Influence of Hyaluronic Acid on the Different Levels of Lysozyme and Peroxidase in the Aspects of Candidacidal Activities

  • Purpose: The purpose of the study was to investigate the influences of hyaluronic acid on the candidacidal activities of lysozyme, the peroxidase system, and the glucose oxidase-mediated peroxidase (GO-PO) system at different concentrations of antimicrobial enzymes. Methods: Hyaluronic acid was used at a final concentration of 0.5 mg/mL. Hen egg-white lysozyme (HEWL) was used at concentrations ranging from 10 to $100{\mu}g/mL$. The peroxidase system included bovine lactoperoxidase (bLPO), potassium thiocyanate (KSCN, 1 mM), and hydrogen peroxide ($100{\mu}M$). The GO-PO system included bLPO, KSCN (1 mM), glucose oxidase (10 units/mL), and glucose ($30{\mu}g/mL$). The final concentration of bLPO in the peroxidase and GO-PO systems ranged from 12.5 to $100{\mu}g/mL$. Candida albicans strains ATCC 10231, 11006, and 18804 were utilized. Candidacidal activities of antimicrobials and the influence of hyaluronic acid on their candidacidal activities were determined based on colony forming units. Results: Candidacidal activities of the peroxidase and GO-PO systems increased with increasing concentrations of bLPO. This tendency was the same in the presence or absence of hyaluronic acid. Candidacidal activity of HEWL was not significantly concentration-dependent. Candidacidal activities of the GO-PO system were higher than those of the corresponding peroxidase system. Candidacidal activity was inhibited in the presence of hyaluronic acid in the following order: HEWL, the peroxidase system, and the GO-PO system. Conclusions: Hyaluronic acid inhibited the candidacidal activities of HEWL, the peroxidase system, and the GO-PO system. The GO-PO system exhibited better candidacidal activity than HEWL and the peroxidase system both in the presence and absence of hyaluronic acid.

  • 21세기 정보사회에서 창의성이 가지는 부가가치는 더 이상의 논의가 필요 없을 정도로 강조되고 있다. 본 연구는 창의적 인재교육에 근간이 되는 유아 통합예술교육 중 비유 놀이(metonymic activities)를 통하여 창의성을 습득하는 프로그램인 에듀드라마(Edu Drama) 행복한 그림책 여행, 맛을 아는 꼬마철학자를 분석하면서 유아 통합예술교육에서 비유 놀이가 가져 올 수 있는 시사점을 살피고자 한다. 이를 위하여 우리나라 유아통합예술교육의 제한적 현실을 정리하고, 비유놀이를 적용하고 있는 국내외 사례를 통하여 앞으로 나아가야 할 유아 통합예술교육의 바람직한 방향을 제시한다. 기존 예술경영 분야가 선도해 온 우리나라 통합예술교육에 대한 연구는 활발하였음에도 불구하고, 유아를 위한 통합예술교육 연구는 상대적으로 미진한 것이 현실이다. 이에 본 연구는 거시적인 우리 삶의 부가가치는 21세기 창의적 인재에 있다는 가정 하에, 비유 놀이를 통하여 유아기에 이루어지는 통합예술교육이 가져오는 가치를 밝히고자 한다.

넙치(Paralichthys olivaceus) 양식산업의 가치사슬 분석 (Value Chain Analysis of the Olive Flounder Paralichthys olivaceus Aquaculture Industry)

  • This study aimed to analyze the structure of the value chain of the olive flounder aquaculture industry to increase the value of this industry. Based on the value chain theory, olive flounder aquaculture industry activities were classified as primary and support activities. The primary activities included seed production, fish production, producer distribution, consumer distribution, and consumption. The support activities were production infrastructure, organization and specialization, R&D, and government policy. A survey was conducted on the costs of seed and fish production in the primary activities to investigate the business structure, and the distribution structure was analyzed to examine distribution costs and margins. In the support activities, the recent trends in R&D and government policy were mainly examined, based on which, a measure to reduce costs and maximize profits was suggested. It is necessary to reduce costs across the production processes by improving seed quality and reducing labor, feed, and management costs, which are strongly associated with support activities. Therefore, lowering costs will be possible in the olive flounder aquaculture industry when R&D outcomes, such as species development, feed quality improvement, and aquaculture system development, are stably diffused and applied in tandem with government policy regarding the industry.

Comparison of TNF-Mediated Glucose Catabolism between the TNF-Sensitive and -Resistant Cell Lines

  • When murine fibrosarcoma L929 cells, a TNF-sensitive cell line, were treated with recombinant human tumor necrosis factor-$\alpha$ (rhTNF-$\alpha$), the activities of glycolytic regulatory enzymes and lactate dehydrogenase increased up to 100-150% compared to the control L929 cells after TNF treatment. By using various metabolic inhibitors and activators, it was found that cAMP-dependent protein kinase is responsible for the increase of activities of the glycolytic enzymes. The activities of glycolytic regulatory enzymes and lactate dehydrogenase of TNF-resistant A549 cells, a human lung carcinoma cell line, did not increase significantly compared to TNF-sensitive L929 cells upon TNF treatment. In contrast, the pyruvate carboxylase activities of A549 cells, but not L929 cells, increased up to 30~40% after TNF treatment. The data suggest that pyruvate carboxylase activity may contribute to the compensation of energy loss mediated by TNF treatment in TNF-resistant A549 cells.

EBM 기반구축을 위한 평위산 전임상 연구 문헌 분석 (Analysis of Studies on Pyeongwi-san to Establish the Fundament for Evidence Based Medicine)

  • To establish the fundament for EBM of Traditional Korean Medicine, the papers on Pyeongwi-san (Pingwei-san) frequently used in medical institutions of Traditional Korean Medicine were analyzed through researching domestic and international papers. The papers were classified by the registration of domestic or international journals, the year of publishment, experimental fields and the kinds of studies on biological activities. Of total 518 papers on Pyeongwi-san (Pingwei-san), 32 volumes were selected according to selection creteria. 20 volumes were published in domestic journals, 11 in Chinese journal and 1 in Japanese journal. The papers on instrumental analyses reported the quantification of standard compounds of herbal medicines in Pyeongwi-san (Pingwei-san) using HPLC, GC-MS. The papers on biological activities of Pyeongwi-san (Pingwei-san) showed improvement of gastrointestinal activity and water-electrolyte metabolism, immune regulation, anti-oxidant, anti-inflammatory, analgesic, anti-convulsant activities, hypnotic duration, blood pressure regulation, hepatic protection, congestive heart failure, anti-cancer activities. Further studies including gastrointestinal activity need to be preceeded to establish the fundament for EBM of Pyeongwi-san (Pingwei-san).

Antiplatelet and Antithrombotic Effects of the Extract of Lindera obtusiloba Leaves

  • Lindera obtusiloba has been used in traditional herbal medicine for the treatment of blood stasis and inflammation. The leaves of Lindera obtusiloba have been reported to exhibit various physiological activities. However, there is little information available on their antiplatelet and antithrombotic activities. Thus, the present study aimed to evaluate the effect of Lindera obtusiloba leaf extract (LLE) on platelet activities, coagulation and thromboembolism. In a platelet aggregation study, LLE significantly inhibited various agonist-induced platelet aggregations in vitro and ex vivo. Furthermore, LLE significantly inhibited collagen-induced thromboxane A2 (TXA2) production in rat platelets. In addition, oral administration of LLE was protective in a mouse model of pulmonary thromboembolism induced by intravenous injection of a mixture of collagen and epinephrine. Interestingly, LLE did not significantly alter prothrombin time (PT) and activated partial thromboplastin time (aPTT). This study indicates that the antithrombotic effects of LLE might be due to its antiplatelet activities rather than anticoagulation. Taken together, these results suggest that LLE may be a candidate preventive and therapeutic agent in cardiovascular diseases associated with platelet hyperactivity.

Activity and Isozyme Profile of Antioxidative Enzymes at Booting Stage of Rice Treated with Cold Water

  • This study was carried out to investigate the antioxidative enzymes and isozymes between chilling-tolerant and -susceptible varieties at the booting stage under cold water stress $(13^{\circ}C)$ in japonica rice. Total SOD, CAT, POX, and GR activities on the basis of protein were found to be important factors to defend cold water stress. Especially, SOD and CAT activities showed distinctive differences between chilling-tolerant and -susceptible varieties. Chilling-tolerant varieties were higher than chilling-susceptible varieties for SOD and CAT activities. One of eight isozyme bands for SOD was a inducible isoform. Three isozymes for CAT and one isozyme for POX were closely correlated with defense to cold water stress. Total GR activities except Stejaree 45 on the basis fresh weight and POX were increased by cold water stress, but there was no difference between chilling-tolerant and -sus­ceptible varieties.

Antitcoagulant and antiplatelet activities of scolymoside

  • Cyclopia subternata is a medicinal plant commonly used in traditional medicine to relieve pain. Here, the anticoagulant effects of scolymoside, an active compound in C. subternata, were examined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT), and the activities of thrombin and activated factor X (FXa). The effects of scolymoside on plasminogen activator inhibitor type 1 (PAI-1) and tissue-type plasminogen activator (t-PA) expression were evaluated in tumor necrosis factor (TNF)-α-activated human endothelial cells. Treatment with scolymoside resulted in prolonged aPTT and PT and the inhibition of thrombin and FXa activities and production. In addition, scolymoside inhibited thrombin-catalyzed fibrin polymerization and platelet aggregation. Scolymoside also elicited anticoagulant effects in mice, including a significant reduction in the PAI-1 to t-PA ratio. Collectively, these findings indicate that scolymoside possesses anticoagulant activities and could be developed as a novel anticoagulant.

국내산 7종 다슬기 추출물의 생리활성 특성 비교 (Biological Activities of Seven Melania Snails in Korea)

  • This study was conducted to compare the biological activities of 7 melania snails from the family Pleuroceridae (Semisulcospira coreana, Koreanomelania nodifila, Semisulcospira forticosta, Koreoleptoxis globus ovalis, Semisulcospira libertina, Semisulcospira tegulata and Semisulcospira gottschei) in Korea. Among the 7 species, S. coreana, Korean. nodifila, S. forticosta and S. gottschei showed over 80% cytotoxicities on three cancer cell lines (SNU-1, A549 and Hep 3B) compared to the non-treatment, whereas S. libertina and S. tegulata showed almost no growth inhibition activities on the same cancer cell lines. In relation to ACE inhibition activity, only S. coreana, Korean. nodifila, and S. forticosta showed over 60% ACE inhibition activities, whereas other melania snails exhibited inhibition activities of lower than 25%. DPPH radical scavenging activities were also determined, and used to categories melania snails into three groups based on Duncan's multiple range test at P<0.05. The amount of TNF-${\alpha}$ produced by in vitro mouse peritoneal macrophage was determined according to bioactivity on L-929 cells. Three melania snails, S. coreana, Korean. nodifila and S. gottschei, exhibited 95.2%, 89.7% and 93.7% cell death(%) on L-929 cells, respectively. Glucose-6-phosphate dehydrogenase inhibitory activity was also obtained in the extract of S. coreana (31.9%) and Korean. nodifila (28.1%), showing that these extracts can be used as supplemental dietary health foods. In conclusion, we believe that the extracts of melania snails should be given due consideration in functional health food development.

한국 진출 외국 R&D 센터의 상호작용 특성 분석 (An Analysis of Interactive Activities of Foreign R&D Centers Operating in Korea)

  • This paper analyzes interactive activities of foreign R&D centers operating in Korea. Interactive activities refer to collaborative R&D activities, R&D outsourcing, and formation of communities between foreign R&D centers and Korean firms and institutions. Active interactions between foreign R&D centers and their Korean partners (Korean firms and universities, research institutes etc.) are essential in promoting spillover effects from foreign R&D centers. Using the survey of 76 foreign R&D centers operating in Korea, this paper found the intensity of interplay between foreign R&D centers and the Korean firms and institutions was not so high. And the partners of foreign R&D centers with the mission of finding new technologies in Korea are mainly customers rather than universities and research institutes. This result is markedly different from previous studies focused on the cases of western countries, where the partners of foreign R&D centers that have the mission of finding new technologies were mainly local universities and research institutes.
