Downscaling of Geophysical Data for Enhanced Resolution by Geostatistical Approach (물리탐사 자료의 해상도 향상을 위한 지구통계학적 다운스케일링)

  • Inversion result of geophysical data given as a block type was geostatistically simulated with borehole observation given as a point type and was applied to the rock classifying map. The geophysical data generally involved secondary information for the target material and were obtained for overall region. In contrast, borehole data provided direct information for the target material, but tended to be effective only for a narrow range of region and were dealt as a point type. Integrated simulation or kriging interpolation of these two different kinds of information required the covariance for point-point, point-block and block-block. Using the Bssim module included in SGeMS software, integrated result of geophysical data and borehole data were obtained. The results were then compared with the method of geostatistical inversion proposed by authors. Downscaling method used in this study showed relatively more flexible than the geostatistical inversion.

In Vivo Spinal Distribution of Cy5.5 Fluorescent Dye after Injection via the Lateral Ventricle and Cisterna Magna in Rat Model

  • Objective : The purpose of this study was to find an optimal delivery route for clinical trials of intrathecal cell therapy for spinal cord injury in preclinical stage. Methods : We compared in vivo distribution of Cy5.5 fluorescent dye in the spinal cord region at various time points utilizing in vivo optical imaging techniques, which was injected into the lateral ventricle (LV) or cisterna magna (CM) of rats. Results : Although CM locates nearer to the spinal cord than the LV, significantly higher signal of Cy5.5 was detected in the thoracic and lumbar spinal cord region at all time points tested when Cy5.5 was injected into the LV. In the LV injection Cy5.5 signal in the thoracic and lumbar spinal cord was observed within 12 hours after injection, which was maintained until 72 hours after injection. In contrast, Cy5.5 signal was concentrated at the injection site in the CM injection at all time points. Conclusion : These data suggested that the LV might be suitable for preclinical injection route of therapeutics targeting the spinal cord to test their treatment efficacy and biosafety for spinal cord diseases in small animal models.

An image enhancement algorithm for detecting the license plate region using the image of the car personal recorder (차량 번호판 검출을 위한 자동차 개인 저장 장치 이미지 향상 알고리즘)

  • We propose an adaptive histogram stretching algorithm for application to a car's personal recorder. The algorithm was used for pre-processing to detect the license plate region in an image from a personal recorder. The algorithm employs a Probability Density Function (PDF) and Cumulative Distribution Function (CDF) to analyze the distribution diagram of the images. These two functions are calculated using an image obtained by sampling at a certain pixel interval. The images were subjected to different levels of stretching, and experiments were done on the images to extract their characteristics. The results show that the proposed algorithm provides less deterioration than conventional algorithms. Moreover, contrast is enhanced according to the characteristics of the image. The algorithm could provide better performance than existing algorithms in applications for detecting search regions for license plates.

Transcription Analysis of Daptomyc in Biosynthetic Genesin Streptomyces roseosporus

  • Insights into gene expression have the potential for improvement of antibiotic yield and the development of robust production hosts for use in recombinant biomolecule production. $Cubicin^{TM}$ (daptomycin for injection) is a recently approved antibiotic active against many Gram(+) pathogens, including those resistant to methicillin, vancomycin, and fluoroquinolones. Daptomycin is produced as a secondary metabolite by Streptomyces roseosporus. A 128 kb region of DNA including the daptomycin biosynthetic gene cluster (dpt) has been cloned. and sequenced. Using a selected array of nucleic acid probes representing this region, we compared the expression levels of the dpt genes between S. roseosporus wild-type (WT) and derived S. roseosporus high-producer of daptomycin (HP). We observed that the majority of the biosynthetic genes were upregulated in HP compared with WT; a total of 12 genes, including those encoding daptomycin synthetase, showed consistently and significantly higher expression levels, at least 5-fold, in HP compared with WT. In contrast, some genes, flanking the dpt cluster, were expressed at higher levels in the WT strain. The expression of housekeeping genes such as S. roseosporus rpsL, rpsG, and 16S (positive controls) and presumptive intergenic regions in the dpt cluster (negative control) were identical in the two strains. In addition, we compared transcription during the early, mid-log, and early-stationary phases of growth in the HP strain. The same set of genes was upregulated and downregulated under all conditions examined; housekeeping genes showed no relative change in expression level over the periods of growth tested. Analyses of this type would be of value in studies of strain improvement and also for the identification of gene regulation processes that are important for secondary metabolite production.

A Study on Classification of Chinese Men's Body Types - Focused in Beijing and Shanghai -

  • The purpose of this study is to provide for some basic data useful to production of the apparels fit and measured well for the Chinese men. For this purpose, 389 men aged between 20 and 49 and living in Beijing and Shanghai, China were sampled to be measured for their constitutions. Then, their constitutions were classified and thereupon, according to the Men's Wear Specifications (GB/T 1335.1-1997), National Standards of People's Republic of China. The collected data were statistically processed using SAS 6.12 for technical statistical analysis, correlation analysis, factor analysis, group-wise analysis. The results of this study can be summarized as follows; 1. As a result of the factor analysis aiming to determine Chinese men's constitutional components, five components could be identified: constitutional obesity, lateral body size, longitudinal body size, shoulder and back width, and shoulder drooping. 2. As a result of classifying Chinese men's constitutions according to drop measurements, four types could be identified. Y type had the lowest obesity and the highest longitudinal body size. A type had a lower obesity and had an average longitudinal body size. B Type had the second highest obesity, the smallest longitudinal body size and shoulders/back width. C Type had the highest obesity, upper body length and shoulders/back width. 3. In terms of distribution, 'B' type (39.10%) of the sample, followed by 'A' type (29.26%), 'C' type (19.95%) and 'Y' type (11.70%). In all, the results of this study suggests that 'B' type represents the Chinese men in contrast with GB specifying that 'A' type represents the Chinese men. On the other hand, Beijing region was dominated most by 'B' type (37.06%), followed by 'A' type (28.82%), 'C' type (22.35%) and 'Y' type (11.76%), while Shanghai region was dominated most by 'B' type (41.13%), followed by 'A' type (31.21%), 'C' type (19.15%) and 'Y' type (8.51%).

Conductivity Imaging of a Canine Head using a 3T MREIT System with a Carbon-Hydrogel Electrode: Postmortem Experiment (3T MREIT 시스템을 이용한 실험견 사체의 두부 도전율 영상)

  • Magnetic Resonance Electrical Impedance Tomography (MREIT) is a new bio-imaging modality providing cross-sectional conductivity images from measurements of internal magnetic flux densities produced by externally injected currents. Recent MREIT studies demonstrated successful conductivity image reconstructions of postmortem and in vivo canine brain. However, the whole head imaging was not achieved due to technical issues related with electrodes and noise in measured magnetic flux density data. In this study, we used a new carbon-hydrogel electrode with a large contact area and injected 30 mA imaging current through a canine head. Using a 3T MREIT system, we performed a postmortem canine experiment and produced high-resolution conductivity images of the entire canine head. Collecting magnetic flux density data inside the head subject to multiple injection currents, we reconstructed cross-sectional conductivity images using the harmonic $B_z$ algorithm. The conductivity images of the canine head show a good contrast not only inside the brain region including white and gray matter but also outside the brain region including the skull, temporalis muscle, mandible, lingualis proprius muscle, and masseter muscle.

Characterization of the xaiF Gene Encoding a Novel Xylanase-activity- increasing Factor, XaiF

  • The DNA sequence immediately following the xynA gene of Bacillus stearothermophilus 236 [about l-kb region downstream from the translational termination codon (TAA) of the xynA gene]was found to have an ability to enhance the xylanase activity of the upstream xynA gene. An 849-bp ORF was identified in the downstream region, and the ORF was confirmed to encode a novel protein of 283 amino acids designated as XaiF (xylanase-activity-increasing factor). From the nucleotide sequence of the xaiF gene, the molecular mass and pI of XaiF were deduced to be 32,006 Da and 4.46, respectively. XaiF was overproduced in the E. coli cells from the cloned xaiF gene by using the T7 expression system. The transcriptional initiation site was determined by primer extension analysis and the putative promoter and ribosome binding regions were also identified. Blast search showed that the xaiF and its protein product had no homology with any gene nor any protein reported so far. Also, in B. subtilis, the xaiF trans-activated the xylanase activity at the same rate as in E. coli. In contrast, xaiF had no activating effect on the co-expressed ${\beta}-xylosidase$ of the xylA gene derived from the same strain of B. stearothermophilus. In addition, the intracellular and extracellular fractions from the E. coli cells carrying the plasmid-borne xaiF gene did not increase the isolated xylanase activity, indicating that the protein-protein interaction between XynA and XaiF was not a causative event for the xylanase activating effect of the xaiF gene.

The effect of depth discontinuity on spreading of motion aftereffect to non-adapted area (비순응 영역으로의 운동 잔여효과의 번짐에 미치는 삼차원 깊이 불연속의 효과)

  • The stationary image appears to move after we view a moving stimulus for a long time. The motion aftereffect(MAE) can spread to an adjacent region if there is no contrast discontinuity between two regions. In this study, it is investigated whether a phenomenon of MAE spreading to an adjacent non adapting area is affected by the depth discontinuity defined by binocular disparity. In the first experiment a disparity defined slanted pattern was presented in an unadapted region, and in the second experiment, a disparity defined pattern with a different depth was presented on the fronto-parallel plane. Although MAE duration in the condition with slanted pattern was not different from that in the non-slanted pattern condition, MAE durations in the pattern presented on pronto-parallel plane was vividly reduced, but not completely disappeared. These results suggest that a phenomenon of MAE spreading might be affected by depth discontinuity, and could be occurred after binocular information converges.

Assessment of Flooding Vulnerability Based on GIS in Urban Area - Focused on Changwon City - (GIS 기반의 도시지역 침수 취약성 평가 - 창원시를 대상으로 -)

  • The purpose of this study is to evaluate flooding vulnerability considering spatial characteristics focused on Changwon-si, Gyeongsangnam-do. Assessment Factors are water cycle area ratio, surface runoff, and precipitation. And construction of assessment factors and vulnerability was analyzed by GIS program. Water cycle ratio and surface runoff were vulnerable in urban area. Precipitation was often distributed in agriculture of the northern region. Results of flooding vulnerability were low in agriculture and forest of the northern region. In contrast, urban area was high because there has covered impervious land cover. Analytical results of flooding vulnerability density using hotspot spatial cluster analysis were high in urban area. And these areas were situated in down stream so flooding were generated. Therefore, flooding vulnerability assessment of this study can help for selecting construction sites of pervious land cover and rainwater management facilities in urban and environmental planning.

Analysis of C43G mutation in the promoter region of the XIST gene in patients with idiopathic primary ovarian insufficiency

  • Objective: The XIST gene is considered to be an attractive candidate gene for skewed X-chromosome inactivation and a possible cause of primary ovarian insufficiency (POI). The purpose of this study was to investigate whether the XIST gene promoter mutation is associated with idiopathic POI in a sample of the Korean population. Methods: Subjects consisted of 102 idiopathic POI patients and 113 healthy controls with normal menstrual cycles. Patients with the following known causes of POI were excluded in advance: cytogenetic abnormalities, prior chemo- or radiotherapy, or prior bilateral oophorectomy. Genotyping was performed using polymerase chain reaction-restriction fragment length polymorphism analysis. Results: The mean age of onset of ovarian insufficiency was $28.7{\pm}8.5years$ and the mean values of serum luteinizing and follicle-stimulating hormones and estradiol in the POI group were $31.4{\pm}18.2mIU/mL$, $74.5{\pm}41.1mIU/mL$, and $30.5{\pm}36.7pg/mL$, respectively. We found no cytosine to guanine (C43G) variation in the XIST gene in both POI patients and controls. Conclusion: The C43G mutation in the promoter region of the XIST gene was not present in the Korean patients with idiopathic POI in our study, in contrast to our expectation, suggesting that the role of XIST in the pathogenesis of POI is not yet clear.
