생후 발달과정동안 남성 생식기의 Efferent Ductules에서 Monocarboxylate Transporters(MCTs)와 Basigin의 발현 양상과 에스트로젠 수용체 α에 의한 MCT1 발현 조절 연구 (Expression of Monocarboxylate Transporters(MCTs) and Basigin and Estrogen Receptor α(ERα)-Mediated Regulation of MCT1 Expression in The Efferent Ductules of Male Reproductive Tract During Postnatal Development)

  • 본 연구에서는 남성 생식 기관인 efferent ductules(ED)에서 monocarboxylate transporter (MCT) isoform과 Basigin(Bsg)의 mRNA 발현을 탐구하고, MCT1의 발현이 에스트로젠 수용체 α에 의해 조절되는지를 연구하였다. 생후 여러 연령대의 백서 ED에서 MCT isoform과 Bsg의 mRNA 발현 여부는 real-time PCR에 의해 조사 되었고, 정상 생쥐와 에스트로젠 수용체 α knockout 생쥐를 사용하여 에스트로젠 수용체 α에 의한 MCT1의 발현 조절 여부는 immunohistochemistry 방법에 의해 간접적으로 알아 보았다. 본 연구 결과는 백서의 ED에서 MCT1, 2, 3, 4와 8 그리고 Bsg 유전자의 발현은 연령에 따라 다르게 나타나며, MCT1의 발현이 ED의 ciliated 세포의 basolateral 지역에서 발현되고 어떤 측면에서 ERα에 의해 조절되어 질 수 있음을 보여 주었다. 따라서 본 연구 결과는 MCT가 monocarboxylate의 세포 내, 외부로 운반을 조절함으로써 남성 생식기의 일부인 ED의 역할을 조절하는데 관여함을 시사한다.

Ontogenetic Expression of Lpin2 and Lpin3 Genes and Their Associations with Traits in Two Breeds of Chinese Fat-tailed Sheep

  • Lipins play dual function in lipid metabolism by serving as phosphatidate phosphatase and transcriptional co-regulators of gene expression. Mammalian lipin proteins consist of lipin1, lipin2, and lipin3 and are encoded by their respective genes Lpin1, Lpin2, and Lpin3. To date, most studies are concerned with Lpin1, only a few have addressed Lpin2 and Lpin3. Ontogenetic expression of Lpin2 and Lpin3 and their associations with traits would help to explore their molecular and physiological functions in sheep. In this study, 48 animals with an equal number of males and females each for both breeds of fat-tailed sheep such as Guangling Large Tailed (GLT) and Small Tailed Han (STH) were chosen to evaluate the ontogenetic expression of Lpin2 and Lpin3 from eight different tissues and months of age by quantitative real-time polymerase chain reaction (PCR). Associations between gene expression and slaughter and tail traits were also analyzed. The results showed that Lpin2 mRNA was highly expressed in perirenal and tail fats, and was also substantially expressed in liver, kidney, reproductive organs (testis and ovary), with the lowest levels in small intestine and femoral biceps. Lpin3 mRNA was prominently expressed in liver and small intestine, and was also expressed at high levels in kidney, perirenal and tail fats as well as reproductive organs (testis and ovary), with the lowest level in femoral biceps. Global expression of Lpin2 and Lpin3 in GLT both were significantly higher than those in STH. Spatiotemporal expression showed that the highest levels of Lpin2 expression occurred at 10 months of age in two breeds of sheep, with the lowest expression at 2 months of age in STH and at 8 months of age in GLT. The greatest levels of Lpin3 expression occurred at 4 months of age in STH and at 10 months of age in GLT, with the lowest expression at 12 months of age in STH and at 8 months of age in GLT. Breed and age significantly influenced the tissue expression patterns of Lpin2 and Lpin3, respectively, and sex significantly influenced the spatiotemporal expression patterns of Lpin3. Meanwhile, Lpin2 and Lpin3 mRNA expression both showed significant correlations with slaughter and tail traits, and the associations appear to be related with the ontogenetic expression as well as the potential functions of lipin2 and lipin3 in sheep.

난자공여를 통한 체외수정 시술에서 성선자극호르몬 유리호르몬 효능제 장기요법과 길항제 단기요법 사이의 임상 결과 비교 (The Comparison of Clinical Outcomes between GnRH Agonist Long Protocol and GnRH Antagonist Short Protocol in Oocyte Donation Cycles)

  • Objective : To assess and compare the clinical outcomes between GnRH agonist long protocol and GnRH antagonist short protocol in oocyte donation program. Materials and Methods: Of total 18 oocyte donation cycles, controlled ovarian hyperstimulation (COH) were performed with GnRH agonist long protocol and GnRH antagonist short protocol in initial 9 cycles and later 9 cycles, respectively. Oral estradiol valerate and progesterone in oil we re administrated to all recipients for endometrial preparation. Oral estradiol administration was started from donor cycle day 1 after full shut down of gonadal axis with GnRH agonist in patients with ovarian function. Progesterone was injected from oocyte retrieval day of donor initially, then continuously till pregnancy 12 weeks if pregnancy was ongoing. We compared the parameters of clinical outcomes, such as number of the retrieved oocytes, fertilization rate, high grade embryo production rate, clinical pregnancy rate, implantation rate, ongoing pregnancy rate, COH duration, total gonadotropin dose for COH between GnRH agonist long protocol group and GnRH antagonist group. Statistical analysis was performed using Mann-Whitney test, p<0.05 was considered as statistically significant. Results: The number of retrieved oocytes, fertilization rate, high grade embryo production rate, clinical pregnancy rate, implantation rate, ongoing pregnancy rate were $14.89{\pm}7.83$, 81%, 64%, 78%, 31%, 78%, respectively in GnRHa long protocol group and $11.22{\pm}8.50$, 79%, 64%, 67%, 34%, 56%, respectively in GnRH antagonist group. There was no significant differences in parameters of clinical outcomes between 2 groups (all p value >0.05). Duration and total gonadotropin dose for COH were $10.94{\pm}1.70$ days and $43.78{\pm}6.8$ vials in 18 cycles, $12.00{\pm}1.73$ days and $48.00{\pm}6.93$ vials in agonist group, $9.88{\pm}0.78$ days and $39.55{\pm}3.13$ vials in antagonist group, respectively. In GnRH agonist long protocol group, significantly longer duration and higher gonadotropin dose for COH were needed (p=0.012). Conclusion: In oocyte donation program, clinical outcomes from controlled ovarian hyperstimulation with GnRH antagonist were comparable to those from GnRH agonist long protocol group, so controlled ovarian hyperstimulation with GnRH antagonist may be effective as GnRH agonist long protocol. At least there may not be harmful effects of GnRH antagonist on oocyte development and quality.

한국 서해산 암컷 비단가리비, Chlamys farreri의 난형성과정 중 생식세포 분화 및 생식주기 (Germ Cell Differentiations during Oogenesis and Reproductive Cycle in Female Jicon Scallop, Chlamys farreri on the West Coast of Korea)

  • 1998년 3월부터 1999년 2월까지 전라남도 대흑산도 연안에서 채집한 비단가리비를 대상으로 생식소중량지수, 생식세포 분화 및 난소주기를 조직, 세포학적 관찰에 의해 조사하였다. 초기 난황형성 난모세포에서, 골지체, 미토콘드리아 및 조면소포체들은 지방적 형성에 관여하였다. 후기난황형성난모세포에서 생식상피상에 존재하는 외인성 물질들 즉, 글리코겐 입자들 및 지방 과립상 물질들이 난황막의 미세융모를 통해서 난모세포의 난질로 통과해 들어갔다. 후기난황형성난모세포에서, 난황과립들과 다포체들은 단백질성 난황과립형성에 관여하였다. 난황형성과정은 내인성 자율합성과 외인성 타가합성에 의해서 일어난다. 보조세포들은 초기단계인, 전난황형성 난모세포 및 초기 난황형성 난모세포들의 형성 및 발달에 영양세포로서 기능을 한다. 생식소 중량지수의 월별 변화는 난소 발달단계들과 밀접한 관련을 가지며 변하였다. 본 종의 생식주기는 초기활성기(1~3월), 후기활성기(3~4월), 완숙기(4~8월), 부분산란기(6~8월), 퇴화 및 비활성기(8~12월)의 5단계로 구분되었다. 산란은 6~8월 사이에 일어나며, 주산란기는 수온이 높은 7~8월이었다.

PRRS 바이러스 Nucleocapsid 단백질 인산화의 기능학적 연구 (Functional Characterization of Phosphorylation of the Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) Nucleocapsid Protein)

  • 돼지생식기호흡기증후군 바이러스를 구성하고 있는 뉴클레오캡시드(N) 단백질은 다양한 기능을 가지고 있는 basic 단백질로써 또한 아직까지 밝혀지지 않은 역할을 하는 serine 인산화 단백질로 알려져 있다. 먼저 바이러스가 복제되는 동안 뉴클레오캡시드 단백질 인산화가 어떤 생물학적 역할을 하는지에 대한 이해를 하기 위하여 mutagenesis 방법으로 단백질 내 모든 serine 잔기들을 alanine으로 대체하여 변이 뉴클레오캡시드 단백질을 구축하였다. 이 재조합 뉴클레오캡시드 단백질은 비인산화 단백질로 확인되었고 이는 뉴클레오캡시드 단백질 인산화에 serine 잔기들이 중요한 역할을 한다는 것을 증명하였다. 돼지 생식기호흡기증후군 바이러스 뉴클레오캡시드 단백질은 세포핵 내 이동과 N-N dimer 형성 등의 특이적인 생물학적 특성들을 보유하고 있으며 이들 각각은 바이러스 감염 시 중요한 역할들을 하는 것으로 알려져 있다. 따라서 본 연구에서는 이 두 가지 뉴클레오캡시드 단백질의 특성들이 인산화 여부에 의해 조절되는지 살펴보았다. 하지만 본 연구의 결과들은 비인산화된 뉴클레오캡시드 단백질이 여전히 transfection된 세포의 핵 또는 핵인에서 발현되었고 더욱이 뉴클레오캡시드 자신과 dimer 형성을 할 수 있었다는 것을 보여주었다. 결론적으로 돼지 생식기호흡기증후군 바이러스 뉴클레오캡시드 단백질의 세포핵 내 수송 및 oligomerization 특성들은 인산화 비의존성으로 조절되는 것으로 보여 진다. 아마도 이 인산화 작용은 뉴클레오캡시드 단백질의 RNA-binding 특성등과 같은 다른 수준의 조절과 관련이 있는 것으로 추측되어 진다.

Reproductive biology of common carp (Cyprinus carpio Linnaeus, 1758) in Lake Hayq, Ethiopia

  • This study was conducted in Lake Hayq between January and December 2018. The objectives of this study were to determine the growth, condition, sex ratio, fecundity, length at first sexual maturity (L50), and spawning seasons of common carp (Cyprinus carpio). Monthly fish samples of C. carpio were collected using gillnets of stretched mesh sizes of 4, 6, 7, 8, 10, and 13 cm and beach seines of mesh size of 6 cm. Immediately after the fish were captured, total length (TL) and total weight (TW) for each individual were measured in centimeters and grams, respectively, and their relationship was determined using power function. Length at first maturity (L50) was determined for both males and females using the logistic regression model. The spawning season was determined from the frequency of mature gonads and variation of gonadosomatic index (GSI) values of both males and females. Fecundity was analyzed from 67 mature female specimens. The length and weight relationship of C. carpio was TW = 0.015TL2.93 for females and TW = 0.018TL2.87 for males that indicate negative allometric growth in both cases. The mean Fulton condition factor (CF) was 1.23 ± 0.013 for females and 1.21 ± 0.011 for males. The value of CF in both cases was > 1 that shows both sexes are in good condition. Among the total 1055 C. carpio collected from Lake Hayq, 459 (43.5%) were females and 596 (56.5%) were males. The chi-square test showed that there was a significant deviation between male and female numbers from 1:1 ratio (χ2= 22, df = 11, P > 0.05) within sampling months. The length at first sexual maturity (L50) for females and males were 21.5 and 17.5 cm, respectively. Males mature at smaller sizes than females. The spawning season of C. carpio was extended from February to April, and the peak spawning season for both sexes was in April. The average absolute fecundity was 28,100 ± 17,462. C. carpio is currently the commercially important fish while Nile tilapia fishery has declined in Lake Hayq. Therefore, this baseline data on growth, condition, and reproductive biology of common carp will be essential to understand the status of the population of carp and design appropriate management systems for the fish stock of Lake Hayq, Ethiopia, and adjacent countries.

Endonuclease G is Upregulated and Required in Testicular Germ Cell Apoptosis after Exposure to 60 Hz at 200 μT

  • Several reports supported that continuous exposure to 60 Hz magnetic field (MF) induces testicular germ cell apoptosis in vivo. We recently evaluated duration- and dose-dependent effects of continuous exposure to a 60 Hz MF on the testes in mice. BALB/c male mice were exposed to a 60 Hz MF at $100{\mu}T$ for 24 hours a day for 2, 4, 6, or 8 weeks, and at 2, 20 or $200{\mu}T$ for 24 hours a day for 8 weeks. To induce the apoptosis of testicular germ cell in mice, the minimum dose is $20{\mu}T$ at continuous exposure to a 60 Hz MF for 8 weeks, and the minimum duration is 6 weeks at continuous exposure of $100{\mu}T$. Continuous exposure to a 60 Hz MF might affect duration- and dose-dependent biological processes including apoptotic cell death and spermatogenesis in the male reproductive system of mice. The safety guideline of the International Commission on Non-Ionizing Radiation Protection (ICNIRP) indicates that the permissible maximum magnetic flux density for general public exposure is $200{\mu}T$ at 60 Hz EMF (ICNIRP Guidelines, 2010). In the present study, we aimed to examine the expression of pro- and anti-apoptotic genes regulated by the continuous exposure to 60 Hz at $200{\mu}T$ in Sprague-Dawley rats for 20 weeks. The continuous exposure to 60 Hz at $200{\mu}T$ does not affect the body and testicular weight in rats. However, exposure to 60 Hz MF significantly affects testicular germ cell apoptosis and sperm count. Further, the apoptosis-related gene was scrutinized after exposure to 60 Hz at $200{\mu}T$ for 20 weeks. We found that the message level of endonuclease G (EndoG) was greatly increased following the exposure to 60 Hz at $200{\mu}T$ compared with sham control. These data suggested that 60 Hz magnetic field induced testicular germ cell apoptosis through mitochondrial protein Endo G.

염산 DDB-S가 랫드의 생식독성에 미치는 영향에 관한 연구 (A Teratogenicity Study on Dimethyl Dimethoxy Biphenylate Derivative (DDB-S) in Rats)

  • A teratogenic study of dimethyl dimethoxy biphenylate derivative (DDB-S) was carried out on Sprague-Dawley rats. DDB-S dissolved in saline was administered to male and female rats by intravenously injection at daily doses of 50 mg/kg, 75 mg/kg, and 100 mg/kg. A half of dams were sacrificed at 20th day of gestation to scrutinize the pregnant performances and fetal development. And the remaining dams were allowed to deliver. The growth, reflex, behaviour and reproductive function of F1 offsprings were examined. There was no treatment-related difference in body weight, food consumption and necropsy findings of dams. No gross, skeletal and visceral abnormalities was observed in F1 fetuses from dams treated with DDB-S. F1 offsprings did not show any treatment-related difference in growth, reflex, behaviour and reproductive performance. At caesarean section of F1 dams, no growth retardation and gross abnormality was observed in F2 fetuses. In conclusion, DDB-S did not show any potential teratogenic effect in rats.

황체호르몬 수용체의 발현이 저반응 환자군의 임신에 미치는 영향 (The effect of LH Receptor in the Pregnancy of Poor Responders)

  • Objectives: To investigate the effect of LH receptor in folliculogenesis, we confirm the expression level of LH receptor (LH-R) mRNA in human granulosa cells (GCs) and its expression levels were analyzed by comparison to embryo developmental rate and pregnancy rate. Materials and Methods: GCs were obtained at the time of oocyte retrieval from the patients undergoing IVF-ET program. The patients were divided into two groups: Group I (n=20) is poor responder (retrieved oocyte(s)$\leq$3ea), Group II (n=80) is normal responder (retrieved oocytes>3ea). After the extraction of total RNA, semiquantitative RT-PCR was performed and the expression level of LH-R mRNA was normalized by $\beta$-actin. Statistical analysis was performed by using $X^2$ test, Student's t-test and Pearson correlation. Results: In Group II, the relative values of LH-R mRNA (0.680 vs. 0.463, p<0.005) and pregnancy rate (54.7% vs. 23.1%, p<0.05) were significantly higher than in Group I. Number of retrieved oocyte(s) was gradually increased when the expression of LH-R mRNA was increased (p<0.05). But the quality of retrieved oocyte and transferred embryo were not related with the expression of LH-R mRNA. When the pregnancy rate was compared with FSH only group and FSH combined with hMG group in the ovarian stimulation protocol, FSH combined with hMG group was significantly higher than FSH only group in Group I (37.5% vs. 0%), and the expression of LH-R mRNA was significantly higher in hMG combined group than FSH only group (p<0.05). Conclusion: Expression level of LH-R mRNA has important role in ovarian function related with the response to gonadotrophin in human folliculogenesis. Furthermore these data might provide the evidence that additional use of hMG is helpful to poor responders.

에스트로겐이 T 림프구 면역기능에 미치는 영향 (The Influence of Estrogen on the Immunologic Function of T Lymphocytes)

  • SLE 등의 자가면역질환 병인론에 있어서 $CD4^+$ T 조력 림프구 (T helper cell)의 기능과 항상성 조절의 이상이 중요하다는 것은 알려진 사실이지만 $CD4^+$ T 조력 림프구 (T helper cell)의 발달과 기능에 대한 성호르몬의 정확한 역할에 대하여는 아직 확립되어 있지 않은 실정이다. 에스트로겐의 $CD4^+$ T 림프구에 대한 여러 방면의 작용에 대한 결과는 에스트로겐 매개 신호가 전달되는 정황에 매우 의존적이라고 할 수 있으며, 또한 에스트로겐이 B 림프구와 항원발현세포들 같이 T 림프구 기능을 변화시킬 수 있는 다른 분획 세포들의 기능에 대해서도 영향을 미칠 수 있다는 사실을 고려한다면, 에스트로겐이 $CD4^+$ T 림프구매개반응에 미치는 작용은 매우 복잡하다고 할 수 있다. 현재로선 이런 과정들과 관련된 에스트로겐의 정확한 기전에 대한 연구 성과는 현재로선 아직걸음마 단계에 지나지 않는다고 볼 수 있다. 일부 분자학적 표적들의 발견에도 불구하고 ER-$\alpha$와 ER-$\beta$, 또는 이런 표적들의 조절에 대한 유전자적 또는 비유전자적 경로의 상대적 역할에 대해서는 현재로선 알려진 바가 많지 않은 것 또한 사실이며, T 림프구 자체의 미세 환경과 ER 매개 전사반응에 영향을 미칠 수 있는 방법 역시 아직까진 알려진 바가 없다. 따라서 에스트로겐의 $CD4^+$ T 림프구에 대한 다각적 영향을 규명하는 데 있어 이런 경로들에 대한 보다 체계적이고 상세한 연구가 반드시 필요하다. 최근 골다공증의 병인에 있어 에스트로겐에 의한 TNF 및 RANK-RANKL계 억제에 대해 연구가 심도 있게 진행됨으로써 결과적으로 다른 시토카인과 면역세포들에 대한 영향이 간접적으로 규명되고 있는 점은 매우 고무적인 현상이라고 여겨진다. 이러한 연구들은 추후 에스트로겐의 면역 및 염증체계에 대한 특징적 작용을 규명하는데 있어서 밑거름이 될 수 있을 것이다. 나아가 호르몬 자체, 또는 SERM 같은 준호르몬제제들이 면역계에 미치는 영향을 분자학적으로 규명함으로써 향후 이러한 지식들이 또한 류마티스성 자가면역질환이나 만성염증성질환, 또는 전신성 감염질환에 있어 면역글로불린이나 기타 다른 기존 치료 약제들을 대체 보완할 수 있는 호르몬 치료로 이어질 수 있는 결정적인 단서를 제공할 수 있을 것이라 기대할 수 있을 것이다.
