Objectives This study is designed to investigate the effects of Jujadokseo-hwan on the brain ability and inducing oxidative stresses. Methods We measured the changes of regional cerebral blood flow and mean arterial blood pressure. Then we analyzed histological examination, immunohistochemistric response and anti-oxidant activity of Jujadokseo-hwan. Results 1. Treatment of Jujadokseo-hwan significantly increased regional cerebral blood flow but decreased mean arterial blood pressure. 2. Treatment of Jujadokseo-hwan-induced increase of regional cerebral blood flow was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. 3. In histological examination through TTC stain, group I was no change, but group II showed that discolored in the most cortical part. Group III showed that decreased discolor in the cortical part. 4. In immunohistochemistric response of BDNF, group II showed that lower response effect. Group III showed that increase response effect. 5. Treatment of Jujadokseo-hwan increased proliferation rates of Glial cell effectively 6. Treatment of Jujadokseo-hwan accelerated proliferation rates of C6 cells in vitro. In addition, protective effects on cell death induced by paraquat, rotenone and hydrogen peroxide. In addition, activity of SOD were increased by treatment with Jujadokseo-hwan. Conclusions In conclusion, Jujadokseo-hwan can improve of the brain ability, learning ability, memory ability and induce ischemic brain injuries.
CheonghunHwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CHTS was CHT adding Schizonepetae Herba. The effects of CHTS on the cerebral blood flow and blood pressure is not known. The purpose of this Study was to investigate effects of CHTS on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(BP), action-mechanism of CHTS-induced changed-rCBF and BP. The changes of rCBF and BP was determinated by Laser-Doppler Flowmetry(LDF). The results were as follows ; CHTS extract was increased significantly rCBF in a dose-dependent, but was not changed BP compared with CHTS non-treated group. Pretreatment with propranolol, indomethacin and methylene blue were inhibited CHTS induced increase of rCBF, propranolol(all CHTS-treated group) and indomethacin(CHTS 0.01 mg/kg) of them were significantly decreased. Pretreatment with propranonol and indomethacin were inhibited CHTS induced increase of BP, but pretreated with methylene blue was significantly accelerated BP in high dosage. This results suggest that CHTS increased rCBF by dilating pial arterial diameter and the action of CHTS is also mediated by adrenergic β -receptor and cyclooxygenase.
This study was performed to provide basic data that predict the application of Saengmaegsan(SMS) as medicinal food. SMS has been used in oriental medicine for many years as a therapeutic agent for cerebral disease. We examined the effects of SMS on physiological function in isolated abdominal aorta and femoral artery from rabbit and measured the changes of regional cerebral hood flow(rCBF), which was continually monitored by laser-doppler flowmeter and pressure transducer in anesthetized adult Spargue-Dawley rats through the data acquisition system composed of MacLab and Macintosh computer. The contraction forces by injection of norephinephrine in isolated abdominal aorta and femoral artery were significantly decreased in each concentration of SMS treatment compared with control. rCBF was increased by SMS in a dose-dependent manner. These results suggest that SMS causes a diverse response of rCBF and arterial diameter. These mechanism in rCBF increase may be mediated by prostaglandis, cyclic GMP and adrenergic $\beta-receptor$. Also mechanism in artery contraction decrease is also mediated by prostaglandis and cyclic GMP. These results indicate that SMS can be nsed as a safe and clinically applicable as a supplementation of diet therapy for cerebral cardiovascular disease patients.
The study was designed to investigate the effects of Leonuri herba extract (LHE) on the change of regional cerebral blood flow (rCBT and cytokines production (IL-1${\beta}$, TNF-${\alpha}$, IL-10, TGF-${\beta}$) in ischemic rats. The results were as follows : The rCBF was significantly and stably increased by LHE (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. In cytokine production of serum by drawing from femoral arterial blood at 1 hr after middle cerebral arterial occlusion, experimental group (LHE 10 mg/kg treated group) was significantly decreased IL-l${\beta}$ production, and increased TGF-${\beta}$ production compared with control group. In cytokine production of serum by drawing from femoral arterial blood at 1 hr after reperfusion, experimental group was significantly decreased IL-1${\beta}$ production compared with control group. IL-10 and TGF-${\beta}$ production of sample group were significantly increased compared with control group. These results suggested that LHE was significantly and stably increased regional cerebral blood flow by inhibited IL-1${\beta}$ production, and accelerated IL-10 and TGF-${\beta}$ production.
Herpes simplex encephalitis (HSE) is one of the fulminant necrotizing, often fatal sporadic form of the encephalitis caused by herpes simplex type I virus. Characteristically, there is early and almost constant involvement of one or both temporal lobes, although there are common additional areas of involvement. Appropriate early treatment following correct diagnosis by clinical findings, CSF study, EEG and several radiological studies including angiography, redionuclide studies, CT or MRI can reduce its mortality and severity of the sequelae. We report a case of HSE diagnosed by adjuvant study of $^{99m}Tc-HMPAO$ regional cerebral blood flow SPECT, which showed a marked increase in bitemporal cerebral blood flow in a 24-year-old man.
Objectives : The purpose of this study is to investigate the effects of $GB_{41}$ supplementation and $BL_{66}$ draining on changes in cerebral blood flow in normal rats. Methods : Regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in normal rats are observed, and those mechanisms were also investigated with pre-treatment of indomethacin(IDM) and methylene blue(MTB) each. Results : In this study, $GB_{41}$ supplementation and $BL_{66}$ draining elevated rCBF in time-dependent manner. Pre-treatment with indomethacin(IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. But pre-treatment with methylene blue(MTB), an inhibitor of guanylate cyclase, didn't affect rCBF levels. In addition, pre-treatment with IDM also decreased MABP levels. Conclusions : In conclusion, these results suggest that $GB_{41}$ supplementation and $BL_{66}$ draining is effective to increase rCBF, and the mechanisms are thought to be related to cyclooxygenase pathways.
뇌혈류 SPECT영상을 사용하여 뇌혈류를 간접적으로 평가하는 방법으로 횡단면 또는 관상면의 단층상을 이용하여 관류 결손 부위의 계수 분포를 평면적으로 조사하는 방법이 주로 사용되어 왔으나, 일반적으로 관류결손 부위의 크기는 다양하며 특히 임의의 형태를 갖는 관류 결손을 포함하는 영상에서는 평면 분석에 의한 결과는 많은 오차를 나타낼 수 있다. 따라서 본 연구는 체적 분석에 의한 평가 방법을 제안하고자 하였으며, 기초연구로서 동일한 뇌혈류 SPECT 영상에 대하여 평면 분석과 체적 분석 방법을 적용하여 비교하므로서 체적 분석 방법의 안정도와 정확도를 검토하였다. 평면 분석 방법에서는 관류 결손 부위의 크기와 형상에 따라 뇌혈류평가 지표인 계수 분포의 변화가 관찰되었으며, 합산에 사용된 단층상의 수에 따라 체적 분석과 상이한 결과를 보였다. 또한 평면 분석은 횡단면을 사용하는 경우와 관상면을 사용하는 경우 각각에서도 상이한 결과를 보여 체적 분석 방법이 뇌혈류 평가지표로서 신뢰도 높은 결과를 산출할 수 있음을 알았다.

CheonghunHwadam-tangGamypang(CHTG) was CHT adding Aurantii Fructus(AF), Gastrodae Rhizoma(GR). The effects of CHT on the cerebral blood flow and cardiacvascular system is not known. The purpose of this Study was to investigate effects of CHT and CHTG on the regional cerebral blood flow(rCBF), mean arterial blood pressure(BP), cardiac muscle contractile force(CMF), heart rate(HR). The changes of cerebral blood flow and movement of cardiacvascular system(BP, CMF, HR) was determinated by Laser-Doppler Rowmetry(LDF). The results were as follows; 1. CHT extract increased rCBF, but decreased BP, HR in a dose-dependent manner. 2. AF extract accelerated rCBF and movement of cardiacvascular system in a dose-dependent manner. 3. GR extract increased significantly rCBF(10.0mg/kg, p<0.05) as well as accelerated BP and rCBF in a dose-dependent manner. 4. CHTG extract increased significantly rCBF(10.0mg/kg, p<0.01) in a dose-dependent, but was not changed movement of cardiacvascular system. This results suggest that CHTG is significantly treated more than CHT in vertigo by wind, fire and phlegm.
Objectives : Folium perillae (FP) can relieve superficial pathogenic factors to dissipate cold and promote the circulation of qi and regulate the function of the stomach and is often used for interior qi-stagnation. We hypothesized that FP could rescue cerebral ischemia in rats. Methods : The present study was carried out to investigate the effects of FP on cerebral ischemia in terms of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. Finally, lactate dehydrogenase (LDH) release was investigated, too. Results : In this study, treatment with FP elevated rCBF and MABP levels in dose-dependent manner. Pre-treatment with indomethacin, an inhibitor of cyclooxygenase, inhibited rCBF increase induced by FP effectively. However, FP did not affect stability during cerebral reperfusion. Finally, FP significantly inhibited LD H activity in vitro Conclusions : These results suggest that FP is useful to treat patient with diseases related to cerebral ischemia, because FP can increase rCBF and MABP.
Amun(GV15), Habkook(LI4) and Shinsu(B23) have been used as a meridian point for apoplexy, hypertention, vertigo etc. The effects of GV 15, LI4 and B23 on the vascular system is not known. The purpose of this Study was to investigate the effect of several meridian points on the regional cerebral bloof flow(rCBF), mean arterial blood pressure(BP) in rats. The changes of BP and rCBF were tested Laser-Doppler Flowmetry(LDF). Results : The results of this Study were obtained as follows ; 1. GV15 and LI4 were increased significantly rCBF. 2. GV15, LI4 and B23 were increased BP, but have not significance. Conclusion : This results suggest that GV15 was increased rCBF by increasing pial arterial diameter, and LI4 was increased rCBF by increasing BP.
