Comparative Characterization of the Bacteria Isolated from Market Milk Treated with ESL and Conventional System (ESL 생산공정에 따른 시유 유래 미생물의 분포 비교 연구)

  • This study was conducted to investigate the characteristics of strains which were isolated from market milk treated with ESL(extended shelf life) and conventional system, and to compare the microbiological quality of ESL milk with conventional milk. In order to characterize the isolated strains, purification, Gram staining, spore staining, catalase, oxidase, motility test, and identification by means of automatic identificator were performed. The results obtained are as follows: total 364 selected strains were analyzed in this study. Depending upon the isolated source, the number of strains from conventional milk was found to be Higher than ESL-milk. By means of grouping of total strains, Bacillus ssp. and Staphylococcus ssp. showed to be predominant. But most of strains were distributed with various groups except Lactobacillus ssp. When the isolates were compared with milk process methods, Enterococcus ssp. was detected much on market milk treated with LTLT pasteurization. Also, Pseudomonas ssp. was detected much on conventional milk treated with UHT pasteurization. By comparison with genus groups depending upon storage temperature of market milk, the higher milk storage temperature increased, the most frequency detected Bacillus ssp. increased. Also, Pseudomonas ssp. was detected most frequently at 10$^{\circ}C$ storage condition. Generally this genus derived from post-contamination during milk processing and related to the quality of market milk during chilled system. In conclusion, it was shown that ESL system reduced post-contamination during milk process, following the improvement of product quality and life cycle during the distribution of market milk.

Adaptive Response Induced by Low Dose Ionizing Radiation in Human Lymphocytes (인체 말초 혈액 림프구에서 저선량 방사선 조사에 의해 유도되는 적응 반응)

  • Adaptive response induced by low dose ${\gamma}-ray$ irradiation in human peripheral lymphocytes was examimed. Human lymphocytes were exposured to low dose of ${\gamma}-ray$ (priming dose, 0.01Gy) followed by high dose (challenging dose, 1.5Gy) after various time intervals (4, 7, 20 hours). Frequencies of micronuclei were enumerated in both primed and unprimed groups. Maximum reduction in frequency of micronuclei was observed when challenging dose irradiation was followed by priming dose after 4hr incubation period. When challenging doses were irradiated 7 or 20hr after priming dose, frequencies of micronuclei were reduced slighty. However, these reduction were not statistically significant. In this study, human peripheral lymphocytes were irradiated at Go phase and they showed adaptive response induced by low dose radiation. Since micronucleus assay is relatively simpler and faster than other methods, it may be a good tool for evaluating radiation-induced adaptive responses.

Effects of 5 Weeks Self-Help Management Program on Reducing Depression and Promoting Activity of Daily Livings, Grasping Power, Hope and Self-Efficacy (재가 뇌졸중환자를 위한 5주간의 자조관리프로그램의 효과에 관한 연구)

  • The purpose of this study was to determine the effects of 5 weeks self-help management program on reducing depression, promoting Activity of Daily Livings(ADLs), Instrumental Activity of Daily Livings(IADLs), grasping power, hope and self-efficacy for post stroke patients visiting public health care centers in Seoul. This was pre-experimental study, and the subjects were 27 post stroke hemiplegic patients. The program was composed of five sessions and each session had health education on stroke, ROM exercise and recreation. Data were collected from May to November, 2000, and all subjects were asked to complete the Questionnaires, be measured vital signs and grasping power. Data were analyzed with frequency, percent, paired t-test, and Pearson's correlation coefficient using SAS(version 6.12) program. The results were as follows : 1) The scores of ADLs were increased from 27.04 to 28.22 after program, and that was statistically significant(p=.005). 2) The scores of IADLs were increased from 18.70 to 19.78 after program, and that was statistically significant(p=.004). 3) The grasping power of right hand were increased from 21.87kg to 26.93kg after program, and that was statistically significant(p=.002). But the grasping power of left hand were statistically insignificant(p=.919). 4) The scores of depression were decreased from 39.63 to 35.30 after program, and that was statistically significant(p=.030). 5) The scores of hope were increased from 30.89 to 34.15 after program, and that was statistically significant(p=.002). 6) The scores of self-efficacy were increased from 67.70 to 76.37 after program, and that was statistically significant(p=.000). According to the results of this study, the scores of Activity of Daily Livings(ADLs), Instrumental Activity of Daily Livings (IADLs), hope, and self-efficacy and the grasping power were improved and depression was reduced in post stroke patients participating in self-help management program. Therefore we recommend to use self-help management programs as a nursing intervention for the post stroke patient.

The Electrochemical Characteristics and Secondary Doping Effects of Poly[Sodium 4-Styrenesulfonate] Doped Polyaniline (폴리아닐린의 이차도핑과 전기화학적 특성)

  • In this study, the polyaniline films of emeraldine base(EB) and lucoemeraldine base(LEB) form chemically doping with poly(sodium-4 styrenesulfonate, PSS) were prepared by casting the mixed solution of chloroform and m-cresol on ITO(indium tin oxide) electrode. By analyzing UV-vis spectra of the mixed solutions, the effects of the secondary doping by m-cresol were obtained. And the conductivity of polyaniline film was increased with increasing m-cresol content. The results suggest that the improvement of conductivity obtained by secondary doping results primarily from interaction of polyaniline and m-cresol. As the results of analyzing cyclic voltammograms, it was known that the redox peak currents of polyaniline electrode prepared from LEB were larger and more reversible than those of polyaniline electrodes prepared from EB. The charge transfer resistances($R_{ct}$) of polyaniline electrodes were reduced with increasing m-cresol content, and LEB/PSS electrodes were smaller than EB/PSS electrodes. This result agrees to the analysis of the redox peak current of cyclic voltammograms. The solution resistance and the capacity of electrical double layer almost unchanged in all prepared polyaniline electrodes. It was confirmed that solution resistance was independent of frequency factor in AC impedance spectra. Also the polyaniline film doping with PSS was revealed pseudo n-type characteristics of conducting polymer.

Multi-hop Routing Protocol based on Neighbor Conditions in Multichannel Ad-hoc Cognitive Radio Networks (인지 무선 애드혹 네트워크에서의 주변 상황을 고려한 협력적 멀티홉 라우팅 방법)

  • During the routing process between nodes on the CR(Cognitive Radio) network conducting for efficient use of limited frequency resources, spectrum handover process due to the appearance of the PU occupies most of the routing latency, and also decreases the reliability of the path. In this paper, a cooperative routing protocol in a multi-channel environment is proposed. The source node broadcasts a message with available channel lists and probability of PU appearance during its route guidance. The intermediate nodes re-transmit the message, received from the source node, and update and maintain the information, status table of the path. The destination node determines the optimal path and sends a reply message to the selected path after it receives the messages from the intermediate nodes. The average probability of the PU appearance and the average time of the PU appearance are updated while transferring data. During data transmission the channel with the lowest probability of appearance of the PU is selected dynamically and if a PU appears on the current channel partial repairment is performed. It is examined that reliability of the selected path considerably is improved and the routing cost is reduced significantly compared to traditional routing methods.

Study of Viral Effects of the Mycovirus (LeV) and Virus-Free Commercial Line in the Edible Mushroom Lentinula edodes

  • dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed an identical sequence to known RdRp genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that, although variations in the growth rate existed among progeny and virus infection was observed in highly actively growing progeny, there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny. This study attempted to cure the edible mushroom L. edodes strain FMRI0339 of the L. edodes mycovirus (LeV) in order to obtain an isogenic virus-free fungal strain as well as a virus-infected strain for comparison. Mycelial fragmentation, followed by being spread on a plate with serial dilutions resulted in a virus-free colony. Viral absence was confirmed with gel electrophoresis after dsRNA-specific virus purification, Northern blot analysis, and PCR using reverse transcriptase (RT-PCR). Once cured, all of fungal cultures remained virus-free over the next two years. Interestingly, the viral titer of LeV varied depending on the culture condition. The titer from the plate culture showed at least a 20-fold higher concentration than that grown in the liquid culture. However, the reduced virus titer in the liquid culture was recovered by transferring the mycelia to a plate containing the same medium. In addition, oxygen-depleted culture conditions resulted in a significant decrease of viral concentration, but not to the extent seen in the submerged liquid culture. Although no $discernable phenotypic changes in colony morphology were observed, virus-cured strains showed significantly higher growth rates and mycelial mass than virus-infected strains. We were also explored effects of LeV on fruiting body formation and mushroom yield. The fruiting body formation yield of virus-free L. edodes was larger than virus-infected L. edodes. These results indicate that LeV infection has a deleterious effect on mycelial growth and fruiting body formation. In addition, we have been investigated host-parasite interaction between L. edodes and its mycovirus interaction to study viral mechanism by establishment of proteomics.

Effect of Ground Roll Suppression Based on Karhunen-Loeve Transform (카루넨-루베 변환을 이용한 탄성파 그라운드 롤 억제 효과)

  • Ground roll is a surface wave which is usually observed in the land seismic data. It is one of the typical coherent noise. During the reflection data processing, ground roll is removed because it is considered as noise. This removal process often causes the loss of reflection signals if the ground roll overlaps reflection signals. In this study, we look over Karhunen-Loeve Transform (KLT) and analyze its effects to suppress the ground roll appropriately while reducing the reflection loss. Numerical tests in homogeneous elastic media show that the ground roll has been properly rejected. However, the field data application reveals that there is no significant suppression of ground roll when compared to band-pass filtering. This can be considered that it is hard to calculate horizontally aligned gathers in the field data because the ground roll contains a wide range of frequency bands. On the contrary, the result of singular value decomposition (SVD) filtering shows that the ground roll has been significantly reduced. It is thought that the SVD filtering performs better in the ground roll suppression than KLT because it is easy to calculate the horizontally aligned gathers in the SVD filtering.

Genetic diversity and structure of Pulsatilla tongkangensis as inferred from ISSR markers (ISSR 표지자에 의한 동강할미꽃(Pulsatilla tongkangensis)의 유전다양성과 구조)

  • The genetic diversity and structure of P. tongkangensis in 5 populations from 3 regions was investigated using 56 markers derived from 6 ISSR primers. Genetic diversity at the species level (P=94.6, SI=0.377, h=0.240) was substantial considering the limited distribution and small size of populations. Genetic differentiation among regions (12%) and among populations (13%) in the region was not clearly evident, which suggested a moderate level of gene flow among adjacent populations. The Mantel test revealed a significant correlation between genetic differentiation (${\Phi}_{ST}$) and geographic distance among populations. This was supported by cluster analysis and principal coordinate analysis (PCoA). The significant difference in marker band frequency at many loci and their fixation in opposite directions in the smallest and most isolated population SC were considered due to genetic drift. Therefore, the genetic diversity of P. tongkangensis could be compromised if the distribution area or the size of the population was further reduced. In particular, small and isolated populations could be at great risk of extinction. Considering this, the unique habitats of P. tongkangensis should be protected and the reduction of population size should be closely monitored. Conservation efforts including the seeding and planting of seedlings should be done carefully based on their genetic and ecological traits. Our data support the argument that establishing an integrated management system for the efficient conservation of P. tongkangensis is critical.

Change of isometric contractile force and muscle activity applying ice and hot according to the time on biceps brachii muscle (온열 및 한랭의 적용시간에 따른 상완이두근의 등척성 수축력과 근활성도 변화)

  • This study has investigated the effect of isometric contractile force and muscle activity applying ice and hot according to the time from the biceps brachii muscle. In this study, 20 university students participants without musculoskeletal and neurological disorders. By applying a hot pack 5 min, 10 min, 20 min and 30 min respectively and ice pack for 5 min, 10 min, and 20 min respectively. After that measurement are skin temperature, contractile force and muscle activity. Skin temperature of the hot and cold 5 min applied that rapidly changing. Increasing the time it takes to apply a variance has been reduced(p<.001). Isometric contractile force was not statistically significant but highest when applying the hot pack 5 minutes and lowest when applying the hot pack 30 minutes(p<.001). Muscle activity and median frequency was highest when applying the hot pack 5 minutes. To analyze the above results, it was found that isometric contractile force and muscle activity changed according to the applying time. These result lead us to the conclusion that this study will be more evidence for changes in muscle contraction to apply hot pack and ice pack on clinic.

THE EFFECT OF SACROOCCIPITAL TECHNIQUE CATEGORY II BLOCKING ON ABDOMINAL MUSCLE ENDURANCE AND SHOULDER PAIN (S.O.T CATEGORY II BLOCKING이 복근지구력과 어깨 통증에 미치는 영향)

  • Objectives: The purpose of this study was to determine whether the sacrooccipital category II blocking technique may improve abdominal muscle endurance and alleviate shoulder pain caused by pelvic tilting. Methods: A total of 50 subjects diagnosed with category II sacroiliac joint instability and lower back pain were randomly assigned to the control or treatment group. The sacrooccipital technique category II blocking procedure (2-min duration) was performed 3 times a week until the category II indicator of joint instability had subsided. The control subjects were subjected to a sham procedure of equal duration and frequency. We assessed abdominal muscle endurance using the partial curl-up test and shoulder pain using a visual analogue scale, before and immediately after the intervention and 2 weeks after the intervention. Results: On two-way analysis of variance with repeated measures on time factor, significant treatment and interaction effects on muscle endurance were found. A significant interaction effect, but not treatment effect, was found for shoulder pain. Post hoc test showed that the shoulder pain was reduced immediately after intervention (treated group) and 2weeks (control and treated group) after the intervention as compared to before the intervention. Conclusions: This study suggests that sacrooccipital category II blocking can be used to alleviate shoulder pain caused by pelvic instability. The mechanisms behind the long-term benefits may include an increase in abdominal muscle endurance.
