. All of fMLS isolates have only erm(A) for the resistance determinant. The growth pattern of the challenged MLS isolate looks intermediate phase between the patterns of inducible MLS resistance and constitutive MLS resistance.
