Resistant cell lines to azetidine-2-carboxylic acid (AZCA) were selected through rice embryo culture after mutagenic treatment of callus irradiated with 30,50,70,90 and 120 Gy. The optimum AZCA concentration for the selection of resistant cell lines was 3 or 4 mM AZCA considering $LD_{50}$ and the fresh weight of callus. Survival rate of the AZCA resistant callus showed remarkable increase in the callus irradiated with 50 and 70 Gy. Regeneration rate of the AZCA resistant callus was much lower on the whole. Ninety and 120 Gy increased the regeneration rate for calli selected from 3 and 4 mM AZCA, respectively. Based on fresh weight, survival rate and regeneration for selection of the AZCA resistant cell line, 50-90 Gy was considered as the optimum range of gamma irradiation. Irradiated calli selected from AZCA were more tolerant to NaCl than those from non-irradiated calli. It suggests that elevated resistance to osmotic stress resulted from mutagenic treatment. The level of free proline content in the AZCA resistant cell line was increased up to 3.5 times compared with that in the control. Proline content in the regenerant derived from the AZCA resistant cell line also increased to 1.7 times that from the control plants regenerated from callus grown in AZCA free medium. Selection of proline overproducing cell lines by in vitro mutagenesis was successful and seems to be useful for improvement of stress tolerance in this crop.
In industrial field, a large volume of regenerants (acid and caustic soda) and their washing effluents are regularly disposed off from the water demineralization plant during regeneration of the ion-exchanger units. Of these waste effluents, a part of the wash water discharged from the single bed Anion and Mixed Bed units can be utilized at a certain stage of their washing cycles when its conductivity is fallen down and becomes considerably less than that of the input raw water. The main aim of this specific waste effluent utilization is to dilute the TDS concentration of the input raw water (fed into the single bed ion-exchanger units) by blending. The achievement is the increase of the longevity of the production cycles of the I.E. units along with the improvement of the production quality and decrease of the regeneration frequencies. As a result, regenerant consumption would be saved because of the reduction of ionic load in feed water which will ultimately reduce the water purification cost. At the same time, the environment pollution will also be protected to a considerable extent. This operational measure is quite effective and useful specially where high TDS water is demineralized only by single bed ion-exchangers. In such case, the water treatment plant is very often found to suffer from both production quality and quantity in addition to carrying out of random and restless regenerations. Proper reuses of the aforesaid wash water effluents of the Anion and MB units excellently minimizes the difficulties experienced in practice. This paper contains the utilities and techniques of reuses of the different kinds of waste effluents of the industrial water treatment plant in addition to the specific reuses of the post-regeneration wash waters of the Anion and MB ion-exchanger units.

Protoplasts from cabbage and radish were isolated and fused symmetrically by PEG treatment. The PEG treated mixture of high concentrated protoplasts produced lots of micro-calli after $2{\sim}3$ weeks. The microcalli developed to normal calli and shoots were regenerated from the calli. A total of 218 shoots were regenerated, but none of them contained the NWB-CMS specific DNA marker, indicating that the transfer of the radish NWB-CMS character into cabbage did not occur. However, ISSR analysis revealed that the cell fusion between protoplasts from radish and cabbage was occurred (3 out of 208 plantlet). The fused regenerants possessed the characteristics of source plants used for protoplast fusion. After vernalization, three regenerants were flowered with white petal color as seen in radish. Only three seeds were able to obtain from one regenerant by backcrossing with the cabbage pollen.

It is difficult to treat wastewater involved in heavy metal in electroplating industry. Recently, many industries adopt the clean technology to prevent production of pollutant in the process or reuse after the appropriate pollutant treatment. In this study, we estimate the ability of recovery of nickel and the efficiency using lab-scale ion exchange and electrodialysis process with electroplating industry wastewater. In the ion exchange experiments with 5 types of resin, the result showed that S 1467(gel-type strong acidic cation exchange resin) has the highest exchange capacity. And it showed that the 4 N HCl has the highest in regeneration efficiency and maximum concentration in the regeneration experiments with various kinds md concentration of the regenerant. During the electrodialysis experiments, we varied the current density, the concentration of electrode rinse solution, the flow rate of concentrate and electrode rinse solution in order to find the optimum operating condition. As a result, we obtained $250A/m^2$ of current density, 2 N $H_2SO_4$ of concentration of electrode rinse solution, 30 mL/min of flow rate of concentrate and electrode rinse solution as the best operating conditions. We performed the scale-up experiments on the basis of ion exchange and electrodialysis experiments. And we obtained the experimental result that exchange capacity of S 1467 was 1.88 eq/L resin, and regeneration efficiency was 93.7% in the ion exchange scale-up experiment, we also got the result that concentration and dilution efficiency increased, and current efficiency kept constant in the scale-up experiments.
To understand the molecular mechanism of hardening process in somatic embryo development and artificial seed germination in celery (Apium graveolens L.), the changes of protein synthesis by ABA or cold teatment at early globular stage were examined. Protein content and nitrate reductase activity in ABA- or cold-treated somatic embryo and seedlings were higher than that in unheated ones. The protein content and nitrate reductase activity were more prominent in somatic embryos than in seedlings. From two-dimensional electrophoresis, several protein spots specific to ABA or cold treatment were identified: 30 KD, 32 KD, 171 KD and 205 KD at heart-shaped stage; and 29 KD, 33 KD, 37 KD, 38 KD, 41 KD, 55 KD, 66 KD, and 110 KD at cotyledonary stage were the most specifically synthesized However the synthesis of certain polypeptides were repressed at heart-shaped or cotyledonary stage: 42 KD, 44 KD, 59 KD, 64 KD, 101 KD, 104 KD, and 190 KD at heart-shaped stage; and 29 KD and 116 KD at cotyledonary stage. The protein pattern changes by ABA or cold treatment occurred simultaneously and mainly in acid-soluble proteins during somatic embryo development and artificial seed germination. Therefore it is suggested that the metabolic changes for adaptation to environmental change occur during somatic embryo development and the germination and growth of seedling from embryo.
The purification of 5'-riboncleotide using ion exchange resins has been studied and the optimum conditions were determined. The amount used of Amberlite IR120 ion exchange resin in 2nd resin tower could be reduced up to 20% by pretreating the hydrolyzed RNA solution in 1st resin tower. The amount used of regenerant could be also reduced up to 20% by desalting the hydrolyzed RNA solution in the 1st tower, because the desalted solution eluted easily by the water in the 2nd tower. The crystal obtained in this experiment was the mixed-crystals of $5'-IMP.Na_2\;and\;5'-GMP.Na_2$. The crystallization of the complexes formed from $5'-IMP.Na_2\;and\;5'-GMP.Na_2$ gave the best result at pH 7.6. The yield of crystal complexes formed from $5'-IMP.Na_2\;and\;5'-GMP.Na_2$ was obtained higher in high MeOH concentration. However, in higher than 60% MeOH concentration the products was amorphous. The higher content of MeOH for the crystallization of the product gave the smaller value of $5'-IMP.Na_{2}/5'-GMP.Na_2$.
