Microbial reduction of hexavalent (VI) to trivalent (III) chromium decreases its toxicity by two orders of magnitude. In order to investigate the nature of Cr-reduction, Cr-resistant Pseudomonas aeruginosa HP014 was isolated and tested for its reduction capability. At the concentration of 0.5 mM Cr(VI), cell growth was not inhibited by the presence of Cr(VI) in a liquid medium, and Cr(VI) reduction was accompanied by ell growth. When cell-free extract was tested, the reduction of Cr(VI) showed a saturation kinetics with the maximum specific activity of 0.33 .mu.mol min$\^$-1/ mg$\^$-1/ cell protein, and an apparent K. of 1.73 mM Cr(VI). The activity required either NADH or NADPH as an electron donor. However, NADPH gave 50% as mush activity as sequently the supernatant and pelleted membrane fractions were tested for Cr(VI) reduction activity. The supernatant of the centrifugation showed almost the same Cr(VI) reduction activity as compared with that of the cell-free extract, indicating that the Cr(VI)-reducing activity of P. aeruginosa HP-14 is due to soluble enzyme. Moreover, the activity appeared to be the highest among the known activities, suggesting that the strain might be useful for remediation of Cr(VI)-contaminated sites.
In this study, phytochemicals extracted from sea mustard (SM) and sea mustard sporophyll (SMS) in ethanol solution have been analyzed and wound healing potential of the phytochemicals was investigated. In the phytochemical screening studies, the extract of SM and SMS includes several phytochemical compounds such as phytol, ascorbic acid, sitgmasta, fucosterol and ergosta. Cytotoxicity studies of the extract of SM and SMS with mouse macrophage RAW 264.7 cells showed on toxicity up to a high concentration of 1.0 mg/mL. Furthermore, the SM and SMS extract significantly reduces the production of nitric oxide (NO) induced lipopolysaccharide on RAW 264.7 cells with a dose-dependent manner. In addition, the extract of SM and SMS has the effect of enhancing the cell migration and invasion of fibroblast. These results demonstrate that the extract of SM and SMS could help to heal wound by reducing NO production and increasing cell migration.
Thermoelectric energy conversion has attracted much attention because it can convert heat into electric power directly through solid state device and vice versa. Current research is aimed at increasing the thermoelectric figure of merit (ZT ) by improving the power factor and reducing the thermal conductivity. Although there have been significant progresses in increasing ZT of material systems composed of Bi, Te, Ge, Pb, and etc. over the last few decades, their relatively high cost, toxicity, and the scarcity have hindered further development of thermoelectrics to expand practical applications. In this paper, we review the current status of research in the fields of nanostructured thermoelectric materials with eco-friendly and low cost elements, such as skutterudites and oxides, for mid-high temperature applications, highlighting the strategies to improve thermoelectric performance.
Myotonic dystrophy type 1 (DM1), which is a dominantly inherited neurodegenerative disorder, results from a CTG trinucleotide repeat expansion in the 3'-untranslated region (3'-UTR) of the myotonic dystrophy protein kinase (DMPK) gene. Retention of mutant DMPK (mDMPK) transcripts in the nuclei of affected cells has been known to be the main cause of pathogenesis of the disease. Thus, reducing the RNA toxicity through elimination of the mutant RNA has been suggested as one therapeutic strategy against DM1. In this study, we suggested RNA replacement with a trans -splicing ribozyme as an alternate genetic therapeutic approach for amelioration of DM1. To this end, we identified the regions of mDMPK 3'-UTR RNA that were accessible to ribozymes by using an RNA mapping strategy based on a trans-splicing ribozyme library. We found that particularly accessible sites were present not only upstream but also downstream of the expanded repeat sequence. Repair or replacement of the mDMPK transcript with the specific ribozyme will be useful for DM1 treatment through reduction of toxic mutant transcripts and simultaneously restore wild-type DMPK or release nucleus-entrapped mDMPK transcripts to the cytoplasm.
This research evaluate antioxidant and skin-whitening effect of Abeliophyllum distichum Nakai by extraction processes. First, antioxidant effects were follows: EE (70% ethanol extract) showed higher DPPH scavenging activity of 69.66% than WE (hot water exract) 59.13% at $0.3mg/m{\ell}$, also UE's (70% ethanol extract by sonication process) higher than EE. Reducing power was that also EE showed higher than WE, and it was the highest value with UE's because of ultrasonic pretreatment. Next, the whitening effect tyrosinase inhibition activity was measured that EE was 23.88%, WE's was 16.69%, and UE was 23.34%. Ultrasonic pretreatment did not influence to tyrosinase inhibition activity. Cell viability showed low cell toxicity in all groups. UE's inhibited melanin synthesis, 55.1%, that is higher than EE and WE, 52.7% and 39.5%, respectively. As a result, we confirmed that antioxidant activities and skin-whitening effect by extraction process. Also, this results confirmed that the Abeliophyllum distichum Nakai extracts worth as cosmetic materials.

The purpose of this study was to investigate the action mechanism of the roots of Adenophora triphylla var. japonica extract (ATE) in 3T3-L1 adipocytes. Cell toxicity test by MTT assay and lipid accumulation was performed to evaluate the inhibitory effect on the differentiation of adipocyte from preadipocytes induced by MDI differentiation medium, while adipogenesis related proteins expression level were evaluated by western blotting. As a result, ATE inhibited MDI-induced adipocyte differentiation in 3T3-L1 cells dose-dependently without cytotoxicity. Our results showed that ATE inhibited the phosphorylation of IRS1, thereby decreasing the expression of PI3K110α and reducing the phosphorylation of AKT and mTOR, resulting in attenuated protein expression of C/EBPα, PPARγ, ap2 and FAS in 3T3-L1 cells. These results suggest anti-adipogenic functions for ATE, and identified IRS1 as a novel target for ATE in adipogenesis.

Thallium and its compounds are a class of highly toxic chemicals that cause wide-ranging symptoms such as gastrointestinal disturbances; polyneuritis; encephalopathy; tachycardia; skin eruptions; hepatic, renal, cardiac, and neurological toxicities; and have mutagenic and genotoxic effects. The present research aimed to evaluate the efficacy of the chelating agents deferasirox (DFX) and deferiprone (L1) in reducing serum and tissue thallium levels after the administration of thallium (III), according to two different dosing regimens, to several groups of Wistar rats for 60 days. It was hypothesized that the two chelators might be more efficient as a combined therapy than as monotherapies in removing thallium (III) from the rats' organs. The chelators were administered orally as either single or combined therapies for a period of 14 days. Serum and tissue thallium (III) and iron concentrations were determined by flame atomic absorption spectroscopy. Serum and tissue thallium (III) levels were significantly reduced by combined therapy with DFX and L1. Additionally, iron concentrations returned to normal levels and symptoms of toxicity decreased.

Brachypodium distachyon is rapidly emerged in biological study and has been currently used as a model system for genetics and functional studies for crop improvement and biofuel production. Phosphinothricin (PPT) has been widely used as a selectable agent, which raises ammonium content and induces toxicity in non-transformed plant cells. However PPT selection is not much effective on Brachypodium callus consequently reducing transformation efficiency. In order to identify the efficient conditions of PPT selection, calli obtained from mature seeds of Brachypodium (PI 254867) were cultured on the callus inducing medium (CIM) or regeneration medium (ReM) containing serial dilutions of the PPT (0, 2, 5, 10, and 15 mg/l) in dark or light condition. Callus growth and ammonium content of each treatment were measured 2 weeks after the treatment. Although callus growth and ammonium content did not show much difference in CIM, slow callus growth and increased ammonium accumulation were found in ReM. No significant difference of ammonium accumulation in response to PPT was found between dark and light conditions. In order to identify major factors affecting increased ammonium accumulation, callus was cultured on the media in combined with phytohormones (2,4-D or kinetin) and carbon sources (sucrose or maltose) containing with PPT (5 mg/l). The highest ammonium content in callus was found in the kinetin and maltose media.
The lead element or its salts are good radiation shielding materials. However, their toxic effects are high. Due to less toxicity of bismuth salts, the radiation shielding properties of the bismuth salts have been investigated and compared to that of lead salts to establish them as a better alternative to radiation shielding material to the lead element or its salts. The transmission geometry was utilized to measure the mass attenuation coefficient (${\mu}/{\rho}$) of different salts containing lead and bismuth using a high-resolution HPGe detector and different energies (between 81 and 1333 keV) emitted from point sources of $^{133}Ba$, $^{57}Co$, $^{22}Na$, $^{54}Mn$, $^{137}Cs$, and $^{60}Co$. The experimental ${\mu}/{\rho}$ results are compared with the theoretical values obtained through WinXCOM program. The theoretical calculations are in good agreement with their experimental ones. The radiation protection efficiencies, mean free paths, effective atomic numbers and electron densities for the present compounds were determined. The bismuth fluoride ($BiF_3$) is found to have maximum radiation protection efficiency among the selected salts. The results showed that present salts are more effective for reducing the intensity of gamma photons at low energy region.
Cardiovascular diseases (CVDs) have posed serious public health problems, accounting for nearly 30% of mortality worldwide and their incidence is still increasing. Therefore, new treatment resources are necessary to prevent or manage the ever-increasing population of patients with CVDs. Sea cucumber is well known for its medical and health benefit effects, but it is not well known what/how effect it has on vascular disease. In the present study, we examined the protect effect of sea cucumber, Apostichopus japonicus 80% ethanol extract (AJE) on zebrafish embryo with the stimulation of free fatty acid, palmitate (PA). In vivo study showed that AJE can attenuate PA-induced toxicity through relieving the rapid heartbeat, increasing the survival rate and reducing the malformation in both wild type and Tg (fli1a:eGFP) transgenic zebrafish lines. Additionally, compare with PA treated embryos, the yolk sac area, body length, axial vascular segment (AVS) and intersegmental vessel (ISV) of the co-treatment group of AJE and PA were comparable to the control group. Moreover, AJE lowered the expression of inducible nitric oxide synthase (iNOS), nitric oxide (NO) and inflammation-related genes induced by PA, and inhibited PA-induced vascular development disorders. Our data preliminarily verify that AJE could be a candidate resource for the prevention or therapy of CVDs.
