  • Phenxoy compounds, 2,4-dichlorophenol acetoxyacid (2,4-D) and 2,4-dichlorophenol (DCP), are widely used as a hormonal herbicide and intermediate for pesticide manufacturing, respectively. We have previously reported the potential of these compounds as androgenic endocrine disruptors using in vivo Hershberger assay and in vitro reporter gene assay.(omitted)

Caspase-11 Promoter-GFP Construct as a Dual Reporter of Cytotoxicity and Inflammation

  • Caspase-11 has been known as a dual regulator of apoptosis and inflammatory response. An unusual feature of caspase-11 is that its expression is induced by apoptotic or proinflammatory stimuli. Utilizing these unusual features of caspase-11, we have developed a simple and sensitive assay method to screen pro- or anti-apoptotic/inflammatory molecules. To develop this assay method, we generated a reporter construct where GFP expression is regulated by caspase-11 promoter. When several types of cultured cells were transfected with this reporter construct and subsequently treated with various apoptotic or proinflammatory molecules, expression of GFP by the activation of caspase-11 promoter was easily detected by fluorescence microscopy or spectrofluorometry. In addition, a reduction of the GFP fluorescence was detected when an agent reported to suppress caspase-11 induction was applied. These results suggest that our reporter system can be used to screen pro- or anti-apoptotic/inflammatory molecules.

  • 우리는 두 가지의 새로운 luciferase reporter plasmid를 개발하였는데 그 하나는 이 plasmid에 promoter조각을 삽입한 다음에 그 promoter의 활성을 측정하는 용도로 사용 될수 있고, 다른 하나는 매우 낮은 basal promoter 활성을 갖고 있기 때문에 진핵생물의 전사 조절인자의 연구에 도움이 될 수 있다. 후자의 reporter plasmid에는 17염기쌍의 initator 와 Spl,GAL4그리고 일부 Drosophila homeodomain protein의 결합우뷔에 해당하는 cis elements등을 들어 있다.그리고 tranciption termination을 촉진할 수 있는 signal을 initiator앞서 삽입하여 이런 reporter plasmid에 존재할 수 있는 cryptic promoter에서 시작된 transcript가 luciferase reporter cDNA로 진행되는 것을 방지하는 개선된 reporter plasmid를 제조하여 promoter활성을 Drosophila Schneider line 2 cells을 이용한 transient transfection assay방법으로 측정하였다. 여기에 사용한 termination 촉진 signal은 SV40 polyadenylation signal의 3연속 조각(AAA)과 tenscription termination signal이 포함된 것으로 믿어지는 mouse c-mos 유전자의 일부조각 (UMS)이다. 기대한으로 이 두가지의 signal을 삽입하였을 때 basal promoter활성이 최대한으로 감소하였으며 이 두 가지 signal을 삽입된 reporter plasmid를 사용하여 promoter의 활성을 보다 sensitive하게 측정하였다. 이 reporter plasmid는 Droiophlla melanogaiter뿐만 아니라 포유동물을 포함한 고등생물의 전사 조절인자 연구의 한 도구로 사용될 수 있을 것이다.

A Novel Rapid Fungal Promoter Analysis System Using the Phosphopantetheinyl Transferase Gene, npgA, in Aspergillus nidulans

  • To develop a convenient promoter analysis system for fungi, a null-pigment mutant (NPG) of Aspergillus nidulans was used with the 4'-phosphopantetheinyl transferase (PPTase) gene, npgA, which restores the normal pigmentation in A. nidulans, as a new reporter gene. The functional organization of serially deleted promoter regions of the A. nidulans trpC gene and the Cryphonectria parasitica crp gene in filamentous fungi was representatively investigated to establish a novel fungal promoter assay system that depends on color complementation of the NPG mutant with the PPTase npgA gene. Several promoter regions of the trpC and crp genes were fused to the npgA gene containing the 1,034-bp open reading frame and the 966-bp 3' downstream region from the TAA, and the constructed fusions were introduced into the NPG mutant in A. nidulans to evaluate color recovery due to the transcriptional activity of the sequence elements. Serial deletion of the trpC and crp promoter regions in this PPTase reporter assay system reaffirmed results in previous reports by using the fungal transformation step without a laborious verification process. This approach suggests a more rapid and convenient system than conventional analyses for fungal gene expression studies.

A New Reporter Vector System Based on Flow-Cytometry to Detect Promoter Activity

  • In this study, we report the development of a new dual reporter vector system for the analysis of promoter activity. This system employs green fluorescence emitting protein, EGFP, as a reporter, and uses red fluorescence emitting protein, DsRed, as a transfection control in a single vector. The expression of those two proteins can be readily detected via flow cytometry in a single analysis, with no need for any further manipulation after transfection. As this system allows for the simultaneous detection of both the control and reporter proteins in the same cells, only transfected cells which express the control protein, DsRed, can be subjected to promoter activity analysis, via the gating out of all un-transfected cells. This results in a dramatic increase in the promoter activity detection sensitivity. This novel reporter vector system should prove to be a simple and efficient method for the analysis of promoter activity.

Detection of Antistaphylococcal and Toxic Compounds by Biological Assay Systems Developed with a Reporter Staphylococcus aureus Strain Harboring a Heat Inducible Promoter - lacZ Transcriptional Fusion

  • Previously it was reported that promoter of groES-groEL operon of Staphylococcus aureus is induced by various cellwall active antibiotics. In order to exploit the above promoter for identifying novel antistaphylococcal drugs, we have cloned the promoter containing region ($P_g$) of groES-groEL operon of S. aureus Newman and found that the above promoter is induced by sublethal concentrations of many antibiotics including cell-wall active antibiotics. A reporter S. aureus RN4220 strain (designated SAU006) was constructed by inserting the $P_g$-lacZ transcriptional fusion into its chromosome. Agarose-based assay developed with SAU006 shows that $P_g$ in single-copy is also induced distinctly by different classes of antibiotics. Data indicate that ciprofloxacin, rifampicin, ampicillin, and cephalothin are strong inducers, whereas, tetracycline, streptomycin and vancomycin induce the above promoter weakly. Sublethal concentrations of ciprofloxacin and ampicilin even have induced $P_g$ efficiently in microtiter plate grown SAU006. Additional studies show for the first time that above promoter is also induced weakly by arsenate salt and hydrogen peroxide. Taken together, we suggest that our simple and sensitive assay systems with SAU006 could be utilized for screening and detecting not only novel antistaphylococcal compounds but also different toxic chemicals.

Luciferase reporter gene assay를 이용한 단삼(丹蔘)추출물의 소염, 진통작용에 대한 in vitro 연구 (In Vitro Study of Anti-inflammatory and Analgesic Effects of Salvia Miltiorrhiza (SM) Extracts Using Luciferase Reporter Gene Assay)

  • Objectives: In order to identify the anti-inflammatory and analgesic properties of Salvia miltiorrhiza (Dan-Sam), widely used in Korean traditional medicine, an in vitro screening system was designed using pGL3, a luciferase reporter vector, and the tumor necrosis factor (TNF)-${\alpha}$ and cyclooxygenase (COX)-2 as target genes. Methods: The promoter regions of each gene were generated by PCR using the human chromosome as template DNA, and inserted into pGL3 vector with Kpn I and Hind III. The final construct was transfected into human myelomonocytic leukemia cells (U-937) that could be differentiated and activated by phorbol 12-myristate 13-acetate (PMA) or lipopolysaccharide (LPS). Using this system, the anti-inflammatory and analgesic effects of several herbal extracts regarded to have the medicinal effects of diminishing body heat and complementing Qi were tested. The chemicals PD98059 and berberine chloride were used as controls of the transcriptional inhibitors of TNF-${\alpha}$ and COX-2, respectively. Results: Salvia miltiorrhiza (Dan-Sam) demonstrated significant decrease of TNF-${\alpha}$ and COX-2 mRNA in the in vitro assay system. In MTT assay, Salvia miltiorrhiza (Dan-Sam) did not significantly inhibit the survival and proliferation of human myelomonocytic leukemia cells (U-937). Conclusions: Salvia miltiorrhiza (Dan-Sam) was found to exhibit the significant medicinal properties of anti-inflammatory and analgesic effects.
