Effect of Vitamin E on Production Performance and Egg Quality Traits in Indian Native Kadaknath Hen

  • This experiment investigated the effects of increasing dietary vitamin E (VE) on production performance and egg quality traits of Indian reared Kadaknath (KN) hens. One hundred and eighty (180), day old female KN chicks were randomly distributed to three dietary treatment groups for a period of 30 weeks. Each treatment comprised three replicates, each containing 20 chicks. The basal diet ($T_1$) contained 15 IU VE/kg and the two experimental diets were supplemented with 150 and 300 IU VE/kg (diets $T_2$ and $T_3$, respectively). DL-${\alpha}$-tocopherol acetate was used as the source of VE. All chicks were provided feed and water ad libitum. Production performance in terms of body weight, egg weight and hatchability did not differ significantly (p>0.05), whereas sexual maturity, egg production and fertility differed significantly (p<0.05) in $T_2$ compared to the other two groups. Egg quality traits in terms of albumin weight, yolk weight, shell thickness, albumin index and yolk index did not differ significantly (p>0.05), whereas the Haugh unit score was significantly higher (p<0.05) in $T_2$ than the control ($T_1$) and high dose treatment group ($T_3$). From this study, it can be concluded that lower levels of dietary VE may be beneficial for production performance and Haugh unit score but have no effect on egg quality traits in Indian reared KN hens.

Behavioral Characteristics of Bos indicus Cattle after a Superovulatory Treatment Compared to Cows Synchronized for Estrus

  • The intensity and duration of sexual behavior in Bos indicus was assessed through the continuous observation of sexual receptivity. Two groups of cows were formed: only synchronized (n = 50) and other group further superovulated (n = 20). An intravaginal implant that released progesterone over 9 d was used. After removing the implant, 25 mg of $PGF_{2{\alpha}}$ was administered. In the superovulated group, the administration of 280 mg (Follicle stimulant hormone) FSH-P1 per cow with a decreasing dosage over 4 d was utilized. In both groups, behavioral observations began at the moment of implant removal. Sexual behavior was analyzed using a Kruskal-Wallis test to compare the mean of hours in estrus, effective mountings and number of mounts/hour during estrus. A nonparametric survival analysis was performed using the time in two ways: i) when an event happened it was placed in a 24 h timeframe and, ii) the time of observation in continuous form (96 h) assessing the difference between curves by the log rank test Chi-square. The only significant difference was the number of mounts/h during receptivity (p<0.05). In the superovulated group three periods of sexual activity during the day were identified, with these events being of greater frequency and duration than the synchronized group (p = 0.02); besides, the superovulated group began estrus before the synchronized group (p = 0.0035) when using the total period. In a simulation study, when the number of observations went from two (06:00-18:00) to three periods (06:00, 12:00 and 18:00) cows detected accurately (<6 h after the onset) increased more than 20%. The results show that superovulated cows presented greater intensity and duration of sexual activity in contrast with only synchronized animals.

Growth and Reproduction of Deep-Water Mud Shrimp (Solenocera melantho) around Geomun Island, Korea (거문도 해역 대롱수염새우(Solencer melantho)의 성자과 성 성숙)

  • This paper describes reproduction, population structure, growth and size at sexual maturity of a deep-water mud shrimp (Solenocera melantho) in the adjacent waters of Geomun Island, Korea. The shrimp was caught by a beam trawl from November 1998 to December 1999. Spawning period was estimated by gonadosomatic index to be from August to early November with a peak in October to early November. Female shrimps were $12\%$ matured at 16 mm in carapace length, $33\%$ matured at 18 mm and $75\%$ matured at 22 mm. The size at $50\%$ maturity for female was estimated to be 20.65 mm in carapace length. Breeding season was estimated to be from August to early November with a peak in October to early November. The size at $50\%$ inseminated was 19.81 mm CL. The fecundity was 87,500-405,200 and related exponentially to carapace length and total weight. This species survived between 25 and 26 months. The maximum carapace length of females and males were 48 mm and 34 mm respectively. Sex ratio was $51.3\%$, and the females were preponderant. Growth was estimated by modified von Bertalanffy growth function incorporating seasonal variation in growth in both years females (K=1.019/yr and $L_{\infty}$=51.731 mm CL) grew faster, reaching a larger size at a given age than males (K=1.848/yr, and $L_{\infty}$=27.499 mm CL). This is indicated by differences in growth performance indices $(\varphi')$ between females (3.291) and males (3.145).

Effect of Diets on Development and Reproduction of Rice Armyworm, Pseudaletia separata (Lepidoptera: Noctuidae) (멸강나방의 발육과 생식에 미치는 먹이의 영향)

  • Development and reproduction of the rice armyworm, Pseudaletia separara, were investigated on different diets (corn leaf, rice leaf, Chinese cabbage leaf, cabbage leaf, sweetpotato leaf, soybean leaf and silkworm artificial diet). Egg and pupal periods were not different among the diets. But larval periods showed significant difference among diets. Developmental period from egg to adult was shortest as 32.5 d on corn leaf and longest as 46.6 d on soybean leaf. At 16 days after hatching, larval weights on rice and silkworm artificial diet were heavier than those on other diets. The larvae reared on cabbage leaf and sweetpotato leaf failed to pupate. Survival rates from larva to adult were highest (70.6%) on com leaf lowest (39.8%) on soybean leaf. The longevity of female adults was not affected by the larval diets. Female adults showed higher fecundity when the larvae were fed with corn leaf, rice leaf and silkworm artificial diet than other diets. Net reproduction rate (R$_{0}$) was highest as 1218.5 on silkworm artificial diet. The intrinsic rate of natural increase (R$_{0}$) ranged from 0.115 to 0.175, and was maximum on com leaf.

A Study on the Occurences and Caused Reproductive Disorders of the Introduced Dariy Cattle in Cheju-do I. The Investigation of Reproductive Performances (제주도입란우의 번식장해원인에 관한 연구 II. 제주도입 유우의 번식상황 조사)

  • The results from a survery on the reproduction of a, pp.oximately 500 dairy cows raised in four different farming areas including Cheju city, Jochun, Hallim and Sung-up are summarized as follows; 1. At the occurrence of first estrus, the average age of the heifers was 15.1 months (330 Kg/B.W.), and at first conception 18.0 months (354Kg/B.W.). There was no regional differences. 2. The average interval of the estrous cycle was 20.4 days. And the repetition of esturs after unsuccessful insemination occurred on an average of 1.6 times, especially 2.0 times in the herds of the Jochun district. 3. The ages at the first calving were similar to the data recorded for the domestic mainland's herds at 27.1 months. But some regional differences were recognized within the four districts: i.e. 28.6 (Cheju city), 27.4(Jochun), 27.1(Hallim) and 25.3 months (Sung-up). There was no significant differences in sex ratio of newborns at 50.4%(♂) and 49.6%(♀). 4. The intervals of esturs recurrence and the following conception after calving were 80.0 and 95.8 days respectively. In the followsing conception intervals after cavling, 90.6-97.4 days were average for the herds of Cheju city and Jochun districts, 62.4-69.7 days for those of Hallim and Sung-up districts. 5. The average gestation period was 279.8 days, and was almost identical in the four districts. 6. The calving intervals was 377.7 days in average, and it was shorter than the data recorded for the domestic mainland's herds. Also it was reduce gradually according to the calving frequency.

Effect of vitamin C on pregnancy rate and 8-OHdG levels during heat stress in post-partum dairy cattle

  • In this study the effect of vitamin C administration on pregnancy rates during summer heat stress in dairy cows was examined. A total of 80 Holstein-Friesian cows were divided into control and treatment groups (n = 40 each). Control group animals were given 10 mL isotonic normal saline, and treatment group, Vitamin C (4 mg/kg) on artificial insemination day (day 0) and 4th, 8th and 12th day post insemination. Pregnancy diagnosis was performed on 30th day post insemination by ultrasonography. Blood samples were randomly taken from 11 animals from each group. Serum P4, GSH, MDA and plasma 8-OHdG levels were determined by using ELISA method. Results showed that 8-OHdG levels were lower in treatment group on day 4, 8 and 12 (p < 0.05) compared with the control group. Similarly, pregnancy rate was higher in treatment group (32.5%) than control (22.5%), respectively. However, MDA, P4 and GSH levels were similar in both groups at 4th, 8th and 12th day. A gradual increase in P4, and MDA levels, and a strong positive correlation between 0, 4th (r = 0.54), 4, 8th (r = 0.59) and 8, 12th (r = 0.51) day was found. Similarly, GSH levels also showed positive correlation at days 0, 4th (r = 0.47) and 4, 8th (r = 0.56). However, a strong negative correlation (r = -0.56) between MDA day 0, and GSH day 8 was found. In conclusion, vitamin C application during insemination period in postpartum cows increases pregnancy rate, and reduces oxidative stress metabolite 8-OHdG levels.

EFFECT OF BREEDING LENGTH ON GENETIC IMPROVEMENT IN JAPANESE HOLSTEIN POPULATION

  • The effect of breeding length of sire on genetic progress was examined in the Holstein dairy cattle population in Japan. Genetic progress was extimated by gene flow method. Breeding length of sires directly influences the replacement rates of sires and the selection intensity of sires because there are a fixed number of progeny tested young bulls per year. As breeding length of sires increased, rate of gene flow decreased and average proportions of genes deriving from selected animals had lower asymptotic values. When breeding length was short, average proportions of genes required a longer period to converge to asymptotic values. Changes of Rcow-sire's(sire to breed recorded cows) and Ncow-sire's(sire to breed non recorded cows) breeding length influenced not only transmission of their genes but also that of genes derived from all other selected animals. Irrespective of whether the discount rate was assumed to be 0 or 6%, longer term (${\geq}$ 20 years) expected total genetic improvement was maximized by a sire breeding length of five years. For shorter term assessment(10 years), genetic improvement was maximized by a sire breeding length of three years. There was a linear increase in the contribution of the sire to bulls pathway to the total genetic improvement, with increase in the term of assessment.

Co-culture of In Vitro Matured and Fertilized Bovine Oocytes with Oviductal Epithelium (체외성숙, 체외수정 우 난포란의 Co-culture에 관한 연구)

  • Bovine oocytes obtained from follicles(2~5mm) of ovaries after slaughter were cultured in TCM 199 medium with 10~20% heat-inactivated estrus cow serum(ECS) for 25~27 hr, at 39$^{\circ}C$ under 5% CO2 in air. At the end of culture period, some oocytes were stained with 1% acetoorcein and examined for the evidence of oocyte maturation. The remainder were used to assess the potential of in vitro fertilization(IVF) with frozen-thawed spermatozoa and subsequent development in media with or without bovine oviduct epithelial cell (BOEC) co-culture. The results obtained were summarized as follows ; 1. The maturation rate of oocyte in vitro in TCM 199 medium with 15% ECS group(76.3) was superior to 10% ECS group(68.3%) and 20% ECS group(64.5%). 2. The IVF rates of oocytes matured in vitro, and formation rate of male and female pronuclei were 63.6%(77/121) and 93.5%(72/77), respectively. The incidence of polyspermy was very low(2.4%). 3. Of 73 oocytes fertilized in vitro and cultured in TCM 199 medium with 10% fetal calf serum for 7 days, 41(56.3%) were cleaved over 2-cell and only 1(2.4%) was developed beyond the 16-cell stage. 4. Of 76 oocytes co-cultured with BOEC, 58(76.3%) were cleavaged and 23(39.7%) were developed to morula and blastocyst stage. The results of this study indicate that co-culture with BOEC deserved a positive effect on the IVF oocyte development through the 16-cell block.

Ultrarapid Freezing of Mouse 2-Cell Embryos (생쥐 2-세포기 수정란의 초급속동결)

  • This study was carried out in order to investigate effects of cryoprotectant concentration and equilibration time on survival of ultrarapidly frozen 2-cell mouse embryos. Mouse 2-cell embryos, following dehydration by exposure to DMSO and sucrose, were directly immersed into liquid nitrogen and thawed in 37$^{\circ}C$ water. Viability was defined by development rate to the blastocyst stage after in vitro culture for 72 hours. The results are summarized as follows ; 1. When 0.25M of sucrose was added into the freezing medium at various concentrations of DMSO and dilution medium, higher development rate of embryo was obtained in 3.0M DMSO concentrations (82.6%). However, when sucrose concentraitons of 0.25 and 0.5 M were added to the freezing medium with 3.0 M DMSO and dilution medium, development rate of embryos were 81.7% and 24.1%, respectively. 2. In the equilibration time at room temperature, higher development rate was attained after short period of time (2.5min) in 3.0 M DMSO+0.25 M sucrose (85.9%). 3. The development rate of embryos at in vitro 2-cell, in vitro 2-cell, solution control and untreated control was 84.6%, 90.9%, 89.9%,, and 89.7%, respectively.

Comparison of Brood Productions in the Cold-Treated Pairing vs. Not Cold-Treated Pairing in a Stenochronous Spider

  • To understand whether experience of cold season in reproductive behaviors in the adults of Amaurobius ferox, the paired adults of a female and a male were exposed under 'cold-treated environment' and 'not cold-treated environment', respectively. I investigated effects of the cold treatment on the brood production of A. ferox. In not cold treatment in which male-female pairs were formed in October at a temperature of $20^{\circ}C$ (${\pm}2^{\circ}C$) and continuously kept under not cold-treated environment, only 3 of 50 pairs successfully reproduced (reproduction was defined as the emergence of spiderlings). In cold treatment where individuals were kept in cold conditions for 3 months prior to pair formation, 57 out of 60 couples succeeded in reproducing. Females which did not experience the low temperature displayed strong aggressiveness toward males. This behavioral inhibition might the primary barrier to copulation of A. ferox that decrease following a period of low temperature. The reproductive inhibition might help the females to allocate the maximum amount of energy in a given environment to reaching the adult stage and to delay reproduction in unfavorable wintering conditions.
