재조합 인간상피세포 성장인자(rhEGF, DWP401)가 랫드의 수태능, 태자와 신생자 발달 및 모체기능에 미치는 영향 (Fertility and Reproductive & Developmental Toxicity Study on Recombinant Human Epidermal Growth Factor (rhEGF, DWP401) in Rats)

  • This study was conducted to investigate for its effects on reproductive and developmental toxicity of recombinant human epidermal growth factor (rhEGF) in Sprague-Dawley rats. Male rats were administered rhEGF at doses of 1, 10, 100, and 1000$\mu$g/kg/day, respective1y, by subcutaneous injection from 63 days before and throughout to mating period until the day before sacrifice. Female rats were administered rhEGF at the same doses from 14 days before mating to day 20 of gestation or to day 21 of lactation. We examined the male and female fertility indices and maternal toxicity of F0 parental animals. Also, we examined the external, visceral, or skeletal malformation of fetuses, growth and development, behavior, and/or reproductive performance of F1 animals. At the highest dose (1,000 $\mu$g/kg), the mean body weights of F0 animals were significantly increased in males and females at 3 or 2 weeks after treatment, respective1y. No clinical signs and food intakes were observed at any time during the experimental period by rhEGF treatment. In autopsy examination, the relative and absolute liver weights significantly increased in both sexes of 1,000 $\mu$g/kg. At the highest dose (1,000 $\mu$g/kg), there was a statistically significant increase of pregnancy period and the number of dead fetuses. Moreover, significant increase of mean fetal body weight and decrease of number of live fetuses, which related to the difficult dilivery were observed in highest dose group. In Fl examination, no adverse effects on external, visceral, and skeletal malformation, physical and functional development, behavior or reproductive ability of Fl animals were observed in any group. Also, there was no significant difference between control and treated groups in copulation or fertility indices of Fl animals. These results indicate that rhEGF had no adverse effect on fertility and reproductive ability of Sprague-Dawley rats.

사료내 동물성 단백질원들이 조피볼락의 성장과 체조성에 미치는 영향 (Effects of Dietary Animal Protein Sources on Growth and Body Composition in Korean Rockfish, Sebastes schlogeli)

  • 본 연구는 조피볼락 사료에 있어서 동물성 단백질원들의 영양적 평가(체조성과 성장)를 결정하기 위하여 수행되었다. 6가지 실험사료 중 북양어분구(WFM), 넙치근육구(FMM), 오징어간분구(SLP)는 한가지, 혈분구(BM)는 혈분과 북양어분을, 카제인구(CG)는 카제인, 젤라틴 및 북양어분을, 그리고 난단백구(EWA)는 난단백, 젤라틴 및 북양어분을 단백질원으로 이용하여 사료내 조단백질 함량을 50%로, 가용에너지는 15.9 KJ/g (physiological fuel value ; 단백질 : 지방 : 탄수화물 = 16.7 KJ/g : 37.7 KJ/g : 16.7 KJ/g)으로 조정하였다. 성장률, 사료효율 및 단백질효율은 북양어분구보다 넙치육분, 오징어간분구보다 혈분구 그리고 난백구보다 카제인구가 유의적으로 우수한 결과를 보였으며(P<0.05), 혈분구는 북양어분을 혼합하여 혈분의 기호성을 높인 결과로 사료된다. 6가지 실험구의 전어체 분석, HSI, Hb, Hematocrit은 다양하게 유의적인 차이를 나타냈다. 따라서, 상기 결과는 조피볼락에 있어서 동결 건조된 넙치근육분이 가장 좋았고, 난단백은 가장 저조한 성장을 보여 주었다.

Chronic Toxicity of a Combined Preparation of Ticlopidine and Ginkgo Biloba Extract (EGb 761) Orally Administered to Rats for 13 Consecutive Weeks

  • Toxicity of a combined preparation of ticlopidine and ginkgo biloba extract (EGb 761) in a ratio of 10: 4 was examined in male and female Sprague-Dawley rats. Rats were treated with the test substance intragastrically at a dose of 0 mg/kg, 17 mg/kg, 52 mg/kg or 156 mg/kg for 91 consecutive days. No death or abnormal clinical sign was observed throughout the administration period. There was no difference in body weight gain, food intake or water consumption among different dose groups. Test sub-stance-related differences were not observed in urinalysis. In hematological results mean corpuscular hemoglobin (MCH) of low and high dose male group was increased. Prothrombin time of medium and high dose female group was decreased. A significant increase in serum total cholesterol was observed in both sexes of rats treated with a daily dose of 156 mg/kg, but all the other values obtained in serum chemistry appeared to be within normal ranges. A dose dependent increase in the relative liver and kidney weights was observed in both male and female rats. There were no gross pathological findings at terminal sacrifice. Microscopic histopathological examination did not show any lesion associated with administration of the test substance. The results suggest that under the conditions employed in this study no observable effect level (NOEL) of the test substance be greater than 17 mg/kg/day, but less than 52 mg/kg/day.

수용성 님추출물이 랫드의 간 독성에 미치는 영향 (Effects of Aqueous Azadirachta indica Extract on Hepatotoxicity in Rats)

  • 님추출물은 유효성분(active ingredient)으로 azadirachtin을 함유하여 전세계적으로 충해방제용 유기농업자재로 널리 사용되고 있다. 그러나, 님추출물은 님 원료부위 및 추출용매에 따라 종류가 매우 다양하고 안전성에 크게 차이가 난다고 보고되고 있다. 본 연구에서는 우리나라에서 유기농업자재 제품의 원제로 사용되는 수용성 님추출물이 주요 독성기관인간에 미치는 영향에 대해서 살펴보고자 SD 랫드를 이용하여 4주 반복경구독성시험을 수행하였다. 시험결과, 님추출물 시험물질의 투여 농도가 증가함에 따라 간의 상대중량이 증가하였다(p <0.05). 혈액 생화학분석 결과, 수컷에서 대조군에 비해 시험물질 처리시 혈중 LDH는 감소하였으나 GOT와 GPT가 증가하였으며(p <0.05), 특히 GPT가 시험물질에 농도 의존적으로 증가함에 따라 수컷에서 간 손상의 가능성을 나타내었다. 또한, 고농도로 님추출물 시료를 처리한 경우 수컷의 혈중 GGT가 급격히 증가하였으며 혈중 GLU도 유의적으로 증가하였으나(p <0.05), 뇨 중 GLU는 님추출물의 처리농도증가에 따른 변화가 나타나지 않았다. 간의 조직병리학적 변화를 살펴본 결과 님추출물 시료 처리에 따른 간의 병변도 확인되지 않았다. 따라서 본 시험에 사용된 수용성 님추출물 시료는 혈액 생화학적 분석 결과 수컷에서 간손상의 가능성은 있었으나 조직병리학적 변화는 관찰되지 않았다. 따라서, 수용성 님추출물 2.0 g/Kg 을 4주간 랫드에 경구투여한 결과 간에 독성을 미치지 않고 안전한 것으로 판단된다.

아플라톡신을 간회 투여한 랫드의 간에서 CYP450 1A1, p53의 발현과 DNA adduct의 형성 (CYP450 1A1 and p53 expression and DNA adduct formation in the liver of rats treated with a single dose of aflatoxins)

  • Aflatoxins are produced mainly by Aspergillus flavus and Aspergillus parasiticus that grow in improperly stored cereals. Aflatoxin B1 ($AFB_1$) is a potent hepatocarcinogen in a variety of experimental animals including human beings. In spite of a high attention to the hepatocarcinogenecity of $AFB_1$, the relative toxicity of aflatoxins ($AFB_2$ and $AFG_1$) is not fully clarified. Sprague-Dawley male rats were orally administered with $AFB_1$, $AFB_2$, and $AFG_1$ at the dose of 250 ${\mu}g/kg$ (additionally including a dose of $1250{\mu}g/kg $ for $AFB_1$) body weight. Animals were then killed at 12, 24 or 48 hrs following aflatoxin exposure. Subsequently the immunohistochemical examination of p53, cytochrome p450 1A1 (CYP450 1A1), and glutathione-S-transferase placental form (GST-P) were performed. The level of the 8-OxodG in the liver was determined. Expressions of CYP450 1A1 and p53 were high in the liver of rats through 48 hrs after treatment of $AFB_1$ at the single dose of $250{\mu}g/kg $. This pattern was more clear as increasing doses. The treatment of $AFB_2$ and $AFG_1$ did not affect the expression of CYP450 1A1 but it caused weak expression of p53. The activity of GST were not found in the liver of rats treated with aflatoxins. The formation of 8-OxodG by $AFB_1$ increased in a dose-dependent manner up to 24 hrs after a single treatment of $AFB_1$ thereafter decreased to the level of control. The treatment of $AFB_2$ and $AFG_1$ did not affect the levels of 8-OxodG in the liver of rats with increasing time. These results in the present study indicate that $AFB_1$ among aflatoxins with low comparable levels is the most toxic as determined by early biomarkers such as CYP450 1A1, p53, GST-P, and 8-OxodG.

Genistein과 Daidzein 급여가 제2형 당뇨동물모델의 적혈구와 조직 중의 항산화방어계에 미치는 영향 (Effect of Genistein and Daidzein on Antioxidant Defense System in C57BL/KsJ-db/db Mice)

  • 제2형 당뇨 동물모델(C57BL/KsJ-db/db)을 대상으로 대두 이소플라본의 주성분인 genistein과 daidzein의 항산화효능을 검증하고자 5주령의 수컷 C57BL/KsJ-db/db 마우스와 그의 이형접합체인 C57BL/KsJ-db/+ 마우스를 2주간 환경에 적응시킨 후 비당뇨군(db/+), 당뇨대조군(db/db), genistein 급여군(db/db-genistein), daidzein 급여군(db/db- daidzein)으로 나누어 6주간 사육하였다. 실험동물의 간, 부고환지방과 신주변지방의 조직무게는 당뇨군(db/db)이 비당뇨군(db/+)에 비해 유의적으로 높았으나, 심장무게는 유의적으로 낮았다. Genistein과 daidzein 급여는 장기무게 변화에 영향을 미치지 않았다. 적혈구의 SOD와 CAT활성은 혈당과 양의 상관성을 보였으나 GSH-Px활성은 음의 상관성을 나타내었다. 따라서 SOD와 CAT활성은 db/db군이 db/+군에 비해 유의적으로 높은 반면, GSH-Px 활성은 유의적으로 낮았다. Genistein과 daidzein 급여로 db/db군의 증가된 CAT활성은 감소되었으며 GSH-Px활성은 높게 나타났다. 적혈구의 GSH함량은 당뇨군들이 비당뇨군에 비해 유의적으로 높았으나 genistein과 daidzein에 의한 영향은 관찰되지 않았다. 간, 신장 및 심장조직 내 SOD활성은 유의적인 변화가 없었으나 간조직 중 CAT와 GSH-Px활성과 신장조직 중의 GSH-Px활성은 db/db군이 db/+군에 비하여 유의적으로 높게 나타난 반면 신장조직 중의 CAT활성과 심장조직 중의 CAT와 GSH-Px활성은 낮았다. 그러나 genistein과 daidzein 급여는 고혈당으로 인한 조직 내 CAT와 GSH- Px활성을 유의적으로 개선하였다. 적혈구를 비롯하여 모든 조직 내 지질과산화물 함량은 db/db군이 db/+군에 비하여 유의적으로 높았으나 genistein과 daidzein 급여로 간, 신장과 심장조직 중의 지질과산화물 생성이 유의적으로 억제되었다. 이와 같이 genistein과 daidzein은 제2형 당뇨동물에서 고혈당으로 야기되는 적혈구와 조직 내 항산화효소 변화를 완화하고 간, 신장 및 심장조직의 지질과산화물을 낮추는 것으로 관찰됨으로써 이들의 항산화작용을 통한 당뇨 합병증을 예방할 것으로 사료된다.

가시오갈피와 두충의 첨가 급여가 브로일러의 생산 능력, 혈장 생화학 지표 및 텔로미어 함량에 미치는 영향 (Effect of Dietary Siberian Ginseng and Eucommia on Broiler Performance, Serum Biochemical Profiles and Telomere Length)

  • 가시오갈피와 두충은 강력한 항산화 작용을 가진 약용 식물로 알려져 있다. 본 연구에서는 상업용 브로일러를 대상으로 가시오갈피 잎과 두충 잎의 첨가 급여(0.5% 및 1%)가 개체의 생산 능력, 장기 무게, 혈장 생화학적 성분 및 텔로미어 함량에 미치는 영향을 알아보고자 하였다. 시험 전체 기간($4{\sim}35$일) 동안 체중, 증체량 및 사료 섭취량에서는 처리구 간 유의적 차이를 보이지 않았으나, 사료 요구율에 있어서는 가시오갈피 첨가 급여구가 대조구 및 두충 첨가구에 비해 다소 저조한 결과를 나타내었다. 장기 무게에 있어서 간, 비장 및 흉선의 무게는 처리구 간 차이가 없었으나, F낭(bursa of Fabricius)의 무게는 가시오갈피 및 두충 첨가 급여가 대조구에 비해 유의적으로 높게 나타났다. 혈액 생화적 성분 분석에서는 혈장 내 albumin, AST, ALT 및 total protein은 대조구와 처리구 간에 거의 비슷한 수치를 나타내었으나 triglycerides, cholesterol 및 gluocose 수준은 생리활성 물질 첨가 급여구가 대조구에 비해 높게 나타났다. 가시오갈피 및 두충 급여에 따른 각 조직 별 텔로미어 함량 분석에 있어 백혈구 내 텔로미어 함유율은 모든 첨가 급여구가 대조구에 비해 현저히 높은 함량을 보이나 간, 심장, 정소 조직 간에는 처리구별 차이가 없었다. 이상의 결과로부터 브로일러에 가시오갈피와 두충의 첨가 급여는 생산의 능력 저하 없이 개체의 면역 활성을 증진시키고, 텔로미어 길이의 감축을 상대적으로 완화시키는 것으로 판단된다.

다이옥신계 TCDD(2,3,7,8-Tetrachlorodibenzo-p-dioxin)에 노출된 흰쥐의 지질대사 및 면역관련 인자에 대하여 키토산 올리고머의 분자량별 섭취효과 (Effects of Various Chitosan Oligomer Molecular Weight Levels on the Disorders of Lipid Metabolism and Immune-related Factors in Rats Treated 2,3,7,8-Tetrachlorodibenzo-p-dioxin)

  • 다이옥신계 TCDD에 노출된 흰쥐의 지질대사 및 면역관련 인자에 대하여 키토산 올리고머의 분자량에 따른 섭취효과를 알아보기 위해 Sprague-Dawley rat(수컷, 6주령)를 이용하여 TCDD를 40 ${\mu}g$/kg BW로 복강 내 주입하고 키토산 올리고머 분자량을 1000 이하, 1000~3000 및 5000~10000으로 수준을 달리하여 4% 첨가하고 3주간 실험하였다. 실험동물의 장기의 무게에서 간의 상대중량은 TCDD 투여군에서 모두 유의적으로 높았다. 뇌와 고환의 상대중량은 대조군과 TCDD 투여군에 비하여 TCDD 투여와 함께 키토산 올리고머를 섭취한 군에서 모두 현저히 증가하였다. 흉선의 상대중량은 대조군에 비하여 TCDD 투여군에서 모두 약 1/4 정도로 감소되었다. 혈액성분에서 적혈구의 수는 TCDD 투여에 의한 감소가 TCDD 투여와 함께 키토산 올리고머를 섭취한 군 모두에서 대조군 수준으로 증가되었고, 백혈구의 수와 헤모글로빈 및 헤마토크리트값은 TCDD 투여에 의한 감소가 TCDD 투여와 함께 키토산 올리고머의 분자량이 1000 이하인 것(COST)과 5000~10000인 것을 섭취한 군(COLT)에서 회복되는 경향을 보였다. 간 기능에 지표가 되는 효소인 혈청 GOT와 ALP는 대조군에 비하여 TCDD 투여군 모두에서 높은 경향을 보였다. 혈청 총콜레스테롤 및 HDL-콜레스테롤함량은 TCDD 투여에 의한 증가가 TCDD 투여와 함께 키토산 올리고머 분자량이 5000~10000인 것을 섭취한 군(COLT)에서 유의적으로 감소되어 콜레스테롤 억제효과가 나타났다. 간 지질에서는 총 지질과 콜레스테롤 및 중성지방이 유사한 경향으로 대조군에 비하여 TCDD 단독투여군에서 현저한 증가를 보여 지질축적이 되었는데 TCDD 투여와 함께 키토산 올리고머를 섭취한 군들에서 현저한 지질 억제효과를 내었다. 특히 TCDD투여와 함께 분자량 1000 이하의 키토산 올리고머를 섭취한 군(COST)이 TCDD 단독 투여 군에 비하여 간 콜레스테롤을 2/3 정도로, 간 중성지방을 1/2 정도로 뚜렷이 억제하였다. 변 중의 총 지질과 콜레스테롤함량은 TCDD 투여로 증가되는 경향이었고 TCDD 투여와 함께 키토산 올리고머 첨가한 군들에서는 TCDD군보다 대체로 낮은 경향이었다. 담즙산 배설은 대조군에 비하여 TCDD 투여군 모두에서 현저하게 낮았다. 간의 현미경적 관찰에서는 TCDD 투여에 의한 간세포의 지방구 침윤과 세포크기의 다양성, 간세포의 과염 색상과 공포변성이 TCDD 투여와 함께 키토산 올리고머를 섭취한 군 모두에서 현저하게 회복되었다. 혈청의 면역인자인 IgA와 IgM의 농도에서는 대조군과 TCDD 단독투여 군보다 TCDD 투여와 함께 키토산 올리고머의 분자량 5000~10000인 것을 섭취한 군(COLT)이 유의적으로 높은 농도를 나타내었다. 이와 같은 결과로 키토산 올리고머의 분자량이 1000 이하인 것은 혈구수의 회복과 간 지질억제에 효과적이었고, 분자량이 5000~10000인 것은 혈구수의 회복과 함께 혈청 콜레스테롤 저하 및 면역인자들 수준을 증가시키는 경향이 있었다. 특히 간의 조직병변이 키토산 올리고머 식이에 의해 정상으로 회복되는 경향을 관찰할 수 있었다. 그런데 키토산 올리고머의 분자량별 효과에 대하여는 앞으로 더 많은 연구가 필요한 것으로 사료된다.

어린이의 식습관(食習慣)이 체위(體位)에 미치는 영향(影響)에 관한 연구(硏究) (The Influence of Food Habits on Body Stature of Children)

  • The Purpose of this study was to determine every possible correlation between food habits and body statures of primary school children, aged ten years old. This study was conducted from July to October of 1975. In July, prepared questionaires concerning life style, anthropometry, food preference, and food behavior were distributed through school teachers to 425 children; 219 boys and 206 girls, in the 5th grade of three elementary schools in the city of Seoul. Then, in October, when subjects had been classified into underweight/obese by statistical analysis, mothers of obese or underweight children were interviewed by the authors to determine weaning history, daily food consumption of their children, and opinions of various snacks for children. Analysis of results in terms of correlation coefficient, chisquare test and percentage calculations, are as follows: 1. Physical growth and development Boys Girls Height (cm) $134.8{\pm}5.74\;134.4{\pm}5.97$ Weight (kg) $30.0{\pm}4.27\;29.5{\pm}5.16$ Chestgirth (cm) $64.1{\pm}3.59\;63.3{\pm}3.81$ Arm circumference (cm) $18.3{\pm}1.61\;18.2{\pm}1.70$ Triceps skinfold thickness (mm) $10.9{\pm}5.13\;12.7{\pm}4.86$ Various indices of nutrition such as relative weight, relative chestgirth, $R{\ddot{o}}hrer's$ index, Kaup index, Vervaeck index were determined. 2. Food habits 1) Food $preference{\cdots}{\cdots}A$ varying number of foods were selected from 60 items were accepted. It was found that the food which children liked best was fruit and snacks were popular one. Lowest ranking among LIKED foods were from strongly flavored vegetables and organ meat. In general, girls had more food dislikes than did boys. Selected as liked foods were fruits, rice noodle soup, biscuits, and peanuts. Disliked foods were liver, green onions, onions, green pepper, mushrooms, oysters, shellfish, and pork. Items which children never ate before were liver, mushrooms, fish cake, boiled rice mixed with sorghum, mayonnaise, and fresh water firsh. Reasons which children gave for dislike were undesirable flavor and odor. 2) Food $behavior{\cdots}{\cdots}It$ was found that boys liked sweet and salty flavors more than did the girls who more often liked sour flavor. The majority of children enjoyed evening meals more than lunch and breakfast. A number of children skipped breakfast because of lack of appetite or lack of time before going to school. Snacks were the most popular food, especially among girls. Their snacking habits were formed by the encouragement of the mother, and the relieve boredom. Educational backgrounds of mothers and family economical levels of children were remarkable correlated with mothers' attitudes toward feeding of children. There were several interesting findings relating body stature to some other responses; such as that the obese child has a small number of brothers, higher birth order, higher educated mothers and higher family cultural background. It was also discovered that food perference, except for fat and oil group foods was not related to body stature. Sweet taste was liked best and pepperly taste was mostly disliked. Sour taste was popular in the group of underweight. Underweight children were more finicky, disliked snacking, and didn't get much attention from their mothers. 3) Correlation between body stature and nutrition during their infancy. The majority of children, both from obese and underweight, were breast fed as infant. Twenty five per cent of obese children and 17.4 per cent of underwight children started weaning at $1{\sim}6$ months old. The most popular supplemental food of weaning was cereal gruel for the obese group, while boiled white rice and cereal gruel were most common for the underweight group. Highly significant relationships were found between stature of parents and their children. In the obese group 47.8 per cent of fathers and 45.9 per cent of mothers were overweight; however, none of the fathers and only one mother was found to be underweight. In daily food consumption patterns, meals consisting of bread or noodle were popular in the obese group but disliked by the underweight group. The study found clear contrast in that the obese group liked meat and fish while the underweight group liked fruits and vegetables, especially kimchee. The obese children desired to eat cereal foods, milk and milk products, and fat foods while the underweight group desired to eat fruits and vegetables. Frequency of snacks per day was much greater in the obese group. Foods which mothers believed to be good for the health were carrots, cucumbers, fruits, milk, potatoes, sweet potatoes, and breads, while sweet foods such as candies, drinks. chocolate were considered not good for the teeth. Watching television was not significantly related to statures of children. Most significant relationships were found beween frequencies of family guest meals/and eating-out, and children's statures. Nutritional problems which have been considered for the malnourished children in addition to those of children who have a tendency toward obesity, must be taken into consideration in the development of proper nutrition education through the channels of regular school teaching and teaching by parents in the homes. Korean standards of anthropometric measurements for children should be revised, current measurements show much higher figures than present Korean standards.

β-용혈성 Streptococcus iniae 불활화백신의 넙치에 대한 효능 평가 (Evaluation on efficacy of β-hemolytic Streptococcus iniae vaccine on olive flounder, Paralichthys olivaceus)

  • Olive flounder, Paralichthys olivaceus is one of the most important cultured fish in Korea, its farming has been negatively impacted by viral, bacterial and parasitic diseases. Streptococcal infection was considered as a serious problem because of significant economic losses in olive flounder farm industry. The development and evaluation of vaccine for protection against infection by this agent were required. We evaluated the safety and efficacy of ${\beta}$-hemolytic Streptococcus (S.) iniae vaccine on olive flounder Three hundreds of flounders (weight $119.8{\pm}20.7g$, body length $22.6{\pm}1.4cm$) were reared in 0.5 tons aquaria in land-marine tank system. Seawater was provided from the sea of Inchon in Korea, and water temperature was set to $22^{\circ}C$ and $25^{\circ}C$ in the vaccination and challenge test, respectively. We used the formalin-inactivated ${\beta}$-hemolytic S. iniae (F2K) vaccine (M VAC INIAE; Kyoritsu seiyaku, Japan) originated in Japan. The vaccine was intraperitoneally administered to fish. Both of vaccinated group and control group were challenged with intraperitoneally injection by virulent S. iniae SI-36 isolates with $1.0{\times}10^7CFU/fish$ at 3 weeks after vaccination. Difference on mortality of control and vaccinated group (90.0 and 15.0%, 76.5 and 8.0% respectively) at two trials were found significant (p<0.05), and relative percent survival were 83.4% and 89.5%, respectively. The dead fishes were showed dark pigmentation of skin, abdominal extension, hemorrhagic ascites, and liver necrosis, and isolated the S. iniae strain from ascites, liver and kidney. We confirmed the safety and efficacy of ${\beta}$-hemolytic S. iniae vaccine by determinations of the optimal management condition and artificial challenge test in olive flounder.
