Knockout of Myostatin by Zinc-finger Nuclease in Sheep Fibroblasts and Embryos

  • Myostatin (MSTN) can negatively regulate the growth and development of skeletal muscle, and natural mutations can cause "double-muscling" trait in animals. In order to block the inhibiting effect of MSTN on muscle growth, we transferred zinc-finger nucleases (ZFN) which targeted sheep MSTN gene into cultured fibroblasts. Gene targeted colonies were isolated from transfected fibroblasts by serial dilution culture and screened by sequencing. Two colonies were identified with mono-allele mutation and one colony with bi-allelic deletion. Further, we introduced the MSTN-ZFN mRNA into sheep embryos by microinjection. Thirteen of thirty-seven parthenogenetic embryos were targeted by ZFN, with the efficiency of 35%. Our work established the technical foundation for generation of MSTN gene editing sheep by somatic cloning and microinjection ZFN into embryos.

이동통신망에서 재생산 단계를 적용한 채널할당 (Channel Allocation Using Mobile Station Network in Reproduction Stage)

  • 이동통신망에서는 이동국에서 채널할당 요청이 있을 때 교환국에서 각 기지국에 속한 이동국에 채널을 할당한다. 채널을 할당하는 방식에는 고정채널할당방식과 동적채널할당방식이 있으며 이를 조합한 하이브리드방식이 주류를 이룬다. 주파수를 잘 할당한다는 것은 그만큼 자원을 효율적으로 사용하고 고객에게 양질의 서비스를 제공하는 길이기도 하다. 본 논문에서는 채널을 할당할 때 채널간 간섭을 최소로 하고 채널을 할당하기까지의 시간과 횟수를 최소화하는 방안을 적용한 방식을 제안한다. 이를 구현하기 위해 유전 알고리즘의 프로세서 단계인 검증단계를 통하여 재생산 단계를 거치므로 제안 방식의 정확성과 효율성을 나타낸다. 또한 시뮬레이션을 통해 다른 방식과 비교 검토하여 제안 방식의 효율성을 검증한다.

더러브렛 암말의 번식기 발정상태가 번식효율에 미치는 영향 (Effects on Reproduction Efficiency of Estrous Status in Thoroughbred Mares During the Breeding Season)

  • The purpose of this study was to ascertain the breeding efficiency in Thoroughbred mare. A total of 106 mares were investigated for the status of follicle (462 cases), ovulation (179 cases) and pregnancy (346 cases). Of total examination, 46.8% was follicle measure to determine breeding time, and mating rate per cases examined was 39.9%. There was no correlation between reproductive results and size of follicles or endometrial edema or degrees of teasing alone. 143 cases were ovulated among 179 cases which were performed ovulation examination, and ovulation rate and fertilization rate per mating times were 79.9% and 39.0%, respectively. The use of hCG(human chorionic gonadotropin), to facilitate ovulation, presented to increase occurrence of double ovulations and twin fertilizations In conclusion, though more examination to estimate the optimal breeding time and higher mating rate was performed, fertilization rate per mating times was lower and then reproductive efficiency also became decreased. Therefore, it seemed that accurate examination of reproductive tracks, appropriate teasing programme and hCG administration before ovulation were of help to improve ovulation rate and fertilization rate.

우유내 요소태 질소 농도와 관련된 우군의 번식성 (Herd Reproductive Performance Related to Urea Nitrogen Concentration in Bulk Milk)

  • The objective of this study were to compare two methods of analysis of milk urea nitrogen (MUN) and to relate conentration of MUN to pregnancy rate in dairy cows. The Azotest and Sigma method for MUN analysis were compared. The Azotest consistently underestimated the Sigma values for same samples, except for those in rank 3. The CV for the Azotest, within each rank, were less than 15%, demonstrating acceptable repeatability of the Azotest within a rank. Concentrations of MUN higher or lower than MUN in rank 3 and 4 were associated with decreased pregnancy rate. Mun concentrations in rank 5 and 6 were associated with a, pp.oximately a 16 percentage point decrease in pregnancy rate. Mun concentrations in rank 5 and 6 were associated with a, pp.oximately a 16 percentage point decrease in pregnancy rate after AI in dairy cow. This results suggest that Azotest may be benificial to dairy producers to monitor urea nitrogen concentration in their herd in efforts to maintain or improve reproductive efficiency.

Factors Involving Reprogramming in Cloned Embryos

  • Although nuclear transfer (NT) techniques are used to clone animals, its efficiency is very low. Moreover, nuclear transfer has resulted in offspring with severe developmental problems, probably due to incomplete nuclear reprogramming. Nuclear reprogramming is characterized by functional modification of the transferred nucleus to allow it to direct normal embryo development with the potential to grow to term. Although the nature of the reprogramming factor(s) in mammals is not clear, various nuclear as well as cytoplasmic components are involved in the processes. In this article we review recent data on factors involved in the nuclear reprogramming of cloned embryos.

Novel Reproductive Techniques in Swine Production - A Review

  • The main objective of modern reproductive technologies in pig reproduction is to increase reproductive efficiency and rates of genetic improvement. They also offer potential for greatly extending the multiplication and transport of genetic materials and the conservation of unique genetic resources in reasonably available forms for possible future use. The development and refinement of these technologies is concentrating on gamete and embryo collection, sorting and preservation, in vitro production of embryos, culturing, manipulation of embryos (splitting, nuclear transfer, production of chimeras, establishment embryo stem cells, and gene transfer) and embryo transfer. Also, the development of these novel technologies is facilitated by modern equipment for ultrasonography, microscopy, cryopreservation, endoscopy, and flow cytometry, microinjectiors, micromanipulators and centrifugation. The real impact on herd productivity will come from combining new reproductive techniques with powerful DNA technologies. The new reproductive techniques will allow a rapid turnover of generations, whereas the DNA technology can provide selection, which does not need phenotypic information when the selection decisions are made.

  • Minipigs are regarded as one of the most important laboratory animal in that anatomical and physiological properties are similar to human and their reproduction efficiency is relatively higher compared to other large animal species. Particularly, several diseases that cannot be mimicked in rodent models are successfully occurred or induced in pig models therefore it has been interested in a valuable model for human diseases. Pigs are also 'standard' species in xenotransplantation research. To maximize experimental outcome using minipigs, establishment and management of proper animal facility, right animal husbandry and control of pathogens are very important. In this review, we summarized several international guidelines related with minipigs published by several companies or governments and discuss optimal conditions for providing informative ideas to the researchers who want to use minipigs in their future studies.

  • As the high density memory is widely used in the various applications, the need for reproduction of memory is increased. In this paper we propose an efficient fault diagnosis algorithm of linear order O(n) that enables the reproduction of memory. The new algorithm can distinguish various fault models and identify all the cells related to the faults. In addition, a new BIST architecture for fault diagnosis is developed. Using the new algorithm, fault diagnosis can be performed efficiently. And the performance evaluation with previous approaches proves the efficiency of the new algorithm.

Reproduction-Nutrition Relationship in Dairy Buffaloes. I. Effect of Intake of Protein, Energy and Blood Metabolites Levels

  • Fifty one Nili-Ravi dairy buffaloes in their last two months of gestation were selected. After parturition, rectal examination of reproductive organs was carried out until the occurrence of the first oestrus (PEI). Milk samples were analyzed for milk progesterone levels (MPL). Ovulation (POI) was confirmed by rectal palpation and MPL. Feed and blood samples were collected fortnightly and analyzed. Body condition score (BCS) was recorded on a scale of 0 to 5. Crude protein (CP) intake varied among different seasons and correlated positively with serum urea levels, POI (p<0.01) and PEI (p<0.05). Excess CPI was lower in the group showing oestrus as compared to those remaining as anoestrus (p<0.05). The dietary ratio of crude protein - metabolizable energy (CP:ME) in the oestrus animals was narrow and constant, while the anoestrus animals had a widely fluctuating one. In normal breeding season (NBS) calvers, mean serum urea level (SUL) was lower than the low breeding season (LBS) calvers. SUL was positively correlated with PEI and POI (p<0.01). Up to six months postpartum, SUL were constantly higher in anoestrus than oestrus buffaloes. Mean metabolizable energy (ME) intake was lower in the NBS calvers than the LBS calvers (p<0.01). BCS and postpartum ovulation interval were correlated with ME intake (p<0.01). Prepartum ME intake was higher in oestrous as compared to anoestrous animals (p<0.05). Higher and lower ME intakes were associated with anoestrus, while a moderate energy intake was associated with a PEI of less than 75 days. Buffaloes with poor BCS belonged to the LBS calving group and most of the NBS calving buffaloes had good BCS. BCS was negatively correlated with PEI (p<0.01) and was higher in oestrous buffaloes than anestrus. It was concluded that excess intake of crude protein, associated with higher serum urea levels and low energy intake, associated with poor body condition, are the key factors for low reproductive efficiency. It may be corrected by adopting a proper feeding strategy.

한우의 번식 효율 향상을 위한 자궁 내 세균 분석 (Analysis of Uterine Bacteria to Increase Reproductive Efficiency in Hanwoo(Korean Native Cattle))

  • The objective of this study was to evaluate several types of uterine bacteria in Hanwoo. uterine bacteria from randomly selected 5 uterus was collected by flushing methods into a sterilized 1.5 ml centrifuge tube and was inoculated onto MacConkey agar and blood agar, respectively. After being incubated for 5% $CO_2$, aerobic or anaerobic condition at $37^{\circ}C$ during 48h, bacterial colonies were selected and re-inoculated onto blood agar plates. Re-cultured colonies were identified by Gram staining and finally identified using Vitek system. The identified bacteria were Staphylococcus lentus, Staphylococcus sciuri, Staphylococcus vitulinus, Staphylococcus warneri of Gram (+) and Rhizobium radiobacter, Sphingomonas paucimobilis of Gram (-) bacteria. Although, pathogenicity of identified bacteria was unclear, the bacteria can have an effect on the uterine microenvironment. Therefore, repetitive research will be required to determine the effects of bacteria in cattle exposed to a various environment.
